    Detection of vitellogenin incorporation into zebrafish oocytes by FITC fluorescence

    <p>Abstract</p> <p>Background</p> <p>Large volumes of lymph can be collected from the eye-sacs of bubble-eye goldfish. We attempted to induce vitellogenin (Vtg) in the eye-sac lymph of bubble-eye goldfish and develop a method for visualizing Vtg incorporation by zebrafish oocytes using FITC-labeling.</p> <p>Methods</p> <p>Estrogen efficiently induced Vtg in the eye-sac lymph of goldfish. After FITC-labeled Vtg was prepared, it was injected into mature female zebrafish.</p> <p>Results</p> <p>Incorporation of FITC-labeled Vtg by zebrafish oocytes was detected in <it>in vivo </it>and <it>in vitro </it>experiments. The embryos obtained from zebrafish females injected with FITC-labeled Vtg emitted FITC fluorescence from the yolk sac and developed normally.</p> <p>Conclusion</p> <p>This method for achieving Vtg incorporation by zebrafish oocytes could be useful in experiments related to the development and endocrinology of zebrafish oocytes.</p

    The efficacy of incretin therapy in patients with type 2 diabetes undergoing hemodialysis

    BACKGROUND: Although incretin therapy is clinically available in patients with type 2 diabetes undergoing hemodialysis, no study has yet examined whether incretin therapy is capable of maintaining glycemic control in this group of patients when switched from insulin therapy. In this study, we examined the efficacy of incretin therapy in patients with insulin-treated type 2 diabetes undergoing hemodialysis. METHODS: Ten type 2 diabetic patients undergoing hemodialysis received daily 0.3 mg liraglutide, 50 mg vildagliptin, and 6.25 mg alogliptin switched from insulin therapy on both the day of hemodialysis and the non-hemodialysis day. Blood glucose level was monitored by continuous glucose monitoring. After blood glucose control by insulin, patients were treated with three types of incretin therapy in a randomized crossover manner, with continuous glucose monitoring performed for each treatment. RESULTS: During treatment with incretin therapies, severe hyperglycemia and ketosis were not observed in any patients. Maximum blood glucose and mean blood glucose on the day of hemodialysis were significantly lower after treatment with liraglutide compared with treatment with alogliptin (p < 0.05), but not with vildagliptin. The standard deviation value, a marker of glucose fluctuation, on the non-hemodialysis day was significantly lower after treatment with liraglutide compared with treatment with insulin and alogliptin (p < 0.05), but not with vildagliptin. Furthermore, the duration of hyperglycemia was significantly shorter after treatment with liraglutide on both the hemodialysis and non-hemodialysis days compared with treatment with alogliptin (p < 0.05), but not with vildagliptin. CONCLUSIONS: The data presented here suggest that patients with type 2 diabetes undergoing hemodialysis and insulin therapy could be treated with incretin therapy in some cases

    How much does emotional valence of action outcomes affect temporal binding?

    Temporal binding refers to the compression of the perceived time interval between voluntary actions and their sensory consequences. Research suggests that the emotional content of an action outcome can modulate the effects of temporal binding. We attempted to conceptually replicate these findings using a time interval estimation task and different emotionally-valenced action outcomes (Experiments 1 and 2) than used in previous research. Contrary to previous findings, we found no evidence that temporal binding was affected by the emotional valence of action outcomes. After validating our stimuli for equivalence of perceived emotional valence and arousal (Experiment 3), in Experiment 4 we directly replicated Yoshie and Haggard’s (2013) original experiment using sound vocalizations as action outcomes and failed to detect a significant effect of emotion on temporal binding. These studies suggest that the emotional valence of action outcomes exerts little influence on temporal binding. The potential implications of these findings are discussed

    Audience effects on the neural correlates of relational reasoning in adolescence

    Adolescents are particularly sensitive to peer influence. This may partly be due to an increased salience of peers during adolescence. We investigated the effect of being observed by a peer on a cognitively challenging task, relational reasoning, which requires the evaluation and integration of multiple mental representations. Relational reasoning tasks engage a fronto-parietal network including the inferior parietal cortex, pre-supplementary motor area, dorsolateral and rostrolateral prefrontal cortices. Using functional magnetic resonance imaging (fMRI), peer audience effects on activation in this fronto-parietal network were compared in a group of 19 female mid-adolescents (aged 14-16 years) and 14 female adults (aged 23-28 years). Adolescent and adult relational reasoning accuracy was influenced by a peer audience as a function of task difficulty: the presence of a peer audience led to decreased accuracy in the complex, relational integration condition in both groups of participants. The fMRI results demonstrated that a peer audience differentially modulated activation in regions of the fronto-parietal network in adolescents and adults. Activation was increased in adolescents in the presence of a peer audience, while this was not the case in adults

    本学の成人看護学における看護技術教育

    報告Report2003年3月、厚生労働省から公表された、「看護基礎教育における技術教育のあり方に関する検討会報告書」を受け、2003・4年度に実施した看護技術教育について検討・評価を行った。2001・2年度入学生の看護基礎教育課程・成人看護学は、成人看護方法論I~IV、成人看護実習I・IIの6科目で構成されている。成人看護学では、看護技術の原理・原則を基盤として、技術の応用と発展的な学習を目標に取り組んだ。成人看護方法論IIでは、心電図検査・肺機能検査を実施した。すべての学生が操作方法を理解し、正確な検査結果を得ていた。臨地実習では、実習前の自己学修課題・期間中の学内演習・臨地実習中に体験した看護技術を調査した。調査結果から、学生が実習中に見学・体験できる看護技術は必ずしも多いとは言えなかった。さらに、学習環境の整備・教育方法の工夫に積極的に取り組む必要がある
