    Evaluation of indigenous Trichoderma isolates from Manipur as biocontrol agent against Pythium aphanidermatum on common beans

    Pythium aphanidermatum is one of the common causal pathogen of damping-off disease of beans (Phaseolus vulgaris L.) grown in Manipur. A total of 110 indigenous Trichoderma isolates obtained from North east India were screened for their biocontrol activity which can inhibit the mycelial growth of P. aphanidermatum, the causal organism of damping-off in beans. Out of the total isolates, 32% of them showed strong antagonistic activity against P. aphanidermatum under in vitro condition and subsequently 20 best isolates were selected based on their mycelial inhibition capacity against P. aphanidermatum for further analysis. Different biocontrol mechanisms such as protease, chitinase, β-1,3-glucanase activity, cellulase and production of volatile and non-volatile compounds were also assayed. Based on their relative biocontrol potency, only three indigenous Trichoderma isolates (T73, T80 and T105) were selected for pot culture experiment against damping-off diseases in common beans. In greenhouse experiment, Trichoderma isolates T-105 significantly reduced the pre- and post-emergence damping-off disease incidence under artificial infection with P. aphanidermatum and showed highest disease control percentage

    Manejo da podridão-de-Sclerotium em pimentão em um argisolo no Amazonas

    A podridão-de-Sclerotium é uma doença comum em plantas da família Solanaceae na Amazônia. Visando avaliar estratégias de manejo para esta doença em pimentão (Capsicum annuum, L. Solanaceae), foi conduzido experimento em campo em blocos casualizados com parcelas subdivididas e seis repetições, em Argissolo Vermelho-Amarelo artificialmente infestado com Sclerotium rolfsii. O tratamento principal foi a cobertura do solo (cobertura do solo com serragem ou solo nu). Os tratamentos secundários consistiram na adição ao solo de: 1) composto vegetal (3 L por cova), 2) arroz colonizado com Trichoderma harzianum (90 g por cova contendo ≈ 1,4 x 10(9) conídios g-1), 3) composto vegetal e T. harzianum nas mesmas proporções descritas anteriormente e 4) testemunha. Todas as plantas receberam apenas adubação orgânica com composto vegetal na proporção de 1,5 L por cova, exceto as dos tratamentos com 3 L de composto por cova. A parcela principal foi constituída de três fileiras com dez plantas de pimentão (0,50 x 1,0 m) e cada subparcela continha três fileiras com cinco plantas. A incidência da podridão-de-Sclerotium foi avaliada duas vezes por semana. A cobertura morta favoreceu significativamente a ocorrência da doença. Nas parcelas com esse tratamento o aumento da intensidade da doença, expressa em área abaixo da curva de progresso da doença (AACPD), foi 35,5% maior, em comparação com as parcelas sem cobertura morta. A aplicação de T. harzianum ou o incremento na quantidade de composto (de 1,5 para 3 L por cova) reduziu a AACPD em 38,1% e 37,5%, respectivamente. A aplicação de T. harzianum ou o incremento na quantidade de composto, mesmo nos tratamentos com cobertura morta, reduziu significativamente a AACPD em 52,8% e em 55,1%, respectivamente, em comparação com o tratamento apenas com cobertura morta. Esses resultados sugerem que a utilização de T. harzianum e o aumento na quantidade de composto por cova são estratégias eficientes de manejo da podridão-de-Sclerotium em pimentão. A cobertura morta com serragem não deve ser utilizada em áreas infestadas com S. rolfsii

    Biofortification is a plant breeding method that introduces increased concentrations of minerals in staple food crops (e.g., legumes, cereal grains), and has shown success in alleviating insufficient Fe intake in various human populations. Unlike other strategies utilized to alleviate Fe deficiency, studies of the gut microbiota in the context of Fe biofortification have not yet been reported, although the consumption of Fe biofortified staple food crops has increased significantly over time. Hence, in this study, we performed a 6-week feeding trial in Gallus gallus (n = 14), aimed to investigate the alterations in the gut microbiome following administration of an Fe biofortified bean-based diet (biofortified, BFe) versus a bean based diet with poorly-bioavailable Fe (standard, SFe). Cream seeded carioca bean based diets were designed in an identical fashion to those used in a recent human clinical trial of Fe biofortified beans in Rwanda. We hypothesized that the different dietary Fe contents in the beans based diets will alter the composition and function of the intestinal microbiome. The primary outcomes were changes in the gut microbiome composition and function analyzed by 16S rRNA gene sequencing. We observed no significant changes in phylogenetic diversity between groups. There were significant differences in the composition of the microbiota between groups, with the BFe group harboring fewer taxa participating in bacterial Fe uptake, increased abundance of bacteria involved in phenolic catabolism, and increased abundance of beneficial butyrate-producing bacteria. Additionally, depletion of key bacterial pathways responsible for bacterial viability and Fe uptake suggest that improvements in Fe bioavailability, in addition to increases in Fe-polyphenol and Fe-phytate complexes due to biofortification, led to decreased concentrations of cecal Fe available for bacterial utilization. Our findings demonstrate that Fe biofortification may improve Fe status without negatively altering the structure and function of the gut microbiota, as is observed with other nutritional methods of Fe supplementation.     Chronic Zinc Deficiency Alters Chick Gut Microbiota Composition and Function

    Zinc (Zn) deficiency is a prevalent micronutrient insufficiency. Although the gut is a vital organ for Zn utilization, and Zn deficiency is associated with impaired intestinal permeability and a global decrease in gastrointestinal health, alterations in the gut microbial ecology of the host under conditions of Zn deficiency have yet to be studied. Using the broiler chicken (Gallus gallus) model, the aim of this study was to characterize distinct cecal microbiota shifts induced by chronic dietary Zn depletion. We demonstrate that Zn deficiency induces significant taxonomic alterations and decreases overall species richness and diversity, establishing a microbial profile resembling that of various other pathological states. Through metagenomic analysis, we show that predicted Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways responsible for macro- and micronutrient uptake are significantly depleted under Zn deficiency; along with concomitant decreases in beneficial short chain fatty acids, such depletions may further preclude optimal host Zn availability. We also identify several candidate microbes that may play a significant role in modulating the bioavailability and utilization of dietary Zn during prolonged deficiency. Our results are the first to characterize a unique and dysbiotic cecal microbiota during Zn deficiency, and provide evidence for such microbial perturbations as potential effectors of the Zn deficient phenotype
