    Rapid tropicalization evidence of subtidal seaweed assemblages along a coastal transitional zone

    Anthropogenic climate change, particularly seawater warming, is expected to drive quick shifts in marine species distribution transforming coastal communities. These shifts in distribution will be particularly noticeable in biogeographical transition zones. The continental Portuguese coast stretches from north to south along 900 km. Despite this short spatial scale, the strong physical gradient intensified by the Iberian upwelling creates a transition zone where seaweed species from boreal and Lusitanian-Mediterranean origin coexist. On the northern coast, kelp marine forests thrive in the cold, nutrient-rich oceanic waters. In the south, communities resemble Mediterranean-type seaweed assemblages and are dominated by turfs. Recent evidence suggests that in these coastal areas, marine intertidal species are shifting their distribution edges as a result of rising seawater temperatures. Taking advantage of previous abundance data collected in 2012 from subtidal seaweed communities, a new sampling program was carried out in the same regions in 2018 to assess recent changes. The results confirmed the latitudinal gradient in macroalgal assemblages. More importantly we found significant structural and functional changes in a short period of six years, with regional increases of abundance of warm-affinity species, small seaweeds like turfs. Species richness, diversity, and biomass increase, all accompanied by an increase of community temperature index (CTI). Our findings suggest that subtidal seaweed communities in this transitional area have undergone major changes within a few years.     High seasonal variability in sediment carbon stocks of cold‐temperate seagrass meadows

    Seagrass meadows have a high ability to capture and store atmospheric CO2 in the plant biomass and underlying sediment and thereby function as efficient carbon sinks. The seagrass Zostera marina is a common species in the temperate Northern Hemisphere, a region with strong seasonal variations in climate. How seasonality affects carbon storage capacity in seagrass meadows is largely unknown, and therefore, in this study, we aimed to assess variations in sedimentary total organic carbon (TOC) content over a 1‐year cycle in seagrass meadows on the Swedish west coast. The TOC was measured in two Z. marina sites, one wave exposed and one sheltered, and at two depths (1.5 and 4 m) within each site, every second month from August 2015 to June 2016. We found a strong seasonal variation in carbon density, with a peak in early summer (June), and that the TOC was negatively correlated to the net community production of the meadows, presumably related to organic matter degradation. There was seasonal variation in TOC content at all sediment sections, indicating that the carbon content down to 30 cm is unstable on a seasonal scale and therefore likely not a long‐term carbon sink. The yearly mean carbon stocks were substantially higher in the sheltered meadow (3,965 and 3,465 g m−2) compared to the exposed one (2,712 and 1,054 g m−2) with similar seasonal variation.     Where the grass is greenest in seagrass seascapes depends on life history and simple species traits of fish

    Tropical seagrass meadows are critical habitats for many fish species, yet few studies have investigated the influence of multiple scale-dependent factors and marine protected areas on seagrass fish species of differing life histories. We assessed the influence of fine-scale seagrass meadow characteristics and seascape-scale variables on the abundance of fish in a seagrass-dominated seascape in the Bazaruto Archipelago, Mozambique, particularly examining patterns of nursery- vs. resident species as well as mobile- vs. sedentary species. We found that fish distribution patterns in this seagrass-dominated seascape were dependent on species' life history characteristics; nursery taxa showed lower abundance in seagrass meadows further from adult reef habitats, while resident species within seagrass meadows occurred in higher abundances far from reefs. For taxa utilizing both mangroves and seagrass meadows as nursery habitat, proximity to mangroves was an important factor. Fish abundances were generally influenced by variables at the seascape scale (km), while sedentary species were predominantly influenced by area variables, and smaller seascapes (<500 m in radius) better explained distribution patterns. The influence of marine protected areas was taxon-specific, with the strongest effects of protection on resident species. Our results indicate that protection efforts in seagrass-dominated seascapes can have varying impacts on fish distribution, depending on the life history of the species present, and the geographical placement of the reserve within the seascape. Further, we suggest that simple species attributes can be utilised to describe generalized abundance patterns of fish in seagrass seascapes

    The Diamond STING server

    Diamond STING is a new version of the STING suite of programs for a comprehensive analysis of a relationship between protein sequence, structure, function and stability. We have added a number of new functionalities by both providing more structure parameters to the STING Database and by improving/expanding the interface for enhanced data handling. The integration among the STING components has also been improved. A new key feature is the ability of the STING server to handle local files containing protein structures (either modeled or not yet deposited to the Protein Data Bank) so that they can be used by the principal STING components: (Java)Protein Dossier ((J)PD) and STING Report. The current capabilities of the new STING version and a couple of biologically relevant applications are described here. We have provided an example where Diamond STING identifies the active site amino acids and folding essential amino acids (both previously determined by experiments) by filtering out all but those residues by selecting the numerical values/ranges for a set of corresponding parameters.     Technological developments for ultra-lightweight, large aperture, deployable mirror for space telescopes

    The increasing interest on space telescopes for scientific applications leads to implement the manufacturing technology of the most critical element, i.e. the primary mirror: being more suitable a large aperture, it must be lightweight and deployable. The presented topic was originally addressed to a spaceborne DIAL (Differential Absorption LIDAR) mission operating at 935.5 nm for the measurement of water vapour profile in atmosphere, whose results were presented at ICSO 2006 and 2008. Aim of this paper is to present the latest developments on the main issues related to the fabrication of a breadboard, covering two project critical areas identified during the preliminary studies: the design and performances of the long-stroke actuators used to implement the mirror active control and the mirror survivability to launch via Electrostatic Locking (EL) between mirror and backplane. The described work is developed under the ESA/ESTEC contract No. 22321/09/NL/RA. The lightweight mirror is structured as a central sector surrounded by petals, all of them actively controlled to reach the specified shape after initial deployment and then maintained within specs for the entire mission duration. The presented study concerns: a) testing the Carbon Fiber Reinforced Plastic (CFRP) backplane manufacturing and EL techniques, with production of suitable specimens; b) actuator design optimisation; c) design of the deployment mechanism including a high precision latch; d) the fabrication of thin mirrors mock-ups to validate the fabrication procedure for the large shells. The current activity aims to the construction of an optical breadboard capable of demonstrating the achievement of all these coupled critical aspects: optical quality of the thin shell mirror surface, actuators performances and back-plane - EL subsystem functionality

    Efficacy of hyaluronic acid binding assay in selecting motile spermatozoa with normal morphology at high magnification

    <p>Abstract</p> <p>Background</p> <p>The present study aimed to evaluate the efficacy of the hyaluronic acid (HA) binding assay in the selection of motile spermatozoa with normal morphology at high magnification (8400x).</p> <p>Methods</p> <p>A total of 16592 prepared spermatozoa were selected and classified into two groups: Group I, spermatozoa which presented their head attached to an HA substance (HA-bound sperm), and Group II, those spermatozoa that did not attach to the HA substance (HA-unbound sperm). HA-bound and HA-unbound spermatozoa were evaluated according to the following sperm forms: 1-Normal morphology: normal nucleus (smooth, symmetric and oval configuration, length: 4.75+/-2.8 ÎŒm and width: 3.28+/-0.20 ÎŒm, no extrusion or invagination and no vacuoles occupied more than 4% of the nuclear area) as well as acrosome, post-acrosomal lamina, neck, tail, besides not presenting a cytoplasmic droplet or cytoplasm around the head; 2-Abnormalities of nuclear form (a-Large/small; b-Wide/narrow; c-Regional disorder); 3-Abnormalities of nuclear chromatin content (a-Vacuoles: occupy >4% to 50% of the nuclear area and b-Large vacuoles: occupy >50% of the nuclear area) using a high magnification (8400x) microscopy system.</p> <p>Results</p> <p>No significant differences were obtained with respect to sperm morphological forms and the groups HA-bound and HA-unbound. 1-Normal morphology: HA-bound 2.7% and HA-unbound 2.5% (P = 0.56). 2-Abnormalities of nuclear form: a-Large/small: HA-bound 1.6% vs. HA-unbound 1.6% (P = 0.63); b-Wide/narrow: HA-bound 3.1% vs. HA-unbound 2.7% (P = 0.13); c-Regional disorders: HA-bound 4.7% vs. HA-unbound 4.4% (P = 0.34). 3.     Kelp carbon sink potential decreases with warming due to accelerating decomposition

    Cycling of organic carbon in the ocean has the potential to mitigate or exacerbate global climate change, but major questions remain about the environmental controls on organic carbon flux in the coastal zone. Here, we used a field experiment distributed across 28° of latitude, and the entire range of 2 dominant kelp species in the northern hemisphere, to measure decomposition rates of kelp detritus on the seafloor in relation to local environmental factors. Detritus decomposition in both species were strongly related to ocean temperature and initial carbon content, with higher rates of biomass loss at lower latitudes with warmer temperatures. Our experiment showed slow overall decomposition and turnover of kelp detritus and modeling of coastal residence times at our study sites revealed that a significant portion of this production can remain intact long enough to reach deep marine sinks. The results suggest that decomposition of these kelp species could accelerate with ocean warming and that low-latitude kelp forests could experience the greatest increase in remineralization with a 9% to 42% reduced potential for transport to long-term ocean sinks under short-term (RCP4.5) and long-term (RCP8.5) warming scenarios. However, slow decomposition at high latitudes, where kelp abundance is predicted to expand, indicates potential for increasing kelp-carbon sinks in cooler (northern) regions. Our findings reveal an important latitudinal gradient in coastal ecosystem function that provides an improved capacity to predict the implications of ocean warming on carbon cycling. Broad-scale patterns in organic carbon decomposition revealed here can be used to identify hotspots of carbon sequestration potential and resolve relationships between carbon cycling processes and ocean climate at a global scale.publishedVersio

    The effects of male age on sperm analysis by motile sperm organelle morphology examination (MSOME)

    <p>Abstract</p> <p>Background</p> <p>This study aimed to investigate the influence of age on sperm quality, as analysed by motile sperm organelle morphology examination (MSOME).</p> <p>Methods</p> <p>Semen samples were collected from 975 men undergoing evaluation or treatment for infertility. Sperm cells were evaluated at 8400× magnification using an inverted microscope equipped with Nomarski (differential interference contrast) optics. Two forms of spermatozoa were considered: normal spermatozoa and spermatozoa with large nuclear vacuoles (LNV, defined as vacuoles occupying > 50% of the sperm nuclear area). At least 200 spermatozoa per sample were evaluated, and the percentages of normal and LNV spermatozoa were determined. The subjects were divided into three groups according to age: Group I, less than or equal to 35 years; Group II, 36-40 years; and Group III, greater than or equal to 41 years.</p> <p>Results</p> <p>There was no difference in the percentages of normal sperm between the two younger (I and II) groups (<it>P ></it>0.05). The percentage of normal sperm in the older group (III) was significantly lower than that in the younger (I and II) groups (<it>P </it>< 0.05). There was no difference in the percentage of LNV spermatozoa between the younger (I and II) groups (<it>P ></it>0.05). The percentage of LNV spermatozoa was significantly higher in the older group (III) than in the younger (I and II) groups (<it>P </it>< 0.05). Regression analysis demonstrated a significant decrease in the incidence of normal sperm with increasing age (<it>P </it>< 0.05; r = -0.10). However, there was a significant positive correlation between the percentage of spermatozoa with LNV and male age (<it>P </it>< 0.05, r = 0.10).</p> <p>Conclusion</p> <p>The results demonstrated a consistent decline in semen quality, as reflected by morphological evaluation by MSOME, with increased age. Considering the relationship between nuclear vacuoles and DNA damage, these age-related changes predict that increased paternal age should be associated with unsuccessful or abnormal pregnancy as a consequence of fertilisation with damaged spermatozoa. Given that sperm nuclear vacuoles can be evaluated more precisely at high magnification, these results support the routine use of MSOME for ICSI as a criterion for semen analysis.</p

    Efficacy of the motile sperm organelle morphology examination (MSOME) in predicting pregnancy after intrauterine insemination

    Background: Although the motile sperm organelle morphology examination (MSOME) was developed merely as a selection criterion, its application as a method for classifying sperm morphology may represent an improvement in the evaluation of semen quality. The aim of this study was to determine the prognostic value of normal sperm morphology using MSOME with regard to clinical pregnancy (CP) after intrauterine insemination (IUI).Methods: A total of 156 IUI cycles that were performed in 111 couples were prospectively analysed. Each subject received 75 IU of recombinant FSH every second day from the third day of the cycle. Beginning on the 10th day of the cycle, follicular development was monitored by vaginal ultrasound. When one or two follicles measuring at least 17 mm were observed, recombinant hCG was administered, and IUI was performed 12-14 h and 36-40 h after hCG treatment. Prior to the IUI procedure, sperm samples were analysed by MSOME at 8400x magnification using an inverted microscope that was equipped with DIC/Nomarski differential interference contrast optics. A minimum of 200 motile spermatozoa per semen sample were evaluated, and the percentage of normal spermatozoa in each sample was determined.Results: Pregnancy occurred in 34 IUI cycles (CP rate per cycle: 21.8%, per patient: 30.6%). Based on the MSOME criteria, a significantly higher percentage of normal spermatozoa was found in the group of men in which the IUI cycles resulted in pregnancy (2.6+/-3.1%) compared to the group that did not achieve pregnancy (1.2+/-1.7%; P = 0.019). Logistic regression showed that the percentage of normal cells in the MSOME was a determining factor for the likelihood of clinical pregnancy (OR: 1.28; 95% CI: 1.08 to 1.51; P = 0.003). The ROC curve revealed an area under the curve of 0.63 and an optimum cut-off point of 2% of normal sperm morphology. At this cut-off threshold, using the percentage of normal sperm morphology by MSOME to predict pregnancy was 50% sensitive with a 40% positive predictive value and 79% specificity with an 85% negative predictive value. The efficacy of using the percentage of normal sperm morphology by MSOME in predicting pregnancy was 65%.Conclusions: The present findings support the use of high-magnification microscopy both for selecting spermatozoa and as a routine method for analysing semen before performing IUI

    Are men universally more dismissing than women? Gender differences in romantic attachment across 62 cultural regions

Contrary to research findings previously reported in Western cultures, we found that men were not significantly more dismissing than women across all cultural regions. Gender differences in dismissing romantic attachment were evident in most cultures, but were typically only small to moderate in magnitude. Looking across cultures, the degree of gender differentiation in dismissing romantic attachment was predictably associated with sociocultural indicators. Generally, these associations supported evolutionary theories of romantic attachment, with smaller gender differences evident in cultures with high–stress and high–fertility reproductive environments. Social role theories of human sexuality received less support in that more progressive sex–role ideologies and national gender equity indexes were not cross–culturally linked as expected to smaller gender differences in dismissing romantic attachment.peer-reviewe
