    Nationwide Seroprevalence Survey of Angiostrongylus vasorum-Derived Antigens and Specific Antibodies in Dogs from Colombia

    Angiostrongylus vasorum is a cardiopulmonary nematode, causing several clinical manifestations in dogs, e.g., severe respiratory signs, coagulopathy, and gastrointestinal or neurological signs. In the last decades, this parasite has been described to spread and emerge in Europe and North America. Scant studies on A. vasorum occurrence in South America exist. Recently, A. vasorum was detected in gastropod intermediate hosts in Colombia, where data on definitive host prevalence, e.g., dogs and wild canids, are still limited. Therefore, the sera of 955 dogs, varying in age and breed from seven different departments all over Colombia, were collected and analysed for A. vasorum antigens and parasite-specific antibodies by ELISA. In total, 1.05 % (n = 10; 95 % CI 0.40–1.69) of the samples were antigen-positive and 2.62 % (n = 25; 95 % CI 1.61–3.63) were antibody-positive. These results confirm the presence of A. vasorum in Colombia, although positive results in antigen and antibody reactions in the same dog were not detected.     Prevalence, associated factors and outcomes of pressure injuries in adult intensive care unit patients: the DecubICUs study

Epidemiologic data is however unavailable. We aimed to provide an international picture of the extent of pressure injuries and factors associated with ICU-acquired pressure injuries in adult ICU patients. Methods: International 1-day point-prevalence study; follow-up for outcome assessment until hospital discharge (maximum 12 weeks). Factors associated with ICU-acquired pressure injury and hospital mortality were assessed by generalised linear mixed-effects regression analysis. Results: Data from 13,254 patients in 1117 ICUs (90 countries) revealed 6747 pressure injuries; 3997 (59.2%) were ICU-acquired. Overall prevalence was 26.6% (95% confidence interval [CI] 25.9–27.3). ICU-acquired prevalence was 16.2% (95% CI 15.6–16.8). Sacrum (37%) and heels (19.5%) were most affected. Factors independently associated with ICU-acquired pressure injuries were older age, male sex, being underweight, emergency surgery, higher Simplified Acute Physiology Score II, Braden score 3 days, comorbidities (chronic obstructive pulmonary disease, immunodeficiency), organ support (renal replacement, mechanical ventilation on ICU admission), and being in a low or lower-middle income-economy. Gradually increasing associations with mortality were identified for increasing severity of pressure injury: stage I (odds ratio [OR] 1.5; 95% CI 1.2–1.8), stage II (OR 1.6; 95% CI 1.4–1.9), and stage III or worse (OR 2.8; 95% CI 2.3–3.3). Conclusion: Pressure injuries are common in adult ICU patients. ICU-acquired pressure injuries are associated with mainly intrinsic factors and mortality. Optimal care standards, increased awareness, appropriate resource allocation, and further research into optimal prevention are pivotal to tackle this important patient safety threat

    Previous studies have demonstrated the existence and expression of genes essential to the process of protein ubiquitination in Giardia intestinalis, indicating that the ubiquitin-proteasome system may be involved in the degradation of proteins during its life cycle of the parasite. In this study, purification of ubiquitin was conducted from protein extracts of G. intestinalis trophozoites. Then, an anti-ubiquitin specific antibody was obtained to standardize an assay for the detection and evaluation of ubiquitination patterns. Finally, HSP90 was identified as an ubiquitinated protein in this protozoan. This post-translational modification could have regulatory effects associated with the functionality of the protein or its turnover to regulate key molecular events during the parasite’s life cycle.Estudios previos han demostrado la existencia y expresión de genes esenciales para el proceso de ubiquitinación de proteínas en Giardia intestinalis, indicando que el sistema ubiquitina-proteosoma puede estar involucrado en el proceso de degradación de proteínas de este parásito durante su ciclo de vida. En el presente trabajo se realizó la purificación de ubiquitina a partir de extractos proteicos de trofozoítos de G. intestinalis, se produjo un anticuerpo anti-ubiquitina específico que permitió la estandarización de un ensayo para la detección y evaluación de los patrones de ubiquitinación, y se identificó la HSP90 como una proteína ubiquitinada en este protozoario. Esta modificación post-transduccional puede tener efectos regulatorios asociados con la funcionalidad de la proteína o con el recambio para regular eventos moleculares claves durante el ciclo de vida del parásito.Estudos anteriores demonstraram a existência e expressão de genes essenciais para o processo de ubiquitinação de proteínas em Giardia intestinalis, indicando que o sistema ubiquitina-proteassoma podem estar envolvidos na degradação de proteínas do parasita durante seu ciclo de vida.     Cross-sectional study of 13 Leptospira serovars in cows in a Colombian dairy region

    Background: Leptospirosis in cattle is recognized as a major cause of reproductive loses due to abortions, early embryonic death, and infertility. The type of Leptospira serovars that are currently circulating in Colombian herds is largely unknown due to the lack of national reports. Objective: The prevalence of antibodies specific to 13 Leptospira spp serovars was investigated in unvaccinated cows from 26 herds in San Pedro de los Milagros (Province of Antioquia) dairy region. Methods: Microscopic agglutination test was used on serum samples, following WHO recommendations. Results: A total of 112/900 (12.4%) of the animals had antibody titers of 1:100 or greater to at least one Leptospira serovar. Of those positive, 34/900 (3,8%) animals seroconverted to two or more serovars. In order of decreasing prevalence, the serovars detected were: hardjo prajitno, pomona, grippotyphosa, tarassovi, copenhageni, canicola, cellodeni and bratislava. There were 28 animals seropositive to serovar hardjo. Conclusion: Our results indicate that leptospirosis should be included as a differential diagnosis for various reproductive problems in this region.Antecedentes: Aleptospirose em bovinos é reconhecida como uma das principais causas de perdas reprodutivas devido a abortos, morte embrionária precoce e infertidade. Os sorotipos de Leptospira que estão atualmente em circulação no rebanho colombiano não se conhecem devido à falta de reportes nacionais. Objetivo: Neste estúdio foi investigada a prevalência de anticorpos específicos para 13 sorovares de Leptospira spp em vacas não vacinadas de 26 rebanhos leiteiros da região de San Pedro, Antioquia. Métodos: Foi empregado o teste de aglutinação microscópica (MAT) em amostras de soro de acordo com as recomendações da OMS. Resultados: Um total de 112/900 (12,4%) animais apresentaram títulos de anticorpos de 1:100 ou superiores para pelo menos um sorotipo de Leptospira. Dos animais positivos, 34/900 (3,8%) soroconverteram para dois ou mais sorotipos. A fim de diminuir a prevalência, foram detectados os sorotipos: hardjo, prajitno, pomona, grippotyphosa, tarassovi, copenhageni, canicola, cellodeni e Bratislava. Um total de28 animais foram soropositivos para o sorotipo hardjo. Conclusão: Os resultados indicam que a leptospirose deveria ser incluída nos diagnósticos diferenciais para o diagnóstico diferencial de problemas em doenças reprodutivas nos bovinos de Antioquia.Antecedentes: La leptospirosis es la mayor causal de pérdidas reproductivas debidas a abortos, muerte embrionaria temprana e infertilidad. Se desconocen los serovares de Leptospira que actualmente se encuentran circulando en el rebaño colombiano debido a la falta de reportes nacionales. Objetivo: Se investigó la prevalencia de anticuerpos específicos para 13 serovares de Leptospira spp en vacas no vacunadas de 26 rebaños en la región lechera de San Pedro de los Milagros (Provincia de Antioquia).     Reduced Efficacy of Commercial Acaricides against Populations of Resistant Cattle Tick Rhipicephalus microplus from Two Municipalities of Antioquia, Colombia

    Two distant Antioquian cattle farms where systemic and topical acaricides had previously failed to control infestations by Rhipicephalus ( Boophilus) microplus were studied. An initial in vivo study was conducted using single subcutaneous injections with a long-acting formulation of ivermectin (630 μg/kg). Injections were made at 3-month intervals on animals at each farm to evaluate the therapeutic and persistent efficacy of ivermectin against R. microplus. Body tick counts and reproductive parameters of semi- or fully engorged females ≥5 mm) were assessed at 10-day intervals, and since no negative control group could be included, values were compared against those for day 0. Although there was an overall reduction of 50%-75% in tick numbers that persisted for 30-40 days, it was not significantly different at one of the farms and not enough to afford protection from severe infestations. The engorgement weight and egg mass weight of ticks from treated animals were significantly lower throughout the 50-day posttreatment period. Egg hatch was not significantly reduced posttreatment and remained at levels of 80%-90%. A random selection of 9 out of 28 commercial formulations of ivermectin sold in Colombia were analyzed by High Performance Liquid Chromatography (HPLC). All were within the expected labeled concentration (∓15% deviation) of 1% and 3.15% ivermectin except for one. A popular unregistered injectable widely used in both farms and labeled as “natural pyrethrin”, was found to contain 10.5% ivermectin. An adult immersion test was conducted to evaluate the efficacy of topical acaricides to recommended concentrations of five commercial products and/or their combinations. Efficacy was determined by comparing the reproductive index of each treated group to that of the control group. Cypermethrin (150 ppm) was completely ineffective at both farms. Amitraz (208 ppm) exhibited low and intermediate efficacies of 14% and 56%. The combination of amitraz (100 ppm) and cypermethrin (150 ppm) was less efficacious than the amitraz alone. A generic product based on amitraz + citronella (208 ppm + 10 ppm, respectively) was shown to be less efficacious than the name-brand amitraz product. Products containing the organophosphate chlorpyrifos or trichlorfon exhibited intermediate efficacies of approximately 60% at the Tarso farm. We conclude that at these two locations, there is a high degree of resistance to many of the acaricides available in Colombia and confirm suspicions that ivermectin is no longer able to eliminate tick infestations

    Aelurostrongylus abstrusus Infections in Domestic Cats (Felis silvestris catus) from Antioquia, Colombia

    Although Aelurostrongylus abstrusus infections in domestic cats (Felis silvestris catus) have sporadically been reported in Colombia, there is still no data available on epidemiology nor on the biology of this neglected lungworm parasite. Thus, this epidemiological study aimed to evaluate the occurrence of patent A. abstrusus infections in domestic cats from the Colombian Federal State of Antioquia. In total, 473 fecal samples of indoor/outdoor domestic cats were collected and analyzed thereafter by the Baermann funnel migration technique for the presence of A. abstrusus first stage larvae 1 (L1). The occurrence of A. abstrusus was confirmed in 0.4% (2/473) of investigated cats. Due to the presence of patent A. abstrusus infections in investigated cats, it is unfailing to include this lungworm within differential diagnoses of feline pulmonary disorders. Despite the fact that the Baermann funnel technique is currently the cheapest and the gold standard diagnostic tool for feline aelurostrongylosis, this technique is still unknown by Colombian veterinary surgeons and rarely utilized in small animal veterinary clinics. The current survey intends to generate awareness on this neglected parasitosis and to be considered as a baseline study for future surveys monitoring feline aelurostrongylosis not only in domestic/stray cats but also in endemic wild felid species of Colombia

    Detección y caracterización del virus de la anemia infecciosa aviar en Colombia

    Objetivo. identificar el virus de la anemia infecciosa aviar (chicken anemia virus, CAV) en granjas avícolas y aves de traspatio en Antioquia, Colombia. Materiales y métodos. Se tomaron muestras de sangre y plumas de gallinas ponedoras; en cada granja se eligieron tres aves de seis edades diferentes (1, 15, 30, 60, 90 y 120 días de edad). También se obtuvieron muestras de aves de traspatio ubicadas cerca de las granjas estudiadas. Se realizó ELISA y PCR para el análisis de las muestras. Resultados. Mediante PCR, el 84% de las aves resultaron positivas al CAV en sangre total y el 66% en muestras de plumas. El 60% de las aves de traspatio dieron positivo en sangre y el 40% en folículo de pluma. Mediante ELISA, el 22% de las aves de las granjas avícolas presentó títulos de anticuerpos altos y el 19% moderados. En las aves de traspatio, el 43% presentó títulos de anticuerpos altos y 29% moderados. Además, los resultados de la prueba de RFLP y la secuenciación mostraron que el virus circulante encontrado en este estudio era diferente del de la cepa vacunal Cux-1 utilizada en el país. Conclusiones. El CAV está presente en Colombia en aves comerciales como de traspatio. Según los hallazgos, un alto porcentaje de las aves dieron positivo para la detección viral, aunque el número de aves positivas por anticuerpos fue bajo. Se requiere determinar las características del virus circulante para explicar la respuesta de anticuerpos obtenida.Objective. identify the presence of chicken anemia virus (CAV) in poultry farms and backyard chickens from Antioquia, Colombia. Materials and Methods. Blood and feather samples were taken from laying chickens; in each farm, three birds of six different ages (1, 15, 30, 60, 90, and 120 days old) were chosen randomly. Backyard chicken samples were also obtained near the research farms. We used serology and molecular techniques to analyze the samples. Results. By serology, the 22% of the poultry farm birds presented high antibody titers and 19% moderate antibody titers. In the backyard chickens, 43% of them presented high antibody titers and 29% moderate antibody titers. In addition, results from the RFLP test and sequencing showed that the circulating virus found in this study was different from the Cux-1 vaccine strain used in Colombia. Conclusions. CAV is present in Colombia in both commercial and backyard chickens. According to the findings, a high percentage of the birds tested positive for viral detection, whereas the number of birds that tested positive for antibodies was low. Thus, the characteristics of the circulating virus need to be determined to explain the antibody response observed in this study
