Metsätuhot vuonna 2022
Luonnonvarakeskus tuottaa vuosittaista Metsätuhot - raporttisarjaa. Raporteissa käydään läpi mitä metsissä, taimitarhoilla sekä puisto- että pihapuilla menneenä vuonna tapahtui eri tuhonaiheuttajien osalta. Lisäksi raportissa käydään läpi muitakin metsien mielenkiintoisiä ilmiÜitä, vaikkeivat ne varsinaisia tuhoja olisivatkaan olleet. Vuosi 2022 oli tältä osin mielenkiintoinen, sillä helteisen kesän 2021 jäljet näkyivät osin vasta tällÜin.
Vuonna 2022 kirjanpainajan (Ips typographus) takia hakattiin metsiä metsäkeskuksen keräämien ilmoitusten perusteella enemmän kuin kertaakaan tilastointihistoriassa (2 834 ha). Laji myÜs parveili aiempaa runsaampana Luken feromoniseurannan pohjoisimmilla alueilla. Kirjanpainajan lisäksi myÜs toinen havupuiden kaarnakuoriainen, kuusentähtikirjaaja, aiheutti näkyviä latvakuolemia kuuselle eri puolilla Suomea. Molempien ilmiÜiden takana vaikutti paitsi kuoriaisille kohtuullinen kesä 2022, niin ennen kaikkea helteinen ja lämmin kesä 2021. IlmiÜ konkretisoikin omalla tavallaan ilmaston lämpenemisen mahdollisia vaikutuksia metsissämme.
Helteiden tuhoja lisäävät vaikutukset eivät kuitenkaan näkyneet valtakunnan metsien inventoinnissa (VMI) samalla tavalla. VMI:ssä havaitut kirjanpainajatuhot jopa laskivat (-14 %) hieman vuoden takaisesta. Tässä on kuitenkin muistettava, että VMI otanta ei ole ajallisesti eikä spatiaaliselta tarkkuudeltaan soveltuva useimpien hyĂśnteis- ja sienituhojen seurantaan. Merkittävimpinä tuhonaiheuttajina VMI:ssä olivat aiempien vuosien tapaan lumi, tuuli ja hirvieläimet â eli tuhot, joita voidaan yleisesti havaita maastossa inventointiajankohdasta riippumatta.
Lämpenevään ilmastoon reagoivat hyÜnteisten lisäksi useat muutkin kotoperäiset taudinaiheuttajat. Ne hyÜtyvät ilmastonmuutoksesta mm. laajentamalla esiintymisaluettaan sekä aiheuttamalla puissa enemmän vaurioita, etenkin kun säät muuttuvat suopeimmaksi niille, mutta huonommiksi niiden isäntäpuille. Kuusenjuurikääpää eteläisempi männynjuurikääpä (Heterobasidion annosum s.s.) raportoitiinkin vuonna 2022 Pohjois-Pohjanmaalta saakka. Juurikääpätuhot myÜs lisääntyivät noin 30 % vuonna 2022 edellisvuoteen verrattuna. Lämpimät ja kuivat kesät ovat myÜs edesauttaneet tulokaslaji havuparikkaan (Diplodia sapinea) aiheuttaman taudin etelänversosurman puhkeamista. Havuparikkaan levinneisyysaluetta kartoitettiin vuonna 2022 kansalaistieteen avulla, ja sienen esiintyminen olikin ennakoitua laajempi. MyÜs tulokaslaji havununna (Lymantria monacha) tavattiin hieman aiempaa runsaampana lajin pohjoisemmilla esiintymisalueilla.
Vuonna 2022 päätään nostivat valitettavasti myÜs ilmastosta riippumattomat uhkatekijät: kansainvälisen kasvikaupan ja turismin mukana leviävät haitalliset vieraslajit. Uusia vierasperäisiä tuhonaiheuttajia lÜydettiin niin taimitarhoilta, kuin puistopuistakin. Koivuilla todettiin Discula betulina-sienen aiheuttamaa versolaikkua, pihdoilta taas lÜydettiin pihtanäppyä (Neonectria macrospora). Haitallisten vieraslajien leviäminen on kansainvälinen ongelma, jolta pohjoinen Suomikaan ei ole turvassa. Luken tutkijat julkaisivat vuonna 2022 suomenkielisen artikkelisarjan mikä sisälsi tietoa useiden eri vieraslajien uhasta. Katsaus muistuttaa, ettei metsiemme valtapuilla useinkaan ole vastustuskykyä näitä uusia tuholaisia vastaan, mikä tekee niistä merkittävän uhan metsien terveydelle. Haitallisten vieraslajien leviämisen estäminen on siis erityisen tärkeää niin nyt kuin tulevaisuudessakin. Tässä kansainvälinen kasvikauppa on pahin uhkatekijä. Metsien ja puutarhojen osalta ulkomaisten taimien ja koristekasvien tuonti muodostaa tehokkaan haitallisten vieraslajien leviämisväylän. Kotimainen taimituotanto taasen tuottaa paitsi terveitä, niin myÜs alkuperältään tunnettuja ja laadultaan varmistettuja taimia. Näitä taimia jokaisen tulisi suosia niin metsänuudistamisessa kuin kotipihoilla, sillä olemassa oleva lainsäädäntÜ ei kansainvälistä kasvikauppaa hillitse.
Luke kerää ja koosta myÜs kansalaisten tekemiä ilmoituksia havaituista puustotuhoista tai muista metsien ilmiÜstä. Vuonna 2022 okakaarnakuoriaisten (Ips acuminatus) takia tehdyt tuhoilmoitukset jatkoivat kasvuaan vuodelta 2021. Ilmoituksissa ja niiden perusteella tehdyissä maastotarkastuksissa mäntyjen todettiin kuolevan etenkin lounaisrannikolla, missä okakaarnakuoriaiset tuntuivat viihtyvän hyvin yhdessä havuparikkaan kanssa. Selvitys näiden kahden tuholaisen mahdollisesta yhteisvaikutuksesta äkillisesti kuivuneiden mäntyjen kuolemiin jatkuu Lukessa. Tämän vuoden raporttiin omaksi tarinakseen nostettiin myÜs ilmoituksia kerännyt lehtikuoriaisten runsas esiintyminen, missä etenkin lepillä esiintyvä idänlehtikuoriainen (Agelastica alni) oli näkyvässä roolissa.
Vaikka vuoden 2022 perusteella tilanne Suomen metsissä on tuhojen suhteen pääosin hyvä, vuosi muistutti meitä konkreettisesti sekä muuttuvan ilmaston että kansainvälisen kasvikaupan meille tuomista uhkista. Sekä vieras- että tulokaslajien aiheuttamat riskit tulevat kasvamaan lämpenevässä ilmastossa, sillä sen ansiosta nämä lajit pystyvät todennäkÜisemmin vakiinnuttamaan asemansa uudella alueellaan. Meillä jo ongelmia aiheuttavien lajien kuten juurikäävän tai kirjanpainajan osalta lämpenevä ilmasto ei niin ikään lupaa missään määrin hyvää metsillemme. Ollaksemme varautuneita mahdolliseen tuhojen äkilliseen lisääntymiseen tarvitsemme paitsi aktiivista seurantaa, niin myÜs perus- ja soveltavaa tutkimusta. sekä tarkempaa analyysiä siitä miten eri tuhonaiheuttajien uhat voivat meillä tulevaisuudessa realisoitua
Metabolic Regulation in Progression to Autoimmune Diabetes
Recent evidence from serum metabolomics indicates that specific metabolic disturbances precede β-cell autoimmunity in humans and can be used to identify those children who subsequently progress to type 1 diabetes. The mechanisms behind these disturbances are unknown. Here we show the specificity of the pre-autoimmune metabolic changes, as indicated by their conservation in a murine model of type 1 diabetes. We performed a study in non-obese prediabetic (NOD) mice which recapitulated the design of the human study and derived the metabolic states from longitudinal lipidomics data. We show that female NOD mice who later progress to autoimmune diabetes exhibit the same lipidomic pattern as prediabetic children. These metabolic changes are accompanied by enhanced glucose-stimulated insulin secretion, normoglycemia, upregulation of insulinotropic amino acids in islets, elevated plasma leptin and adiponectin, and diminished gut microbial diversity of the Clostridium leptum group. Together, the findings indicate that autoimmune diabetes is preceded by a state of increased metabolic demands on the islets resulting in elevated insulin secretion and suggest alternative metabolic related pathways as therapeutic targets to prevent diabetes
Generation of self-reactive, shared T-cell receptor ? chains in the human thymus
The T-cell receptor (TCR) repertoire is generated in a semistochastic process of gene recombination and pairing of TCR? to TCR? chains with the estimated total TCR diversity of >108. Despite this high diversity, similar or identical TCR chains are found to recur in immune responses. Here, we analyzed the thymic generation of TCR sequences previously associated with recognition of self- and nonself-antigens, represented by sequences associated with autoimmune diabetes and HIV, respectively. Unexpectedly, in the CD4+ compartment TCR? chains associated with the recognition of self-antigens were generated in significantly higher numbers than TCR? chains associated with the recognition of nonself-antigens. The analysis of the circulating repertoire further showed that these chains are not lost in negative selection nor predominantly converted to the regulatory T-cell lineage. The high abundance of self-reactive TCR? chains in multiple individuals suggests that the human thymus has a predilection to generate self-reactive TCR? chains independently of the HLA-type and that the individual risk of autoimmunity may be modulated by the TCR? repertoire associated with these chains.Peer reviewe
Autoantibodies to N-terminally truncated GADââ (96â585):HLA associations and predictive value for type 1 diabetes
Abstract
Objective: To evaluate the role of autoantibodies to N-terminally truncated glutamic acid decarboxylase GADââ
(96â585) (t-GADA) as a marker for type 1 diabetes (T1D) and to assess the potential human leukocyte antigen (HLA) associations with such autoantibodies.
Methods: In this cross-sectional study combining data from the Finnish Pediatric Diabetes Register, the Type 1 Diabetes Prediction and Prevention study, the DIABIMMUNE study, and the Early Dietary Intervention and Later Signs of Beta-Cell Autoimmunity study, venous blood samples from 760 individuals (53.7% males) were analyzed for t-GADA, autoantibodies to full-length GAD65 (f-GADA), and islet cell antibodies. Epitope-specific GAD autoantibodies were analyzed from 189 study participants.
Results: T1D had been diagnosed in 174 (23%) participants. Altogether 631 (83%) individuals tested positive for f-GADA and 451 (59%) for t-GADA at a median age of 9.0 (range 0.2â61.5) years. t-GADA demonstrated higher specificity (46%) and positive predictive value (30%) for T1D than positivity for f-GADA alone (15% and 21%, respectively). Among participants positive for f-GADA, those who tested positive for t-GADA carried more frequently HLA genotypes conferring increased risk for T1D than those who tested negative for t-GADA (77% vs 53%; P < 0.001).
Conclusions: Autoantibodies to N-terminally truncated GAD improve the screening for T1D compared to f-GADA and may facilitate the selection of participants for clinical trials. HLA class II-mediated antigen presentation of GAD(96â585)-derived or structurally similar peptides might comprise an important pathomechanism in T1D

Heterogeneity in the presentation of clinical type 1 diabetes defined by the level of risk conferred by human leukocyte antigen class II genotypes
Abstract
Objectives: The association between human leukocyte antigen (HLA) class II genotypes and susceptibility to type 1 diabetes (T1D) is well established. This study aimed at examining whether there are differences in the presentation of T1D depending on the HLA genotype.
Research Design and Methods: We divided the study participants (N = 5798) in the Finnish Pediatric Diabetes Register into two groups based on the T1D risk conferred by their HLA genotype (high and moderate-risk genotypes, Group 1 vs. other genotypes, Group 2). We then examined differences in clinical, metabolic, and immunological characteristics. Children included in the study were 0â14-year-old and diagnosed between January 2003 and December 2019.
Results: Participants in Group 1 were younger at the time of diagnosis (Pâ<â0.001) and had more frequently family members affected by T1D (Pâ<â0.001). Diabetic ketoacidosis (DKA) was more frequent among participants in Group 2 (P = 0.014) who also had a longer duration of symptoms before diagnosis (Pâ<â0.001) and higher hemoglobin A1c (P = 0.001) at diagnosis. The HLA genotype was not, however, directly related to the DKA frequency. The frequency of islet cell antibodies (Pâ<â0.003), insulin autoantibodies (Pâ<â0.001), and islet antigen 2 autoantibodies (Pâ<â0.001) was higher in Group 1 whereas glutamic acid decarboxylase autoantibodies were more frequent (Pâ<â0.001) in Group 2. Group 1 had more participants with multiple autoantibodies (P = 0.027) whereas antibody negativity was more frequent in Group 2 (P = 0.003).
Conclusions: These findings indicate disease heterogeneity in relation to both clinical disease presentation and humoral autoimmunity, in particular. This heterogeneity is, at least partly, defined by HLA Class II genotypes
HLA-DR-DQ haplotypes and specificity of the initial autoantibody in islet specific autoimmunity
Abstract
Objective: We aimed to clarify the association of various HLA risk alleles with different types of autoantibodies initiating islet specific autoimmunity.
Methods: Follow-up cohorts from the Finnish Type 1 Diabetes Prediction and Prevention (DIPP) study and children diagnosed with type 1 diabetes (T1D) from the Finnish Pediatric Diabetes Register (FPDR) were analyzed for the presence of autoantibodies to insulin (IAA), glutamic acid decarboxylase (GADA), IA-2 antigen (IA-2A), and zinc transporter 8 (ZnT8A); and genotyped for HLA DR/DQ alleles. In the DIPP study, autoantibodies were regularly analyzed from birth up to 15âyears of age.
Results: In the DIPP cohort, 621 children developed one single persistent autoantibody, GADA in 284, IAA in 268, and IA-2A in 40 cases. Highly significant differences in the specificity of the first autoantibody were observed between HLA genotypes. Homozygotes for the DR3-DQ2 haplotype had almost exclusively GADA as the first autoantibody, whereas a more even distribution between GADA and IAA was found in DR3-DQ2/DR4-DQ8 as well as DR3-DQ/x and DR4-DQ8/x genotypes (x referring to neutral haplotypes). In DR4-DQ8 positive genotypes with the DRB1*04:01 allele IAA was more often the first autoantibody than in DRB1*04:04 positive genotypes. Various neutral haplotypes also significantly affected the relative proportions of different initial autoantibodies. These findings were confirmed and expanded in a series of 1591 T1D children under the age of 10âyears from FPDR.
Conclusions: These results emphasize the importance of HLA class II polymorphisms in the recognition of autoantigen epitopes in the initiation of various pathways of the autoimmune response
Fungal dysbiosis and intestinal inflammation in children with beta-cell autoimmunity
Abstract
Although gut bacterial dysbiosis is recognized as a regulator of beta-cell autoimmunity, no data is available on fungal dysbiosis in the children at the risk of type 1 diabetes (T1D). We hypothesized that the co-occurrence of fungal and bacterial dysbiosis contributes to the intestinal inflammation and autoimmune destruction of insulin-producing beta-cells in T1D. Fecal and blood samples were collected from 26 children tested positive for at least one diabetes-associated autoantibody (IAA, GADA, IA-2A or ICA) and matched autoantibody-negative children with HLA-conferred susceptibility to T1D (matched for HLA-DQB1 haplotype, age, gender and early childhood nutrition). Bacterial 16S and fungal ITS2 sequencing, and analyses of the markers of intestinal inflammation, namely fecal human beta-defensin-2 (HBD2), calprotectin and secretory total IgA, were performed. Anti-Saccharomyces cerevisiae antibodies (ASCA) and circulating cytokines, IFNG, IL-17 and IL-22, were studied. After these analyses, the children were followed for development of clinical T1D (median 8 years and 8 months). Nine autoantibody positive children were diagnosed with T1D, whereas none of the autoantibody negative children developed T1D during the follow-up. Fungal dysbiosis, characterized by high abundance of fecal Saccharomyces and Candida, was found in the progressors, i.e., children with beta-cell autoimmunity who during the follow-up progressed to clinical T1D. These children showed also bacterial dysbiosis, i.e., increased Bacteroidales and Clostridiales ratio, which was, however, found also in the non-progressors, and is thus a common nominator in the children with beta-cell autoimmunity. Furthermore, the progressors showed markers of intestinal inflammation detected as increased levels of fecal HBD2 and ASCA IgG to fungal antigens. We conclude that the fungal and bacterial dysbiosis, and intestinal inflammation are associated with the development of T1D in children with beta-cell autoimmunity
