    Practice pointer: Using the new UK-WHO growth charts

    The new UK growth charts for children aged 0-4 years (designed using data from the new WHO standards) describe the optimal pattern of growth for all children, rather than the prevailing pattern in the UK (as with previous charts). The new charts are suitable for all ethnic groups and set breast feeding as the norm. UK children match the new charts well for length and height, but after age 6 months fewer children will be below the 2nd centile for weight or show weight faltering, and more will be above the 98th centile. The new charts look different: they have a separate preterm section, no lines between 0 and 2 weeks, and the 50th percentile is no longer emphasised. The charts give clear instructions on gestational correction, and there is a new chart for infants born before 32 weeks’ gestation. The instructions advise on when and how to measure and when a measurement or growth pattern is outside the normal range. The charts include a “look-up” tool for determining the body mass index centile from height and weight centiles without calculation and aid for predicting adult height.     Maximum Running Speed of Captive Bar-Headed Geese Is Unaffected by Severe Hypoxia

    While bar-headed geese are renowned for migration at high altitude over the Himalayas, previous work on captive birds suggested that these geese are unable to maintain rates of oxygen consumption while running in severely hypoxic conditions. To investigate this paradox, we re-examined the running performance and heart rates of bar-headed geese and barnacle geese (a low altitude species) during exercise in hypoxia. Bar-headed geese (n = 7) were able to run at maximum speeds (determined in normoxia) for 15 minutes in severe hypoxia (7% O2; simulating the hypoxia at 8500 m) with mean heart rates of 466±8 beats min�1. Barnacle geese (n = 10), on the other hand, were unable to complete similar trials in severe hypoxia and their mean heart rate (316 beats.min�1) was significantly lower than bar-headed geese. In bar-headed geese, partial pressures of oxygen and carbon dioxide in both arterial and mixed venous blood were significantly lower during hypoxia than normoxia, both at rest and while running. However, measurements of blood lactate in bar-headed geese suggested that anaerobic metabolism was not a major energy source during running in hypoxia. We combined these data with values taken from the literature to estimate (i) oxygen supply, using the Fick equation and (ii) oxygen demand using aerodynamic theory for bar-headed geese flying aerobically, and under their own power, at altitude. This analysis predicts that the maximum altitude at which geese can transport enough oxygen to fly without environmental assistance ranges from 6,800 m to 8,900 m altitude, depending on the parameters used in the model but that such flights should be rare

    Cross-talk between human airway epithelial cells and 3T3-J2 feeder cells involves partial activation of human MET by murine HGF

    There is considerable interest in the ex vivo propagation of primary human basal epithelial stem/progenitor cells with a view to their use in drug development, toxicity testing and regenerative medicine. These cells can be expanded in co-culture with mitotically inactivated 3T3-J2 murine embryonic feeder cells but, similar to other epithelial cell culture systems employing 3T3-J2 cells, the aspects of cross-talk between 3T3-J2 cells and human airway basal cells that are critical for their expansion remain largely unknown. In this study, we investigated secreted growth factors that are produced by 3T3-J2 cells and act upon primary human airway basal cells. We found robust production of hepatocyte growth factor (HGF) from fibroblast feeder cells following mitotic inactivation. Consistent with the limited cross-species reactivity of murine HGF on the human HGF receptor (MET; HGFR), MET inhibition did not affect proliferative responses in human airway basal cells and HGF could not replace feeder cells in this culture system. However, we found that murine HGF is not completely inactive on human airway epithelial cells or cancer cell lines but stimulates the phosphorylation of GRB2-associated-binding protein 2 (GAB2) and signal transducer and activator of transcription 6 (STAT6). Although HGF induces phosphorylation of STAT6 tyrosine 641 (Y641), there is no subsequent STAT6 nuclear translocation or STAT6-driven transcriptional response. Overall, these findings highlight the relevance of cross-species protein interactions between murine feeder cells and human epithelial cells in 3T3-J2 co-culture and demonstrate that STAT6 phosphorylation occurs in response to MET activation in epithelial cells. However, STAT6 nuclear translocation does not occur in response to HGF, precluding the transcriptional activity of STAT6

    Nuclear Star Clusters across the Hubble Sequence

    Over the last decade, HST imaging studies have revealed that the centers of most galaxies are occupied by compact, barely resolved sources. Based on their structural properties, position in the fundamental plane, and spectra, these sources clearly have a stellar origin. They are therefore called ``nuclear star clusters'' (NCs) or ``stellar nuclei''. NCs are found in galaxies of all Hubble types, suggesting that their formation is intricately linked to galaxy evolution.     A technological framework for data-driven IoT systems: application on landslide monitoring

    The emergence of the paradigm of the Internet of Things has underpinned the development of data-driven cyber-physical systems that collect and process data that is dense both in space and time. The application areas of such data-driven IoT systems are numerous and their socio-economic impact of great importance as they enable the monitoring and management of processes in sectors ranging from urban management to management of the natural environment. In this work, we introduce and detail an end-to-end technological framework for data-driven IoT systems for landslide monitoring. The framework is articulated in three tiers – namely data acquisition, data curation and data presentation. For each tier we present and detail its design and development aspects; from the IoT hardware design and the wireless communication technologies of choice, to how Big Data infrastructure and Machine Learning components can be combined to support a sophisticated presentation tier that delivers the true added value of a system to its final users. The framework is validated, extended and fine-tuned by means of two pilots at locations experiencing the impact of different landslide types and activity. This work qualitatively improves upon existing methods of landslide monitoring and showcases how data-driven IoT systems can pave new pathways for interdisciplinary research as well as generate positive impact on modern societies

    Protease-activated receptor 4 protects mice from Coxsackievirus B3 and H1N1 influenza A virus infection

    PAR4 is expressed by a variety of cells, including platelets, cardiac, lung and immune cells. We investigated the contribution of PAR4 to viral infections of the heart and lung. Toll-like receptor (TLR) 3-dependent immune responses were analyzed after co-stimulation of PAR4 in murine bone-marrow derived macrophages, embryonic fibroblasts and embryonic cardiomyocytes. In addition, we analyzed Coxsackievirus B3 (CVB3) or H1N1 influenza A virus (H1N1 IAV) infection of PAR4−/− (ΔPAR4) and wild-type (WT) mice. Lastly, we investigated the effect of platelet inhibition on H1N1 IAV infection. In vitro experiments revealed that PAR4 stimulation enhances the expression of TLR3-dependent CXCL10 expression and decreases TLR3-dependent NFκB-mediated proinflammatory gene expression. Furthermore, CVB3-infected ΔPAR4 mice exhibited a decreased anti-viral response and increased viral genomes in the heart leading to more pronounced CVB3 myocarditis compared to WT mice. Similarly, H1N1 IAV-infected ΔPAR4 mice had increased immune cell numbers and inflammatory mediators in the lung, and increased mortality compared with infected WT controls.     Non-linear regression models for Approximate Bayesian Computation

    Approximate Bayesian inference on the basis of summary statistics is well-suited to complex problems for which the likelihood is either mathematically or computationally intractable. However the methods that use rejection suffer from the curse of dimensionality when the number of summary statistics is increased. Here we propose a machine-learning approach to the estimation of the posterior density by introducing two innovations. The new method fits a nonlinear conditional heteroscedastic regression of the parameter on the summary statistics, and then adaptively improves estimation using importance sampling. The new algorithm is compared to the state-of-the-art approximate Bayesian methods, and achieves considerable reduction of the computational burden in two examples of inference in statistical genetics and in a queueing model.Comment: 4 figures; version 3 minor changes; to appear in Statistics and Computin
