Bytový dům se nachází na pozemku svažujícím se k jihu, s hlavní příjezdovou komunikací z ulice Tomečkova. Pozemek se nachází v jihovýchodní části panelového sídliště Lesná, zároveň je v těsném sousedství drobné zástavby rodinnými domy, tzv. Divišovy kolonie. Architektonicky je bytový dům řešen jako dva samostatné celky spojené v jeden funkční blok. Záměrem bylo využít svažujícího se pozemku, proto vznikl terasový dům s postupně ubírající hmotou, z důvodu lepšího oslunění a vzniku teras. Navržená bílá fasáda s tmavými rámy oken doplňuje gradaci objektu. V jižní části pozemku je navrhnut předprostor se zelení a přístupovými komunikacemi pro pěší. Veškeré pěší trasy kolem objektu jsou vydlážděny.Residential house is located on site sloping to the south. Main access road is from the street ‘Tomečkova’. The place is located in the south-eastern part of the housing estate ‘Lesná’ and it is also close to the group of small houses, called ‘Divišova kolonie’. Building is designed as two separate units connected in one functional block. The intention was to use the sloping land to create a terraced house with gradually diminishing material, for better exposure to sun and the formation of terraces. Proposed white façade, with dark window frames complements object’s gradation. The southern part of the site was designed as a front place with vegetation and access roads for pedestrians. All footpaths around the building are paved.

Optimization of recombinant protein production in animal cell culture

V této diplomové práci je popsána přechodná transfekce buněk 293 HEK adaptovaných na růst při suspenzní kultivaci bez přítomnosti séra za použití polyethyleniminů (PEI). Buňky byly transfekovány plasmidem pcDNA5/SEAP, který exprimuje sekretovanou formu lidské placentální alkalické fosfatázy. K porovnání účinnosti jednotlivých transfekcí byla měřena koncentrace exprimované fosfatázy v buněčném supernatantu. Cílem této práce bylo optimalizovat různé faktory ovlivňující účinnost transfekcí s důrazem na nalezení optimálního poměru DNA:PEI.The thesis describes successful transient transfections of suspension-adapted HEK 293 cells with linear polyethyleneimines (PEIs) in serum-free medium. The cells were transfected with pcDNA5/SEAP plasmid expressing the secreted form of human placental alkaline phosphatase (SEAP). SEAP concentrations in cell culture supernatants were determined in order to compare efficiencies of individual transfections. The goal of the thesis was to optimize various factors affecting transfection efficiencies, primarily the DNA:PEI ratio.

Analysis of Service Quality in the Restaurant Chain

Cílem této práce je sjednotit úroveň poskytovaných služeb restauračního řetězce, a to takovým způsobem, který bude vyhovovat současným požadavkům zákazníků Radegastoven. Analyzovány byly pobočky okresních měst Moravskoslezského kraje. Analýza proběhla kombinací kvantitativní a kvalitativní metody. Kvantitativní analýza proběhla formou elektronického dotazování a kvalitativní část metodou Mystery shopping. Provedeným výzkumem byly zjištěny preference, očekávání a spokojenost zákazníků Radegastoven. Zákazníci v Radegastovně oceňují skvěle načepované pivo a moderní interiér, ale nejsou moc spokojeni s personálem, který velmi ovlivňuje jejich spokojenost. Pro sjednocení úrovní poboček, je tedy potřeba zlepšit především kvalitu personálu. V rámci odlišností jednotlivých poboček, je třeba zaměřit se na pobočku v Karviné. Pro tuto pobočku je zásadní zajistit především školení personálu, dotažení vzhledu interiéru, obstarání stejnokroje pro obsluhu a zajištění informací o pobočce na webových stránkách.Aim of the thesis is to unify the level of services in the restaurant chain of Radegastovna, in a way that will meet the current requirements of their customers. The analyze was realized in district towns of the Moravian-Silesian Region. The analysis was performed by a combination of quantitative and qualitative methods. Quantitative analysis was realized with a questionnaire and the qualitative part was realized with Mystery Shopping. The research revealed the preferences, expectations and satisffaction of Radegastovna's customers. These customers appreciate well-drafted beer and modern interior, but they are not very satisfied with the employees, which greatly affects their satisfaction. In order to unify the levels of branches, it is necessary to improve the quality of employees. Within the differences of individual branches, it is necessary to focus on the branch in Karviná city. Assessment of Financial Products for Retirement in the Czech Republic

Předmětem této bakalářské práce je prozkoumání finančního trhu a výběr vhodných variant produktů pro zabezpečení na stáří v České republice. V první části je podrobně probrán současný důchodový systém v České republice, základní fungování systému, veličiny pro výpočet důchodu, podmínky pro splnění nároku na výplatu důchodu a možné druhy důchodových dávek. V druhé části je práce zaměřena na jednotlivé produkty, které se dají na zabezpečení na stáří využít, převážně na finančním trhu. Důkladněji rozvedeny jsou program doplňkové penzijní spoření, který nahradil dřívější produkt penzijní připojištění, dále varianty životního pojištění z nich je nejvíce rozebrána varianta investičního životního pojištění a třetí je investování do podílových fondů. V třetí praktické části jsou pro tři různé věkové kategorie zhodnoceny vždy dva porovnatelné produkty a na základě srovnání těchto produktů je zvolen ten, který je pro danou věkovou kategorii více vhodný.The subject of this bachelor thesis is to examination the financial market and to select the appropriate options of products for retirement in the Czech Republic. In the first part, there are a detailed description of the current pension system of Czech Republic, his basic functioning, parameters for the calculation of pensions, the conditions of qualifying for a retirement pension a possible types of retirement benefits. In the second part, there are individual products that we can use to retirement, exclusively on the Financial market. In more detail, they are described the complimentary pension savings, which replaced the supplementary pension contribution, next there we can find different variants of life instances, special the investment life insurance and the third is the investment to the mutual funds. 5-Minutes Neighbourhood

Řešená lokalita se nachází v městské části Brno-Židenice, v těsné blízkosti železniční tratě spojující Brno s Českou Třebovou. Území je vymezeno z jihu ulicí Bubeníčkova, z východu ulicí Koperníkova, ze severu Lazaretní a ze západu vymezuje území silnice kopírující železniční trať. Základní hmota projektovaného objektu je rozdělena průchozími osami na tři bloky, jež jsou propojené společným podzemním podlažím, kde je situováno především parkování. Bloky mají až šest nadzemních podlaží. V prvním nadzemním podlaží jsou různé komerční prostory pro obchody, restaurace a ve střední části se nachází supermarket. V severním objektu jsou v druhém a vyšším podlaží situovány různé funkce občanské vybavenosti (školka, různé mimoškolní aktivity pro děti, wellness, zábavní centrum, jazyková škola). V posledním podlaží je řešeno bydlení. V prostředním bloku je od druhého podlaží řešeno bydlení, v jižním bloku nalezneme komerční prostory, polikliniku, administrativu či knihovnu.The locality is situated in the city quarter Brno-Židenice in close proximity of the railway line connecting Brno and Česká Třebová. The area is delimited by the Bubeníčkova Street from the South, the Koperníkova Street from the East, by Lazaretní Street from the North and by a road copying the railway line from the West. The elemental form of the projected compound is divided into three blocks by pass-through axes. The blocks are interconnected with a common basement where underground car park is located. The blocks have up to six floors above ground. On the first floor, there is a variety of commercial spaces for shops and restaurants and in the middle there is a supermarket. In the northern object, there are situated different types of amenities (kinder-garden, leisure activities for children, spa) on the second and higher floors. On the top floor of this object we can find the flats as well as in the middle building from the second floor. The commercial areas, offices and library are situated in the southern block.

Working with Synonyms in Teaching of Russian Language at High School

The aim of this thesis is to evaluate the usage of synonyms in teaching Russian language at high schools. The theoretical part of this thesis outlines the issues of synonyms and synonymous series from the linguistic (paradigmatic relations, semantics, stylistic) and linguodidactic point of view. The theoretical part also includes the issue of vocabulary from the didactic point of view. Furthermore, in the practical part, the diversity of synonyms used in curriculum of Russian language teaching are evaluated as well as the methods of demonstration and practising of such curriculum. In the final part of this thesis my own concepts for working with synonyms in teaching Russian language at high schools are suggested. Bakalářská práce se zabývá konstrukčním návrhem ruční pračky s řemenovým převodem. Cílem je navrhnout pračku tak, aby splňovala zadané podmínky. V úvodní části je zpracován krátký přehled zabývající se historií praní prádla a vývojem praček. V další části jsou příklady současných ručních praček. Práce pokračuje popisem a potřebnými výpočty již navrženého zařízení. V práci jsou také uvedeny pevnostní kontroly hřídelí a šroubových spojů. Součástí práce je výkresová dokumentace.This bachelor thesis deals with the design of a hand-operated washing machine with a belt drive. The aim of the thesis is to design the device to meet the specified conditions. In the introductory part, there is a short overview dealing with the history of laundry washing and the development of washing machines. In the next section, there are examples of current hand washers. The work continues with a description and necessary calculations of the already designed equipment. Design of a bending tool for U-bending of sheet metal

Bakalářská práce s názvem Návrh ohýbacího nástroje pro U-ohyb plechu, zpracovaná v rámci bakalářského studia oboru B-STG, předkládá návrh technologie výroby lišty do tvaru U. Zadaná součást je vyrobena z konstrukční oceli S235 JR o tloušťce 2 mm. Navržený ohýbací nástroj bude sloužit k výrobě lišty z ocelového pozinkovaného plechu pro montáž regálů pro spotřebitelský průmysl. Součástí návrhu ohýbacího nástroje pro U-ohyb jsou technologické výpočty, výkresová dokumentace a technicko-ekonomické zhodnocení.The Bachelor thesis entitled The proposal of bending tool for U-sheet-bending, worked out under the bachelor studies of B-STG branch, presents a proposal of technology of production of a batten in U-form. The assigned part is made from construct steel S235 JR of 2 mm thickness. The proposed bending tool is designed to be used for production of a batten from galvanized steel sheet for assembly of shelves in consumer industry. This proposal of bending tool for U-bending includes technological calculations, drawing documents and technical-economic evaluation.

Test equipment for testing glued joints.

Diplomová práce se zabývá konstrukčním návrhem zkušebního zařízení na testování lepených spojů. To bude sloužit pro experimentální výuku a pro výzkumnou činnost. V úvodní částí práce je zpracován přehled druhů lepených spojů a typů lepidel. Následuje rešerše z norem zabývajících se testováním lepených spojů. Práce pokračuje základními výpočty vstupních hodnot a několika variantami řešení upínacích čelistí a přenosu síly. Dále jsou uvedeny pevnostní kontroly dílů, způsob snímání a záznamu síly a návrh metodiky provádění zkoušek.This diploma thesis deals with the construction design of the tensile testing machine for glued joints. This equipment will be used for experimental teaching and for various research activities. In the indtroductory part of the thesis there is a summary of types of glued joints and sorts of adhesives. The research of standards dealing with glued joints testing follows. The thesis continues with the basic calculations of input values and a few options ot the possible solution of fixing jaws and force transfer. Valuation of the Company in the Non-alcoholic Beverage Industry by the Yield Methods

Cílem diplomové práce je ocenění podniku Kofola a.s. a stanovení její tržní hodnoty pomocí výnosových metod. Ocenění bylo provedeno pomocí dvoufázových metod DCF-Entity a EVA entity. Srovnání bylo provedeno, také s metodou účetní. Práce je rozčleněna do 5 kapitol, včetně úvodu a závěru. Druhá kapitola se týká metodiky související s oceňováním podniku. V třetí kapitole je daná metodika aplikovaná na vybraný podniku a čtvrtá kapitola obsahuje zhodnocení dosažených výsledků a analýzu rizika pomocí citlivostní analýzy a scénářů vývoje podniku.The aim of the thesis is the valuation of the company Kofola a.s. and to establish its market value with using the yield method. The valuation was made by a two-phased method od DFC-Entity and EVA entity. The comparison was also made with the accounting method. The thesis is divided into five chapters, including the introduction and the conclusion. The second chapter concerns to the methodology of the valuation of the company.
