    Differential ABC transporter gene expression in adult Dirofilaria immitis males and females following in vitro treatment with ivermectin, doxycycline or a combination of both

    BACKGROUND: Combination doxycycline/macrocyclic lactone (ML) protocols have been shown to provide a more rapid adulticidal and microfilaricidal effect than either MLs or doxycycline alone, although female worms were reported to have a higher tolerance to treatments compared to male worms. The present study aimed to evaluate how ABC transporters may be involved in the synergic effect of the combination treatment. Adult worms of D. immitis were treated in vitro for 24 hours with doxycycline (DOXY), ivermectin (IVM) and a combination of both, and changes in the modulation of ABC transporter genes were measured. Levels of doxycycline inside different treatment media, post-treatment, were determined through HPLC analysis. RESULTS: Quantitative RT-PCR analysis showed the presence of changes in the modulation of ABC transporter genes evaluated in this study. In particular, in female worms, the combination treatment induced a substantial increase in gene expressions, especially of Dim-pgp-10 and Dim-haf-4; whereas in male worms, the greatest increase in gene expression was observed for Dim-pgp-10 and Dim-pgp-11 when treated with DMSO\u2009+\u2009IVM and DMSO\u2009+\u2009DOXY/IVM. HPLC analysis of the DOXY concentrations in the media after in vitro treatments of male worms showed a slight difference between the DMSO\u2009+\u2009DOXY samples and the combination (DMSO\u2009+\u2009DOXY\u2009+\u2009IVM), while no difference was observed among females. CONCLUSIONS: Further studies are required to explain whether the modulation of cellular efflux plays a role, even partially, in the adulticide effect of doxycycline/macrocyclic lactone combinations in heartworm-infected dogs. To the authors' knowledge, this is the first study to evaluate P-gp expression in adult D. immitis

    Brave New World versus Island — Utopian and Dystopian Views on Psychopharmacology

    Aldous Huxley’s Brave New World is a famous dystopia, frequently called upon in public discussions about new biotechnology. It is less well known that 30 years later Huxley also wrote a utopian novel, called Island. This paper will discuss both novels focussing especially on the role of psychopharmacological substances. If we see fiction as a way of imagining what the world could look like, then what can we learn from Huxley’s novels about psychopharmacology and how does that relate to the discussion in the ethical and philosophical literature on this subject? The paper argues that in the current ethical discussion the dystopian vision on psychopharmacology is dominant, but that a comparison between Brave New World and Island shows that a more utopian view is possible as well. This is illustrated by a discussion of the issue of psychopharmacology and authenticity. The second part of the paper draws some further conclusions for the ethical debate on psychopharmacology and human enhancement, by comparing the novels not only with each other, but also with our present reality. It is claimed that the debate should not get stuck in an opposition of dystopian and utopian views, but should address important issues that demand attention in our real world: those of evaluation and governance of enhancing psychopharmacological substances in democratic, pluralistic societies

    Measurement of the neutron magnetic form factor from inclusive quasielastic scattering of polarized electrons from polarized 3He

    We report a measurement of the asymmetry in spin-dependent quasielastic scattering of longitudinally polarized electrons from a polarized 3He target. The neutron magnetic form factor GMn has been extracted from the measured asymmetry based on recent PWIA calculations using spin-dependent spectral functions. Our determination of GMn at Q2=0.19 (GeV/c)2 agrees with the dipole parametrization. This experiment represents the first measurement of the neutron magnetic form factor using spin-dependent electron scattering

    Measurement of the Neutron Spin Structure Function g1ng_1^n with a Polarized ^3He Target

    Results are reported from the HERMES experiment at HERA on a measurement of the neutron spin structure function g1n(x,Q2)g_1^n(x,Q^2) in deep inelastic scattering using 27.5 GeV longitudinally polarized positrons incident on a polarized 3^3He internal gas target. The data cover the kinematic range 0.023<x<0.60.023<x<0.6 and 1(GeV/c)2<Q2<15(GeV/c)21 (GeV/c)^2 < Q^2 <15 (GeV/c)^2. The integral 0.0230.6g1n(x)dx\int_{0.023}^{0.6} g_1^n(x) dx evaluated at a fixed Q2Q^2 of 2.5(GeV/c)22.5 (GeV/c)^2 is 0.034±0.013(stat.)±0.005(syst.)-0.034\pm 0.013(stat.)\pm 0.005(syst.). Assuming Regge behavior at low xx, the first moment Γ1n=01g1n(x)dx\Gamma_1^n=\int_0^1 g_1^n(x) dx is 0.037±0.013(stat.)±0.005(syst.)±0.006(extrapol.)-0.037\pm 0.013(stat.)\pm 0.005(syst.)\pm 0.006(extrapol.).Comment: 4 pages TEX, text available at http://www.krl.caltech.edu/preprints/OAP.htm

    Virtual Compton Scattering and Neutral Pion Electroproduction in the Resonance Region up to the Deep Inelastic Region at Backward Angles

    We have made the first measurements of the virtual Compton scattering (VCS) process via the H(e,ep)γ(e,e'p)\gamma exclusive reaction in the nucleon resonance region, at backward angles. Results are presented for the WW-dependence at fixed Q2=1Q^2=1 GeV2^2, and for the Q2Q^2-dependence at fixed WW near 1.5 GeV. The VCS data show resonant structures in the first and second resonance regions. The observed Q2Q^2-dependence is smooth. The measured ratio of H(e,ep)γ(e,e'p)\gamma to H(e,ep)π0(e,e'p)\pi^0 cross sections emphasizes the different sensitivity of these two reactions to the various nucleon resonances. Finally, when compared to Real Compton Scattering (RCS) at high energy and large angles, our VCS data at the highest WW (1.8-1.9 GeV) show a striking Q2Q^2- independence, which may suggest a transition to a perturbative scattering mechanism at the quark level.Comment: 20 pages, 8 figures.     Measurement of the Proton Spin Structure Function g1p with a Pure Hydrogen Target

    A measurement of the proton spin structure function g1p(x,Q^2) in deep-inelastic scattering is presented. The data were taken with the 27.6 GeV longitudinally polarised positron beam at HERA incident on a longitudinally polarised pure hydrogen gas target internal to the storage ring. The kinematic range is 0.021<x<0.85 and 0.8 GeV^2<Q^2<20 GeV^2. The integral Int_{0.021}^{0.85} g1p(x)dx evaluated at Q0^2 of 2.5 GeV^2 is 0.122+/-0.003(stat.)+/-0.010(syst.).Comment: 7 pages, 3 figures, 1 table, RevTeX late

    Observation of exclusive DVCS in polarized electron beam asymmetry measurements

    We report the first results of the beam spin asymmetry measured in the reaction e + p -> e + p + gamma at a beam energy of 4.25 GeV. A large asymmetry with a sin(phi) modulation is observed, as predicted for the interference term of Deeply Virtual Compton Scattering and the Bethe-Heitler process. The amplitude of this modulation is alpha = 0.202 +/- 0.028. In leading-order and leading-twist pQCD, the alpha is directly proportional to the imaginary part of the DVCS amplitude.Comment: 6 pages, 5 figure

    Electron Scattering From High-Momentum Neutrons in Deuterium

    We report results from an experiment measuring the semi-inclusive reaction d(e,eps)d(e,e'p_s) where the proton psp_s is moving at a large angle relative to the momentum transfer. If we assume that the proton was a spectator to the reaction taking place on the neutron in deuterium, the initial state of that neutron can be inferred. This method, known as spectator tagging, can be used to study electron scattering from high-momentum (off-shell) neutrons in deuterium. The data were taken with a 5.765 GeV electron beam on a deuterium target in Jefferson Laboratory's Hall B, using the CLAS detector. A reduced cross section was extracted for different values of final-state missing mass WW^{*}, backward proton momentum ps\vec{p}_{s} and momentum transfer Q2Q^{2}. The data are compared to a simple PWIA spectator model. A strong enhancement in the data observed at transverse kinematics is not reproduced by the PWIA model. This enhancement can likely be associated with the contribution of final state interactions (FSI) that were not incorporated into the model. A ``bound neutron structure function'' F2neffF_{2n}^{eff} was extracted as a function of WW^{*} and the scaling variable xx^{*} at extreme backward kinematics, where effects of FSI appear to be smaller. For ps>400p_{s}>400 MeV/c, where the neutron is far off-shell, the model overestimates the value of F2neffF_{2n}^{eff} in the region of xx^{*} between 0.25 and 0.6.     A Bayesian analysis of pentaquark signals from CLAS data

    We examine the results of two measurements by the CLAS collaboration, one of which claimed evidence for a Θ+\Theta^{+} pentaquark, whilst the other found no such evidence. The unique feature of these two experiments was that they were performed with the same experimental setup. Using a Bayesian analysis we find that the results of the two experiments are in fact compatible with each other, but that the first measurement did not contain sufficient information to determine unambiguously the existence of a Θ+\Theta^{+}. Further, we suggest a means by which the existence of a new candidate particle can be tested in a rigorous manner.Comment: 5 pages, 3 figure
