Hormony tarczycy i otyłość: znana, lecz słabo rozpoznana relacja
Hormony tarczycy (thyroid hormones, TH) są zaangażowane w wiele różnych procesów biologicznych, wliczając rozwój układu nerwowego, regulację metabolizmu pośredniego oraz zużycie energii. Aktywnie uczestniczą w podstawowym zużyciu energii i termogenezie adaptacyjnej i z tego względu mogą mieć wpływ na masę ciała w przebiegu chorób tarczycy. Otyłość to niezakaźna, przewlekła, zapalna choroba metaboliczna, która implikuje dodatni bilans energetyczny. Tkanka tłuszczowa produkuje szereg hormonów i adipocytokin, takich jak leptyna, które mogą wpływać na stan tarczycy na różnych poziomach. Istnieją dowody na to, że dysfunkcja tarczycy może predysponować do otyłości i odwrotnie, istnieją dowody sugerujące, że otyłość powoduje zmiany dotyczące tarczycy. Celem tej pracy było opisanie związku między układem tarczycy a otyłością. Ponadto w pracy zaprezentowano hipotetyczny model podkreślający znaczenie obwodowej dejodynacji hormonów tarczycy i jego rolę w ustanowieniu dodatniego bilansu energetycznego. Podsumowując, możemy stwierdzić, że relacja między układem tarczycy a otyłością i nadwagą jest złożona i obejmuje wiele poziomów interakcji. Ponadto, poddając ocenie otyłego pacjenta, powinno się rozważyć ocenę funkcji tarczycy, aby uzyskać lepsze i spersonalizowane efekty leczenia

An oleuropein β-glucosidase from olive fruit is involved in determining the phenolic composition of virgin olive oil
Phenolic composition of virgin olive oil is determined by the enzymatic and/or chemical reactions that take place during olive fruit processing. Of these enzymes, β-glucosidase activity plays a relevant role in the transformation of the phenolic glycosides present in the olive fruit, generating different secoiridoid derivatives. The main goal of the present study was to characterize olive fruit β-glucosidase genes and enzymes responsible for the phenolic composition of virgin olive oil. To achieve that, we have isolated an olive β-glucosidase gene from cultivar Picual (OepGLU), expressed in Nicotiana benthamiana leaves and purified its corresponding recombinant enzyme. Western blot analysis showed that recombinant OepGLU protein is detected by an antibody raised against the purified native olive mesocarp β-glucosidase enzyme, and exhibits a deduced molecular mass of 65.0 kDa. The recombinant OepGLU enzyme showed activity on the major olive phenolic glycosides, with the highest levels with respect to oleuropein, followed by ligstroside and demethyloleuropein. In addition, expression analysis showed that olive GLU transcript level in olive fruit is spatially and temporally regulated in a cultivar-dependent manner. Furthermore, temperature, light and water regime regulate olive GLU gene expression in olive fruit mesocarp. All these data are consistent with the involvement of OepGLU enzyme in the formation of the major phenolic compounds present in virgin olive oil
Imatinib therapy of chronic myeloid leukemia restores the expression levels of key genes for DNA damage and cell-cycle progression
[EN] Background
Chronic myeloid leukemia (CML) is a malignant clonal disorder of the hematopoietic system caused by the expression of the BCR/ABL fusion oncogene. It is well known that CML cells are genetically unstable. However, the mechanisms by which these cells acquire genetic alterations are poorly understood. Imatinib mesylate is the standard therapy for newly diagnosed CML patients. Imatinib mesylate targets the oncogenic kinase activity of BCR-ABL.
Objective
To study the gene expression profile of bone marrow hematopoietic cells in the same patients with CML before and 1 month after imatinib therapy.
Methods
Samples from patients with CML were analyzed using Affymetrix GeneChip Expression Arrays.
Results
A total of 594 differentially expressed genes, most of which (393 genes) were downregulated, as a result of imatinib therapy were observed.
Conclusion
The blockade of oncoprotein Bcr-Abl by imatinib could cause a decrease in the expression of key DNA repair genes and substantially modify the expression profile of the bone marrow cells in the first days of therapy
Cytogenetic and genomic analysis of a patient with turner syndrome and t(2;12): a case report
Background: Turner syndrome is a genetic disorder that afects women. It is caused by an absent or incomplete X chromosome, which can be presented in mosaicism or not. There are 12 cases of Turner syndrome patients who present structural alterations in autosomal chromosomes.
Case presentation: The present case report describes a patient with a reciprocal, maternally inherited translocation between chromosomes 2 and 12 with a mosaicism of X monosomy 45,X,t(2;12)(p13;q24)[95]/46,XX,t(2;12)(p13;q24) [5]. Through genetic mapping arrays, altered genes in the patient were determined within the 23 chromosome pairs. These genes were associated with the patient’s clinical features using a bioinformatics tool
Conclusion: To our knowledge, this is the frst case in which a translocation (2;12) is reported in a patient with Turner syndrome and confrmed by conventional cytogenetics, FISH and molecular genetics. Clinical features of our patient are closely related with the loss of one X chromosome, however mild intellectual disability can be likely explained by autosomal genes. The presence of familial translocations was a common fnding, thus emphasizing the need for familiar testing for further genetic counselling
Structural changes after videogame practice related to a brain network associated with intelligence
Here gray and white matter changes after four weeks of videogame practice were analyzed using optimized voxel-based morphometry (VBM), cortical surface and cortical thickness indices, and white matter integrity computed from several projection, commissural, and association tracts relevant to cognition. Beginning with a sample of one hundred young females, twenty right handed participants were recruited for the study and assigned to a practice or a control group carefully matched by their general cognitive ability scores. After the first scan, the practice group played ‘Professor Layton and The Pandora's Box’ 4 h per week during four weeks. A second scan was obtained at the end of practice and intelligence was measured again. Image analyses revealed gray and white matter changes in the practice group. Gray matter changes theoretically relevant for intelligence were observed for the practice group mainly in frontal clusters (Brodmann areas 9 and 10) and also in smaller parietal and temporal regions. White matter findings were focused in the hippocampal cingulum and the inferior longitudinal fasciculus. These gray and white matter changes presumably induced by practice did not interact with intelligence tests' scores
