    Not So Revisionary: The Regressive Treatment of Gender in Alan Moore\u27s Watchmen

    While Alan Moore’s comic book Watchmen is often hailed as a revisionary text for introducing flawed superheroes and political anxiety to the genre, it is also remarkably regressive in its treatment of gender. Some critics do argue that women are given a newfound voice in Watchmen, but this interpretation neglects to examine character Laurie Jupiter adequately, or the ways in which other female characters\u27 appearance and dialogue are limited and/or based on their sexuality and relationships with male characters. Watchmen\u27s main female characters, mother and daughter Sally and Laurie Jupiter, lack autonomy and their identities are completely intertwined with and dependent on the male superheros that the two women interact with. This effect culminates with Earth\u27s fate being dependent on Laurie’s relationship with fellow hero Jon - a fate that is determined through a conversation in which Laurie has no say. Written only as submissive, and depicted only as highly sexualized, Watchmen\u27s female characters thus embody misogynistic, outdated ideas about women, even for the 1980s

    Establishing RNAi in a non-model organism: The Antarctic nematode Panagrolaimus sp. DAW1

    The Antarctic nematode Panagrolaimus sp. DAW1 is one of the only organisms known to survive extensive intracellular freezing throughout its tissues. Although the physiological mechanisms of this extreme adaptation are partly understood, the molecular mechanisms remain largely unknown. RNAi is a method that allows the examination of gene function in a direct, targeted manner, by knocking out specific mRNAs and revealing the effects on the phenotype. In this study we have explored the viability of RNAi in Panagrolaimus sp. DAW1. In the first trial, nematodes were fed E. coli expressing Panagrolaimus sp. DAW1 dsRNA of the embryonic lethal genes rps-2 and dhc, and the blister gene duox. Pd-rps-2(RNAi)-treated nematodes showed a significant decrease in larval hatching. However, qPCR showed no significant decrease in the amount of rps-2 mRNA in Pd-rps-2(RNAi)-treated animals. Several soaking protocols for dsRNA uptake were investigated using the fluorescent dye FITC. Desiccation-enhanced soaking showed the strongest uptake of FITC and resulted in a significant and consistent decrease of mRNA levels of two of the four tested genes (rps-2 and tps-2a), suggesting effective uptake of dsRNA-containing solution by the nematode. These findings suggest that RNAi by desiccation-enhanced soaking is viable in Panagrolaimus sp. DAW1 and provide the first functional genomic approach to investigate freezing tolerance in this non-model organism. RNAi, in conjunction with qPCR, can be used to screen for candidate genes involved in intracellular freezing tolerance in Panagrolaimus sp. DAW1

    Expressive vocabulary predicts non-verbal executive function: a 2-year longitudinal study of deaf and hearing children

    Numerous studies suggest an association between language and executive function (EF), but evidence of a developmental relationship remains inconclusive. Data were collected from 75 deaf/hard-of-hearing (DHH) children and 82 hearing age-matched controls. Children were 6-11 years old at first time of testing, and completed a battery of nonverbal EF tasks and a test of expressive vocabulary. These tasks were completed again two years later. Both groups improved their scores on all tasks over this period. DHH children performed significantly less well than hearing peers on some EF tasks and the vocabulary test at both time points. Cross-lagged panel models showed that vocabulary at Time 1 predicted change in EF scores for both DHH and hearing children but not the reverse

    An exploratory study looking at the relationship marketing techniques used in the music festival industry

    There are current issues and trends in the music festival market, which may affect the success of an event, and market saturation is at the forefront of these issues. Previous literature, maintaining the need for a marketing approach to festivals, identifi es the need for maintaining strong stakeholder relationships in order to succeed in a business environment; attention has been focused to the theory of relationship marketing (RM) because of the recognition that this practice is complementary to the marketing of festivals. The very nature of the music festival as an annual, usually, 4-day event means that effective marketing is needed to keep connections with the consumer throughout the year. This article focuses on the RM techniques utilised within the music festival industry from the viewpoint of the festival organiser in an attempt to establish how festival organisations value and monitor organisational relationships. This article explores the extent to which these relationships are valued and managed; furthermore, the variations between these intricate relationships are considered by focusing on those held with the organisation ’ s consumers and sponsors, the results of which have provided the ability to establish the importance and relevance of RM to the industry and further identify the marketing communication methods employed to establish and maintain such relationships. In-depth, convergent interviews have been conducted with a segment of music festival organisers from a range of events. The results have been integrated with the study of current literature to best exemplify these issues. It has been established that RM has a strong role in today ’ s commercial and independent music festival industry; technological advances are enabling the organiser to support online relationships further and increase consumer loyalty. There is a need to expand the research further because of the complexity of organisational relationships and the varying categories of festivals

    Provider Counseling, Health Education, and Community Health Workers: The Arizona WISEWOMAN Project

    Background: The Arizona Well-Integrated Screening and Evaluation for Women Across the Nation (WISEWOMAN) project used provider counseling, health education, and community health workers (CHWs) to target chronic disease risk factors in uninsured, primarily Hispanic women over age 50. Methods: Participants were recruited from two Tucson clinics participating in the National Breast and Cervical Cancer Early Detection Program (NBCCEDP). Women were randomly assigned into one of three intervention groups: (1) provider counseling, (2) provider counseling and health education, or (3) provider counseling, health education, and CHW support. At baseline and 12 months (1998–2000), participants were measured for height, weight, waist and hip circumference, and blood pressure. Blood tests were conducted to check blood glucose, cholesterol, and triglyceride levels. At each time point, participants also completed 24-hour dietary recalls and questionnaires focusing on their physical activity levels. Results: A total of 217 women participated in baseline and 12-month follow-up. Three fourths were Hispanic. All three intervention groups showed an increase in self-reported weekly minutes of moderate-to-vigorous physical activity, with no significant differences between the groups. Significantly more women who received the comprehensive intervention of provider counseling, health education, and CHW support progressed to eating five fruits and vegetables per day, compared with participants who received only provider counseling or provider counseling plus health education. Conclusions: All three interventions increased moderate-to-vigorous physical activity but not fruit and vegetable consumption. The intervention group with provider counseling, health education, and CHW support significantly increased the number of women meeting national recommendations for fruit and vegetable consumption

    Investigating trehalose synthesis genes after cold acclimation in the Antarctic nematode Panagrolaimus sp. DAW1

    Panagrolaimus sp. DAW1 is a freeze-tolerant Antarctic nematode which survives extensive intracellular ice formation. The molecular mechanisms of this extreme adaptation are still poorly understood. We recently showed that desiccation-enhanced RNAi-soaking can be used - in conjunction with qPCR - to screen for phenotypes associated with reduced expression of candidate genes in Panagrolaimus sp. DAW1. Here we present the use of this approach to investigate the role of trehalose synthesis genes in this remarkable organism. Previous studies have showed that acclimating Panagrolaimus sp. DAW1 at 5 °C before freezing or desiccation substantially enhances survival. In this study the expression of tps-2 and other genes associated with trehalose metabolism, as well as lea-1, hsp-70 and gpx-1 in cold-acclimated and non-acclimated nematodes was analyzed using qPCR. Pd-tps-2 and Pd-lea-1 were significantly up-regulated after cold-acclimation, indicating an inducible expression in the cold-adaptation of Panagrolaimus sp. DAW1. The role of trehalose synthesis genes in Panagrolaimus sp. DAW1 was further investigated by RNAi. Compared to the controls, Pd-tps-2a(RNAi)-treated and cold-acclimated nematodes showed a significant decrease in mRNA, but no change in trehalose content or freezing survival. The involvement of two other trehalose synthesis genes (tps-2b and gob-1) was also investigated. These findings provide the first functional genomic investigation of trehalose synthesis genes in the non-model organism Panagrolaimus sp. DAW1. The presence of several trehalose synthesis genes with different RNAi sensitivities suggests the existence of multiple backup systems in Panagrolaimus sp. DAW1, underlining the importance of this sugar in preparation for freezing

    Identifying reports of randomized controlled trials (RCTs) via a hybrid machine learning and crowdsourcing approach

    OBJECTIVES: Identifying all published reports of randomized controlled trials (RCTs) is an important aim, but it requires extensive manual effort to separate RCTs from non-RCTs, even using current machine learning (ML) approaches. We aimed tomake this process more efficient via a hybrid approach using both crowdsourcing andML. METHODS: We trained a classifier to discriminate between citations that describe RCTs and those that do not. We then adopted a simple strategy of automatically excluding citations deemed very unlikely to be RCTs by the classifier and deferring to crowdworkers otherwise. RESULTS: Combining ML and crowdsourcing provid es a highly sensitive RCT identification strategy (our estimates suggest 95%-99% recall) with substantially less effort (we observed a reduction of around 60%-80%) than relying on manual screening alone. CONCLUSIONS: Hybrid crowd-ML strategies warrant further exploration for biomedical curation/annotation tasks

    Searching for AGN Outflows: Spatially Resolved Chandra HETG Spectroscopy of the NLR Ionization Cone in NGC 1068

    We present initial results from a new 440-ks Chandra HETG GTO observation of the canonical Seyfert 2 galaxy NGC 1068. The proximity of NGC 1068, together with Chandra’s superb spatial and spectral resolution, allow an unprecedented view of its nucleus and circumnuclear NLR. We perform the first spatially resolved high-resolution X-ray spectroscopy of the ‘ionization cone’ in any AGN, and use the sensitive line diagnostics offered by the HETG to measure the ionization state, density, and temperature at discrete points along the ionized NLR. We argue that the NLR takes the form of outflowing photoionized gas, rather than gas that has been collisionally ionized by the small-scale radio jet in NGC 1068. We investigate evidence for any velocity gradients in the outflow, and describe our next steps in modeling the spatially resolved spectra as a function of distance from the nucleus

    The InfraRed Imaging Spectrograph (IRIS) for TMT: latest science cases and simulations

    The Thirty Meter Telescope (TMT) first light instrument IRIS (Infrared Imaging Spectrograph) will complete its preliminary design phase in 2016. The IRIS instrument design includes a near-infrared (0.85 - 2.4 micron) integral field spectrograph (IFS) and imager that are able to conduct simultaneous diffraction-limited observations behind the advanced adaptive optics system NFIRAOS. The IRIS science cases have continued to be developed and new science studies have been investigated to aid in technical performance and design requirements. In this development phase, the IRIS science team has paid particular attention to the selection of filters, gratings, sensitivities of the entire system, and science cases that will benefit from the parallel mode of the IFS and imaging camera. We present new science cases for IRIS using the latest end-to-end data simulator on the following topics: Solar System bodies, the Galactic center, active galactic nuclei (AGN), and distant gravitationally-lensed galaxies. We then briefly discuss the necessity of an advanced data management system and data reduction pipeline.Comment: 15 pages, 7 figures, SPIE (2016) 9909-0
