Trinucleotide cassettes increase diversity of T7 phage-displayed peptide library
<p>Abstract</p> <p>Background</p> <p>Amino acid sequence diversity is introduced into a phage-displayed peptide library by randomizing library oligonucleotide DNA. We recently evaluated the diversity of peptide libraries displayed on T7 lytic phage and M13 filamentous phage and showed that T7 phage can display a more diverse amino acid sequence repertoire due to differing processes of viral morphogenesis.</p> <p>Methods</p> <p>In this study, we evaluated and compared the diversity of a 12-mer T7 phage-displayed peptide library randomized using codon-corrected trinucleotide cassettes with a T7 and an M13 12-mer phage-displayed peptide library constructed using the degenerate codon randomization method.</p> <p>Results</p> <p>We herein demonstrate that the combination of trinucleotide cassette amino acid codon randomization and T7 phage display construction methods resulted in a significant enhancement to the functional diversity of a 12-mer peptide library. This novel library exhibited superior amino acid uniformity and order-of-magnitude increases in amino acid sequence diversity as compared to degenerate codon randomized peptide libraries. Comparative analyses of the biophysical characteristics of the 12-mer peptide libraries revealed the trinucleotide cassette-randomized library to be a unique resource.</p> <p>Conclusion</p> <p>The combination of T7 phage display and trinucleotide cassette randomization resulted in a novel resource for the potential isolation of binding peptides for new and previously studied molecular targets.</p
Takotsubo cardiomyopathy and sepsis: a systematic review
Takotsubo cardiomyopathy (TTC) is characterized by a systolic dysfunction localized in the apical and medial aspect of
the left ventricle. It is usually related to physical or emotional stress. Recent evidence highlighting the role of infection
led us to analyze the links between TTC and sepsis. A systematic review of the literature was undertaken to assess any
trends in clinical findings, diagnosis, and outcomes in such patients. We identified 23 selected papers reporting a total of
26 patients, having sepsis, in whom TTC occurred. For each case, we collected data identifying population characteristics,
source of sepsis, clinical disease description, and the results of cardiovascular investigations. The majority of patients
were females (n = 16), mean age was 62.8 (14.0 standard deviation) years, and clinical outcome was favorable in 92.3% of
the cases once the management of sepsis was initiated. A better understanding of the mechanisms of sepsis-associated
TTC may generate novel strategies to treat the complications of this cardiomyopathy and may even help predict and
prevent its occurrence

Medium-term seed storage of 50 genera of forage legumes and evidence-based genebank monitoring intervals
Genebanks maintaining seeds for long-term genetic resources conservation monitor seed lots to detect early loss in viability. Monitoring is costly and depletes valuable seed. Three decades of genebank seed germination test results of diverse forage species from 50 legume genera in the International Livestock Research Instituteâs medium-term store (circa 8° C with 5 % moisture content) were analysed to determine whether advice on seed monitoring intervals could be derived. Cumulative normal distributions were fitted by probit analysis for each seed lot and compared within each genus. Six patterns of within-genus variation were identified: no detectable trend in germination test results during storage (4 genera); detectable trends, but variable (positive to negative) amongst lots (5); consistent slope of loss in viability amongst lots (17); consistent slope of increase in ability to germinate amongst lots (21); common loss in viability amongst lots (2); common increase in ability to germinate amongst lots (1). Seed lot monitoring intervals for the medium-term store were derived for each of 19 genera with consistent loss in viability across seed lots: three genera provided comparatively rapid deterioration, five met the general expectations for a medium-term store (2-10 yearsâ maintenance of high viability), whilst 11 provided much better survival. Moreover, 26 further genera provided no evidence as yet of seed deterioration; of these, 22 improved in ability to germinate during storage indicating confounding of hardseededness with viability in germination tests
Protocol for studying cough frequency in people with pulmonary tuberculosis.
INTRODUCTION: Cough is a key symptom of tuberculosis (TB) as well as the main cause of transmission. However, a recent literature review found that cough frequency (number of coughs per hour) in patients with TB has only been studied once, in 1969. The main aim of this study is to describe cough frequency patterns before and after the start of TB treatment and to determine baseline factors that affect cough frequency in these patients. Secondarily, we will evaluate the correlation between cough frequency and TB microbiological resolution. METHODS: This study will select participants with culture confirmed TB from 2 tertiary hospitals in Lima, Peru. We estimated that a sample size of 107 patients was sufficient to detect clinically significant changes in cough frequency. Participants will initially be evaluated through questionnaires, radiology, microscopic observation drug susceptibility broth TB-culture, auramine smear microscopy and cough recordings. This cohort will be followed for the initial 60â
days of anti-TB treatment, and throughout the study several microbiological samples as well as 24â
h recordings will be collected. We will describe the variability of cough episodes and determine its association with baseline laboratory parameters of pulmonary TB. In addition, we will analyse the reduction of cough frequency in predicting TB cure, adjusted for potential confounders. ETHICS AND DISSEMINATION: Ethical approval has been obtained from the ethics committees at each participating hospital in Lima, Peru, Asociación Benéfica PRISMA in Lima, Peru, the Universidad Peruana Cayetano Heredia in Lima, Peru and Johns Hopkins University in Baltimore, USA. We aim to publish and disseminate our findings in peer-reviewed journals. We also expect to create and maintain an online repository for TB cough sounds as well as the statistical analysis employed
Constraint Release mechanisms for H-Polymers moving in Linear Matrices of varying molar masses
We investigate the influence of the environment on the relaxation dynamics of well-defined H-polymers diluted in a matrix of linear chains. The molar mass of the linear chain matrix is systematically varied and the relaxation dynamics of the H-polymer is probed by means of linear viscoelastic measurements, with the aim to understand its altered motion in different blends, compared to its pure melt state. Our results indicate that short unentangled linear chains accelerate the relaxation of both the branches and the backbone of the H-polymers by acting as an effective solvent. On the other hand, the relaxation of the H-polymer in an entangled matrix is slowed-down, with the degree of retardation depending on the entanglement number of the linear chains. We show that this retardation can be quantified by considering that the H-polymers are moving in a dilated tube at the rhythm of the motion of the linear matrix
Risk and sensitivity quantification of fracture failure employing cohesive zone elements
Many structures are subjected to the risk of fatigue failure. For their reliability-based design, it is thus important to calculate the probability of fatigue failure and assess the relative importance of the involved parameters. Although various studies have analyzed the fatigue failure, the stage of fracture failure has been less focused. In particular, the risk analysis of fracture failure needs to be conducted considering its importance in actual structures. This article proposes a new probabilistic framework for the risk and sensitivity analysis of structural fatigue failure employing cohesive zone elements. The proposed framework comprises three steps, namely finite element analysis using cohesive zone elements, response surface construction, and risk and sensitivity analysis of fatigue failure, which require several mathematical techniques and algorithms. The proposed framework is tested by applying it to an illustrative example, and the corresponding analysis results of fracture failure probability with different threshold values of a limit-state function are presented. In addition, the sensitivities of failure risk with respect to the statistical parameters of random variables are presented and their relative importance is discussed
Mechanical Metamaterials with Negative Compressibility Transitions
When tensioned, ordinary materials expand along the direction of the applied
force. Here, we explore network concepts to design metamaterials exhibiting
negative compressibility transitions, during which a material undergoes
contraction when tensioned (or expansion when pressured). Continuous
contraction of a material in the same direction of an applied tension, and in
response to this tension, is inherently unstable. The conceptually similar
effect we demonstrate can be achieved, however, through destabilisations of
(meta)stable equilibria of the constituents. These destabilisations give rise
to a stress-induced solid-solid phase transition associated with a twisted
hysteresis curve for the stress-strain relationship. The strain-driven
counterpart of negative compressibility transitions is a force amplification
phenomenon, where an increase in deformation induces a discontinuous increase
in response force. We suggest that the proposed materials could be useful for
the design of actuators, force amplifiers, micro-mechanical controls, and

Field Emission of ITO-Coated Vertically Aligned Nanowire Array
An indium tin oxide (ITO)-coated vertically aligned nanowire array is fabricated, and the field emission characteristics of the nanowire array are investigated. An array of vertically aligned nanowires is considered an ideal structure for a field emitter because of its parallel orientation to the applied electric field. In this letter, a vertically aligned nanowire array is fabricated by modified conventional UV lithography and coated with 0.1-ÎŒm-thick ITO. The turn-on electric field intensity is about 2.0 V/ÎŒm, and the field enhancement factor, ÎČ, is approximately 3,078 when the gap for field emission is 0.6 ÎŒm, as measured with a nanomanipulator in a scanning electron microscope
Discovery of the Acoustic Faraday Effect in Superfluid 3He-B
We report the discovery of the acoustic Faraday effect in superfluid 3He-B.
The observation of this effect provides the first direct evidence for
propagating transverse acoustic waves in liquid 3He, a mode first predicted by
Landau in 1957. The Faraday rotation is large and observable because of
spontaneously broken spin-orbit symmetry in 3He-B. We compare the experimental
observations with a simulation of the transverse acoustic impedance that

Dynamic upregulation of CD24 in pre-adipocytes promotes adipogenesis
The development of mature adipocytes from pre-adipocytes is a highly regulated process. CD24 is a
glycophosphatidylinositol-linked cell surface receptor that has been identified as a critical cell surface marker for identifying pre-adipocytes that are able to reconstitute white adipose tissue (WAT) in vivo. Here, we examined the role and regulation of CD24 during adipogenesis in vitro. We found that CD24 mRNA and protein expression is upregulated early during adipogenesis in the 3T3-L1 pre-adipocytes and in murine primary pre-adipocytes isolated from subcutaneous and visceral WAT, followed by downregulation in mature adipocytes. CD24 mRNA expression was found to be dependent on increased transcription due to increased promoter activity in response to activation of a preexisting transcriptional regulator. Furthermore, either intracellular cAMP or dexamethasone were sufficient to increase expression in pre-adipocytes, while both additively increased CD24 expression. Preventing the increase in CD24 expression, by siRNA-mediated knock-down, resulted in fewer mature lipid-laden adipocytes and decreased expression of mature adipogenic genes. Therefore, conditions experienced during adipogenesis in vitro are sufficient to increase CD24 expression, which is necessary for differentiation. Overall, we conclude that the dynamic upregulation of CD24 actively promotes adipogenesis in vitro
