    Rhodobacter johrii sp. nov., an endospore-producing cryptic species isolated from semi-arid tropical soils

    An oval to rod shaped phototrophic purple nonsulfur bacterium, strain JA192T was isolated from an enrichment culture of a pasteurized rhizosphere soil sample of jowar crop collected from Godumakunta village, near Hyderabad, India. Strain JA192T is Gram-negative, motile and produces endospores. Phylogenetic analysis on the basis of 16S rRNA gene sequences showed that the strain JA192T is closely related to Rhodobacter sphaeroides DSM 158T (99.9% sequence similarity), Rhodobacter megalophilus JA194T (99.8%) and Rhodobacter azotoformans JCM 9340T (98.1%) and clusters with other species of the genus Rhodobacter of the family Rhodobacteraceae. However, DNA-DNA hybridization with Rba. sphaeroides DSM 158T, Rba. megalophilus JA194T and Rba. azotoformans JCM 9320T showed a relatedness of only 38-57% with respect to JA192T. On the basis of 16S rRNA gene sequence analysis, DNA-DNA hybridization data, morphological, physiological and chemotaxanomic characters, strain JA192T represents a novel species of the genus Rhodobacter, for which the name Rhodobacter johrii sp. nov. is proposed.     Microbacterium invictum sp. nov., isolated from homemade compost

    Strain DC-200T was isolated from homemade compost produced from kitchen refuse and characterized using a polyphasic approach. The isolate was a Gram-positive motile short rod, facultatively aerobic, catalase-positive and oxidase-negative, and was able to grow at 10–37 6C, pH 6.0–9.5 and with up to 5% of NaCl. The peptidoglycan was of the type B1 alpha and the muramic acid residues were glycolylated. The major fatty acids were anteiso-C15 : 0 and anteiso- C17 : 0. The predominant respiratory menaquinones were MK-11 and MK-12. The G+C content of the genomic DNA was 70 mol%. Based on the analysis of the 16S rRNA gene sequence, the closest phylogenetic neighbours of strain DC-200T were Microbacterium lacus A5E-52T (98.7 %) and Microbacterium aoyamense KV-492T (98.2 %). The phenetic characterization of the isolate supports its inclusion within the genus Microbacterium; however, its distinctive phenotypic features and the results from the 16S rRNA gene sequence analysis and the DNA–DNA hybridization study suggest that the isolate represents a novel species. The name Microbacterium invictum sp. nov. is proposed. The type strain is DC-200T (5DSM 19600T5LMG 24557T)

    Complete genome sequence and description of Salinispira pacifica gen. nov., sp. nov., a novel spirochaete isolated form a hypersaline microbial mat

    During a study of the anaerobic microbial community of a lithifying hypersaline microbial mat of Lake 21 on the Kiritimati atoll (Kiribati Republic, Central Pacific) strain L21-RPul-D2(T) was isolated. The closest phylogenetic neighbor was Spirochaeta africana Z-7692(T) that shared a 16S rRNA gene sequence identity value of 90% with the novel strain and thus was only distantly related. A comprehensive polyphasic study including determination of the complete genome sequence was initiated to characterize the novel isolate. Cells of strain L21-RPul-D2(T) had a size of 0.2 - 0.25 x 8-9 mu m, were helical, motile, stained Gram-negative and produced an orange carotenoid-like pigment. Optimal conditions for growth were 35 degrees C, a salinity of 50 g/l NaCl and a pH around 7.0. Preferred substrates for growth were carbohydrates and a few carboxylic acids. The novel strain had an obligate fermentative metabolism and produced ethanol, acetate, lactate, hydrogen and carbon dioxide during growth on glucose. Strain L21-RPul-D2(T) was aerotolerant, but oxygen did not stimulate growth. Major cellular fatty acids were C-14:0, iso-C-15:0, C-16:0 and C-18:0. The major polar lipids were an unidentified aminolipid, phosphatidylglycerol, an unidentified phospholipid and two unidentified glycolipids. Whole cell hydrolysates contained L-ornithine as diagnostic diamino acid of the cell wall peptidoglycan. The complete genome sequence was determined and annotated. The genome comprised one circular chromosome with a size of 3.78 Mbp that contained 3450 protein-coding genes and 50 RNA genes, including 2 operons of ribosomal RNA genes. The DNA G + C content was determined from the genome sequence as 51.9 mol%. There were no predicted genes encoding cytochromes or enzymes responsible for the biosynthesis of respiratory lipoquinones. Based on significant differences to the uncultured type species of the genus Spirochaeta, S. plicatilis, as well as to any other phylogenetically related cultured species it is suggested to place strain L21-RPul-D2(T) (=DSM 27196(T) = JCM 18663(T)) in a novel species and genus, for which the name Salinispira pacifica gen. nov., sp. nov. is proposed

    Paenibacillus tylopili sp.nov., a chitinolytic bacterium isolated from the mycorhizosphere of Tylopilus felleus

    Two chitinolytic bacterial strains (designated MK2T and V7) were isolated from the mycorhizosphere of the fungus Tylopilus felleus. The strains were facultatively anaerobic G+ endospore formers. Physiological analysis and 16S rRNA gene PCR-RFLP assays revealed nearly identical profiles for both strains, demonstrating their relationship at the species level. Sequences specific for the genus Paenibacillus were found within the 16S rRNA gene sequence of the strain MK2T. The 16S rRNA gene sequence showed the highest similarity to the sequences of Paenibacillus amylolyticus, P. pabuli and P. xylanilyticus. DNA-DNA relatedness of the strain with the type strain of P. amylolyticus was 4.95 %, of P. pabuli 38.0 %, and of P. xylanilyticus 46.3 %, indicating no relatedness between MK2T and any of them at the species level. The most abundant fatty acids in strains MK2T and V7 were anteiso-C15:0, iso-C16:0, iso-C15:0 and n-C16:0. DNA-DNA relatedness, morphological, physiological and chemotaxonomic analyses, and phylogenetic data based on 16S rRNA gene sequencing made it possible to describe both strains as the novel species of the genus Paenibacillus, for which the name Paenibacillus tylopili is proposed, the type strain being MK2T (DSM 18927T, LMG 23975T)
