    DEFYING CONVENTION: ATYPICAL PERSPECTIVES OF SLAVERY IN ANTEBELLUM NEW ORLEANS

    During the first half of the nineteenth century, slavery became a vital economic component upon which the success of the southern states in America rested. Cotton was king, and slavery was the peculiar institution that ensured its dominance in the domestic and international markets of America. Popular portrayals, however, often neglect the complicated dynamics of American slavery and instead depict the institution in simplistic terms. The traditional view has emphasized an image of white southerners as slaveholders and blacks as slaves. In New Orleans, the lives of three menā€”all of whom were tied to slavery in varying capacitiesā€”reveal a much more nuanced picture of American slavery. John McDonogh, a white slaveowner and member of the American Colonization Society (ACS), proposed an emancipation plan to his slaves by which they could gradually purchase their freedom on the condition that, once freed, they were repatriated to Liberia. Andrew Durnford, a homme de couleur libre (free man of color) and slaveowner, was a business associate and friend of McDonogh who showed little to no interest in emancipating his slaves. Washington Watts McDonogh, a college-educated former slave of John McDonogh, was a minister in Liberia who supported the ACS repatriation plan at a time when many free blacks in America did not support the African colonization movement, preferring to remain in the United States. The experiences of three men reveal how slavery in nineteenth-century New Orleans was a much more nuanced institution that did not resemble the traditional narrative that the public has come to know

    Long-term propagule pressure overwhelms early community determination of invader success in a serpentine grassland

    The role of plant diversity in reducing invasions has generated decades of debate. Diverse communities might be more resistant to invasion because the communities contain resident species that are functionally similar to the invader (limiting similarity/sampling effect), or the residents use the range of available resources more effectively (complementarity) than single species. However, the resistance of diverse communities to invasion appears to decline with increasing spatial and temporal scale, in a phenomenon called the ā€œinvasion paradox.ā€ I addressed two groups of hypotheses related to this paradox, broadly that: (1) functional diversity and functional identity resist invasion initially, via complementarity or limiting similarity; and (2) disturbance and propagule pressure interact to weaken the effects of functional diversity and identity on invader success through time. Using long-term data from experimental assemblages in a serpentine grassland of California, I examined the abundance of a high impact invader, yellow starthistle (Centaurea solstitialis), in response to functional diversity (functional dispersion), functional dissimilarity, extent of pocket gopher disturbance, and propagule pressure. Neither diversity, nor dissimilarity, nor disturbance significantly impacted the success of C. solstitialis at any point in the study. Instead, propagule pressure was the single most important predictor of C. solstitialis abundance. These findings suggest that continued dispersal may eventually outweigh community resistance when considering a longer-term view of invasion. Thus, conservation efforts focused on preventing the introduction and establishment of invasive species should recognize that even diverse communities are susceptible to invasion over time

    Land use influences disease emergence by changing the ecological dynamics of humans, wildlife, domestic animals, and pathogens. This is a central tenet of One Health, and one that is gaining momentum in wildlife management decision-making in the United States. Using almost 2000 serological samples collected from non-native wild pigs (Sus scrofa) throughout Florida (U.S.), we compared the prevalence and exposure risk of two directly transmitted pathogens, pseudorabies virus (PrV) and Brucella spp., to test the hypothesis that disease emergence would be positively correlated with one of the most basic wildlife management operations: Hunting. The seroprevalence of PrV-Brucella spp. coinfection or PrV alone was higher for wild pigs in land management areas that allowed hunting with dogs than in areas that culled animals using other harvest methods. This pattern did not hold for Brucella alone. The likelihood of exposure to PrV, but not Brucella spp., was also significantly higher among wild pigs at hunted sites than at sites where animals were culled. By failing to consider the impact of dog hunting on the emergence of non-native pathogens, current animal management practices have the potential to affect public health, the commercial livestock industry, and wildlife conservation

    Quantification of Proteins Using Peptide Immunoaffinity Enrichment Coupled with Mass Spectrometry

    There is a great need for quantitative assays in measuring proteins. Traditional sandwich immunoassays, largely considered the gold standard in quantitation, are associated with a high cost, long lead time, and are fraught with drawbacks (e.g. heterophilic antibodies, autoantibody interference, 'hook-effect').1 An alternative technique is affinity enrichment of peptides coupled with quantitative mass spectrometry, commonly referred to as SISCAPA (Stable Isotope Standards and Capture by Anti-Peptide Antibodies).2 In this technique, affinity enrichment of peptides with stable isotope dilution and detection by selected/multiple reaction monitoring mass spectrometry (SRM/MRM-MS) provides quantitative measurement of peptides as surrogates for their respective proteins. SRM/MRM-MS is well established for accurate quantitation of small molecules 3, 4 and more recently has been adapted to measure the concentrations of proteins in plasma and cell lysates.5-7 To achieve quantitation of proteins, these larger molecules are digested to component peptides using an enzyme such as trypsin. One or more selected peptides whose sequence is unique to the target protein in that species (i.e. "proteotypic" peptides) are then enriched from the sample using anti-peptide antibodies and measured as quantitative stoichiometric surrogates for protein concentration in the sample. Hence, coupled to stable isotope dilution (SID) methods (i.e. a spiked-in stable isotope labeled peptide standard), SRM/MRM can be used to measure concentrations of proteotypic peptides as surrogates for quantification of proteins in complex biological matrices. The assays have several advantages compared to traditional immunoassays. The reagents are relatively less expensive to generate, the specificity for the analyte is excellent, the assays can be highly multiplexed, enrichment can be performed from neat plasma (no depletion required), and the technique is amenable to a wide array of proteins or modifications of interest.8-13 In this video we demonstrate the basic protocol as adapted to a magnetic bead platform

    Dispersal and Land Cover Contribute to Pseudorabies Virus Exposure in Invasive Wild Pigs

    We investigated the landscape epidemiology of a globally distributed mammal, the wild pig (Sus scrofa), in Florida (U.S.), where it is considered an invasive species and reservoir to pathogens that impact the health of people, domestic animals, and wildlife. Specifically, we tested the hypothesis that two commonly cited factors in disease transmission, connectivity among populations and abundant resources, would increase the likelihood of exposure to both pseudorabies virus (PrV) and Brucella spp. (bacterial agent of brucellosis) in wild pigs across the Kissimmee Valley of Florida. Using DNA from 348 wild pigs and sera from 320 individuals at 24 sites, we employed population genetic techniques to infer individual dispersal, and an Akaike information criterion framework to compare candidate logistic regression models that incorporated both dispersal and land cover composition. Our findings suggested that recent dispersal conferred higher odds of exposure to PrV, but not Brucella spp., among wild pigs throughout the Kissimmee Valley region. Odds of exposure also increased in association with agriculture and open canopy pine, prairie, and scrub habitats, likely because of highly localized resources within those land cover types. Because the effect of open canopy on PrV exposure reversed when agricultural cover was available, we suggest that small-scale resource distribution may be more important than overall resource abundance. Our results underscore the importance of studying and managing disease dynamics through multiple processes and spatial scales, particularly for non-native pathogens that threaten wildlife conservation, economy, and public health

    Social Networks and Friendships at School: Comparing Children With and Without ASD

    Self, peer and teacher reports of social relationships were examined for 60 high-functioning children with ASD. Compared to a matched sample of typical children in the same classroom, children with ASD were more often on the periphery of their social networks, reported poorer quality friendships and had fewer reciprocal friendships. On the playground, children with ASD were mostly unengaged but playground engagement was not associated with peer, self, or teacher reports of social behavior. Twenty percent of children with ASD had a reciprocated friendship and also high social network status. Thus, while the majority of high functioning children with ASD struggle with peer relationships in general education classrooms, a small percentage of them appear to have social success

    Should we welcome robot teachers?

    Abstract Current uses of robots in classrooms are reviewed and used to characterise four scenarios: (s1) Robot as Classroom Teacher; (s2) Robot as Companion and Peer; (s3) Robot as Care-eliciting Companion; and (s4) Telepresence Robot Teacher. The main ethical concerns associated with robot teachers are identified as: privacy; attachment, deception, and loss of human contact; and control and accountability. These are discussed in terms of the four identified scenarios. It is argued that classroom robots are likely to impact childrenā€™sā€™ privacy, especially when they masquerade as their friends and companions, when sensors are used to measure childrenā€™s responses, and when records are kept. Social robots designed to appear as if they understand and care for humans necessarily involve some deception (itself a complex notion), and could increase the risk of reduced human contact. Children could form attachments to robot companions (s2 and s3), or robot teachers (s1) and this could have a deleterious effect on their social development. There are also concerns about the ability, and use of robots to control or make decisions about childrenā€™s behaviour in the classroom. It is concluded that there are good reasons not to welcome fully fledged robot teachers (s1), and that robot companions (s2 and 3) should be given a cautious welcome at best. The limited circumstances in which robots could be used in the classroom to improve the human condition by offering otherwise unavailable educational experiences are discussed
