    Skrining antikanker menggunakan metode BST (Brine Shrimp Lethality Test) pada ekstrak metanol daun saga (Abrus precatorius L.) dengan partisi etanol

    Latar belakang : Pengembangan senyawa antikanker dari bahan alami perlu dilakukan untuk meminimalisir efek samping dari penanganan penyakit kanker yang saat ini banyak dilakukan. Suatu senyawa dapat diketahui potensi antikankernya dengan pendekatan menggunakan metode BST (Brine Shrimp Lethality Test). Ekstrak metanol daun saga (Abrus precatorius L.) bersifat toksik terhadap larva A. salina Leach.   Tujuan : Mengetahui bagian teraktif dari ekstrak metanol daun saga yang dipartisi (dipisahkan) menggunakan pelarut etanol.   Metode : Ekstrak metanol daun saga dipartisi dengan pelarut etanol hingga terbentuk bagian larut dan tidak larut. Kedua bagian tersebut diuji menggunakan metode BST dengan 5 kali ulangan dan 3 replikasi yang masing-masing menggunakan 10  ekor larva A.salina.   Hasil : Dari perhitungan didapatkan nilai LC50 bagian larut etanol sebesar 144,544 ppm sedangkan nilai LC50 bagian tidak larut etanol sebesar 151,356 ppm.   Kesimpulan : Bagian larut etanol menyebabkan persentase kematian yang lebih tinggi daripada bagian tidak larut. Nilai LC50 bagian larut etanol lebih rendah dibanding bagian tidak larutnya. Semakin rendah nilai LC50 senyawa, maka semakin berpotensi pula untuk dikembangkan sebagai agen antikanker.   Kata kunci : toksisitas, Abrus precatorius L., Artemia salina Leach., antikanker, partisi.&nbsp

    Dampak dari mengonsumsi kopi instan dengan kadar gula yang tinggi akan menyebabkan peningkatan kadar glukosa dalam darah dan membuat hepar bekerja lebih keras untuk memetabolismenya. Metabolisme yang tidak sempurna dapat menyebabkan kerusakan sel-sel hepar. Tujuan penelitian ini untuk mengetahui pengaruh kopi instan tinggi gula dosis bertingkat terhadap gambaran histologi hepar tikus putih (Rattus norvegicus) galur wistar. Penelitian ini dilakukan secara eksperimental dari bulan April-Juli 2020. Sebanyak 24 ekor tikus putih (Rattus norvegicus) dibagi menjadi 4 kelompok perlakuan. Hasil penelitian dianalisis dengan uji Chi Square. Hasil penelitian diketahui adanya hubungan antara dosis kopi instan tinggi gula dengan kerusakan histologi hepar p<0,000. Terdapat perbedaan gambaran histologi hepar tikus antara kelompok kontrol dengan kelompok perlakuan yang diberikan dosis 4,5 ml/250 g BB/hari, 9 ml/250 g BB/hari dan 13,5 ml/250 g BB/hari. Hasil ini dipengaruhi oleh faktor penggangu cacing Taenia taeniaeformis pada organ hepar. Kata Kunci : kopi instan, gula, histologi, hepa

    Apple Juice to Reduce Total Cholesterol Level in Women Over 40 Years Old

    Cholesterol is a fatty substance in the human body’s cells. A person who has high cholesterol levels may develop coronary heart disease (CHD), which increases their risk of developing atherosclerosis. Due to pregnancy, hormonal, and menopausal reasons, women over the age of 40 are more likely to have elevated cholesterol levels than younger women. Apples' fiber content has been shown to reduce blood cholesterol levels. This study aimed to determine the effect of apple juice on cholesterol levels.  A pretest-posttest group design with a pre-experimental methodology was used in this study. 38 women between the ages of 40 and 66 made up the sample of this study. Every morning for seven days, respondents received apple juice. The study was carried out in August 2020. According to this study, apple juice has a positive effect in lowering total cholesterol levels. Prior to receiving apple juice, the mean total cholesterol level was 226.68 17.230 mg/dL, while the mean cholesterol level after apple juice treatment was 184.39 13.771 mg/dL. Total cholesterol levels before and after receiving apple juice are known to differ by 42.29 mg/dL. The Paired T-Test analysis of the data revealed a value of P = 0.000 (P 0.05), indicating that there was an effect of giving apple juice on lowering respondents' total cholesterol levels.Cholesterol is a fatty substance in the human body’s cells.     Limbah B3 adalah sisa suatu usaha dan/atau kegiatan yang mengandung B3. Beberapa unsur yang  masuk dalam kategori limbah B3 adalah logam berat seperti timbal (Pb), tembaga (Cu), cadmium (Cd), merkuri (Hg), besi (Fe), dan yang lainnya. Timbal merupakan racun lingkungan multitarget yang mampu menyebabkan berbagai penyakit akut atau kronis pada manusia. Penelitian ini dilakukan secara observasional deskriptif untuk mengetahui kadar timbal (Pb) dan tembaga (Cu) pada air sumur dan urine masyarakat yang tinggal di sekitar tempat pengolahan limbah Bahan Berbahaya dan Beracun (B3) Desa Karangdawa Kabupaten Tegal. Hasil analisis kualitatif pada sampel air sumur dan urine dari semua sampel adalah negatif mengandung Pb, namun hasil analisis kuantitatif sampel urine menggunakan Atomic Absorbtion Spectrophotometer (AAS) adalah semua sampel memiliki kadar Pb dalam urine di atas nilai normal yang ditentukan oleh Kementerian Kesehatan yaitu sebesar ≤ 0,15 mg/L.   Kata Kunci: Air Sumur, Timbal (Pb), Urin

    Deteksi Endoparasit Cacing pada Hepar Tikus Laboratorium (Rattus norvegicus) dari Sentra Peternak di Kabupaten Banyumas dan Kabupaten Purbalingga

    The presence of laboratory rats that are maintained and bred for laboratory purposes or laboratory observations is very necessary. The presence of endoparasite in laboratory rats will have an impact on the result of the research or laboratory observations. This study aims to detect helminth endoparasites in the liver of laboratory rats (Rattus norvegicus) from animal breeders in Banyumas and Purbalingga Districts. This research was an observational study with a cross-sectional design. A total of 52 laboratory rats were used in the study. Rats are killed with chloroform, liver surgery then identify the presence of worm larvae. Out of the 52 rats obtained, 7 (29.17%) from 24 laboratory rats in Banyumas District and 5 (17.86%) from 28 laboratory rats in Purbalingga District were infected with Taenia taeniaeformis. It is necessary to control helminth infections in laboratory rats, such as laboratory animal quarantine, health monitoring, and antihelmintic treatment. It is important to handle carefully during travel to assure the results of research or laboratory observations using the animals

    The development of transportation technology has resulted an increase of public need for economic activities in the form of vehicles. This condition causes an increase in the number of workshops to repair and maintain vehicles. Workshop activities result in waste containing lead (Pb) such as used lubricating oil (oil), gasoline, and used batteries. Lead is a heavy metal that is classified as a Hazardous and Toxic Material (B3). It can be toxic to the human body when it binds to erythrocytes, it will also inhibit heme synthesis which results in decreased production of hemoglobin (Hb). The objective of this study was to determine the correlation between blood lead (Pb) levels and hemoglobin in motorcycle workshop mechanics in Purwokerto. The type of this research is analytic observational using a case control study research design with a sample of 15 respondents obtained using purposive sampling method. The examination of metals in the blood using the Atomic Absorption Spectrophotometer (AAS), while hemoglobin using a Hematology Analyzer (HA). The results of the study indicated that low lead levels were 0 g/dl and the high lead levels were 7.7 g/dl., while low hemoglobin levels were 12.7 g/dl and high lead levels were 16.2 g/dl. The results of the analysis using the correlation spearmen  test obtained a significant value (p) = 0.833 and a correlation value (r) = 0.060. The conclusion is that Ho is accepted, so there is no correlation between blood lead levels and hemoglobin levels in motorcycle workshop mechanics in Purwokert

    Klebsiella pneumoniae is a gram-negative rod-shaped bacterium that is a member of the Enterobacteriaceae group of bacteria and is pathogenic and can infect humans. Currently, many K. pneumoniae bacteria are resistant to antibiotics, giving rise to new problems in the health sector that must be addressed immediately by utilizing biodiversity in the form of medicinal plants such as coconut trees, which can produce coconut shells that can be made into charcoal. The results of the phytochemical analysis show that coconut shell charcoal contains many active compounds that have been used in the cosmetic and health industries. The purpose of this study was to obtain an optimal and effective coconut shell charcoal extract gel formula for inhibiting the growth of K. pneumoniae bacteria. This study included a true experimental post-only control group with different gel concentration groups, namely 3%, 6%, and 9%. The optimal formulation of coconut shell charcoal extract gel is a gel with a concentration of 9%, and the results of the one-way ANOVA statistical analysis show that the treatment of the inhibition test on K. pneumoniae bacteria from gels with different concentrations shows that there is a mean difference in the area of the inhibition zone with a significant value of 0.033 (p<0.05). The results of the LSD post-hoc test showed that several treatment groups had significant values (p <0.05). Based on the results of the study, it can be concluded that the optimal formulation of shell charcoal extract gel is a gel with a concentration of 9%, while the most effective gel for inhibiting the growth of K. pneumoniae bacteria is a gel with a concentration of 3%

    Staphylococcus aureus is a species of bacteria that is often identified as a source of infection and has increased the number of cases of resistance. Resistance to Penicillin-class antibiotics in S. aureus is associated with plasmids containing the blah-Z gene encoding ß-Lactamase. Some factors that can be modified to obtain optimal test results in PCR are annealing temperature and primary concentration. This study aims to identify the primary concentration and annealing temperature that produce PCR products with the best band in detecting the blaZ gene in Metichilin Resistant Staphylococcus aureus (MRSA) bacteria. The research design used used experimental methods. The results of the study obtained were PCR products at primary concentrations of 10 μM were clearly visible, thick and clean, compared to primary concentrations of 5 μM. Annealing temperature with the best band results at temperatures of 61.8 °C and 58.7°C.  This study concluded the primary concentration to obtain a clearly visible band result of 10 μM. The annealing temperature of 61.8°C produced the best band in the sample code R.PG. U4, R.V.U2, and R.151.U2 temperatures. While the sample code R.IGD. U5 band results best at annealing temperature 58.7°C
