    Pseudomonas prosekii isolated in Antarctica inhibits plantpathogenic strains of Pseudomonas viridiflava and Pseudomonas fluorescens

    Pseudomonas-caused plant diseases are present worldwide and affect most of the major lineages of higher plants which, as a consequence, may result in significant economic losses. Despite the use of bacteriocins produced by rhizosphere and soil bacteria has been nowadays considered as novel crop protection approach, antagonistic interactions of cold-adapted isolates toward agriculturally important phytopathogenic bacteria have not been studied yet. In this study, we tested inhibition activity of Antarctic Pseudomonas spp. against phytopathogenic pseudomonads. Four Antarctic stains (P. prosekii CCM 8878, CCM 8879, and CCM 8881 and Pseudomonas sp. CCM 8880) inhibited several phytopathogenic strains of P. viridiflava and P. fluorescens. Based on inhibition zone character and previous genome research we suggest that L-pyocin activity was responsible for this effect against P. viridiflava strains and that tailocin inhibited P. fluorescens isolate

    Agapé: Moral Attitudes through the Prism of the Movie Subtitles

    We deal with the terms subtitling, guilt and innocence. Moreover, we describe the influence that the translator of the subtitles can have on our/spectator’s opinion about moral attitudes in film. In this article we do not describe the visual part of the film or the sound, we focus our attention only on the text (oral or written). We demonstrate it by comparing two sets of Czech subtitles of the Croatian movie Agape (2017, Schmidt). The first set was made with an intermediator (via English subtitles), while the second one was made without it. We point out that the language based misunderstanding can cause a misleading perception of the original message of the movie, and in the described case it can influence our perception of guilt or innocence of the main character

    Screening of growth phases of Antarctic algae and cyanobacteria cultivated on agar plates by chlorophyll fluorescence imaging

    Recently, chlorophyll fluorescence imaging is frequently used non-invasive method to monitor the metabolic state and photosynthetic activities of vascular plants and other autotrophic organisms. In our study, we used the measurements of chlorophyll fluorescence kinetics to follow the development of culture of Antarctic algae (Macrochloris rubrioleum, Zygnema sp.) and cyanobacteria (Hassalia antarctica, Nostoc commune). On the cultures grown on agar plates, Bold´s Basal Medium (BBM), slow Kautsky kinetics supplemented with saturation pulses were measured repeatedly in a week interval. On the kinetics, typical points (OPSMT) were distinguished and species-specific and time of cultivation-dependent differences in shape of the OPSMT kinetics evaluated. We tested sensitivity of various chlorophyll fluorescence parameters to cultivation time on agar plates. In the algae, the most pronounced changes were the decrease in maximum quantum yield of photosystem II (FV/FM) and quenching of basal chlorophyll fluorescence qF0 (M. rubrioleum, Zygnema sp.). In cyanobacteria, chlorophyll fluorescence parameters did not show clear trends with the time of cultivation. F0 quenching (qF0) reached positive values in H. antarctica, while it was negative in N. commune. In both cases, however, qF0 showed an increase with cultivation time. The differences are discussed as well as the potential of the emerging area of the application of chlorophyll fluorescence imaging for evaluation of photosynthetic performance of algal/cyanobacterial cultures on agar plates

    Vitality and growth rate of agar plate-cultivated Antarctic microautotrophs: Analysis of PSII functioning by chlorophyll fluorescence parameters

    In our study, we focused on the growth of three different microautotrophs isolated from Antarctic lichens (Placopsis contortuplicata, Solorina spongiosa) and cryptoendolithic algal vegetation. The isolates were purified and inoculated on agar plates, Bold´s Basal Medium (BBM). The growth of the cultures and the markers of physiological (photosynthetic) activity were monitored by chlorophyll fluorescence in 1 week intervals for 3 months after inoculation. For the assessment of photosynthetic activity, the method of slow Kautsky kinetics supplemented with saturation pulses was applied. Four chlorophyll fluorescence parameters calculated: (1) maximum quantum yield of PSII  (FV/FM), (2) effective quantum yield of photosynthetic processes in PSII (ΦPSII), (3) non-photochemical quenching of chlorophyll fluorescence, and (4) background chlorophyll fluorescence ratio (F0/F0´). Troughout the cultivation period, the maximum quantum yield of PSII (FV/FM) showed high values in all three autotrophs with only slight increase in the first part of the cultivation period, followed by slight decrease in the second part. The ΦPSII values showed a rapid decline within the first 4 weeks of cultivation followed by more or less constant values in the isolates from P. contortuplicata and cryptoendolithic alga. Contrastingly, time course of ΦPSII rather showed an increase followed by a decrease in S. spongiosa isolate. NPQ (related to the activation of protective mechanisms) increased in the second part of cultivation period, the rate of increase and maximum values were species-specific. The species-specific differences in chlorophyll fluorescence parameters are discussed as well as their potential for evaluation of photosynthetic performance of in vitro cultivated algal/cyanobacterial cultures on agar plates

    Urinary tract infections caused by Pseudomonas aeruginosa present a serious complication in the context of urinary catheter use. P. aeruginosa are equipped with various virulence determinants that may result in increased resistance. That contributes to higher morbidity and mortality in vulnerable patients. Pyocins are proteinaceous agents produced by pseudomonads with an antimicrobial effect targeting closely related bacteria. In this study, we examined the inhibition interactions among a set of 135 P. aeruginosa clinical isolates originated from the catheter-associated urinary tract infections. Togerther with pyocinogeny, the co-occurrence of other virulence factors and their co-association with pyocins were examined. The strong antagonistic interactions and high prevalence of pyocin genes among the isolates were observed. The overall competitive interactions provide promising findings in the area of alternative antibiotic approaches

    Cílem práce tedy je pokusit se aplikovat historické, kulturní a etnografické poznatky o Chodsku do výuky matematiky. Stěžejní náplní bude vlastní tvorba matematických úloh využívajících poznatky o chodském regionu, jejich praktická realizace v edukačním procesu a následná evaluace, zejména reflexe ze strany žáků zapojených do výuky. V teoretické části diplomové práce jsou zmiňovány motivace a interdisciplinarita jako důležité složky procesu výuky a zároveň představen region Chodsko. Charakterizuji historii této etnografické oblasti a popisuji současnou podobu (včetně detailního popisu jedenácti vesnic tvořící Chodsko), tradice a zvyky. V praktické části je prezentována realizace nevržených matematických úloh ve všech ročnících prvního stupně Základní školy Horšovský Týn, kde momentálně vyučuji. Úlohy vymýšlím tak, aby děti zaujaly a bavily. Mým cílem je také vymyslet úkoly, pomocí kterých si děti nejen zopakují matematiku, ale rovněž si prohloubí a zafixují vlastivědné učivo, a navíc získají nové poznatky o regionu, který se nachází poblíž jejich domovů, a dodnes ovlivňuje kulturu celého Domažlicka.ObhájenoThe main aim of the diploma thesis was to design teaching units of mathematics, in which there will be a noticeable overlap with other subjects of compulsory school attendance. The lessons are connected with local history, specifically with knowledge of the historical ethnographic area around the town of Domažlice called Chodsko. My work has two parts. The theoretical part contains knowledge about motivation and intersubject relationships, at the same time the region of Chodsk is examined here from a historical and contemporary point of view. Here I also cover culture, interesting places we can visit, famous people who are connected to this place, and also sports. Theoretical knowledge is used in the practical part and when designing mathematical problems. The aim of the practical part is to design, implement and describe in detail mathematics lessons. There are a total of 5 teaching units for all grades of the first grade

Jitky Kučerové, která předmět Výchova ke zdraví vyučuje. Výsledná práce se zaměřuje na předání informací o komplexním zdraví žákům ve věku 14-15 let. Součástí práce jsou také doprovodné plakáty a animace, které podporují celkovou prezentaci projektu. V práci je popsána podrobná rešerše daného tématu, tvůrčí záměr a průběh zpracovávání nové vizuální podoby. Výsledná práce je na ukázkách zasazena do reálného prostředí. Výsledkem je vizuální styl plakátů, programové brožury a webové bannery.
