    多重荧光PCR同时检测转基因成分35S和Nos方法的建立

    根据商品化转基因作物中常用的花椰菜花叶病毒启动子 (CaMV 35S)和根癌农杆菌终止子 (Nos)的序列特点 ,设计并合成了两对引物和相对应的荧光双链探针 ,建立一种应用荧光双链探针的多重荧光PCR同时检测转基因成分 35S启动子和Nos终止子的方法 .并利用该方法对马铃薯、大豆、玉米、甜椒、番茄等 11份实物样品进行了检测 ,其中有 5份样品结果阳性 .结果表明所建立的多重荧光PCR方法能同时检测出 35S和Nos双组分 ,较常规PCR技术更为简便、快速、准确 ,有很好的应用前景

    新疆塔里木盆地北部碳酸盐裂缝性储层测井资料综合评价方法研究

    该成果为在“七五”攻关基础上所研制的测井综合评价方法和计算机处理系统。通过岩芯分析结果与测井资料的相关对比,特别是通过综合地质录井信息得到的综合裂缝概率与岩芯分析的裂缝孔隙度的相关对比,建立了利用测井资料裂缝发育程度和裂缝分级的方法,效果较好,有明显的实用价值。采用改进的有限元法进行了裂缝模型的正演模拟,为研究裂缝带及其分级提供了理论根据。建立了包含原“七五”的解释模型(CFIS)的碳酸盐岩裂缝性储层综合评价系统(FRAC)和相应程序。该系统具有岩性判别与测井解释参数选择、裂缝带识别、储层参数估算和油水性质判别等功能;综合了包括声波全波列在内的多种测井参数,拾取多种裂缝指标,并把地质录井信息综合到神经网络技术之中,将如此众多的参数和先进解释技术同时用于碳酸盐岩裂缝性储层测井评价,取得了明显效果

    目的建立注意缺陷多动障碍患者(AdHd)哌甲酯缓释片清除率的群体药动学模型。方法前瞻性收集闽南地区服用哌甲酯缓释片治疗的78例AdHd患者的消除相血药浓度(n=165),采用非线性混合效应模型(nOnMEM)法分析数据。结果最终模型为Cl(l·H-1)=254·(WT/34.7)0.663·0.884(如果合用丙戌酸钠,否则为1)·EXP(ETA(Cl)),WT为体质量(kg)。结论成功建立AdHd患者哌甲酯缓释片治疗的群体药动学模型,根据患者的生理用药资料,可估算其清除率,为制定给药方案提供依据。AIM To establish a clearance model for methylphenidate prolonged-release tablets(MPH)in patients with attention deficit/hyperactivity disorder(ADHD)by population pharmacokinetic method.METHODS Serum concentration data(n=165)in elimination phase were collected prospectively from 78 ADHD patients in south Fujian during their routine clinical treatment.Nonlinear mixed effects modeling(NONMEM)was employed to establish the population pharmacokinetic model of the MPH clearance(CL).RESULTS The final model was:CL(L·h-1)=254·(WT/34.7)0.663·0.884(if taking sodium valproate,otherwise it equaled to 1)·EXP(ETA(CL))in which WT was weight(kg).CONCLUSION A population pharmacokinetic model is established successfully and proposed to estimate the individual CL for patients taking MPH in terms of patients' characteristics and dosing history,and to establish a prior dosage regimen.福建省卫生厅青年科研课题(编号2013-2-02); 厦门市卫生局科研立项(编号3502Z20077079

    目的评价结核分枝杆菌注射类二线药耐药突变检测试剂盒MElTPrO Tb/Sl的分析性能以及临床性能。方法首先使用企业参考品对试剂盒的突变检出能力进行考察,之后将野生型基因组dnA以10倍梯度稀释后考察试剂盒的最低检测限,使用野生型质粒以及含常见耐药突变的质粒以不同突变比例混合考察试剂盒检测不均一耐药样本的能力,并使用23株非结核分枝杆菌标准株考察试剂盒的分析特异性,最后使用241份临床分离株对试剂盒进行临床评价。结果该试剂盒可以将9种耐药相关突变与1种野生型多态性与野生型进行区分。可以检测到低至5个拷贝的野生型基因组dnA。当突变比例为30%或更高时,试剂盒可以将其与野生型进行区分。23种非结核分枝杆菌的结果显示该试剂盒有良好的特异性。和比例法药敏试验进行对比,该试剂盒的临床灵敏度为卡那霉素90.0%(9/10),阿米卡星80.0%(8/10),卷曲霉素85.7%(6/7);特异性为卡那霉素98.8%(85/86),阿米卡星98.8%(85/86),卷曲霉素96.6%(86/89)。试剂盒检测结果与测序结果一致。结论该试剂盒灵敏度高、特异性好,可以快速、准确地检测结核分枝杆菌注射类二线药常见的耐药突变,有望应用于临床。The MeltPro TB/SL assay was first evaluated by a reference panel containing 10 plasmids of different mutations to evaluate the analytical and clinical performances.The limit of detection(LOD)was studied by purified genomic DNA of a reference strain H37 Rv that was serially diluted in ten-fold.The wild-type and mutant plasmids of different mutation types were mixed with serial percentages of mutation to evaluate the detection ability of mixed bacterial samples of this kit.The analytical specificity was evaluated using 23non-tuberculous mycobacterium(NTM)isolates.The clinical performance was evaluated by analyzing 241 clinical strains.Ten plasmids of different mutations could be clearly identified from the wild-type plasmid.The LOD was 5copies of wild-type Mycobacterium tuberculosis(MTB)genomic DNA per reaction.The assay could detect30% mutant DNA in wild-type background.All the 23 NTM strains showed no cross reactivity with M.tuberculosis strain.Clinical study showed that the clinical sensitivity of the assay was 90.0%for kanamycin,80.0%for amikacin,85.7%for capreomycin,and the clinical specificity was 98.8%for kanamycin,98.8%for amikacin,96.6%for capreomycin,respectively.The MeltPro TB/SL assay gave a 100% concordance with sequencing analysis.The MeltPro TB/SL assay was able to reliably detect common mutations associated with injectable second-line drug-resistance.The assay has the potential to be used for rapid screening of injectable second-line drug-resistant mutations in MTB strains.国家“十一五”重大专项(No.2008ZX10003-004)资助~

    Electrochemical Performance of Al-Doped Spinel LiMn2O4 Cathode Material for Li-ion Batteries

    尖晶石LiMn2O4作为锂离子电池正极可大电流放电,且成本低、环境友好. 采用溶胶-凝胶法制备尖晶石LiMn2O4及Al掺杂材料. 使用X-射线衍射(XRD)和扫描电子显微镜(SEM)观察材料结构与形貌. 结果表明,复合材料颗粒尺寸300 ~ 500 nm,呈类球形. 电化学恒流充放电测试表明,Al掺杂尖晶石LiMn2O4电极的循环性明显提高,Al掺杂5%LiMn2O4(by mass,下同)正极在1C倍率充放电100周期循环后的容量保持率为98.2%,1C倍率充电、5C倍率放电下,100周期循环后其容量保持率为99.0%,表现出较优的电化学循环性能.Spinel LiMn2O4, a cathode material for Li-ion batteries, is featured as good high-rate performance, low cost and environmental friendly. The pristine and Al-doped LiMn2O4 samples were prepared by sol-gel method. The structure and micromorphology of the as-prepared samples were characterized by X-ray diffraction (XRD) and scanning electron microscopy (SEM). The results demonstrated that the as-prepared LiMn2O4 samples had a typical cubic spinel structure and were composed of sphere-like particles with a diameter range of 300 ~ 500 nm. Galvanostatic charge-discharge tests showed that the LiMn2O4 samples doped with Al element exhibited an enhanced cycling performance.
