    Doppler Aliasing Reduction in Wide-Angle Synthetic Aperture Radar Using Phase Modulated Random Stepped-Frequency Waveforms

    This research effort examines the theory, application and results of side-looking airborne radar operation in hot clutter. Hot clutter is an electronic counter-measure used to degrade the performance of airborne radar. Hot clutter occurs by illuminating the ground with an airborne jammer at some velocity, azimuth, elevation, and range from the airborne radar. When the received RCS scattered hot clutter waveform is perfectly coherent with the radar waveform, the radar believes the returns created by the hot clutter jammer resulted from the transmitting radar. Hot clutter degrades radar performance at locations in azimuth and Doppler. The effect of hot clutter is examined for side-looking airborne radar using adaptive and non-adaptive processing. Factored Time Space and Joint Domain Localized adaptive filters are shown to improve radar performance 32 to 36 dB per element per pulse, respectively, over non-adaptive processing in the mainbeam jammer normalized Doppler location when the jammer is not in the radar look direction in azimuth

    In Vitro and In Vivo Characterization of the Alkaloid Nuciferine

    RationaleThe sacred lotus (Nelumbo nucifera) contains many phytochemicals and has a history of human use. To determine which compounds may be responsible for reported psychotropic effects, we used in silico predictions of the identified phytochemicals. Nuciferine, an alkaloid component of Nelumbo nucifera and Nymphaea caerulea, had a predicted molecular profile similar to antipsychotic compounds. Our study characterizes nuciferine using in vitro and in vivo pharmacological assays.MethodsNuciferine was first characterized in silico using the similarity ensemble approach, and was followed by further characterization and validation using the Psychoactive Drug Screening Program of the National Institute of Mental Health. Nuciferine was then tested in vivo in the head-twitch response, pre-pulse inhibition, hyperlocomotor activity, and drug discrimination paradigms.ResultsNuciferine shares a receptor profile similar to aripiprazole-like antipsychotic drugs. Nuciferine was an antagonist at 5-HT2A, 5-HT2C, and 5-HT2B, an inverse agonist at 5-HT7, a partial agonist at D2, D5 and 5-HT6, an agonist at 5-HT1A and D4 receptors, and inhibited the dopamine transporter. In rodent models relevant to antipsychotic drug action, nuciferine blocked head-twitch responses and discriminative stimulus effects of a 5-HT2A agonist, substituted for clozapine discriminative stimulus, enhanced amphetamine induced locomotor activity, inhibited phencyclidine (PCP)-induced locomotor activity, and rescued PCP-induced disruption of prepulse inhibition without induction of catalepsy.ConclusionsThe molecular profile of nuciferine was similar but not identical to that shared with several approved antipsychotic drugs suggesting that nuciferine has atypical antipsychotic-like actions

    Detection of a Fourth Orbivirus Non-Structural Protein

    The genus Orbivirus includes both insect and tick-borne viruses. The orbivirus genome, composed of 10 segments of dsRNA, encodes 7 structural proteins (VP1–VP7) and 3 non-structural proteins (NS1–NS3). An open reading frame (ORF) that spans almost the entire length of genome segment-9 (Seg-9) encodes VP6 (the viral helicase). However, bioinformatic analysis recently identified an overlapping ORF (ORFX) in Seg-9. We show that ORFX encodes a new non-structural protein, identified here as NS4. Western blotting and confocal fluorescence microscopy, using antibodies raised against recombinant NS4 from Bluetongue virus (BTV, which is insect-borne), or Great Island virus (GIV, which is tick-borne), demonstrate that these proteins are synthesised in BTV or GIV infected mammalian cells, respectively. BTV NS4 is also expressed in Culicoides insect cells. NS4 forms aggregates throughout the cytoplasm as well as in the nucleus, consistent with identification of nuclear localisation signals within the NS4 sequence. Bioinformatic analyses indicate that NS4 contains coiled-coils, is related to proteins that bind nucleic acids, or are associated with membranes and shows similarities to nucleolar protein UTP20 (a processome subunit). Recombinant NS4 of GIV protects dsRNA from degradation by endoribonucleases of the RNAse III family, indicating that it interacts with dsRNA. However, BTV NS4, which is only half the putative size of the GIV NS4, did not protect dsRNA from RNAse III cleavage. NS4 of both GIV and BTV protect DNA from degradation by DNAse. NS4 was found to associate with lipid droplets in cells infected with BTV or GIV or transfected with a plasmid expressing NS4

    Detection of Batrachochytrium dendrobatidis in amphibians imported into the UK for the pet trade

    There is increasing evidence that the global spread of the fungal pathogen Batrachochytrium dendrobatidis (Bd) has been facilitated by the international trade in amphibians. Bd was first detected in the UK in 2004, and has since been detected in multiple wild amphibian populations. Most amphibians imported into the UK for the pet trade from outside the European Union enter the country via Heathrow Animal Reception Centre (HARC), where Bd positive animals have been previously detected. Data on the volume, diversity and origin of imported amphibians were collected for 59 consignments arriving at HARC between November 2009 and June 2012, along with a surveillance study to investigate the prevalence of Bd in these animals. Forty three amphibian genera were recorded, originating from 12 countries. It was estimated that 5000 – 7000 amphibians are imported through HARC into the UK annually for the pet trade. Bd was detected in consignments from the USA and Tanzania, in six genera, resulting in an overall prevalence of 3.6%. This suggests that imported amphibians are a source of Bd within the international pet trade

    Genome sequencing reveals Zika virus diversity and spread in the Americas

    Although the recent Zika virus (ZIKV) epidemic in the Americas and its link to birth defects have attracted a great deal of attention, much remains unknown about ZIKV disease epidemiology and ZIKV evolution, in part owing to a lack of genomic data. Here we address this gap in knowledge by using multiple sequencing approaches to generate 110 ZIKV genomes from clinical and mosquito samples from 10 countries and territories, greatly expanding the observed viral genetic diversity from this outbreak. We analysed the timing and patterns of introductions into distinct geographic regions; our phylogenetic evidence suggests rapid expansion of the outbreak in Brazil and multiple introductions of outbreak strains into Puerto Rico, Honduras, Colombia, other Caribbean islands, and the continental United States. We find that ZIKV circulated undetected in multiple regions for many months before the first locally transmitted cases were confirmed, highlighting the importance of surveillance of viral infections. We identify mutations with possible functional implications for ZIKV biology and pathogenesis, as well as those that might be relevant to the effectiveness of diagnostic tests

    Environmental detection of Batrachochytrium dendrobatidis in a temperate climate

    The aetiological agent of amphibian chytridiomycosis Batrachochytrium dendrobatidis is a primary cause of amphibian population declines. Current surveillance is based on the detection of B. dendrobatidis in its host but in vitro work suggests infective stages may survive in the abiotic environment for at least 3 mo. We describe here a surveillance system using filtration and quantita tive PCR that can detect B. dendrobatidis in small (<1 l) volumes of water. After assessing the analyt ical sensitivity of the protocol for both water and sediment samples in the laboratory, we analyzed environmental samples from the Sierra de Guadarrama mountain range in Spain at locations associ ated with chytrid-related die-offs and at other sites across Spain. B. dendrobatidis was detected in samples from 64% of the ponds in the Sierra de Guadarrama and at 2 sites outside this region, show ing that levels of amphibian exposure to B. dendrobatidis are spatially heterogeneous. In experimen tal microcosms, we detected B. dendrobatidis for up to 12 wk, though we found no evidence for an overall increase in biomass. Our results emphasise the need to further investigate the life cycle of B. dendrobatidis to more completely understand the epidemiology of this emerging pathogen.El agente etiológico de la quitridiomicosis de los anfibios, Batrachochytrium dendrobatidis, es una de las principales causas de la disminución de las poblaciones de anfibios. La vigilancia actual se basa en la detección de B. dendrobatidis en su huésped, pero los trabajos in vitro sugieren que los estadios infecciosos pueden sobrevivir en el entorno abiótico durante al menos 3 meses. Aquí describimos un sistema de vigilancia que utiliza la filtración y la PCR cuantitativa que puede detectar B. dendrobatidis en pequeños volúmenes de agua (<1 l). Después de evaluar la sensibilidad analítica del protocolo tanto para muestras de agua como de sedimentos en el laboratorio, analizamos muestras ambientales de la Sierra de Guadarrama en España en lugares asociados con la muerte de quitridios y en otros lugares de España. Se detectó B. dendrobatidis en muestras del 64% de las charcas de la Sierra de Guadarrama y en 2 lugares fuera de esta región, lo que demuestra que los niveles de exposición de los anfibios a B. dendrobatidis son espacialmente heterogéneos. En los microcosmos experimentales, detectamos B. dendrobatidis hasta 12 semanas, aunque no encontramos evidencias de un aumento global de la biomasa. Nuestros resultados subrayan la necesidad de seguir investigando el ciclo vital de B. dendrobatidis para comprender mejor la epidemiología de este patógeno emergente.Universidad Nacional, Costa Rica.Escuela de Medicina Veterinari
