    A pimenta-do-reino Piper nigrum é um dos principais produtos agrícolas do Pará gerando sustentabilidade para uma porcentagem considerável dos agricultores do estado. A Embrapa (PA) desenvolve trabalhos com a espécie como uma coleção de germoplasma. Com o objetivo de conhecer os potenciais recursos genéticos da espécie, foi sequenciado o transcriptoma de P. nigrum. Uma plataforma de sequenciamento de nova geração foi usada para gerar as sequencias dos genes e o trabalho foi analisado com recursos de bioinformática.     Defining best practices in the management of geothermal exploration data

    International audienceThe objective of this work is to define best practices in the management of geothermal exploration data. This study builds on a questionnaire to survey the geothermal data management practices in mature geothermal markets. The inquiry targeted public Regulatory entities with overview of geothermal resources as well as public and private developers. Topics covered in the questionnaire range from the country status to the database set up. The questionnaire focused on the specifications, usage and investments required for installing/maintaining information systems capable of managing exploration data. In addition, information on the different regulatory frameworks and company policies for managing/sharing exploration data has been gathered to identify the requirements imposed on the design of information systems. The responses were analyzed to identify commonalities in data management practices. They reveal that installing an Information System (IS) is the best practice to systematically and securely manage exploration data. They also provide recommendations with respect to the regulatory framework, data types, data collection methodologies, data storage, data quality control, data accessibility and dissemination, IS architecture, financial investments and human resources required to develop a state-of-the art IS. These results will guide the design of future technical assistance programs for beneficiaries of World Bank support to geothermal exploration activities and it is our belief that they will be beneficial for the geothermal sector at large

    Neste trabalho nós extraímos o RNA total e isolamos o mRNA de raízes de Piper nigrum L. infectada com o fungo Fusarium solani f.sp. piperis na fase mais severa da infecção, para construção de uma biblioteca de fragmentos utilizando o método RNA-Seq. A biblioteca gerada foi sequenciada utilizando a plataforma de nova geração ?SOLiDTM 3 Plus System? (Applied Biosystems, EUA) produzindo 67.452.415 leituras com 50 pb. As leituras obtidas foram pré-processadas com softwares de Bioinformática para obtenção de leituras de alta qualidade. Para avaliar os dados pré-processados estatisticamente, foi utilizado o software FASTQC. Os resultados obtidos foram leituras com alta qualidade que serão usadas no processo de montagem, a fim de obter uma lista de transcritos de Planta expressos em resposta a infecção pelo patógeno

    First genomic microsatellite markers developed for Platonia insignis (Clusiaceae), a Brazilian fruit tree.

    Platonia insignis is a fruit tree native of Brazil with allogamous and asexual reproduction. The production of fruits is mainly obtained by exploitation of natural populations and the impact of genetic structuring on plant production may be evaluated. For this purpose, codominant and multiallelic markers such as microsatellite are the most suitable, but they need to be developed for this species. Thus, the aim of this work was to develop and validate microsatellite markers for P. insignis. We used Roche 454 GS FLX sequencing platform of a single P. insignis genotype and 1702 microsatellite sequences were identified. Based on some pre-requisites, we could develop 50 primer pairs to be tested. Twenty-two primer pairs successfully amplified fragments and they were tested in 31 genotypes of P. insignis that belong to a germplasm bank and were sampled in the northeast of Pará State, Brazil. Thirteen primers were polymorphic and the number of alleles per loci varied from 5 (PI18 and PI27) to 2 (PI08, PI25, PI31, PI33 and PI 37). Expected heterozygosity (HE) varied from 0.74 (PI27) to 0.12 (PI31)b and observed heterozygosity (HO) varied from 1.00 (PI25) to 0.00 (PI08, PI31, PI33 and PI37). Principal coordinates could separate the genotypes of P. insignis in clusters and we can conclude that the primers can estimate the genetic diversity of P. insignis populations.Short Comunication

    Distinct triterpene synthases in the laticifers of Euphorbia lathyris

    Abstract Euphorbia lathyris was proposed about fifty years ago as a potential agroenergetic crop. The tremendous amounts of triterpenes present in its latex has driven investigations for transforming this particular biological fluid into an industrial hydrocarbon source. The huge accumulation of terpenes in the latex of many plant species represent a challenging question regarding cellular homeostasis. In fact, the enzymes, the mechanisms and the controllers that tune the amount of products accumulated in specialized compartments (to fulfill ecological roles) or deposited at important sites (as essential factors) are not known. Here, we have isolated oxidosqualene cyclases highly expressed in the latex of Euphorbia lathyris. This triterpene biosynthetic machinery is made of distinct paralogous enzymes responsible for the massive accumulation of steroidal and non-steroidal tetracyclic triterpenes. More than eighty years after the isolation of butyrospermol from shea butter (Heilbronn IM, Moffet GL, and Spring FS J. Chem. Soc. 1934, 1583), a butyrospermol synthase is characterized in this work using yeast and in folia heterologous expression assays

    Ultra-Deep Sequencing Reveals the microRNA Expression Pattern of the Human Stomach

    Background: While microRNAs (miRNAs) play important roles in tissue differentiation and in maintaining basal physiology, little is known about the miRNA expression levels in stomach tissue. Alterations in the miRNA profile can lead to cell deregulation, which can induce neoplasia. Methodology/Principal Findings: A small RNA library of stomach tissue was sequenced using high-throughput SOLiD sequencing technology. We obtained 261,274 quality reads with perfect matches to the human miRnome, and 42% of known miRNAs were identified. Digital Gene Expression profiling (DGE) was performed based on read abundance and showed that fifteen miRNAs were highly expressed in gastric tissue. Subsequently, the expression of these miRNAs was validated in 10 healthy individuals by RT-PCR showed a significant correlation of 83.97% (P<0.05). Six miRNAs showed a low variable pattern of expression (miR-29b, miR-29c, miR-19b, miR-31, miR-148a, miR-451) and could be considered part of the expression pattern of the healthy gastric tissue. Conclusions/Significance: This study aimed to validate normal miRNA profiles of human gastric tissue to establish a reference profile for healthy individuals.     Mapping the geometry of volcanic systems with magnetotelluric soundings: Results from a land and marine magnetotelluric survey performed during the 2018–2019 Mayotte seismovolcanic crisis

    A major seismovolcanic crisis has afflicted the islands of Mayotte, Comoros Archipelago, since May 2018, although the origin is debated. Magnetotellurics (MT), which is sensitive to hydrothermal and/or magmatic fluids and can map the subsurface electrical resistivity structure, can provide insight by revealing the internal structure of the volcanic system. In this paper, we report the results of a preliminary land and shallow marine MT survey performed on and offshore the island nearest the crisis. The 3D inversion-derived electrical resistivity model suggests that the island is underlain by a shallow ~500-m-thick conductive layer atop a deeper, more resistive layer, possibly associated with a high-temperature geothermal system. At depths of ~15 km, the resistivity drops by almost two orders of magnitude, possibly due to partial melting. Further petrophysical and geophysical studies are underway for confirmation and to map the geometry and evolution of the volcanic system
