    Host-Adaptation of Francisella tularensis Alters the Bacterium's Surface-Carbohydrates to Hinder Effectors of Innate and Adaptive Immunity

    The gram-negative bacterium Francisella tularensis survives in arthropods, fresh water amoeba, and mammals with both intracellular and extracellular phases and could reasonably be expected to express distinct phenotypes in these environments. The presence of a capsule on this bacterium has been controversial with some groups finding such a structure while other groups report that no capsule could be identified. Previously we reported in vitro culture conditions for this bacterium which, in contrast to typical methods, yielded a bacterial phenotype that mimics that of the bacterium's mammalian, extracellular phase.SDS-PAGE and carbohydrate analysis of differentially-cultivated F. tularensis LVS revealed that bacteria displaying the host-adapted phenotype produce both longer polymers of LPS O-antigen (OAg) and additional HMW carbohydrates/glycoproteins that are reduced/absent in non-host-adapted bacteria. Analysis of wildtype and OAg-mutant bacteria indicated that the induced changes in surface carbohydrates involved both OAg and non-OAg species. To assess the impact of these HMW carbohydrates on the access of outer membrane constituents to antibody we used differentially-cultivated bacteria in vitro to immunoprecipitate antibodies directed against outer membrane moieties. We observed that the surface-carbohydrates induced during host-adaptation shield many outer membrane antigens from binding by antibody. Similar assays with normal mouse serum indicate that the induced HMW carbohydrates also impede complement deposition. Using an in vitro macrophage infection assay, we find that the bacterial HMW carbohydrate impedes TLR2-dependent, pro-inflammatory cytokine production by macrophages. Lastly we show that upon host-adaptation, the human-virulent strain, F. tularensis SchuS4 also induces capsule production with the effect of reducing macrophage-activation and accelerating tularemia pathogenesis in mice.F. tularensis undergoes host-adaptation which includes production of multiple capsular materials. These capsules impede recognition of bacterial outer membrane constituents by antibody, complement, and Toll-Like Receptor 2. These changes in the host-pathogen interface have profound implications for pathogenesis and vaccine development

    QUBIC: A Fizeau Interferometer Targeting Primordial B-Modes

    Q and U Bolometric Interferometer for Cosmology (QUBIC) is a Fizeau interferometer sensitive to linear polarisation, to be deployed at the Antarctic station of Dome C. This experiment in its final configuration will be operated at 97, 150 and 220 GHz and is intended to target CMB primordial B-modes in a multipole window 20<â„“<150. A sensitivity of r=0.05 (95 % CL) can be reached by the first module alone, after 2 years of operation. Here we review in particular its working principles, and we show how the QUBIC interferometric configuration can be considered equivalent to a pupil-plane filtered imaging system. In this context, we show how our instrument can be self-calibrated. Finally, we conclude by showing an overview of the first dual-band module (150/220 GHz), which will serve also as a demonstrator for the subsequent units, and review the technological choices we made for each subsystem, with particular emphasis on the detection system

    Acromegaly in the elderly

    UNLABELLED: Multicentric registers such as The French Acromegaly Register provide data on rare disorders that are otherwise difficult to obtain, so avoiding extrapolation from limited data sets. This study focuses on clinical, hormonal and therapy characteristics of acromegaly in people over 70 years old. The objective was to compare this population with the youngest to disclose if the medical management was similar. PATIENTS AND METHODS: The data were obtained from the 30 centres that have registered patients in the Acromegaly Register since 1999. RESULTS: The register listed 644 acromegaly patients on 1st January 2005, of whom 68 (22 men and 46 women) were over 70 years old, independently of the diagnosis date of their disease. Their average age was 76.8 + or - 5 years (range: 70-95) and they had suffered from acromegaly for 11 + or - 6 years (compared to 7 years in those aged less than 70). Their BMI were similar. Diabetes and hypertension were more frequent than in younger acromegalic patients and in the general French population matched for age. Circulating GH and IGF-1 concentrations were lower than in the younger acromegalic patients on inclusion and 1 year after treatment. There was no significant difference in the tumor size. Only 44% of the patients over 70 underwent surgery, against 90% of patients under 70 years. However, the fractions of young and elderly patients with a controlled disease 1 year after inclusion were the same (51%). CONCLUSION: In the limits of the use of register, these data reveal a high prevalence of diabetes and hypertension in the eldest acromegalic patients. Despite much less frequent surgical intervention, patients' disease under control is equivalent to the younger population 1 year after the initial interview, confirming the effectiveness of the choices of treatment

    Tracing the sources of suspended sediment and particle-bound trace metal elements in an urban catchment coupling elemental and isotopic geochemistry, and fallout radionuclides

    International audienceThe excessive supply of contaminants from urban areas to rivers during the last centuries has led to deleterious impacts on aquatic ecosystems. The sources, the behavior and the dynamics of these contaminants must be better understood in order to reduce this excessive anthropogenic pollution. Accordingly, the current research investigated the particle-bound trace element (TE) contamination of the 900-km² Orge River (Seine basin, France) and the potential sources of these particles (agricultural or forest soils, channel banks, road deposited sediments), through the analysis of multiple fallout radionuclides, elemental geochemistry and lead isotopic composition on suspended particulate matter (SPM) collected during a hydrological year at four stations following an increasing urbanization gradient (300 to 5000 inhab.km-2). Fallout radionuclide measurements showed an increasing contribution of recently eroded particles from urban areas to the SPM in downstream direction. However, this contribution varied depending on hydrological conditions. A greater contribution of particles originating from urban areas was observed during low stage periods. On the contrary, the contribution of agricultural soils and channel banks that are less enriched in contaminants and fallout radionuclides was higher during seasonal floods, which explained the dilution of radionuclide contents in sediment transiting the river during those events. Trace element contamination of SPM in Cu, Zn, Pb and Sb increased from moderate to significant levels with urban pressure in downstream direction (with corresponding enrichment factors raising from 2 to 6). In addition, Pb isotopic ratios indicated that the main source of Pb corresponded to the "urban" signature found in road deposited sediments. The low variations in lead isotope ratios found in the SPM for contrasting hydrological conditions demonstrated the occurrence of a single source of Pb contamination. These results demonstrate the need to better manage urban runoff during both flood and low precipitation events to prevent the supply of diffuse particle-bound contamination to rivers draining urban areas

    Street Dust: Source and Sink of Heavy Metals To Urban Environment

    Air-transferred solid material accumulated for 40 years in different places in an urban area, Grenoble city, France. An appropriate fractionation procedure allowed to separate: 1) a coarse inorganic fraction, 2) a coarse organic fraction, 3) a slowly depositing organo-clay fraction and 4) very fine particles. The composition of each fraction was determined for 20 elements and for isotopic lead signature. The organo-clay fraction was especially rich in Ag, Cd, Co, Cr, Ni and Ti. The sand fraction showed very high concentrations specifically in Cu, Pb and Fe. In contrast, Cd and Zn were mainly accumulated in the coarse organic fraction. The SEM-EDX study of the coarse inorganic fraction showed the presence of red particles associating Pb and Fe, black particles rich in Cu and typical fly ashes originating mostly from iron industry. This sand fraction is suspected to contribute to the contamination of the organo-clay fraction through adsorption. The Pb-Fe contamination likely originates from the neighbouring road surface contaminated by car traffic for several decades. The 206Pb/207Pb ratio showed that these street dust samples may contain up to 50% of lead originated from leaded gasoline additives, twelve years after their prohibition. All these features clearly differ from the composition found for the deposit on tree bark in the same place. These results demonstrate that the deposition over several decades is a very complex phenomenon which requires multi-techniques investigations to be understood thoroughly. They also show that large dense particles which can only be transported on a limited distance by high magnitude events may play a major role in the long-term contamination of urban soils
