    3-磷酸甘油醛脱氢酶(glyCErAldEHydE-3-PHOSPHATEdE HydrOgEnASE,gAPdH)是维持生命最基本活动的关键酶之一。采用rT-PCr、rACE等技术,获得了拟穴青蟹gAPdH基因全长CdnA序列。该序列全长1 440 bP,开放阅读框(Orf)为1 008 bP,编码335个氨基酸残基。同源分析显示,该基因编码的蛋白与其他一些物种具有很高相似性,推测gAPdH基因具有很高的保守性。经荧光定量PCr检测,gAPdH基因在拟穴青蟹多个组织中均有表达,且在胸神经团、眼柄神经节、卵巢、表皮中表达量较高。在拟穴青蟹卵巢发育过程中,gAPdH基因在卵巢发育早期(Ⅱ期)表达量最高,在卵巢发育成熟期(V期)表达量最低,由此推测gAPdH主要参与了卵巢的细胞分裂增殖过程。Glyceraldehyde-3-phosphatede hydrogenase(GAPDH)is one of the basic enzymes that maintain the life activities.In this paper,gapdh gene was isolated from the mud crab,Scylla paramamosain using RT-PCR and RACE methods.The obtained full-length cDNA of GAPDH was 1 440 bp with an open reading frame of 1 008 bp encoding a putative peptide of 335 amino acids.By alignment,the amino acid sequence of S.paramamosain GAPDH showed high homology with those of some other animals.It suggested gapdh was highly conservative.Real-time PCR showed that the gapdh gene was expressed in various tissues,and highly expressed in thoracic ganglia,eyestalk ganglia,ovary and epidermis.During the ovarian development,the expression of gapdh mRNA was significantly higher at early-developing stage(Ⅱstage)and lowest at ripe stage(Ⅴstage).It indicated that GAPDH might play an important role in cell mitosis in ovary of the mud crab.国家自然科学基金项目(40406030;41076081;31272632

    通过转录组测序,获得在接种ErM真菌的云锦杜鹃苗根系中显著差异表达的基因,其中硝酸根转运蛋白(nrT)基因是硝态氮吸收转运的关键基因。利用生物信息学方法,分析云锦杜鹃根转录组的硝酸根转运蛋白(nrT)基因序列,对其推导的氨基酸的理化性质、亲水性/疏水性、跨膜结构、导肽、二级结构、高级结构进行预测,并对硝酸根转运蛋白的氨基酸做进化发育分析。为进一步了解nrT基因在云锦杜鹃接种苗根系氮素吸收的作用奠定了基础。We used next generation sequencing technology to investigate the transcriptomes between inoculated roots and uninoculated roots of the Rhododendron fortunei and obtained many differentially expressed genes.In this paper,the nucleic acid sequences and amino acid sequences of nitrate transporters fromR.fortunei,were analyzed by bioinformatics tools.Several parameters of these sequences,including sequences composition,physicochemical property,leader peptide,topological structure of transmembrane regions,hydrophobicity or hydrophilicity,secondary structures,functional domains and protein structures,were predicted.Phylogenetic tree was reconstructed for the nitrate transporters protein family.Provide the bioinformatics foundation to understand NRTgene's function in inoculated seedling roots.国家自然科学基金(30972409

    Pharmaceutical salts have been traditionally used in drug formulation. As a salt former, acesulfame (AH), an aliphatic calorie-free sweetener, is actively being employed. Acesulfamates of active pharmaceutical ingredients (APIs) may provide a pleasant sweet taste along with modified physicochemical properties. In this paper, four quinine (QN) salts were obtained with AH, including two 1:1 anhydrous forms (QNAH11a and QNAH11b), one monohydrate of 1:1 salt (QNAH111), and one 1:2 anhydrous forms (QNAH12). The resulting salts were fully characterized by a range of analytical methods. Crystal structures of QNAH11a, QNAH111, and QNAH12 were determined by single-crystal X-ray diffraction, and the crystal structure of QNAH11b was solved from powder X-ray diffraction data by Rietveld refinement. Ionization states for all samples were confirmed by <sup>13</sup>C solid-state NMR spectra. The mutual transformation and thermodynamic relationships of these solid forms were revealed via slurry conversion experiments and differential scanning calorimetry measurements. Additionally, enhanced solubility was observed for each acesulfamate when compared with the pure free base.     西藏羌塘盆地鸭湖地区天然气水合物成藏条件

    近年来中国陆域冻土区天然气水合物调查研究结果表明,气源条件是制约羌塘盆地天然气水合物找矿突破的关键因素。为明确鸭湖地区天然气水合物成藏潜力,基于近年来的钻探调查成果,从陆域冻土区天然气水合物成藏系统理论出发,系统分析了影响天然气水合物成藏的冻土、气源、储集、构造等地质因素。分析结果显示,鸭湖地区局部具有较好的冻土、地温、气源、储集、构造及水源条件,具备一定的天然气水合物成藏潜力,继续寻找充足的烃类气源是下一步天然气水合物调查的主要方向。同时,选取钻探调查获取的地温梯度、气体组分等参数,结合音频大地电磁测深(AMT)冻土厚度调查成果,对鸭湖地区天然气水合物稳定带的厚度和底界深度进行了预测。结果显示,当甲烷为85%、乙烷为9%、丙烷为6%时,天然气水合物稳定带厚度与冻土厚度分布变化基本一致,稳定带厚度400~630m,底界深度400~680m。当甲烷为98%、乙烷为2%时,天然气水合物稳定带厚度急剧减薄,大部分地区仅有0~30m,最厚仅有150m,局部地区稳定带底界最深仅为240m。结合气测录井结果,认为渐新世唢呐湖组比上三叠统土门格拉组更具备天然气水合物成藏潜力,土门格拉组自身具备较强的生排烃能力,可作为寻找常规油气或页岩气的一个重要层位
