What can legacy datasets tell us about soil quality trends? Soil acidity in Victoria
Purpose-built soil monitoring networks have been established in many countries to identify where soil functionality is threatened and to target remediation initiatives. An alternative to purpose-built soil monitoring networks is to use legacy soils information. Such information yields almost instant assessments of soil change but the results should be interpreted with caution since the information was not collected with monitoring in mind. We assess the threat of soil acidification in Victoria using two legacy datasets: (i) the Victorian Soils Information System (VSIS) which is a repository of the results of soil analyses conducted for scientific purposes since the 1950s and (ii) a database of 75 000 routine soil test results requested by farmers between 1973 and 1993. We find that the VSIS measurements are clustered in space and time and are therefore suitable for local rather than broad-scale assessments of soil change. The farmers' results have better spatial and temporal coverage and space-time models can be used to quantify the spatial and temporal trends in the pH measurements. However, careful validation of these findings is required since we do not completely understand how the measured paddocks were selected and we cannot be certain that sampling or laboratory protocols have not changed with time
Assessment of endocytic traffic and ocrl function in the developing zebrafish neuroepithelium
Endocytosis allows cells to internalise a wide range of molecules from their environment and to maintain their plasma membrane composition. It is vital during development and for maintenance of tissue homeostasis. The ability to visualise endocytosis in vivo requires suitable assays to monitor the process. Here, we describe imaging-based assays to visualise endocytosis in the neuroepithelium of living zebrafish embryos. Injection of fluorescent tracers into the brain ventricles followed by live imaging was used to study fluid-phase or receptor-mediated endocytosis, for which we used receptor-associated protein (RAP, encoded by Lrpap1) as a ligand for low-density lipoprotein receptor-related protein (LRP) receptors. Using dual-colour imaging combined with expression of endocytic markers, it is possible to track the progression of endocytosed tracers and to monitor trafficking dynamics. Using these assays, we reveal a role for the Lowe syndrome protein Ocrl in endocytic trafficking within the neuroepithelium. We also found that the RAP-binding receptor Lrp2 (encoded by lrp2a) appears to contribute only partially to neuroepithelial RAP endocytosis. Altogether, our results provide a basis to track endocytosis within the neuroepithelium in vivo and support a role for Ocrl in this process

Enzymatic Synthesis of Diverse Heterocycles by a Noncanonical Nonribosomal Peptide Synthetase
Nonribosomal peptide synthetases (NRPSs) are typically multimodular enzymes that assemble amino acids or carboxylic acids into complex natural products. Here, we characterize a monomodular NRPS, PvfC, encoded by the Pseudomonas virulence factor (pvf) gene cluster that is essential for virulence and signaling in different bacterial species. PvfC exhibits a unique adenylation-thiolation-reductase (ATR) domain architecture that is understudied in bacteria. We show that the activity of PvfC is essential in the production of seven leucine-derived heterocyclic natural products, including two pyrazines, a pyrazinone, and a rare disubstituted imidazole, as well as three pyrazine N-oxides that require an additional N-oxygenation step. Mechanistic studies reveal that PvfC, without a canonical peptide-forming domain, makes a dipeptide aldehyde intermediate en route to both the pyrazinone and imidazole. Our work identifies a novel biosynthetic route for the production of pyrazinones, an emerging class of signaling molecules and virulence factors. Our discovery also showcases the ability of monomodular NRPSs to generate amino acid- and dipeptide-aldehydes that lead to diverse natural products. The diversity-prone biosynthesis by the pvf-encoded enzymes sets the stage for further understanding the functions of pvf in bacterial cell-to-cell signaling
The Gaseous Environments of Radio Galaxies
X-ray emission traces the gaseous environments of radio sources. The medium
must be present for jet confinement, but what are its influence on jet
fuelling, dynamics, propagation, and disruption? The observational situation is
both complicated and enriched by radio sources being multi-component X-ray
emitters, with several possible regions of non-thermal emission. Recent work,
primarily based on sensitive ROSAT pointings, is used to contrast the X-ray
emission and environments of radio sources with (a) low power, (b) high power
at high redshift, (c) high power at lower redshift, and (d) GHz peaked spectrum
emission. The trends in external gas density and pressure near extended radio
structures are reviewed. Imminently-available X-ray measurements with vastly
improved resolution and sensitivity have great potential for resolving many

Unveiling Soft Gamma-Ray Repeaters with INTEGRAL
Thanks to INTEGRAL's long exposures of the Galactic Plane, the two brightest
Soft Gamma-Ray Repeaters, SGR 1806-20 and SGR 1900+14, have been monitored and
studied in detail for the first time at hard-X/soft gamma rays.
This has produced a wealth of new scientific results, which we will review
here. Since SGR 1806-20 was particularly active during the last two years, more
than 300 short bursts have been observed with INTEGRAL. and their
characteristics have been studied with unprecedented sensitivity in the 15-200
keV range. A hardness-intensity anticorrelation within the bursts has been
discovered and the overall Number-Intensity distribution of the bursts has been
determined. In addition, a particularly active state, during which ~100 bursts
were emitted in ~10 minutes, has been observed on October 5 2004, indicating
that the source activity was rapidly increasing. This eventually led to the
Giant Flare of December 27th 2004, for which a possible soft gamma-ray (>80
keV) early afterglow has been detected.
The deep observations allowed us to discover the persistent emission in hard
X-rays (20-150 keV) from 1806-20 and 1900+14, the latter being in a quiescent
state, and to directly compare the spectral characteristics of all Magnetars
(two SGRs and three Anomalous X-ray Pulsars) detected with INTEGRAL.Comment: 8 pages, 7 figures, Presented at the conference "Isolated Neutron

Systematic Cu-63 NQR studies of the stripe phase in La(1.6-x)Nd(0.4)Sr(x)CuO(4) for 0.07 <= x <= 0.25
We demonstrate that the integrated intensity of Cu-63 nuclear quadrupole
resonance (NQR) in La(1.6-x)Nd(0.4)Sr(x)CuO(4) decreases dramatically below the
charge-stripe ordering temperature T(charge). Comparison with neutron and X-ray
scattering indicates that the wipeout fraction F(T) (i.e. the missing fraction
of the integrated intensity of the NQR signal) represents the charge-stripe
order parameter. The systematic study reveals bulk charge-stripe order
throughout the superconducting region 0.07 <= x <= 0.25. As a function of the
reduced temperature t = T/T(charge), the temperature dependence of F(t) is
sharpest for the hole concentration x=1/8, indicating that x=1/8 is the optimum

Recent glitches detected in the Crab pulsar
From 2000 to 2010, monitoring of radio emission from the Crab pulsar at
Xinjiang Observatory detected a total of nine glitches. The occurrence of
glitches appears to be a random process as described by previous researches. A
persistent change in pulse frequency and pulse frequency derivative after each
glitch was found. There is no obvious correlation between glitch sizes and the
time since last glitch. For these glitches and
span two orders of magnitude. The pulsar suffered the
largest frequency jump ever seen on MJD 53067.1. The size of the glitch is
6.8 Hz, 3.5 times that of the glitch occured in
1989 glitch, with a very large permanent changes in frequency and pulse
frequency derivative and followed by a decay with time constant 21 days.
The braking index presents significant changes. We attribute this variation to
a varying particle wind strength which may be caused by glitch activities. We
discuss the properties of detected glitches in Crab pulsar and compare them

Genomic Deletion Marking an Emerging Subclone of Francisella tularensis subsp. holarctica in France and the Iberian Peninsula
P. 7465-7470Francisella tularensis subsp. holarctica is widely disseminated in North America and the boreal and temperate
regions of the Eurasian continent. Comparative genomic analyses identified a 1.59-kb genomic deletion specific
to F. tularensis subsp. holarctica isolates from Spain and France. Phylogenetic analysis of strains carrying this
deletion by multiple-locus variable-number tandem repeat analysis showed that the strains comprise a highly
related set of genotypes, implying that these strains were recently introduced or recently emerged by clonal
expansion in France and the Iberian PeninsulaS
Optical Spectra of SNR Candidates in NGC 300
We present moderate-resolution (<5A) long-slit optical spectra of 51 nebular
objects in the nearby Sculptor Group galaxy NGC 300 obtained with the 2.3 meter
Advanced Technology Telescope at Siding Spring Observatory, Australia. Adopting
the criterion of [SII]/Ha>=0.4 to confirm supernova remnants (SNRs) from
optical spectra, we find that of 28 objects previously proposed as SNRs from
optical observations, 22 meet this criterion with six showing [SII]/Ha of less
than 0.4. Of 27 objects suggested as SNRs from radio data, four are associated
with the 28 previously proposed SNRs. Of these four, three (included in the 22
above) meet the criterion. In all, 22 of the 51 nebular objects meet the
[SII]/Ha criterion as SNRs while the nature of the remaining 29 objects remains
