    G-133: A soft x ray solar telescope

    The GOLDHELOX Project, NASA payload number G-133, is a robotic soft x ray solar telescope designed and built by an organization of undergraduate students. The telescope is designed to observe the sun at a wavelength of 171 to 181 A. Since we require observations free from atmospheric interference, the telescope will be launched in a NASA Get-Away-Special (GAS) canister with a Motorized Door Assembly (MDA). In this paper we primarily discuss the most important elements of the telescope itself. We also elaborate on some of the technical difficulties associated with doing good science in space on a small budget (about $100,000) and mention ways in which controlling the instrument environment has reduced the complexity of the system and thus saved us money

    The identification and use of robust transaminases from a domestic drain metagenome

    Transaminases remain one of the most promising biocatalysts for use in chiral amine synthesis, however their industrial implementation has been hampered by their general instability towards, for example, high amine donor concentrations and organic solvent content. Herein we describe the identification, cloning and screening of 29 novel transaminases from a household drain metagenome. The most promising enzymes were fully characterised and the effects of pH, temperature, amine donor concentration and co-solvent determined. Several enzymes demonstrated good substrate tolerance as well as an unprecedented robustness for a wild-type transaminase. One enzyme in particular readily accepted IPA as an amine donor giving the same conversion with 2–50 equivalents, as well as being tolerant to a number of co-solvents, and operational in up to 50% DMSO – a characteristic as yet unobserved in a wild-type transaminase. This work highlights the value of using metagenomics for biocatalyst discovery from niche environments, and here has led to the identification of one of the most robust native transaminases described to date, with respect to IPA and DMSO tolerance

    The KPNO International Spectroscopic Survey (KISS) is an objective-prism survey designed to detect extragalactic emission-line objects. It combines many of the features of previous slitless spectroscopic surveys with the advantages of modern CCD detectors, and is the first purely digital objective-prism survey for emission-line galaxies (ELGs). Here we present the third list of ELG candidates selected from our red spectral data, which cover the wavelength range 6400 to 7200 A. In most cases, the detected emission line is H-alpha. The current survey list covers the region of the NOAO Deep Wide-Field Survey (NDWFS). This survey covers two fields; the first is 3 x 3 degrees square and located at RA = 14h 30m, DEC = 34.5 deg (B1950), the second is 2.3 x 4.0 degrees and centered at RA = 2h 7.5m, DEC = -4.75 deg. A total area of 19.65 deg^2 is covered by the KISS data. A total of 261 candidate emission-line objects have been selected for inclusion in the survey list (13.3 per deg^2). We tabulate accurate coordinates and photometry for each source, as well as estimates of the redshift, emission-line flux and line equivalent width based on measurements of the digital objective-prism spectra. The properties of the KISS ELGs are examined using the available observational data. When combined with the wealth of multi-wavelength data already available for the NDWFS fields, the current list of KISS ELGs should provide a valuable tool for studying star-formation and nuclear activity in galaxies in the local universe.Comment: 18 pages, including 10 figures and 4 tables.     The KPNO International Spectroscopic Survey (KISS) is an objective-prism survey for extragalactic emission-line objects. It combines many of the features of previous slitless spectroscopic surveys with the advantages of modern CCD detectors, and is the first purely digital objective-prism survey for emission-line galaxies. Here we present the second list of emission-line galaxy candidates selected from our red spectral data, which cover the wavelength range 6400 to 7200 A. In most cases, the detected emission line is H-alpha. The current survey list covers a 1.6-degree-wide strip located at Dec(1950) = 43d 30' and spans the RA range 11h 55m to 16h 15m. The survey strip runs through the center of the Bootes Void, and has enough depth to adequately sample the far side of the void. An area of 65.8 sq. deg. is covered. A total of 1029 candidate emission-line objects have been selected for inclusion in the survey list (15.6 per sq. deg.). We tabulate accurate coordinates and photometry for each source, as well as estimates of the redshift and emission-line flux and equivalent width based on measurements of the digital objective-prism spectra. The properties of the KISS emission-line galaxies are examined using the available observational data. Although the current survey covers only a modest fraction of the total volume of the Bootes Void, we catalog at least twelve objects that appear to be located within the void.     Concordance of sibling's recall of measures of childhood socioeconomic position

    <p>Abstract</p> <p>Background</p> <p>Studies of socioeconomic determinants of health often rely on recalled information on childhood socioeconomic position, despite limited evidence of the validity of this information. This study examined concordance between siblings of recalled measures of childhood socioeconomic position.</p> <p>Methods</p> <p>This cross-sectional study examined reports by 1280 adult sibling pairs in the National Survey of Midlife Development in the United States of seven measures of childhood socioeconomic position: father's occupation (in 9 categories), father having a professional occupation, father being a supervisor at work, father's education level, mother's education level, receipt of welfare payments, and subjective appraisal of being better or worse off financially than others.</p> <p>Results</p> <p>Concordance was high for father's professional occupation (0.97; 95% confidence interval (CI) 0.96, 0.98), father's occupation in 9 categories (0.76; 95% CI 0.73, 0.80), and receipt of welfare payments (0.95; 95% CI 0.93, 0.97). Concordance was lower for father's and mother's education level, and lowest for subjective appraisal of socioeconomic position (0.60; 95% CI 0.57, 0.64). Concordance of parental education was lower for sibling pairs with high school educations or less.</p> <p>Conclusion</p> <p>Concordance of recalled measures of childhood socioeconomic position by siblings is generally but not uniformly high.</p

    Pharmaceutical Metabolism in Fish: Using a 3-D Hepatic In Vitro Model to Assess Clearance

    At high internal doses, pharmaceuticals have the potential for inducing biological/pharmacological effects in fish. One particular concern for the environment is their potential to bioaccumulate and reach pharmacological levels; the study of these implications for environmental risk assessment has therefore gained increasing attention. To avoid unnecessary testing on animals, in vitro methods for assessment of xenobiotic metabolism could aid in the ecotoxicological evaluation. Here we report the use of a 3-D in vitro liver organoid culture system (spheroids) derived from rainbow trout to measure the metabolism of seven pharmaceuticals using a substrate depletion assay. Of the pharmaceuticals tested, propranolol, diclofenac and phenylbutazone were metabolised by trout liver spheroids; atenolol, metoprolol, diazepam and carbamazepine were not. Substrate depletion kinetics data was used to estimate intrinsic hepatic clearance by this spheroid model, which was similar for diclofenac and approximately 5 fold higher for propranolol when compared to trout liver microsomal fraction (S9) data. These results suggest that liver spheroids could be used as a relevant and metabolically competent in vitro model with which to measure the biotransformation of pharmaceuticals in fish; and propranolol acts as a reproducible positive control

    Patient-reported outcomes in the ProtecT randomized trial of clinically localized prostate cancer treatments: Study design, and baseline urinary, bowel and sexual function and quality of life

    Objectives: To present the baseline patient-reported outcome measures (PROMs) in the Prostate Testing for Cancer and Treatment (ProtecT) randomized trial comparing active monitoring, radical prostatectomy and external-beam conformal radiotherapy for localized prostate cancer and to compare results with other populations. Materials and Methods: A total of 1643 randomized men, aged 50-69 years and diagnosed with clinically localized disease identified by prostate-specific antigen (PSA) testing, in nine UK cities in the period 1999-2009 were included. Validated PROMs for disease-specific (urinary, bowel and sexual function) and condition-specific impact on quality of life (Expanded Prostate Index Composite [EPIC], 2005 onwards; International Consultation on Incontinence Questionnaire-Urinary Incontinence [ICIQ-UI], 2001 onwards; the International Continence Society short-form male survey [ICSmaleSF]; anxiety and depression (Hospital Anxiety and Depression Scale [HADS]), generic mental and physical health (12-item short-form health survey [SF-12]; EuroQol quality-of-life survey, the EQ-5D-3L) were assessed at prostate biopsy clinics before randomization. Descriptive statistics are presented by treatment allocation and by men's age at biopsy and PSA testing time points for selected measures. Results: A total of 1438 participants completed biopsy questionnaires (88%) and 77-88% of these were analysed for individual PROMs. Fewer than 1% of participants were using pads daily (5/754). Storage lower urinary tract symptoms were frequent (e.g. nocturia 22%, 312/1423). Bowel symptoms were rare, except for loose stools (16%, 118/754). One third of participants reported erectile dysfunction (241/735) and for 16% (118/731) this was a moderate or large problem. Depression was infrequent (80/1399, 6%) but 20% of participants (278/1403) reported anxiety. Sexual function and bother were markedly worse in older men (65-70 years), whilst urinary bother and physical health were somewhat worse than in younger men (49-54 years, all P < 0.001). Bowel health, urinary function and depression were unaltered by age, whilst mental health and anxiety were better in older men (P < 0.001). Only minor differences existed in mental or physical health, anxiety and depression between PSA testing and biopsy assessments. Conclusion: The ProtecT trial baseline PROMs response rates were high. Symptom frequencies and generic quality of life were similar to those observed in populations screened for prostate cancer and control subjects without cancer
