    The first histidine triad motif of phtd is critical for zinc homeostasis in Streptococcus pneumoniae

    Streptococcus pneumoniae is the world's foremost human pathogen. Acquisition of the first row transition metal ion zinc is essential for pneumococcal colonization and disease. Zinc is acquired via the ATP-binding cassette transporter AdcCB and two zinc-binding proteins, AdcA and AdcAII. We have previously shown that AdcAII is reliant upon the polyhistidine triad (Pht) proteins to aid in zinc recruitment. Pht proteins generally contain five histidine (His) triad motifs that are believed to facilitate zinc binding and therefore play a significant role in pneumococcal metal ion homeostasis. However, the importance and potential redundancy of these motifs have not been addressed. We examined the effects of mutating each of the five His triad motifs of PhtD. The combination of in vitro growth assays, active zinc uptake, and PhtD expression studies show that the His triad closest to the protein's amino terminus is the most important for zinc acquisition. Intriguingly, in vivo competitive infection studies investigating the amino- and carboxyl-terminal His triad mutants indicate that the motifs have similar importance in colonization. Collectively, our new insights into the contributions of the individual His triad motifs of PhtD, and by extension the other Pht proteins, highlight the crucial role of the first His triad site in zinc acquisition. This study also suggests that the Pht proteins likely play a role beyond zinc acquisition in pneumococcal virulence

    Aeroacoustic Analysis of Fan Noise Reduction With Increased Bypass Nozzle Area

    An advanced model turbofan was tested in the NASA Glenn 9-by 15-Foot Low Speed Wind Tunnel (9x15 LSWT) to explore far field acoustic effects of increased bypass nozzle area. This fan stage test was part of the NASA Glenn Fan Broadband Source Diagnostic Test, second entry (SDT2) which acquired aeroacoustic results over a range of test conditions. The baseline nozzle was sized to produce maximum stage performance at cruise condition. However, the wind tunnel testing is conducted near sea level condition. Therefore, in order to simulate and obtain performance at other operating conditions, two additional nozzles were designed and tested one with +5 percent increase in weight flow (+5.4 percent increase in nozzle area compared with the baseline nozzle), sized to simulate the performance at the stage design point (takeoff) condition, and the other with a +7.5 percent increase in weight flow (+10.9 percent increase in nozzle area) sized for maximum weight flow with a fixed nozzle at sea level condition. Measured acoustic benefits with increased nozzle area were very encouraging, showing overall sound power level (OAPWL) reductions of 2 or more dB while the stage thrust actually increased by 2 to 3 percent except for the most open nozzle at takeoff rotor speed where stage performance decreased. Effective perceived noise levels for a 1500 ft engine flyover and 3.35 scale factor showed a similar noise reduction of 2 or more EPNdB. Noise reductions, principally in the level of broadband noise, were observed everywhere in the far field. Laser Doppler Velocimetry measurements taken downstream of the rotor showed that the total turbulent velocity decreased with increasing nozzle flow, which may explain the reduced rotor broadband noise levels

    The Effect of Bypass Nozzle Exit Area on Fan Aerodynamic Performance and Noise in a Model Turbofan Simulator

    The design of effective new technologies to reduce aircraft propulsion noise is dependent on identifying and understanding the noise sources and noise generation mechanisms in the modern turbofan engine, as well as determining their contribution to the overall aircraft noise signature. Therefore, a comprehensive aeroacoustic wind tunnel test program was conducted called the Fan Broadband Source Diagnostic Test as part of the NASA Quiet Aircraft Technology program. The test was performed in the anechoic NASA Glenn 9- by 15-Foot Low Speed Wind Tunnel using a 1/5 scale model turbofan simulator which represented a current generation, medium pressure ratio, high bypass turbofan aircraft engine. The investigation focused on simulating in model scale only the bypass section of the turbofan engine. The test objectives were to: identify the noise sources within the model and determine their noise level; investigate several component design technologies by determining their impact on the aerodynamic and acoustic performance of the fan stage; and conduct detailed flow diagnostics within the fan flow field to characterize the physics of the noise generation mechanisms in a turbofan model. This report discusses results obtained for one aspect of the Source Diagnostic Test that investigated the effect of the bypass or fan nozzle exit area on the bypass stage aerodynamic performance, specifically the fan and outlet guide vanes or stators, as well as the farfield acoustic noise level. The aerodynamic performance, farfield acoustics, and Laser Doppler Velocimeter flow diagnostic results are presented for the fan and four different fixed-area bypass nozzle configurations. The nozzles simulated fixed engine operating lines and encompassed the fan stage operating envelope from near stall to cruise. One nozzle was selected as a baseline reference, representing the nozzle area which would achieve the design point operating conditions and fan stage performance. The total area change from the smallest to the largest nozzle was 12.9 percent of the baseline nozzle area. The results will show that there are significant changes in aerodynamic performance and farfield acoustics as the fan nozzle area is increased. The weight flow through the fan model increased between 7 and 9 percent, the fan and stage pressure dropped between 8 and 10 percent, and the adiabatic efficiency increased between 2 and 3 percent--the magnitude of the change dependent on the fan speed. Results from force balance measurements of fan and outlet guide vane thrust will show that as the nozzle exit area is increased the combined thrust of the fan and outlet guide vanes together also increases, between 2 and 3.5 percent, mainly due to the increase in lift from the outlet guide vanes. In terms of farfield acoustics, the overall sound power level produced by the fan stage dropped nearly linearly between 1 dB at takeoff condition and 3.5 dB at approach condition, mainly due to a decrease in the broadband noise levels. Finally, fan swirl angle survey and Laser Doppler Velocimeter mean velocity and turbulence data obtained in the fan wake will show that the swirl angles and turbulence levels within the wake decrease as the fan nozzle area increases, which helps to explain the drop in the fan broadband noise at all fan speeds

    Subsonic and Transonic Wind Tunnel Testing of Two Inflatable Aerodynamic Decelerators

    Two inflatable aerodynamic decelerator designs were tested in the Transonic Dynamics Tunnel at the NASA Langley Research Center: a tension cone and an isotensoid. The tension cone consists of a flexible tension shell attached to a torus and the isotensoid employs a ram-air inflated envelope. Tests were conducted at Mach numbers from 0.3 to 1.08 and Reynolds numbers from 0.59 to 2.46 million. The main objective of these tests was to obtain static aerodynamic coefficients at subsonic and transonic speeds to supplement supersonic aerodynamic data for these same two designs. The axial force coefficients of both designs increased smoothly from subsonic through transonic Mach numbers. Dynamic data show significant oscillation of the tension cone and minimal oscillation of the isotensoid. The transonic and subsonic data will be used to assemble an inflatable decelerator aerodynamic database for use in computational analyses and system studies

    An opportunity for diagonal development in global surgery: cleft lip and palate care in resource-limited settings

    Global cleft surgery missions have provided much-needed care to millions of poor patients worldwide. Still, surgical capacity in low- and middle-income countries is generally inadequate. Through surgical missions, global cleft care has largely ascribed to a vertical model of healthcare delivery, which is disease specific, and tends to deliver services parallel to, but not necessarily within, the local healthcare system. The vertical model has been used to address infectious diseases as well as humanitarian emergencies. By contrast, a horizontal model for healthcare delivery tends to focus on long-term investments in public health infrastructure and human capital and has less often been implemented by humanitarian groups for a variety of reasons. As surgical care is an integral component of basic healthcare, the plastic surgery community must challenge itself to address the burden of specific disease entities, such as cleft lip and palate, in a way that sustainably expands and enriches global surgical care as a whole. In this paper, we describe a diagonal care delivery model, whereby cleft missions can enrich surgical capacity through integration into sustainable, local care delivery systems. Furthermore, we examine the applications of diagonal development to cleft care specifically and global surgical care more broadly

    Akt regulates centrosome migration and spindle orientation in the early Drosophila melanogaster embryo

    Correct positioning and morphology of the mitotic spindle is achieved through regulating the interaction between microtubules (MTs) and cortical actin. Here we find that, in the Drosophila melanogaster early embryo, reduced levels of the protein kinase Akt result in incomplete centrosome migration around cortical nuclei, bent mitotic spindles, and loss of nuclei into the interior of the embryo. We show that Akt is enriched at the embryonic cortex and is required for phosphorylation of the glycogen synthase kinase-3β homologue Zeste-white 3 kinase (Zw3) and for the cortical localizations of the adenomatosis polyposis coli (APC)–related protein APC2/E-APC and the MT + Tip protein EB1. We also show that reduced levels of Akt result in mislocalization of APC2 in postcellularized embryonic mitoses and misorientation of epithelial mitotic spindles. Together, our results suggest that Akt regulates a complex containing Zw3, Armadillo, APC2, and EB1 and that this complex has a role in stabilizing MT–cortex interactions, facilitating both centrosome separation and mitotic spindle orientation

    Comparison of Intraoperative Fluoroscopy to Postoperative Weight-Bearing Radiographs Obtained 4 to 6 Weeks After Bunion Repair With A Chevron Osteotomy

    Background: During operative treatment of bunions, an attempt is made to correct the hallux valgus angle (HVA) and the intermetatarsal angle (IMA). In this study, the HVA and the IMA were measured using intraoperative C-arm fluoroscopic images obtained during surgical treatment of a bunion with chevron osteotomy. These angles were again measured using weight-bearing radiographs obtained 4 to 6 weeks postoperatively. Methods: At our institution, we reviewed medical records of patients who underwent a bunion repair with chevron osteotomy between January 2013 and October 2017. A total of 26 feet from 24 patients were included. Three authors (ALP, TMH, and RAM) measured the HVA and IMA using intraoperative fluoroscopic images and postoperative weight-bearing radiographs (4 measurements per foot; total, 104 measurements). The authors were blinded to their previous angular measurements and to measurements made by the others. An intraclass correlation coefficient was calculated for the HVA and IMA measurements between groups (ie, intraoperative fluoroscopic images and postoperative radiographs) to determine interobserver reliability. We compared the angles measured by the authors between groups and used a paired t test for statistical evaluation. Results: Interobserver difference of the HVA and IMA was low between intraoperative fluoroscopic images and postoperative weight-bearing radiographs (0.98 and 0.79; 0.78 and 0.95, respectively). The measured IMAs were relatively consistent between groups (6.21° and 6.37°, respectively); only two patients had a difference \u3e 3°. There was a greater difference in HVAs between groups (11.5° and 14.2°, respectively). In 11 feet, the HVA was \u3e 5° (range, 5.3-12.7°) in the postoperative radiograph compared to the fluoroscopic image. In one foot, we noted a 7° decrease of the HVA on the postoperative radiograph. The average difference of HVA between groups was 2.6° (P \u3c 0.0001), whereas the IMA was 0.16° (P = 0.002). Conclusions: Interobserver measurements of the HVA and IMA were reliable on both the intraoperative fluoroscopic images and the postoperative weightbearing radiographs. The IMA was similar between groups; however, the HVA was often greater on the postoperative weight-bearing radiographs
