    Developing research skills in training sports professionals: a reflective approach

    Sport and physical exercise are vital components of preventive measures against illnesses, promoting well-being and a high quality of life within communities. To achieve these goals, educational actions through sports practise must be implemented, considering the unique characteristics and needs of the participants. Understanding the true nature of the sports educational process requires in-depth investigation, leading to interventions that incorporate knowledge and techniques from the sports sciences. As a result, nurturing research competencies among sports professionals within their educational paths becomes crucial. This study reflects on recent methodological approaches in educational-sports research and their potential in the academic settings of Sports and Exercise Sciences degree courses. One approach that stands out is Mixed Methods Research, which combines qualitative and quantitative methods to gain a comprehensive understanding of complex phenomena in sports education. By utilising both types of data, researchers can explore the nuances of participants' experiences and gather statistical evidence to support their findings. This integrative approach allows for a more holistic analysis, strengthening the credibility and reliability of the research outcomes. Moreover, the inclusion of critical-reflexive components in educational-sports research adds depth and context to the findings. By encouraging researchers to critically examine their assumptions, biases, and positionality, this approach acknowledges the influence of personal perspectives on the research process and outcomes. It fosters a more self-aware and accountable approach to research, promoting transparency and authenticity in reporting results. The academic contexts of Sports and Exercise Sciences degree courses offer fertile ground for fostering research competencies among aspiring sports professionals. Incorporating research-oriented modules, seminars, and workshops into the curriculum can provide students with essential tools and skills for conducting rigorous and insightful research. By engaging students in hands-on research projects, they can apply theoretical knowledge to real-world situations, developing their analytical and problem-solving abilities. Integrating research competencies into the educational paths of sports professionals is vital to advancing the field of sports education. The adoption of Mixed Methods Research and critical-reflexive components enhances the quality and depth of educational-sports research. By nurturing research skills in academic settings and fostering collaboration with established researchers, we can cultivate a new generation of sports professionals who are equipped to contribute meaningfully to the well-being and development of individuals and communities through sports and physical exercise

    Biomaterials in Neurodegenerative Disorders : A Promising Therapeutic Approach

    Neurodegenerative disorders (i.e., Alzheimer's disease, Parkinson's disease, amyotrophic lateral sclerosis, and spinal cord injury) represent a great problem worldwide and are becoming prevalent because of the increasing average age of the population. Despite many studies having focused on their etiopathology, the exact cause of these diseases is still unknown and until now, there are only symptomatic treatments. Biomaterials have become important not only for the study of disease pathogenesis, but also for their application in regenerative medicine. The great advantages provided by biomaterials are their ability to mimic the environment of the extracellular matrix and to allow the growth of different types of cells. Biomaterials can be used as supporting material for cell proliferation to be transplanted and as vectors to deliver many active molecules for the treatments of neurodegenerative disorders. In this review, we aim to report the potentiality of biomaterials (i.e., hydrogels, nanoparticles, self-assembling peptides, nanofibers and carbon-based nanomaterials) by analyzing their use in the regeneration of neural and glial cells their role in axon outgrowth. Although further studies are needed for their use in humans, the promising results obtained by several groups leads us to suppose that biomaterials represent a potential therapeutic approach for the treatments of neurodegenerative disorders

    Evaluating GAIA performances on eclipsing binaries. II. Orbits and stellar parameters for V781 Tau, UV Leo and GK Dra

    The orbits and physical parameters of three close, double-lined G0 eclipsing binaries have been derived combining H_P, V_T, B_T photometry from the Hipparcos/Tycho mission with 8480-8740 \AA ground-based spectroscopy. The setup is mimicking the photometric and spectroscopic observations that should be obtained by GAIA. The binaries considered here are all of G0 spectral type, but each with its own complications: V781 Tau is an overcontact system with components of unequal temperature, UV Leo shows occasional surface spots and GK Dra contains a delta-Scuti variable. Such peculiarities will be common among binaries to be discovered by GAIA.     Optical coherence tomography features of the repair tissue following RPE tear and their correlation with visual outcomes

    To assess the optical coherence tomography (OCT) features of the repair tissue after retinal pigment epithelial (RPE) tear in neovascular age-related macular degeneration. Retrospective, observational study. Medical and imaging records of patients that developed tears after starting anti-VEGF treatment and with at least 12 months of follow-up were reviewed. OCT reflectivity of the RPE-subretinal hyperreflective tissue (SHT) complex was measured at 6, 12 and 18 months (when available). Reflectivity of the adjacent unaffected RPE-Bruch’s membrane was taken as internal reference. Other variables: grade and rip occurrence (early/late); number of intravitreal injections; type of macular neovascularization; sub-macular hemorrhage (SMH) at onset. Forty-nine eyes (age: 76.1 ± 7.0 years; VA: 0.54 ± 0.27 LogMAR) were included. Thirty-eight eyes had OCT signs of healing during the follow-up, with 21 showing SMH at baseline. Final VA positively correlated with the number of injections and negatively correlated with the RPE-SHT reflectivity and the presence of SMH (p < 0.001). Reflectivity of the RPE-SHT complex was positively associated with time and SMH at baseline (p < 0.05). In our study, most eyes showed signs of tissue repair after RPE tear. The reflectivity of repair tissue, the SMH presence and the number of anti-VEGF injections appeared to be major predictors of visual outcomes

    Lung cryobiopsy for the diagnosis of interstitial lung diseases: A series contribution to a debated procedure

    Introduction: Transbronchial cryobiopsy is an alternative to surgical biopsy for the diagnosis of fibrosing interstitial lung diseases, although the role of this relatively new method is rather controversial. Aim of this study is to evaluate the diagnostic performance and the safety of transbronchial cryobiopsy in patients with fibrosing interstitial lung disease. Materials and methods: The population in this study included patients with interstitial lung diseases who underwent cryobiopsy from May 2015 to May 2018 at the Division of Pneumology of San Giuseppe Hospital in Milan and who were retrospectively studied. All cryobiopsy procedures were performed under fluoroscopic guidance using a flexible video bronchoscope and an endobronchial blocking system in the operating room with patients under general anaesthesia. The diagnostic performance and safety of the procedure were assessed. The main complications evaluated were endobronchial bleeding and pneumothorax. All cases were studied with a multidisciplinary approach, before and after cryobiopsy. Results: Seventy-three patients were admitted to this study. A specific diagnosis was reached in 64 cases, with a diagnostic sensitivity of 88%; 5 cases (7%) were considered inadequate, 4 cases (5%) were found to be non-diagnostic. Only one major bleeding event occurred (1.4%), while 14 patients (19%) experienced mild/moderate bleeding events while undergoing bronchoscopy; 8 cases of pneumothorax (10.9%) were reported, of which 2 (2.7%) required surgical drainage. Conclusions: When performed under safe conditions and in an experienced center, cryobiopsy is a procedure with limited complications having a high diagnostic yield in fibrotic interstitial lung disease

    Doxorubicin and congo red effectiveness on prion infectivity in golden Syrian hamster

    The effect of doxorubicin and Congo Red on prion protein (PrP) infectivity in experimental scrapie was studied to better understand the effect of these compounds in prion diseases and to establish whether a dose-response correlation exists for Congo Red. This was performed in order to test the effectiveness of compounds that may easily be used in human prion diseases. Brain homogenate containing membrane bound PrPSc monomers was used as inoculum and was previously incubated with doxorubicin 10(-3) M and with increasing concentrations of Congo Red ranging from 10(-7) to 10(-2) M. This study shows for the first time that doxorubicin, and confirms that Congo Red, may interact with pathological PrP monomers modifying their infectious properties. Pre-incubation of infected brain homogenate with Congo Red resulted in prolonged incubation time and survival, independently of Congo Red concentration (p&lt;0.05). Doxorubicin and Congo Red effects do not depend upon interaction with PrP amyloid material

    Subcutaneous Adipose Tissue Transcriptome Highlights Specific Expression Profiles in Severe Pediatric Obesity: A Pilot Study

    The prevalence of pediatric obesity is rising rapidly worldwide, and "omic" approaches are helpful in investigating the molecular pathophysiology of obesity. This work aims to identify transcriptional differences in the subcutaneous adipose tissue (scAT) of children with overweight (OW), obesity (OB), or severe obesity (SV) compared with those of normal weight (NW). Periumbilical scAT biopsies were collected from 20 male children aged 1-12 years. The children were stratified into the following four groups according to their BMI z-scores: SV, OB, OW, and NW. scAT RNA-Seq analyses were performed, and a differential expression analysis was conducted using the DESeq2 R package. A pathways analysis was performed to gain biological insights into gene expression. Our data highlight the significant deregulation in both coding and non-coding transcripts in the SV group when compared with the NW, OW, and OB groups. A KEGG pathway analysis showed that coding transcripts were mainly involved in lipid metabolism. A GSEA analysis revealed the upregulation of lipid degradation and metabolism in SV vs. OB and SV vs. OW. Bioenergetic processes and the catabolism of branched-chain amino acids were upregulated in SV compared with OB, OW, and NW. In conclusion, we report for the first time that a significant transcriptional deregulation occurs in the periumbilical scAT of children with severe obesity compared with those of normal weight or those with overweight or mild obesity
