    Effect of venlafaxine on bone loss associated with ligature-induced periodontitis in Wistar rats

    <p>Abstract</p> <p>Background</p> <p>The present study investigated the effects of venlafaxine, an antidepressant drug with immunoregulatory properties on the inflammatory response and bone loss associated with experimental periodontal disease (EPD).</p> <p>Materials and Methods</p> <p>Wistar rats were subjected to a ligature placement around the second upper left molar. The treated groups received orally venlafaxine (10 or 50 mg/kg) one hour before the experimental periodontal disease induction and daily for 10 days. Vehicle-treated experimental periodontal disease and a sham-operated (SO) controls were included. Bone loss was analyzed morphometrically and histopathological analysis was based on cell influx, alveolar bone, and cementum integrity. Lipid peroxidation quantification and immunohistochemistry to TNF-α and iNOS were performed.</p> <p>Results</p> <p>Experimental periodontal disease rats showed an intense bone loss compared to SO ones (SO = 1.61 ± 1.36; EPD = 4.47 ± 1.98 mm, p < 0.001) and evidenced increased cellular infiltration and immunoreactivity for TNF-α and iNOS. Venlafaxine treatment while at low dose (10 mg/kg) afforded no significant protection against bone loss (3.25 ± 1.26 mm), a high dose (50 mg/kg) caused significantly enhanced bone loss (6.81 ± 3.31 mm, p < 0.05). Venlafaxine effectively decreased the lipid peroxidation but showed no significant change in TNF-α or iNOS immunoreactivity.</p> <p>Conclusion</p> <p>The increased bone loss associated with high dose venlafaxine may possibly be a result of synaptic inhibition of serotonin uptake.</p

    Do steroids trigger mania in multiple sclerosis? Not always!

    This paper consists of an unusual case history and an overview of the relationship between multiple sclerosis, manic episode and corticosteroid treatment. Bipolar disorder and multiple sclerosis co-occur at a relatively higher rate. A 25-year old woman with a 7-year history of multiple sclerosis admitted to our hospital with right side weakness. During examination she showed some manic behavior patterns and diagnosed as ‘bipolar disorder-manic episode’ by psychiatrists. She was suggested to receive high dose steroids for her attack and antipsychotics for the manic episode but refused the latter. Unexpectedly at the third day of steroid treatment, instead of being triggered, almost all of the manic symptoms disappeared. This is the first report of a multiple sclerosis patient with bipolar disorder whose manic symptoms were cured after receiving steroids

    PubMed: 15688268BACKGROUND OBJECTIVE: To evaluate the results we obtained from 37 patients referred to our clinics with resorbable plate-screw systems which have been claimed as biodegradable fixation materials and used in craniofacial, maxillofacial and orthopedic reconstructive procedures owing to their advantages such as biocompatibility, adequate biomechanical resistance, longer dwelling time, elimination through physiological routes without causing any foreign body reaction and/or significant sequaleae. METHODS: Resorbable plate-screw systems used in 37 patients between 2000-2003 for various craniofacial reconstructive procedures were evaluated as for their efficacy, and biocompatibility. RESULTS: Adequate fixation was obtained in all patients, excluding one case operated for mandibula fracture whose reconstruction was completed with titanium plate-screw system. No serious complication was seen except a granuloma and sinus formation on subciliary incision in one patient. CONCLUSION: Owing to their improved polylactic acid/polyglycolic acid ratio resorbable plate-screw systems which contain varying compositions of polylactic acid and polyglycolic acid copolymers are ideal fixation materials used favourably in maxillofacial, craniofacial and orthopedic reconstructive surgery in that they make effective fixation and have further advantages such as biocompatibility, adequate biomechanical resistance against distraction and compression forces in the early postoperative period, longer dwelling time and elimination from the body through physiological routes without causing any foreign body reaction or significant sequaleae

    Restless legs syndrome (RLS) is a relatively common sensory-motor disorder. However, it is frequently unrecognized. The main clinical characteristics of RLS are “an urge to move” and “uncomfortable and unpleasant sensations” in the legs, which can affect quality of daily life and sleep. The diagnosis is based on clinical findings. Polysomnography can reveal its impact on sleep physiology. Dopaminergic dysfunction and a change in the regulation of iron homeostasis leading to iron depletion in the central nervous system (CNS) are thought to be involved in the pathophysiology of RLS. The aim of this paper is to point out the side effects with off-label use of antipsychotics and call attention to often overlooked RLS by describing an elderly patient who developed RLS symptoms after addition of 50 mg/d quetiapine to preexisting 20 mg/d citalopram treatment.Huzursuz bacaklar sendromu (HBS) nispeten sık görülen ancak sıklıkla tanınmayan ve etyolojisi bilinmeyen duyusal-motor bozukluktur. HBS’nin temel klinik belirtisi hastaların bacaklarında ortaya çıkan, günlük yaşam ve uyku kalitesini etkileyebilen, “hareket ettirme dürtüsü” ve “hoş olmayan hisler”dir. HBS’nin tanısı klinik bulgulara dayanılarak konulur. Polisomniografi, rahatsızlığın uyku fizyolojisi üzerindeki etkilerini göstermede faydalı olabilir. Merkezi sinir sistemindeki (MSS) dopaminerjik bir disfonksiyonun veya bir demir hemostazında deplesyona yol açacak bir değişikliğin, HBS’nin fizyopatolojisinde etkili olabileceği düşünülmektedir. Bu makalede, 20mg/gün dozunda sitalopram tedavisine 50 mg/gün dozunda ketiapin eklenmesinden sonra HBS semptomları geliştiren yaşlı bir erkek hastanın klinik tablosu tanımlanarak, genellikle gözden kaçırılan HBS‘ye ve antipsikotiklerin normal endikasyonları dışında kullanımları esnasında ortaya çıkabilecek yan etkilere dikkat çekilmesi amaçlanmıştır

    The reaction of hexachlorocyclotriphosphazene, N3P3Cl6, with tyramine podand (2) afforded partly substituted spiro-cyclotriphosphazene (3). Amine-substituted spiro-cyclotriphosphazenes 4a–g were prepared by substitution of the Cl–atoms in 3 with pyrrolidine, piperidine, morpholine, 1,4-dioxa-8-azaspiro[4,5]decane, 1-(2-aminoethyl)pyrrolidine, 1-(2-aminoethyl) piperidine, and 4-(2-aminoethyl)morpholine, respectively. All of the cyclotriphosphazene derivatives were characterized by elemental analysis, FTIR, MS, 1D 1H, 13C and 31P NMR and 2D HSQC techniques, and the crystal structures of 3 and 4b were verified by X-ray diffraction analysis. The relationships ?POPN shifts with exocyclic OPN (?') and endocyclic NPN (?) bond angles, and electron density transfer parameters ?(P-N) for spiro-cyclotriphosphazenes were presented. The DNA cleavage activity of cyclotriphosphazene derivatives (3, and 4a–g) was studied on double-stranded pBR322 DNA using gel electrophoresis experiments. It was found that 4e and 4f caused the highest level of DNA damage. The interactions of 3 and 4e with calf thymus DNA were also investigated using absorption spectrometry.
