    Serum Dioxin-like Activity Is Associated with Reproductive Parameters in Young Men from the General Flemish Population

    BACKGROUND: 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) and some related environmental contaminants are aryl hydrocarbon receptor (AhR) ligands that exert reproductive and developmental toxicity in laboratory animals. In humans, fertility-related effects are less documented. OBJECTIVE: The aim of this study was to investigate the relationship between dioxin-like biological activity in serum and parameters of reproductive status in men from the general population 5 months after a polychlorinated biphenyl and dioxin food-contamination episode in Belgium. DESIGN: In the framework of the cross-sectional Flemish Environment and Health Study (FLEHS), we recruited 101 men 20–40 years of age and evaluated sperm parameters, measured sex hormones, and gathered information on a number of lifestyle factors. In addition, we determined the AhR-mediated enzymatic response elicited by individual serum samples and expressed it as TCDD equivalent concentrations (CALUX-TEQs) using an established transactivation assay. RESULTS: Age (p = 0.04) and the frequency of fish (p = 0.02) and egg (p = 0.001) consumption were independent positive determinants of serum dioxin-like activity. After correcting for possible confounders, we found that a 2-fold increase in CALUX-TEQ > 16 pg/L was associated with a 7.1% and 6.8% (both p = 0.04) decrease in total and free testosterone, respectively. We also observed a more pronounced drop in semen volume of 16.0% (p = 0.03), whereas sperm concentration rose by 25.2% (p = 0.07). No relationship was found with total sperm count or sperm morphology. CONCLUSIONS: These data suggest an interaction of dioxin-like compounds with the secretory function of the seminal vesicles or prostate, possibly indirectly through an effect on testosterone secretion, at levels not affecting spermatogenesis as such

    Stargardt disease:monitoring incidence and diagnostic trends in the Netherlands using a nationwide disease registry

    PURPOSE: To assess the incidence of Stargardt disease (STGD1) and to evaluate demographics of incident cases. METHODS: For this retrospective cohort study, demographic, clinical and genetic data of patients with a clinical diagnosis of STGD1 were registered between September 2010 and January 2020 in a nationwide disease registry. Annual incidence (2014-2018) and point prevalence (2018) were assessed on the basis of this registry. RESULTS: A total of 800 patients were registered, 56% were female and 83% were of European ancestry. The incidence was 1.67-1.95:1,000,000 per year and the point prevalence in 2018 was approximately 1:22,000-1:19,000 (with and without 10% of potentially unregistered cases). Age at onset was associated with sex (p = 0.027, Fisher's exact); 1.9x more women than men were observed (140 versus 74) amongst patients with an age at onset between 10 and 19 years, while the sex ratio in other age-at-onset categories approximated one. Late-onset STGD1 (≥45 years) constituted 33% of the diagnoses in 2014-2018 compared to 19% in 2004-2008. Diagnostic delay (≥2 years between the first documentation of macular abnormalities and diagnosis) was associated with older age of onset (p = 0.001, Mann-Whitney). Misdiagnosis for age-related macular degeneration (22%) and incidental STGD1 findings (14%) was common in patients with late-onset STGD1. CONCLUSION: The observed prevalence of STGD1 in real-world data was lower than expected on the basis of population ABCA4 allele frequencies. Late-onset STGD1 was more frequently diagnosed in recent years, likely due to higher awareness of its phenotype. In this pretherapeutic era, mis- and underdiagnosis of especially late-onset STGD1 and the role of sex in STGD1 should receive special attention

    With increasing dependency on fossil fuels, biomass is a key to ensure the security of supply of sustainable energy. Biomass currently accounts for around half (44-65%) of all renewable energy in the EU, meeting 6% of the EU’s energy needs. Based on the Renewable Energy Action Plans for NWE Member States in 2010, it is estimated that biomass production for energy in NWE could increase by 69% to 2020, but only if significant improvements to the supply chain are made, e.g. through the energetic use of currently disposed waste streams or the sustainable production of biomass on non-agricultural land

    Degradation of branched octylphenol was studied in a bacterial culture of a Sphingomonas sp. strain. Octylphenol is considered to be the most stable degradation intermediate formed from the corresponding nonionic octylphenol polyethoxylates surfactants during biological wastewater treatment. Since octylphenol can exert estrogenic effects in wildlife, a detailed study of its biodegradation is warranted. The aerobic microbiological transformation of octylphenol was examined with and without the addition of the easily assimilable sodium acetate. In both cases the formation of the metabolite 2,4,4-trimethyl-2-pentanol, representing the intact alkyl chain as a tertiary alcohol, was observed. Since the octylphenol degradation rate was not affected by the presence of acetate, this strain did not show any diauxic metabolic behaviour when incubated with octylphenol and sodium acetate as the sources of carbon and energy. As a result of the biotransformation of octylphenol, its estrogenic potency was removed because it is the phenolic moiety that interacts with the estrogen receptors. This feature opens perspectives for the use of this strain in the framework of an adequate treatment of wastewater with high levels of alkylphenol polyethoxylates
