Centronuclear Myopathy Caused by Defective Membrane Remodelling of Dynamin 2 and BIN1 Variants

    Get PDF
Centronuclear myopathy (CNM) is a congenital myopathy characterised by centralised nuclei in skeletal myofibers. T-tubules, sarcolemmal invaginations required for excitation-contraction coupling, are disorganised in the skeletal muscles of CNM patients. Previous studies showed that various endocytic proteins are involved in T-tubule biogenesis and their dysfunction is tightly associated with CNM pathogenesis. DNM2 and BIN1 are two causative genes for CNM that encode essential membrane remodelling proteins in endocytosis, dynamin 2 and BIN1, respectively. In this review, we overview the functions of dynamin 2 and BIN1 in T-tubule biogenesis and discuss how their dysfunction in membrane remodelling leads to CNM pathogenesis

3,3′,4,4′,5-Pentachlorobiphenyl Inhibits Drug Efflux Through P-Glycoprotein in KB-3 Cells Expressing Mutant Human P-Glycoprotein

The effects on the drug efflux of 3,3′,4,4′,5-pentachlorobiphenyl (PCB-126), the most toxic of all coplanar polychlorinated biphenyls (Co-PCBs), were examined in KB-3 cells expressing human wild-type and mutant P-glycoprotein in which the 61st amino acid was substituted for serine or phenylalanine (KB3-Phe(61)). In the cells expressing P-glycoproteins, accumulations of vinblastine and colchicine decreased form 85% to 92% and from 62% to 91%, respectively, and the drug tolerances for these chemicals were increased. In KB3-Phe(61), the decreases in drug accumulation were inhibited by adding PCB-126 in a way similar to that with cyclosporine A: by adding 1 μM PCB-126, the accumulations of vinblastine and colchicine increased up to 3.3- and 2.3-fold, respectively. It is suggested that PCB-126 decreased the drug efflux by inhibiting the P-glycoprotein in KB3-Phe(61). Since there were various P-glycoproteins and many congeners of Co-PCBs, this inhibition has to be considered a new cause of the toxic effects of Co-PCBs

Aquaporin 1 expression in tissues of canines possessing inherited high K+ erythrocytes

We investigated the expression of aquaporin 1 (AQP1) in tissues from canines with an inherited anomaly that causes their erythrocytes to have high K+. Northern blot analysis revealed abundant AQP1 expression in lung and kidney, though little expression was found in spleen. Using anti-C-terminus for dog AQP1, abundant expression was shown in kidney, trachea, and eye, but little expression was shown in pancreas and cerebrum, indicating that AQP1 expression in canine tissues is similar to that noted in other mammals

Whales as Indicators of Historical and Current Changes in the Marine Ecosystem of the Indo-Pacific Sector of the Antarctic

We review the scientific information on whales that could be indicative of historical and current changes in the ecosystem in the Indo-Pacific sector of the Antarctic. The increased krill availability in the middle of the past century as a result of the heavy harvesting of the larger baleen whale species could have been translated into better nutritional conditions for the Antarctic minke whale, resulting in a decreasing trend in the age at sexual maturity and an increasing trend in recruitment rate and hence total population size between approximately 1940 and 1970. This nutritional condition has deteriorated more recently, as revealed by a decrease in energy storage and stomach content weight since the 1980’s; these changes coincide with appreciable increases in the abundances of humpback and fin whales, which were heavily harvested in the first half of the past century. The historical demographic changes observed in the Antarctic minke whale are consistent with the pattern to be expected under the krill surplus hypothesis, with minke whales now again competing with other (recovering) baleen whale species for krill. However, these minke whales could also be using alternative feeding areas (e.g. polynias within the pack-ice) in response to the increase in abundance and geographical expansion of these other large whale species. This could provide an alternative explanation for indications from sighting surveys and population models of a decrease and then re-stabilisation of minke whale abundance in open water areas since the 1970s

The author has presented part of a collection of Olympic memorabilia relating to the 1928 Amsterdam Olympic Games. The fourth report includes the autographed photo of Mikio Oda who became the first Japanese Olympic gold medalist, the autographed writing book by Kinue Hitomi who became the first Japanese woman to be an Olympian and a medalist, and the autographed photo of Leni Junker who was a German female sprinter and won the bronzemedal in the 4×100m relay. In addition, the commemorative photo album owned by an officer of Japanese team, the Olympic memorial shield presented to Kintaro Usuda who participated in boxing, and a commemorative report on the welcome party for Olympic athletes were showed. Finally, Olympic spectators, including the media, should watch the competitions without undue expectations and pressure on athletes, reflecting on what happened when Hitomi lost in the women’s 100m semifinal

Study of LEO-SAT microwave link for broad-band mobile satellite communication system

In the field of mobile satellite communications, a system based on low-earth-orbit satellites (LEO-SAT's) such as the Iridium system has been proposed. The LEO-SAT system is able to offer mobile telecommunication services in high-latitude areas. Rain degradation, fading and shadowing are also expected to be decreased when the system is operated at a high elevation angle. Furthermore, the propagation delay generated in the LEO-SAT system is less pronounced than that in the geostationary orbit satellite (GEO-SAT) system and, in voice services, the effect of the delay is almost negligible. We proposed a concept of a broad-band mobile satellite communication system with LEO-SAT's and Optical ISL. In that system, a fixed L-band (1.6/1.5 GHz) multibeam is used to offer narrow band service to the mobile terminals in the entire area covered by a LEO-SAT and steerable Ka-band (30/20 GHz) spot beams are used for the wide band service. In this paper, we present results of a study of LEO-SAT microwave link between a satellite and a mobile terminal for a broad-band mobile satellite communication system. First, the results of link budget calculations are presented and the antennas mounted on satellites are shown. For a future mobile antenna technology, we also show digital beamforming (DBF) techniques. DBF, together with modulation and/or demodulation, is becoming a key technique for mobile antennas with advanced functions such as antenna pattern calibration, correction, and radio interference suppression. In this paper, efficient DBF techniques for transmitting and receiving are presented. Furthermore, an adaptive array antenna system suitable for this LEO-SAT is presented
