Abstract

Background

The ubiquitin proteasome system (UPS) is a key player in regulating many cellular processes via proteasomal degradation of ubiquitinated proteins. Recently published data show that Jab1/CSN5 interacts with p97/VCP and controls the ubiquitination status of proteins bound to p97/VCP in mouse and human cells. However, coexpression of p97/VCP and Jab1/CSN5 in the developing rat testis and epididymis has not previously been studied.

Methods

Testicular and epididymal tissues from 5-, 15-, 30-, and 60-day-old rats were examined by immunohistochemistry and Western blotting. Colocalisation of proteins was determined by immunofluorescence microscopy.

Results

In the 5-day-old rat testis, p97/VCP and Jab1/CSN5 were specifically expressed in gonocytes. The expression of p97/VCP and Jab1/CSN5 significantly increased at day 15 and was found in spermatogonia, Sertoli cells and spermatocytes. In 30- and 60-day-old rat testes, p97/VCP indicated moderate to strong expression in Sertoli cells, spermatogonia, round and elongating spermatids. However, moderate to weak expression was observed in spermatocytes. Jab1/CSN5 showed strong expression in spermatogonia and spermatocytes, while relatively moderate expression was observed in round and elongating spermatids in 30- and 60-day-old rat testes. In contrast, in the epididymis, the expression of both proteins gradually increased from 5 to 60 days of age. After rats reached 2 weeks of age, the expression of both proteins was mostly restricted to the basal and principal cells of the caput epididymis.

Conclusions

Our study suggests that p97/VCP and Jab1/CSN5 could be an important part of the UPS in the developing rat testis and epididymis and that both proteins may be involved in the regulation of spermatogenesis and epididymal epithelial functions.</p

Background/aim: Several studies have demonstrated that cigarette smoke has detrimental effects on testicular function. However, it is unknown whether melatonin or BQ-123 has beneficial effects on the rat testis damage caused by cigarette smoke. The aim of the present study was to investigate the beneficial effects of melatonin or BQ-123 on the testicular damage caused by cigarette smoke. Materials and methods: Twenty Wistar rats were randomly divided into 4 equal groups: control group (n = 5), cigarette smoke group (n = 5), melatonin group (n = 5), and BQ-123 group (n = 5). At the end of 4 weeks, all the rats were sacrificed for histopathological evaluation and subsequent stereological analysis. The optical fractionator counting method, the most efficient and unbiased method, was used to estimate the total number of spermatogonia and spermatocytes. Results: All the control testes demonstrated complete spermatogenesis. There was a significant decrease in the germ cells of rats exposed to cigarette smoke for 4 weeks. After the application of melatonin or BQ-123, the total number of spermatogonia and spermatocytes in the testes was significantly higher. Conclusion: Based on these findings, melatonin and BQ-123 are able to minimize the degenerative effects of cigarette smoke by increasing the germ cell count.Gaziosmanpasa University Research FundGaziosmanpasa University [2006/19]This study was supported by Gaziosmanpasa University Research Fund Project no. 2006/19. SUMMARY Leptomeningeal carcinomatosis (LCM) is a rare complication of gastric cancer. It usually occurs late in advanced stage of disease and is sometimes misdiagnosed as toxicity of chemotherapeutic agents. Here we report a rare case of gastric cancer that developed LCM in follow-up. A 28-year-old woman with signet ring cell gastric cancer associated with Krukenberg tumor was admitted with persistent headache, nausea, vomiting, vertigo, and diplopia. Linear appearance of contrast enhancement in cerebellar fissures and around cranial nerves was seen in magnetic resonance imaging and cerebrospinal fluid was hypercellular with numerous carcinoma cells. LMC was confirmed and treated with intrathecal methotrexate and additional whole-brain irradiation. LCM is a rare complication, but occurs more often than expected and is often misdiagnosed. If patient who is being treated for gastric cancer presents with neurological symptoms, LCM should be kept in mind. Clinical improvement can be achieved with current treatment modalities, including radiotherapy, chemotherapy, or targeted molecules

Patolojik inceleme sonucu kitlelerin endometrium dokusuna sahip endometrioma oldugu saptandı.Endometriosis is a common clinical problem of the child-bearing women. Incisional endometrioma is very rare. Patients frequently presented with a history of previous gynecological surgery such as cesarean section or hysterectomy. Although being a gynecological disorder, it may seem like an incisional hernia or abdominal wall tumor. Therefore, it is frequently referred to general surgery outpatient clinics. Here, we report two cases with previous histories of cesarean sections and questinable clinical and laboratory findings of malignancy. The masses located on the previous incision scars were excised widely, and pathological examination of surgical specimens revealed the diagnosis of endometrioma in both cases

Typically, DC patients present with disease features such as nail dystrophy, oral leukoplakia, and abnormal skin pigmentation along with peripheral pancytopenia and marrow hypoplasia with strong predisposition to cancer.1 In DC, hematopoietic failure occurs due to critical shortening of telomeres,2,3 which enhances the DNA damage response4,5 and leads to premature senescence of hematopoietic stem cellsMedical Research Council (grant MR/P018440/1), Bloodwise (grant 14032), and Children with Cancer UK (grant 2013/144)

Beta1-Adrenoceptor Polymorphism Predicts Flecainide Action in Patients with Atrial Fibrillation

BACKGROUND: Antiarrhythmic action of flecainide is based on sodium channel blockade. Beta(1)-adrenoceptor (beta(1)AR) activation induces sodium channel inhibition, too. The aim of the present study was to evaluate the impact of different beta(1)AR genotypes on antiarrhythmic action of flecainide in patients with structural heart disease and atrial fibrillation. METHODOLOGY/PRINCIPAL FINDINGS: In 145 subjects, 87 with atrial fibrillation, genotyping was performed to identify the individual beta(1)AR Arg389Gly and Ser49Gly polymorphism. Resting heart rate during atrial fibrillation and success of flecainide-induced cardioversion were correlated with beta(1)AR genotype. The overall cardioversion rate with flecainide was 39%. The Arg389Arg genotype was associated with the highest cardioversion rate (55.5%; OR 3.30; 95% CI; 1.34-8.13; p = 0.003) compared to patients with Arg389Gly (29.5%; OR 0.44; 95% CI; 0.18-1.06; p = 0.066) and Gly389Gly (14%; OR 0.24; 95% CI 0.03-2.07; p = 0.17) variants. The single Ser49Gly polymorphism did not influence the conversion rate. In combination, patients with Arg389Gly-Ser49Gly genotype displayed the lowest conversion rate with 20.8% (OR 0.31; 95% CI; 0.10-0.93; p = 0.03). In patients with Arg389Arg variants the heart rate during atrial fibrillation was significantly higher (110+/-2.7 bpm; p = 0.03 vs. other variants) compared to Arg389Gly (104.8+/-2.4 bpm) and Gly389Gly (96.9+/-5.8 bpm) carriers. The Arg389Gly-Ser49Gly genotype was more common in patients with atrial fibrillation compared to patients without atrial fibrillation (27.6% vs. 5.2%; HR 6.98; 95% CI; 1.99-24.46; p<0.001). CONCLUSIONS: The beta(1)AR Arg389Arg genotype is associated with increased flecainide potency and higher heart rate during atrial fibrillation. The Arg389Gly-Ser49Gly genotype might be of predictive value for atrial fibrillation

An effective method for videorecording the nasal-dorsal part of a rhinoplasty - a multiple case study

    No full text
WOS: 000391177400005PubMed ID: 27797290Rhinoplasty is a difficult procedure to record for several reasons which include the narrow, dark and deep surgical field cluttered with many instruments, and the heads and hands of the surgical team often blocking the view. This study discusses 13 rhinoplasty cases that were recorded using the authors' technique. A three-minute, unedited segment was extracted from the mid-portion of each surgical case, and that footage was evaluated by three specialist examiners. Results indicate that the authors' technique can offer a effective solution in the video recording of the nasal-dorsal part of a rhinoplasty

An assessment of indoor air concentrations and health risks of volatile organic compounds in three primary schools

    No full text
Concentrations of volatile organic compounds (VOCs), including formaldehyde, in classrooms, kindergartens, and outdoor playgrounds of three primary schools were measured in spring, winter, and fall terms in Izmir, Turkey. A health-risk assessment was conducted for odor detection, sensory irritation, chronic toxic effects, and cancer. Active sampling was applied for VOCs and formaldehyde on Tenax TA and DNPH tubes, respectively. VOCs were analyzed in a thermal desorption-GC-MS system. Formaldehyde analysis was performed using an HPLC instrument. Benzene, toluene, and formaldehyde were the most abundant compounds with 95th percentile indoor air concentrations of 29, 87, and 106μg/m3, respectively. Naphthalene and xylenes followed them with an order of magnitude lower concentrations. Two isomers of dichlorobenzene (1,3 and 1,4) were the other notable compounds. The concentrations were utilized to classify the indoor air pollutants with respect to potential health effects. In addition, carcinogenic and chronic toxic risks were estimated using Monte-Carlo simulation. Formaldehyde appears to be the most concerning pollutant with high chronic toxic and carcinogenic risk levels according to the health assessment followed by naphthalene, benzene, and toluene due to their chronic effects
