    Intracellular localization of tyrosine kinase substrates beneath crosslinked surface immunoglobulins in B cells

    Crosslinking of surface immunoglobulins (sIg) in B cells led to the accumulation of submembranal phosphotyrosine, which was followed morphologically with the PY20 antiphosphotyrosine monoclonal antibody. Phosphotyrosine was not detected before sIg crosslinking. After sIg crosslinking, phosphotyrosine-containing proteins were redistributed from scattered small clusters near the plasma membrane to a juxtanuclear region, where immunofluorescent staining decreased with time. Double immunofluorescent staining of individual cells showed accumulation of phosphotyrosine beneath crosslinked sIg molecules at the cell surface. The sIg molecules were subsequently internalized more rapidly than the phosphotyrosine-containing molecules were redistributed. Genistein, a protein tyrosine kinase (PTK) inhibitor, blocked intracellular tyrosine phosphorylations but not cell surface patching of crosslinked sIg. When polyacrylamide beads coated with anti-Ig antibodies were added to the cells, intracellular tyrosine phosphorylation occurred beneath the regions of contact with the beads. This study provides an independent line of evidence confirming recent biochemical experiments that show that crosslinking of the antigen receptor induces PTK activity in B cells, and that components of the newly described sIg complex are among the PTK substrates. The surprising finding that the bulk of the induced phosphotyrosine remains associated with crosslinked sIg for many minutes suggests a role for complex local protein interactions in phosphotyrosine-mediated signal transduction through the antigen receptor of B cells

    A J-Like Protein Influences Fatty Acid Composition of Chloroplast Lipids in Arabidopsis

    A comprehensive understanding of the lipid and fatty acid metabolic machinery is needed for optimizing production of oils and fatty acids for fuel, industrial feedstocks and nutritional improvement in plants. T-DNA mutants in the poorly annotated Arabidopsis thaliana gene At1g08640 were identified as containing moderately high levels (50–100%) of 16∶1Δ7 and 18∶1Δ9 leaf fatty acids and subtle decreases (5–30%) of 16∶3 and 18∶3 (http://www.plastid.msu.edu/). TLC separation of fatty acids in the leaf polar lipids revealed that the chloroplastic galactolipids monogalactosyldiacylglycerol (MGDG) and digalactosyldiacylglycerol (DGDG) were the main lipid types affected by this mutation. Analysis of the inferred amino acid sequence of At1g08640 predicted the presence of a transit peptide, three transmembrane domains and an N-terminal J-like domain, and the gene was named CJD1 for Chloroplast J-like Domain 1. GFP reporter experiments and in vitro chloroplast import assays demonstrated CJD1 is a chloroplast membrane protein. Screening of an Arabidopsis cDNA library by yeast-2-hybrid (Y2H) using the J-like domain of CJD1 as bait identified a plastidial inner envelope protein (Accumulation and Replication of Chloroplasts 6, ARC6) as the primary interacting partner in the Y2H assay. ARC6 plays a central role in chloroplast division and binds CJD1 via its own J-like domain along with an adjacent conserved region whose function is not fully known. These results provide a starting point for future investigations of how mutations in CJD1 affect lipid composition

    The relationship between web enjoyment and student perceptions and learning using a web-based tutorial

    Web enjoyment has been regarded as a component of system experience. However, there has been little targeted research considering the role of web enjoyment alone in student learning using web-based systems. To address this gap, this study aims to examine the influence of web enjoyment on learning performance and perceptions by controlling system experience as a variable in the study. 74 students participated in the study, using a web-based tutorial covering subject matter in the area of 'Computation and algorithms'. Their learning performance was assessed with a pre-test and a post-test and their learning perceptions were evaluated with a questionnaire. The results indicated that there are positive relationships between the levels of web enjoyment and perceived usefulness and non-linear navigation for users with similar, significant levels of system experience. The implications of these findings in relation to web-based learning are explored and ways in which the needs of students who report different levels of web enjoyment might be met are discussed

    Comparative transcriptomics and metabolomics reveal specialized metabolite drought stress responses in switchgrass (<i>Panicum virgatum</i> L.)

    ⋅ Switchgrass (Panicum virgatum) is a bioenergy model crop valued for its energy efficiency and drought tolerance resilience. The related monocot species rice (Oryza sativa) and maize (Zea mays) deploy species-specific, specialized metabolites as core stress defenses. By contrast, specialized chemical defenses in switchgrass are largely unknown. ⋅ To investigate specialized metabolic drought responses in switchgrass, we integrated tissue-specific transcriptome and metabolite analyses of the genotypes Alamo and Cave-in-Rock that feature different drought tolerance. ⋅ The more drought-susceptible Cave-in-Rock featured an earlier onset of transcriptomic changes and significantly more differentially expressed genes in response to drought compared to Alamo. Specialized pathways showed moderate differential expression compared to pronounced transcriptomic alterations in carbohydrate and amino acid metabolism. However, diterpenoid-biosynthetic genes showed drought-inducible expression in Alamo roots, contrasting largely unaltered triterpenoid and phenylpropanoid pathways. Metabolomic analyses identified common and genotype-specific flavonoids and terpenoids. Consistent with transcriptomic alterations, several root diterpenoids showed significant drought-induced accumulation, whereas triterpenoid abundance remained predominantly unchanged. Structural analysis of drought-responsive root diterpenoids verified these metabolites as oxygenated furanoditerpenoids. ⋅ Drought-dependent transcriptome and metabolite profiles provide the foundation to understand the molecular mechanisms underlying switchgrass environmental resilience. Accumulation of specialized root diterpenoids and corresponding pathway transcripts supports a role in drought stress tolerance for these compounds

    Increased occurrence of the jellyfish Periphylla periphylla in the European high Arctic

The final authenticated version is available online at https://doi.org/10.1007/s00300-018-2368-4.The jellyfish Periphylla periphylla, which can have strong ecological impacts on its environment, is ubiquitous in the Norwegian Sea and its range was predicted to extend northwards. The occurrence of P. periphylla in the northern Barents Sea increased since 2014 and, for the first time, several individuals were collected within a high Arctic fjord (> 78°N) in western Spitsbergen in January 2017. The low solar irradiance prevailing during the polar night and an increased inflow of relatively warm Atlantic water in the European Arctic since the last decade likely provide suitable conditions for the medusa to colonize Svalbard’s fjords during the winter months. However, light avoidance constrains the photophobic P. periphylla to deeper offshore areas during the midnight sun period. The current occurrence of P. periphylla in high Arctic fjords during the polar night will have a limited impact on marine ecosystems in the short term, but long-term effects are more uncertain if its abundance continues to increase

    The diagnosis of bilateral primary renal paragangliomas in a cat

    A 9-year-old sterilised female domestic short-hair cat was referred with a history of vomiting and anorexia of 3 months’ duration. Biochemistry, full-blood counts, thoracic radiographs, feline pancreatic-specific lipase, abdominal ultrasonography and feline immunodeficiency virus/feline leukaemia virus (FIV/FeLV) SNAP tests had been performed. Mild hypochloraemia and moderate hypokalaemia were evident on initial presentation. Abdominal ultrasonography initially revealed unilateral renal nodules on the left side. These were subjected to fine-needle aspiration and cytological evaluation. A neuroendocrine tumour was suspected, and biopsies via midline coeliotomy were taken to confirm the diagnosis. Initial histopathology diagnosed primary renal carcinomas or neuroendocrine neoplasia; however, the definitive diagnosis became renal paragangliomas after immunohistochemistry and transmission electron microscopy were performed. The cat was regularly monitored with serum biochemistry parameters, blood pressure determinations, thoracic radiographs and subsequent abdominal ultrasonography. Biochemistry, radiography and blood pressures remained normal over a 24-week follow-up period, while subsequent ultrasonography revealed tumour progression in both number and size in both kidneys. Primary neuroendocrine tumours of the kidney are frequently incorrectly diagnosed as other renal tumours such as renal cell carcinoma, mesonephric tumours or undifferentiated carcinomas.     The impact of environmental variability on Atlantic mackerel Scomber scombrus larval abundance to the west of the British Isles

    The value of the Continuous Plankton Recorder (CPR) fish larvae dataset, with its extensive spatio-temporal coverage, has been recently demonstrated with studies on long-term changes over decadal scales in the abundance and distribution of fish larvae in relation to physical and biological factors in the North Sea. We used a similar approach in the west and southwest area of the UK shelf and applied a principal component analysis (PCA) using 7 biotic and abiotic parameters, combined with Hierarchical Cluster Analysis (HCA), to investigate the impact of environmental changes in the west and southwest area of the UK shelf on mackerel larvae during the period 1960–2004. The analysis revealed 3 main periods of time (1960–1968; 1969–1994; 1995–2004) reflecting 3 different ecosystem states. The results suggest a transition from an ecosystem characterized by low temperature, high salinity, high abundances of zooplankton and the larger phytoplankton groups, to a system characterized by higher temperature, lower salinities, lower abundances of zooplankton and larger phytoplankton and higher abundances of the small phytoplankton species. Analysis revealed a very weak positive correlation between the Second principal component and mackerel larvae yearly abundance, attributed to the North Atlantic Oscillation (NAO). The results presented here are in broad accord with recent investigations that link climatic variability and dynamics of mackerel reproduction. However, the growing body of literature that documents statistical correlations between environment and mackerel needs to be supplemented by local process studies, to gain more insight and to be able to predict mackerel response to climate change scenarios. Utilising the strength of the CPR dataset, namely its unique temporal coverage, in an analysis where other data (such as egg surveys) are drawn in to compensate for the spatial issues could prove to be the way forward

    Mobility as a contradictory resource: Peripatetic Qur’anic students in Kano, Nigeria

    Mobility is a powerful resource young people can draw on to improve their lives, but it can also entail risks. This paper explores how mobility becomes a contradictory resource for peripatetic Qur'anic students (almajirai) in Kano State in northern Nigeria. Moving to urban areas allows the young almajirai to escape difficult conditions and to access educational and income opportunities absent in their rural homes. It makes it possible for them to adopt self-conceptions as migrants in search of sacred knowledge who were once widely respected. However, economic decline has made survival in the city more difficult. Lacking the economic and cultural resources to participate in displays of status, and without social superiors to speak for them, the almajirai feel they have become fair game for those searching for scapegoats
