    Changes in the West African landscape at the end of the African Humid Period

    Existing pollen datasets from northern Africa stored in the African Pollen Database were used to assess changes in landscape physiognomy at the end of the African Humid Period (AHP) from 5000 cal yr BP to the present using arboreal pollen percentages. The thirty-six sites available were used to map changes in arboreal cover at a sub-continental scale. Based on their location in present-day forested and non-forested areas and their relatively higher temporal resolution eight of them were selected to examine the timing and amplitude of the vegetation response in more detail, and particularly in the Sahel. In spite of low pollen production and dispersal of many tropical plants, which lead to the under representation of most of the trees relative to their abundance in the landscape, we were able to distinguish the geographical pattern and timing of vegetation changes. The landscape response to the end of the AHP was far from homogeneous particularly in the Sahel where a clear east-west gradient of changing tree cover is indicated with the central Sahel being notably species poor. In areas where forests were well developed during the AHP, i.e. in the south and west, the establishment of the modern landscape was abrupt with a threshold crossed between 3300 and 2500 cal yr BP according to local conditions. Elsewhere in northern Africa the switch from tree (C3) to grass (C4) dominated landscapes occurred more gradually during the same period. This review shows that the timing of the ecosystem response at the end of the AHP was remarkably synchronous throughout northern Africa

    Assimilation of shrimp farm sediment by Holothuria scabra: a coupled fatty acid and stable isotope approach

    Deposit-feeding sea cucumbers are efficient nutrient recyclers and have the potential to contribute to the limitation of organic matter load in polyculture or integrated aquaculture systems. Assessing how they assimilate organic matter originating from other farmed species is therefore important for the development of such multi-species farming systems. Here, a coupled stable isotope − fatty acid approach was used to characterize the assimilation of organic matter from shrimp (Penaeus stylirostris) farming by Holothuria scabra in an experimental culture system. H. scabra were reared in mesocosms on shrimp farming-originating sediment with and without additional food sources (maize and fish meals). Although fatty acid results did indicate that shrimp-farming sediment was assimilated by holothurids, we found no evidence of maize waste and fish meal contribution to H. scabra organic carbon (no effect on δ13C, no accumulation of meal-specific fatty acids). However, a strong effect of fish meal on H. scabra δ15N was observed, suggesting that this additional food source could represent an alternative source of nitrogen for holothurids. Finally, this study supports the culture of H. scabra as a perspective to reduce sedimentary organic matter excess associated with shrimp farms, and suggest that the addition of selected food sources might contribute to increasing the content in some nitrogen organic compounds in holothurid tissues

    Sustained Progression-Free Survival Benefit of Rituximab Maintenance in Patients With Follicular Lymphoma : Long-Term Results of the PRIMA Study

    PURPOSE The PRIMA study (ClinicalTrials.gov identifier: NCT00140582) established that 2 years of rituximab maintenance after first-line immunochemotherapy significantly improved progression-free survival (PFS) in patients with follicular lymphoma compared with observation. Here, we report the final PFS and overall survival (OS) results from the PRIMA study after 9 years of follow-up and provide a final overview of safety. METHODS Patients (> 18 years of age) with previously untreated high-tumor-burden follicular lymphoma were nonrandomly assigned to receive one of three immunochemotherapy induction regimens. Responding patients were randomly assigned (stratified by induction regimen, response to induction treatment, treatment center, and geographic region) 1:1 to receive 2 years of rituximab maintenance (375 mg/m(2), once every 8 weeks), starting 8 weeks after the last induction treatment, or observation (no additional treatment). All patients in the extended follow-up provided their written informed consent (data cutoff: December 31, 2016). RESULTS In total, 1,018 patients completed induction treatment and were randomly assigned to rituximab maintenance (n = 505) or observation (n = 513). Consent for the extended follow-up was provided by 607 patients (59.6%) of 1,018 (rituximab maintenance, n = 309; observation, n = 298). After data cutoff, median PFS was 10.5 years in the rituximab maintenance arm compared with 4.1 years in the observation arm (hazard ratio, 0.61; 95% CI, 0.52 to 0.73; P <.001). No OS difference was seen in patients randomly assigned to rituximab maintenance or observation (hazard ratio, 1.04; 95% CI, 0.77 to 1.40; P = .7948); 10-year OS estimates were approximately 80% in both study arms. No new safety signals were observed. CONCLUSION Rituximab maintenance after induction immunochemotherapy provides a significant long-term PFS, but not OS, benefit over observation.Peer reviewe

    Immunity Traits in Pigs: Substantial Genetic Variation and Limited Covariation

    BACKGROUND: Increasing robustness via improvement of resistance to pathogens is a major selection objective in livestock breeding. As resistance traits are difficult or impossible to measure directly, potential indirect criteria are measures of immune traits (ITs). Our underlying hypothesis is that levels of ITs with no focus on specific pathogens define an individual's immunocompetence and thus predict response to pathogens in general. Since variation in ITs depends on genetic, environmental and probably epigenetic factors, our aim was to estimate the relative importance of genetics. In this report, we present a large genetic survey of innate and adaptive ITs in pig families bred in the same environment. METHODOLOGY/PRINCIPAL FINDINGS: Fifty four ITs were studied on 443 Large White pigs vaccinated against Mycoplasma hyopneumoniae and analyzed by combining a principal component analysis (PCA) and genetic parameter estimation. ITs include specific and non specific antibodies, seric inflammatory proteins, cell subsets by hemogram and flow cytometry, ex vivo production of cytokines (IFNα, TNFα, IL6, IL8, IL12, IFNγ, IL2, IL4, IL10), phagocytosis and lymphocyte proliferation. While six ITs had heritabilities that were weak or not significantly different from zero, 18 and 30 ITs had moderate (0.1<h2≤0.4) or high (h2>0.4) heritability values, respectively. Phenotypic and genetic correlations between ITs were weak except for a few traits that mostly include cell subsets. PCA revealed no cluster of innate or adaptive ITs. CONCLUSIONS/SIGNIFICANCE: Our results demonstrate that variation in many innate and adaptive ITs is genetically controlled in swine, as already reported for a smaller number of traits by other laboratories. A limited redundancy of the traits was also observed confirming the high degree of complementarity between innate and adaptive ITs. Our data provide a genetic framework for choosing ITs to be included as selection criteria in multitrait selection programmes that aim to improve both production and health traits

    Different Transcriptional Control of Metabolism and Extracellular Matrix in Visceral and Subcutaneous Fat of Obese and Rimonabant Treated Mice

    BACKGROUND: The visceral (VAT) and subcutaneous (SCAT) adipose tissues play different roles in physiology and obesity. The molecular mechanisms underlying their expansion in obesity and following body weight reduction are poorly defined. METHODOLOGY: C57Bl/6 mice fed a high fat diet (HFD) for 6 months developed low, medium, or high body weight as compared to normal chow fed mice. Mice from each groups were then treated with the cannabinoid receptor 1 antagonist rimonabant or vehicle for 24 days to normalize their body weight. Transcriptomic data for visceral and subcutaneous adipose tissues from each group of mice were obtained and analyzed to identify: i) genes regulated by HFD irrespective of body weight, ii) genes whose expression correlated with body weight, iii) the biological processes activated in each tissue using gene set enrichment analysis (GSEA), iv) the transcriptional programs affected by rimonabant. PRINCIPAL FINDINGS: In VAT, "metabolic" genes encoding enzymes for lipid and steroid biosynthesis and glucose catabolism were down-regulated irrespective of body weight whereas "structure" genes controlling cell architecture and tissue remodeling had expression levels correlated with body weight. In SCAT, the identified "metabolic" and "structure" genes were mostly different from those identified in VAT and were regulated irrespective of body weight. GSEA indicated active adipogenesis in both tissues but a more prominent involvement of tissue stroma in VAT than in SCAT. Rimonabant treatment normalized most gene expression but further reduced oxidative phosphorylation gene expression in SCAT but not in VAT. CONCLUSION: VAT and SCAT show strikingly different gene expression programs in response to high fat diet and rimonabant treatment. Our results may lead to identification of therapeutic targets acting on specific fat depots to control obesity

    Autoantibodies against type I IFNs in patients with critical influenza pneumonia

    In an international cohort of 279 patients with hypoxemic influenza pneumonia, we identified 13 patients (4.6%) with autoantibodies neutralizing IFN-alpha and/or -omega, which were previously reported to underlie 15% cases of life-threatening COVID-19 pneumonia and one third of severe adverse reactions to live-attenuated yellow fever vaccine. Autoantibodies neutralizing type I interferons (IFNs) can underlie critical COVID-19 pneumonia and yellow fever vaccine disease. We report here on 13 patients harboring autoantibodies neutralizing IFN-alpha 2 alone (five patients) or with IFN-omega (eight patients) from a cohort of 279 patients (4.7%) aged 6-73 yr with critical influenza pneumonia. Nine and four patients had antibodies neutralizing high and low concentrations, respectively, of IFN-alpha 2, and six and two patients had antibodies neutralizing high and low concentrations, respectively, of IFN-omega. The patients' autoantibodies increased influenza A virus replication in both A549 cells and reconstituted human airway epithelia. The prevalence of these antibodies was significantly higher than that in the general population for patients 70 yr of age (3.1 vs. 4.4%, P = 0.68). The risk of critical influenza was highest in patients with antibodies neutralizing high concentrations of both IFN-alpha 2 and IFN-omega (OR = 11.7, P = 1.3 x 10(-5)), especially those <70 yr old (OR = 139.9, P = 3.1 x 10(-10)). We also identified 10 patients in additional influenza patient cohorts. Autoantibodies neutralizing type I IFNs account for similar to 5% of cases of life-threatening influenza pneumonia in patients <70 yr old

    International audienceLe phénomène de l'amok, accès soudain et inexpliqué de folie meurtrière dont les manifestations ont commencé à passionner les psychiatres occidentaux dans la seconde moitié du XIXe siècle, fournit le fil conducteur du roman de Reinhard Jirgl, "Abtrünnig - Roman aus der nervösen Zeit "(titre français: "Renégat - Roman du temps nerveux", Quidam, 2010). La phénoménologie de l'amok, que caractérisent entre autres une course effrénée et désordonnée puis un oubli qui le rend inaccessible, est ici rapprochée des formes élaborées par Reinhard Jirgl, en particulier des encadrés et des renvois qui jalonnent son texte et lui confèrent une allure labyrinthique. La combinaison des multiples chemins de lecture possibles avec l'existence de plages narratives closes sur elles-mêmes, combinaison particulièrement apte à rendre le caractère à la fois agité et forclos de l'amok, n'est pas sans évoquer la forme de l'arabesque en peinture, à partir de laquelle Friedrich Schlegel a élaboré sa théorie de la forme poétique
