Watershed rainfall forecasting using neuro-fuzzy networks with the assimilation of multi-sensor information
The complex temporal heterogeneity of rainfall coupled with mountainous physiographic context makes a great challenge in the development of accurate short-term rainfall forecasts. This study aims to explore the effectiveness of multiple rainfall sources (gauge measurement, and radar and satellite products) for assimilation-based multi-sensor precipitation estimates and make multi-step-ahead rainfall forecasts based on the assimilated precipitation. Bias correction procedures for both radar and satellite precipitation products were first built, and the radar and satellite precipitation products were generated through the Quantitative Precipitation Estimation and Segregation Using Multiple Sensors (QPESUMS) and the Precipitation Estimation from Remotely Sensed Information using Artificial Neural Networks-Cloud Classification System (PERSIANN-CCS), respectively. Next, the synthesized assimilated precipitation was obtained by merging three precipitation sources (gauges, radars and satellites) according to their individual weighting factors optimized by nonlinear search methods. Finally, the multi-step-ahead rainfall forecasting was carried out by using the adaptive network-based fuzzy inference system (ANFIS). The Shihmen Reservoir watershed in northern Taiwan was the study area, where 641 hourly data sets of thirteen historical typhoon events were collected. Results revealed that the bias adjustments in QPESUMS and PERSIANN-CCS products did improve the accuracy of these precipitation products (in particular, 30-60% improvement rates for the QPESUMS, in terms of RMSE), and the adjusted PERSIANN-CCS and QPESUMS individually provided about 10% and 24% contribution accordingly to the assimilated precipitation. As far as rainfall forecasting is concerned, the results demonstrated that the ANFIS fed with the assimilated precipitation provided reliable and stable forecasts with the correlation coefficients higher than 0.85 and 0.72 for one- and two-hour-ahead rainfall forecasting, respectively. The obtained forecasting results are very valuable information for the flood warning in the study watershed during typhoon periods. © 2013 Elsevier B.V
Evaluation of Dynamic Cell Processes and Behavior Using Video Bioinformatics Tools
Just as body language can reveal a personâs state of well-being, dynamic changes in cell behavior and
morphology can be used to monitor processes in cultured cells. This chapter discusses how CL-Quant
software, a commercially available video bioinformatics tool, can be used to extract quantitative data on:
(1) growth/proliferation, (2) cell and colony migration, (3) reactive oxygen species (ROS) production, and
(4) neural differentiation. Protocols created using CL-Quant were used to analyze both single cells and
colonies. Time-lapse experiments in which different cell types were subjected to various chemical
exposures were done using Nikon BioStations. Proliferation rate was measured in human embryonic stem
cell colonies by quantifying colony area (pixels) and in single cells by measuring confluency (pixels).
Colony and single cell migration were studied by measuring total displacement (distance between the
starting and ending points) and total distance traveled by the colonies/cells. To quantify ROS production,
cells were pre-loaded with MitoSOX Redâą, a mitochondrial ROS (superoxide) indicator, treated with
various chemicals, then total intensity of the red fluorescence was measured in each frame. Lastly, neural
stem cells were incubated in differentiation medium for 12 days, and time lapse images were collected
daily. Differentiation of neural stem cells was quantified using a protocol that detects young neurons. CLQuant
software can be used to evaluate biological processes in living cells, and the protocols developed in
this project can be applied to basic research and toxicological studies, or to monitor quality control in
culture facilities

Mapping the UK renal psychosocial workforce : the first comprehensive workforce survey
Background: Emerging evidence of psychosocial problems in CKD patients has led to an acceptance that a focus
on the emotional wellbeing of the patient should be included in the provision of comprehensive CKD care. It is
unclear if an increased attention for psychosocial needs in guidelines and policy documents has led to a rise in
psychosocial staffing levels or change in composition of staff since the last workforce mapping in 2002. This paper
offers a critical analysis and in-depth discussion of findings and their implications, in addition to providing an
international perspective and exposing gaps in current knowledge.
Methods: Data on psychosocial staffing levels was taken from a survey based on the Scottish Renal Associationâs
(SRA) staffing survey that was sent to all units in England, Wales and Northern-Ireland in 2016. In addition, data
from a psychosocial staffing survey designed by and distributed via psychosocial professional groups was used. This data was then completed with Freedom of Information (FOI) requests and collated to describe the current renal
psychosocial workforce in all 84 UK renal units. This was compared to results from the last renal workforce mapping
in 2002.
Results: The results from this mapping show great variability in models of service provision, significant exceeding of benchmarks for staffing levels, and a change in staffing patterns over the past 15 years. Adult psychology services have increased in number, but provision remains low due to increased patient numbers, whereas adult social work and paediatric services have decreased.
Conclusion: A lack in the provision of renal psychosocial services has been identified, together with the absence of
a general service provision model. These findings provide a valuable benchmark for units, a context from which to
review and monitor provision alongside patient need. Along with recommendations, this paper forms a foundation
for future research and workforce planning. Research into best practice models of service provision and the
psychosocial needs of CKD patients lies at the heart of the answers to many identified questions
Gender Inequity Norms Are Associated with Increased Male-Perpetrated Rape and Sexual Risks for HIV Infection in Botswana and Swaziland
There is limited empirical research on the underlying gender inequity norms shaping gender-based violence, power, and HIV risks in sub-Saharan Africa, or how risk pathways may differ for men and women. This study is among the first to directly evaluate the adherence to gender inequity norms and epidemiological relationships with violence and sexual risks for HIV infection.Data were derived from population-based cross-sectional samples recruited through two-stage probability sampling from the 5 highest HIV prevalence districts in Botswana and all districts in Swaziland (2004-5). Based on evidence of established risk factors for HIV infection, we aimed 1) to estimate the mean adherence to gender inequity norms for both men and women; and 2) to model the independent effects of higher adherence to gender inequity norms on a) male sexual dominance (male-controlled sexual decision making and rape (forced sex)); b) sexual risk practices (multiple/concurrent sex partners, transactional sex, unprotected sex with non-primary partner, intergenerational sex).A total of 2049 individuals were included, nâ=â1255 from Botswana and nâ=â796 from Swaziland. In separate multivariate logistic regression analyses, higher gender inequity norms scores remained independently associated with increased male-controlled sexual decision making power (AORmenâ=â1.90, 95%CI:1.09-2.35; AORwomenâ=â2.05, 95%CI:1.32-2.49), perpetration of rape (AORmenâ=â2.19 95%CI:1.22-3.51), unprotected sex with a non-primary partner (AORmenâ=â1.90, 95%CI:1.14-2.31), intergenerational sex (AORwomenâ=â1.36, 95%CI:1.08-1.79), and multiple/concurrent sex partners (AORmenâ=â1.42, 95%CI:1.10-1.93).These findings support the critical evidence-based need for gender-transformative HIV prevention efforts including legislation of women's rights in two of the most HIV affected countries in the world
A canine BCAN microdeletion associated with episodic falling syndrome
Episodic falling syndrome (EFS) is a canine paroxysmal hypertonicity disorder found in Cavalier King Charles spaniels. Episodes are triggered by exercise, stress or excitement and characterized by progressive hypertonicity throughout the thoracic and pelvic limbs, resulting in a characteristic 'deer-stalking' position and/or collapse. We used a genome-wide association strategy to map the EFS locus to a 3.48 Mb critical interval on canine chromosome 7. By prioritizing candidate genes on the basis of biological plausibility, we found that a 15.7 kb deletion in BCAN, encoding the brain-specific extracellular matrix proteoglycan brevican, is associated with EFS. This represents a compelling causal mutation for EFS, since brevican has an essential role in the formation of perineuronal nets governing synapse stability and nerve conduction velocity. Mapping of the deletion breakpoint enabled the development of Multiplex PCR and Multiplex Ligation-dependent Probe Amplification (MLPA) genotyping tests that can accurately distinguish normal, carrier and affected animals. Wider testing of a larger population of CKCS dogs without a history of EFS from the USA revealed that carriers are extremely common (12.9%). The development of molecular genetic tests for the EFS microdeletion will allow the implementation of directed breeding programs aimed at minimizing the number of animals with EFS and enable confirmatory diagnosis and pharmacotherapy of affected dogs

composite neutrino scenario. For a certain class of composite neutrino
theories, these sterile neutrinos naturally have the appropriate mixing angles
to be resonantly produced warm dark matter (WDM). Alternatively, we show these
sterile neutrinos can be WDM produced by an entropy-diluted thermal freeze-out,
with the necessary entropy production arising not from an out-of-equilibrium
decay, but rather from the confinement of the composite neutrino sector,

An AP-MS- and BioID-compatible MAC-tag enables comprehensive mapping of protein interactions and subcellular localizations
Protein-protein interactions govern almost all cellular functions. These complex networks of stable and transient associations can be mapped by affinity purification mass spectrometry (AP-MS) and complementary proximity-based labeling methods such as BioID. To exploit the advantages of both strategies, we here design and optimize an integrated approach combining AP-MS and BioID in a single construct, which we term MAC-tag. We systematically apply the MAC-tag approach to 18 subcellular and 3 sub-organelle localization markers, generating a molecular context database, which can be used to define a protein's molecular location. In addition, we show that combining the AP-MS and BioID results makes it possible to obtain interaction distances within a protein complex. Taken together, our integrated strategy enables the comprehensive mapping of the physical and functional interactions of proteins, defining their molecular context and improving our understanding of the cellular interactome.Peer reviewe
Breed-Specific Hematological Phenotypes in the Dog: A Natural Resource for the Genetic Dissection of Hematological Parameters in a Mammalian Species
Remarkably little has been published on hematological phenotypes of the domestic dog, the most polymorphic species on the planet. Information on the signalment and complete blood cell count of all dogs with normal red and white blood cell parameters judged by existing reference intervals was extracted from a veterinary database. Normal hematological profiles were available for 6046 dogs, 5447 of which also had machine platelet concentrations within the reference interval. Seventy-five pure breeds plus a mixed breed control group were represented by 10 or more dogs. All measured parameters except mean corpuscular hemoglobin concentration (MCHC) varied with age. Concentrations of white blood cells (WBCs), neutrophils, monocytes, lymphocytes, eosinophils and platelets, but not red blood cell parameters, all varied with sex. Neutering status had an impact on hemoglobin concentration, mean corpuscular hemoglobin (MCH), MCHC, and concentrations of WBCs, neutrophils, monocytes, lymphocytes and platelets. Principal component analysis of hematological data revealed 37 pure breeds with distinctive phenotypes. Furthermore, all hematological parameters except MCHC showed significant differences between specific individual breeds and the mixed breed group. Twenty-nine breeds had distinctive phenotypes when assessed in this way, of which 19 had already been identified by principal component analysis. Tentative breed-specific reference intervals were generated for breeds with a distinctive phenotype identified by comparative analysis. This study represents the first large-scale analysis of hematological phenotypes in the dog and underlines the important potential of this species in the elucidation of genetic determinants of hematological traits, triangulating phenotype, breed and genetic predisposition
Obstructive sleep apnea in Treacher Collins syndrome
The aim of the present study was to investigate the prevalence of obstructive sleep apnea syndrome (OSAS) among the Norwegian population with Treacher Collins syndrome (TCS). A secondary aim was to establish whether TCS phenotype severity is associated with OSAS severity. A prospective case study design was used. Individuals who were 5Â years old and above with a known diagnosis of TCS in Norway were invited to participate in a study. The study included genetic testing, medical and dental examinations and polysomnography. All participants demonstrated disturbed respiration during sleep; 18/19 met the diagnostic criteria for OSAS. Subjectively evaluated snoring was not a reliable predictor of OSAS. We found no significant association between TCS phenotype severity and the severity of OSAS. OSAS is common in TCS, but there is no association with the phenotype severity. Individuals diagnosed with TCS must undergo sleep studies to identify the presence of OSAS
