    Purification and characterization of a calmodulin-dependent protein kinase that is highly concentrated in brain

    A calcium and calmodulin-dependent protein kinase has been purified from rat brain. It was monitored during the purification by its ability to phosphorylate the synaptic vesicle-associated protein, synapsin I. A 300-fold purification was sufficient to produce kinase that is 90-95% pure as determined by scans of stained sodium dodecyl sulfate-polyacrylamide gels and has a specific activity of 2.9 mumol of 32P transferred per min/mg of protein. Thus, the kinase is a relatively abundant brain enzyme, perhaps comprising as much as 0.3% of the total brain protein. The Stokes radius (95 A) and sedimentation coefficient (16.4 S) of the kinase indicate a holoenzyme molecular weight of approximately 650,000. The holoenzyme is composed of three subunits as judged by their co-migration with kinase activity during the purification steps and co-precipitation with kinase activity by a specific anti-kinase monoclonal antibody. The three subunits have molecular weights of 50,000, 58,000, and 60,000, and have been termed alpha, beta', and beta, respectively. The alpha- and beta-subunits are distinct peptides, however, beta' may have been generated from beta by proteolysis. All three of these subunits bind calmodulin in the presence of calcium and are autophosphorylated under conditions in which the kinase is active. The subunits are present in a ratio of about 3 alpha-subunits to 1 beta/beta'-subunit. We therefore postulate that the 650,000-Da holoenzyme consists of approximately 9 alpha-subunits and 3 beta/beta'-subunits. The abundance of this calmodulin-dependent protein kinase indicates that its activation is likely to be an important biochemical response to increases in calcium ion concentration in neuronal tissue

    Past research has demonstrated a negative relationship between ambition, or the desire to get ahead, and job satisfaction. In the present paper, age was hypothesized to moderate the relationship between ambition and job satisfaction such that the relationship between ambition and satisfaction is more negative for older employees than for younger employees. Three studies, with three criterion variables (promotion satisfaction, extrinsic job satisfaction, overall job satisfaction), were used to test the hypothesis. Results indicated support for the hypothesized interaction. The discussion focuses on the implications of the results for organizational and individual career management strategies

    High-resolution width-modulated pulse rebalance electronics for strapdown gyroscopes and accelerometers

    Three different rebalance electronic loops were designed, implemented, and evaluated. The loops were width-modulated binary types using a 614.4 kHz keying signal; they were developed to accommodate the following three inertial sensors with the indicated resolution values: (1) Kearfott 2412 accelerometer - resolution = 260 micro-g/data pulse, (2) Honeywell GG334 gyroscope - resolution = 3.9 milli-arc-sec/data pulse, (3) Kearfott 2401-009 accelerometer - resolution = 144 milli-g/data pulse.     This research aims to prove the effect of Application of Performance-Based Budgeting to performance SKPD Riau province, then Effect of System of Financial Management Information on the performance of skpd Riau province, and the Effect of Quality of Human Resources to performance SKPD Riau province. The population in this study is the Regional Working Units (on education) in the Riau provincial government on education in 38 provinces comprising Region 1 Secretariat Riau, Riau Province Inspectorate 1, 1 Secretary DPRD Riau, Riau KORPRI 1 Secretary, Civil Service Police Unit 1, 2 Hospital (Hospital and RSJ), Agency 14, and 17 Department. Samples from this study are echelon III and IV at Department and Agency of Riau Province by 86 people. Analysis of the data in this study using tools SPSS software version 17.     Considering long-memory when testing for changepoints in surface temperature:a classification approach based on the time-varying spectrum

    Changepoint models are increasingly used to represent changes in the rate of warming in surface temperature records. On the opposite hand, a large body of literature has suggested long‐memory processes to characterize long‐term behavior in surface temperatures. While these two model representations provide different insights into the underlying mechanisms, they share similar spectrum properties that create “ambiguity” and challenge distinguishing between the two classes of models. This study aims to compare the two representations to explain temporal changes and variability in surface temperatures. To address this question, we extend a recently developed time‐varying spectral procedure and assess its accuracy through a synthetic series mimicking observed global monthly surface temperatures. We vary the length of the synthetic series to determine the number of observations needed to be able to accurately distinguish between changepoints and long‐memory models. We apply the approach to two gridded surface temperature data sets. Our findings unveil regions in the oceans where long‐memory is prevalent. These results imply that the presence of long‐memory in monthly sea surface temperatures may impact the significance of trends, and special attention should be given to the choice of model representing memory (short versus long) when assessing long‐term changes

    Quantum Field Theory of Forward Rates with Stochastic Volatility

    In a recent formulation of a quantum field theory of forward rates, the volatility of the forward rates was taken to be deterministic. The field theory of the forward rates is generalized to the case of stochastic volatility. Two cases are analyzed, firstly when volatility is taken to be a function of the forward rates, and secondly when volatility is taken to be an independent quantum field. Since volatiltiy is a positive valued quantum field, the full theory turns out to be an interacting nonlinear quantum field theory in two dimensions. The state space and Hamiltonian for the interacting theory are obtained, and shown to have a nontrivial structure due to the manifold moving with a constant velocity. The no arbitrage condition is reformulated in terms of the Hamiltonian of the system, and then exactly solved for the nonlinear interacting case.Comment: 7 Figure

    Exactness of the Fock space representation of the q-commutation relations

    We show that for all q in the interval (-1,1), the Fock representation of the q-commutation relations can be unitarily embedded into the Fock representation of the extended Cuntz algebra. In particular, this implies that the C*-algebra generated by the Fock representation of the q-commutation relations is exact. An immediate consequence is that the q-Gaussian von Neumann algebra is weakly exact for all q in the interval (-1,1).Comment: 20 page

    Effects of transport on fecal glucocorticoid levels in captive-bred cotton-top tamarins (Saguinus oedipus)

    The relocation of animals can induce stress when animals are placed in novel environmental conditions. The movement of captive animals among facilities is common, especially for non-human primates used in research. The stress response begins with the activation of the hypothalamic-pituitary-adrenal (HPA) axis which results in the release of glucocorticoid hormones (GC), which at chronic levels could lead to deleterious physiological effects. There is a substantial body of data concerning GC levels affecting reproduction, and rank and aggression in primates. However, the effect of transport has received much less attention. Fecal samples from eight (four male and four female) captive-bred cotton-top tamarins (Saguinus oedipus) were collected at four different time points (two pre-transport and two post-transport). The fecal samples were analyzed using an immunoassay to determine GC levels. A repeated measures analysis of variance (ANOVA) demonstrated that GC levels differed among transport times (p = 0.009), but not between sexes (p = 0.963). Five of the eight tamarins exhibited an increase in GC levels after transport. Seven of the eight tamarins exhibited a decrease in GC levels from three to six days post-transport to three weeks post-transport. Most values returned to pre-transport levels after three weeks. The results indicate that these tamarins experienced elevated GC levels following transport, but these increases were of short duration. This outcome would suggest that the negative effects of elevated GC levels were also of short duration
