    Reisen - damit verbinden wir fast alle etwas Positives und doch jeder etwas Anderes. FĂŒr den Einen bedeutet Reisen Erholung pur, fĂŒr den Anderen Strand und Party, fĂŒr manche vor allem Bildung in unterhaltsamer Verpackung und fĂŒr immer mehr Abenteuer in der „Wildnis“ und das Erleben unbekannter, vermeintlich „ursprĂŒnglicher, unzivilisierter“ Kulturen. Die Angebotspalette der Tourismusindustrie ist so vielfĂ€ltig wie die möglichen Assoziationen zum Thema Reisen und versucht gestressten „Westlern“ zu bieten, wonach sie suchen. Kapitel II.1. dieser Arbeit widmet sich dieser Vielfalt des Tourismus und leitet ĂŒber zu Fragen, mit denen sich viele Touristen gar nicht oder zu wenig auseinandersetzen: Welche Bilder bringen „wir“, die Vertreter westlicher Industriegesellschaften, von der Gastgeberkultur bereits mit, bevor wir sie ĂŒberhaupt erlebt haben? Wie beeinflusst das unsere Begegnung? Welche Konsequenzen hat Tourismus fĂŒr die Kultur der Besuchten? Die Tourismusforschung hat dazu Überlegungen angestellt und Studien veröffentlicht, von denen einige ausgewĂ€hlte in Kapitel II.2. kurz vorgestellt werden. Ich selbst habe wĂ€hrend meines viermonatigen Auslandspraktikums im Amazonasgebiet Ecuadors begonnen, mich fĂŒr diese Frage zu interessieren. Das Praktikum absolvierte ich im Rahmen meines Studiums der Mehrsprachigen Kommunikation an der FH Köln. Als Tourdolmetscherin in einer Dschungel-Lodge war eine meiner Aufgaben, zwischen Besuchern aus Nordamerika oder Europa und den besuchten Quichua vom oberen Napo sprachlich zu vermitteln. Die Kultur dieser indigenen Gemeinschaft hat mich sehr fasziniert und mich zu der Frage gefĂŒhrt, ob der Tourismus zu einer Wiederbelebung ihrer Kultur beitragen kann. Diese Frage soll im Mittelpunkt meiner Arbeit stehen. Im Zusammenhang dazu wird in Kapitel III. der Ökotourismus oder nachhaltige Tourismus vorgestellt, der neue Anforderungen an die Tourismusentwicklung stellt und im Amazonasgebiet Ecuadors - mehr oder weniger konsequent umgesetzt - bereits weit verbreitet ist. In diesem Kapitel soll verdeutlicht werden, welche Faktoren mit dem Erhalt einer indigenen Kultur wie der der Quichua verknĂŒpft sind. Kapitel IV. soll dem Leser einen Einblick in ausgewĂ€hlte Aspekte der Kultur und Geschichte der Quichua des oberen Napo gewĂ€hren, die im lokalen Ökotourismus von Bedeutung sind. Das letzte Kapitel, Kapitel V. stellt das Projekt RICANCIE vor, in dem Quichua-Gemeinden selbst Ökotourismus anbieten. Mit diesem Beispiel soll gezeigt werden, dass Tourismus tatsĂ€chlich dazu beitragen kann, die WertschĂ€tzung der Quichua-Kultur bei GĂ€sten und Gastgebern zu steigern, sofern alle fĂŒr das Gelingen eines solchen Projekts notwendigen Faktoren beachtet werden

    Effects of canagliflozin on cardiovascular biomarkers in older adults with type 2 diabetes

    Background: Sodium glucose co-transporter 2 (SGLT2) inhibitors may reduce cardiovascular and heart failure risk in patients with type 2 diabetes mellitus (T2DM). Objectives: To examine the effects of canagliflozin on cardiovascular biomarkers in older patients with T2DM. Methods: In 666 T2DM patients randomized to receive canagliflozin 100 or 300 mg or placebo, we assessed median percent change in serum N-terminal pro-B type natriuretic peptide (NT-proBNP), high-sensitivity troponin I (hsTnI) , soluble (s)ST2, and galectin-3 from baseline to 26, 52, and 104 weeks. Results: Both serum NT-proBNP and serum hsTnI levels increased in placebo recipients but remained largely unchanged in those randomized to canagliflozin. Hodges-Lehmann estimates of the difference in median percent change between pooled canagliflozin and placebo were –15.0%, –16.1%, and –26.8% for NT-proBNP, and –8.3%, –11.9%, and –10.0% for hsTnI at weeks 26, 52, and 104, respectively (all P <0.05). Serum sST2 was unchanged with canagliflozin and placebo over 104 weeks. Serum galectin-3 modestly increased from baseline with canagliflozin versus placebo, with significant differences observed at 26 and 52 weeks but not at 104 weeks. These results remained unchanged when only patients with complete samples were assessed. Conclusions: Compared to placebo, treatment with canagliflozin delayed rise in serum NT-proBNP and hsTnI over 2 years in older T2DM patients. These cardiac biomarker data provide support for beneficial cardiovascular effect of SGLT2 inhibitors in T2DM

    Activation-induced cytidine deaminase (AID) linking immunity, chronic inflammation, and cancer

    Activation-induced cytidine deaminase (AID) is critically involved in class switch recombination and somatic hypermutation of Ig loci resulting in diversification of antibodies repertoire and production of high-affinity antibodies and as such represents a physiological tool to introduce DNA alterations. These processes take place within germinal centers of secondary lymphoid organs. Under physiological conditions, AID is expressed predominantly in activated B lymphocytes. Because of the mutagenic and recombinogenic potential of AID, its expression and activity is tightly regulated on different levels to minimize the risk of unwanted DNA damage. However, chronic inflammation and, probably, combination of other not-yet-identified factors are able to create a microenvironment sufficient for triggering an aberrant AID expression in B cells and, importantly, in non-B-cell background. Under these circumstances, AID may target also non-Ig genes, including cancer-related genes as oncogenes, tumor suppressor genes, and genomic stability genes, and modulate both genetic and epigenetic information. Despite ongoing progress, the complete understanding of fundamental aspects is still lacking as (1) what are the crucial factors triggering an aberrant AID expression/activity including the impact of Th2-driven inflammation and (2) to what extent may aberrant AID in human non-B cells lead to abnormal cell state associated with an increased rate of genomic alterations as point mutations, small insertions or deletions, and/or recurrent chromosomal translocations during solid tumor development and progression

    The human side of animal experimentation: A qualitative, exploratory study into workrelated stress and coping in animal experimenters

    Besides the pervasive controversy of animal experimentation in society, ethics and science, the human experimenter side of laboratory animal studies is a relatively underrepresented topic in human-animal interaction research. Few studies have addressed scientists’ stress responses to animal experiments. The main aim of this study was to assess work-related stress by means of salivary cortisol secretion, coping strategies, self-esteem, pet attitude and personality traits in academic researchers who regularly perform invasive animal experiments. Invitation to participate in the study resulted in a response rate of 15.4% of 65 invited scientists, of which only four (6.15%) completed data collection. Study participants carried out saliva sampling on working days with and without animal experiments, completed a semi-structured qualitative interview and psychological questionnaires. Salivary cortisol (SC) was measured via enzyme immunoassay. The results indicate that animal experimenters used problem-focused and emotion-focused coping strategies. Three participants reached above average values in self-esteem. Pet attitudes scores were moderately positive. Three out of four animal experimenters reached high scores on the personality dimensions "openness to experience", "agreeableness" and "conscientiousness". In the absence of an acute increase in SC related to animal experimentation, two out of four participants exhibited an altered circadian pattern of SC secretion only on working days with animal experiments. Although and as a matter of fact because only four of 65 invited scientists volunteered to participate, we discussed the seemingly low willingness of researchers to participate in such a study based on a theoretical analysis, particularly highlighting the concept of deindividuation and provide suggestions for future research
