SYNTHES OF ROBUST ACTIVE SHIELDING SYSTEMS OF MAGNETIC FIELD GENERATED BY GROUP OF HIGH-VOLTAGE POWER LINES
Aim. The synthesis of robust active shielding system of magnetic field, generated by group of high voltage power lines for reducing the induction of the initial magnetic field to the sanitary standards level and reducing the sensitivity of the system to variations in the plant parameters is given. Methodology. The synthesis is based on the solution of a multi-criteria stochastic game, in which the gain vector is calculated on the basis of the Maxwell equations solutions in the quasi-stationary approximation. The equilibrium state of the game is based on the stochastic multiagent optimization algorithms of the multiswarm particles. The initial parameters for the synthesis of active shielding system are the location of the high voltage power lines with respect to the protected from transmission line space, geometry and number of cables, operating currents, as well as the size of the protected space and normative value magnetic field induction, which should be achieved as a result of screening. The objective of the synthesis of the active shielding system is to determine their number, configuration, spatial arrangement, wiring diagrams and compensation cables currents, setting algorithm of the control systems as well as the resulting value of the induction magnetic field at the points of the protected space. Results. Robust active shielding system synthesis results for reduction of a magnetic field generated by group of high voltage power lines is given. The possibility of a significant reduction in the level of induction of the magnetic field source within and reducing the sensitivity of the system to variations in the plant parameters is given. Originality. For the first time carried out the synthesis of the robust active shielding systems of magnetic field generated by group of high voltage power lines within a given region of space. Practicalvalue. Practical recommendations on reasonable choice of the number and spatial arrangement of compensating cables of robust active shielding systems of the magnetic field generated by the group of high voltage power lines is given
Doebner-type pyrazolopyridine carboxylic acids in an Ugi four-component reaction
Substituted 1H-pyrazolo[3,4-b]pyridine-4- and 1H-pyrazolo[3,4-b]pyridine-6-carboxamides have been synthetized through a Doebner–Ugi multicomponent reaction sequence in a convergent and versatile manner using diversity generation strategies: combination of two multicomponent reactions and conditions-based divergence strategy. The target products contain as pharmacophores pyrazolopyridine and peptidomimetic moieties with four points of diversity introduced from readily available starting materials including scaffold diversity. A small focused compound library of 23 Ugi products was created and screened for antibacterial activity
Informative significance of serum cytokines and their importance for development of metabolic syndrome with arterial hypertension in elderly persons
Metabolic syndrome (MS) is among the main public health challenges worldwide, leading to significant labor losses, increased costs for treatment and rehabilitation of the patients. The aim of the present study was to identify the informative serum interleukins, by determining the odds ratio in elderly patients with MS and hypertension. The main group of 86 patients with MS and arterial hypertension (AH) aged 60-75 years was examined under clinical conditions. The inclusion criteria were as follows: age of 60-75 years, presence of MS, primary hypertension (grade II-III), absence of acute myocardial infarction, malignant neoplasms, disorders of cerebral circulation, kidney failure over last 6 months. Diagnostics of MS and hypertension was carried out in accordance with Expert Guidelines from the Russian Research Society of Cardiology on the MS Diagnosis and Treatment. Our first study of a large range of serum interleukins in elderly patients with MS and hypertension allowed us to reveal the inversely directed changes in pro- and anti-inflammatory cytokine contents. Combined AH/MS in elderly persons is accomplished by sufficient increase of the most proinflammatory cytokines, and vice versa, by significant decrease in anti-inflammatory cytokines in blood serum. This finding clearly points to importance of immunological regulatory systems for initiation of AH with MS at older age. Pro- and anti-inflammatory serum interleukins are actively involved into the AH/MS development in elderly accompanied by their pronounced imbalance. The mentioned immune reactions could underlie the MS/AH condition. High risk of this disorder is connected with changed production of proinflammatory cytokines (IL-8, IL-1β), like as anti-inflammatory serum interleukins (IL-4, IL-10), with predominance of the former. The above interleukins should be considered dominant diagnostic markers of AH/MS in elderly persons. Measurement of serum interleukins and discriminant-based approach allows highly reliable differentiation of elderly patients with AH/MS from similar individuals without this disorder

БЕЛКИ КЛЕТОЧНОЙ ПОДВИЖНОСТИ В КАРЦИНОМЕ ЭНДОМЕТРИЯ И ЭНДОМЕТРИАЛЬНОЙ ГИПЕРПЛАЗИИ: СВЯЗЬ С РИСКОМ РАЗВИТИЯ РАКА
Introduction. Proteins associated with cellular motility are known to play an important role in invasion and metastasis of cancer, however there is no evidence of their association with the development of malignant tumors including endometrial cancer (EC).The aim of the present study was to investigate the levels of actin-binding proteins, p45-Ser-β-catenin, and calpain activity in endometrial hyperplasia and in EC.Material and Methods. Total calpain activity, p45-Ser β-catenin, Arp3, gelsolin, cofillin and thymosin β-4 levels were evaluated in 43 postmenopausal patients with stage I–II endometrioid EC and 40 endometrial hyperplasia patients. Flow cytometry and Western blotting were used for expression determination of p45 Ser β-catenin and actin-biding proteins. Total calpain activity was estimated by fluorimetric method.Results. Levels of cofilin-1, thymosin β-4 and calpain activity were higher in cancer tissues than in endometrial hyperplasia. Cofilin-1 and thymosin β-4 levels were associated with the depth of myometrial invasion. The thymosin β-4 expression was correlated with the presence of tumor cervical invasion. Revealed correlations between the actin-binding proteins, p45-Ser-β-catenin and total calpain activity in endometrial hyperplasia tissue, but not in the tissue of cancer, is evidence of the involvement of these proteases in regulation of cell migration in endometrial hyperplasia. Levels of thymosin β-4, cofilin and total calpain activity are independent cancer risk factors in patients with endometrial hyperplasia.Conclusion. The level of actin-binding proteins as well as the total calpain activity were enhanced in endometrium carcinoma tissues compared to endometrial hyperplasia. The levels of thymosinβ-4, cofilin and total calpain activity in endometrial hyperplasia tissues are associated with a hyperplasia transition to cancer and may be considered as predictive biomarkers. Известно, что белки, связанные с клеточной подвижностью, играют важную роль в инвазии и метастазировании злокачественных опухолей, тем не менее нет данных об их связи с развитием новообразований, в том числе рака эндометрия (РЭ).Целью исследования было изучение уровня актин-связывающих белков, p45-Ser-β-катенина и активности кальпаинов при гиперплазии эндометрия и при РЭ.Материал и методы. Общая активность кальпаинов, уровни p45-Ser β-катенина, Arp3, гельзолина, кофиллина и тимозина β-4 были оценены у 43 больных раком эндометрия I–II стадии в постменопаузе и у 40 пациенток с гиперплазией эндометрия. Проточную цитометрию и вестерн-блоттинг использовали для определения экспрессии p45 Ser β-катенина и актин-связывающих белков. Общая активность кальпаинов оценивалась флуориметрическим методом.Результаты. Уровни кофилина-1, тимозина β-4 и активность кальпаинов были выше в тканях злокачественных опухолей, чем при гиперплазии эндометрия. Уровни кофилина-1 и тимозина β-4 были связаны с глубиной инвазии рака эндометрия в миометрий. Содержание тимозина β-4 коррелировало с наличием инвазии опухоли в шейку матки. Выявленные корреляции между актин-связывающими белками, р45-Ser-β-катенином и общей активностью кальпаинов в ткани гиперплазии эндометрия, но не в ткани рака свидетельствуют об участии этих протеаз в регуляции клеточной миграции при гиперплазии эндометрия. Уровни тимозина-4, кофилина и общая активность кальпаинов являются независимыми факторами риска развития рака у пациентов с гиперплазией эндометрия.Заключение. Уровень актин-связывающих белков, а также общая активность кальпаинов были повышены в тканях карциномы эндометрия по сравнению с гиперплазией эндометрия. Уровни тимозина-4, кофилина и общая активность кальпаинов в тканях гиперплазии эндометрия могут быть независимыми факторами риска развития рака.
Oxylipins from both pathogen and host antagonize jasmonic acid-mediated defence via the 9-lipoxygenase pathway in Fusarium verticillioides infection of maize
Oxylipins are a newly emerging group of signals that serve defence roles or promote virulence. To identify specific host and fungal genes and oxylipins governing the interactions between maize and Fusarium verticillioides, maize wild-type and lipoxygenase3 (lox3) mutant were inoculated with either F. verticillioides wild-type or linoleate-diol-synthase 1-deleted mutant (DFvlds1D). The results howed that lox3 mutants were more resistant to F. verticillioides. The reduced colonization on lox3 was associated with reduced fumonisin production and with a stronger and earlier induction of ZmLOX4, mLOX5 and ZmLOX12. In addition to the reported defence function of ZmLOX12, we showed that lox4 and lox5 mutants were more susceptible to F. verticillioides and possessed decreased jasmonate levels during infection, suggesting that these genes are essential for jasmonic acid (JA)-mediated defence. Oxylipin profiling revealed a dramatic reduction in fungal linoleate diol synthase 1 (LDS1)-derived oxylipins, especially 8-HpODE (8-hydroperoxyoctadecenoic acid), in infected lox3 kernels, indicating the importance of this molecule in virulence. Collectively, we make the following conclusions: (1) LOX3 is a major susceptibility factor induced by fungal LDS1-derived oxylipins to suppress JA-stimulating 9-LOXs; (2) LOX3-mediated signalling promotes the biosynthesis of virulence-promoting oxylipins in the fungus; and (3) both fungal LDS1- and host LOX3-produced oxylipins are essential for the normal infection and colonization processes of maize seed by F. verticillioides
