    Isotopic Fingerprinting of Shallow and Deep Groundwaters in Southwestern Ontario and its Applications to Abandoned Well Remediation

    Abandoned hydrocarbon wells in southwestern Ontario can act as conduits for Sulphur water, brines, and hydrocarbons from deep Paleozoic bedrock aquifers. Such leakage may pose a threat to shallow groundwater and the environment. Cost-effective plugging of these wells requires knowledge of the sources of the leaking fluids. This study characterizes the isotopic compositions (δ18OH2O, δ2HH2O, δ34SSO4, δ18OSO4, δ13CDIC, 87Sr/86Sr) of groundwaters in the region, which are distinct in different bedrock formations. A Bayesian mixing model was applied to these data to develop a tool for identifying the source(s) of leaking fluids. The geochemical data also improve our understanding of groundwater origin and evolution. Shallow (~\u3c350m) aquifers are recharged by recent meteoric water. At greater depths, brine aquifers contain residual evaporated Paleozoic seawater, modified by rock-water interaction and mixing with meteoric water. These brines are likely related to long-distance fluid migration from deeper portions of the adjacent Michigan and Appalachian basins

    A Pedagogical Venturing into the Three Sisters' Garden: Lessons of Attunement and Reciprocity in Education

    This paper explores the connections the authors make between their experiences in the classroom and the powerful work of Robin Wall Kimmerer (2013) in Braiding Sweetgrass: Indigenous Wisdom, Scientific Knowledge, and the Teaching of Plants; specifically, her chapter entitled “The Three Sisters.” Through Kimmerer’s work, we interpret our own experiences within the classroom. We also draw upon Hans-Georg Gadamer’s philosophical hermeneutics in general and his notion of Erfahrung in particular. We were inspired by the author’s insights into how she happened upon a “new teacher” for her students. As a result, this paper explores her work, as it provides an image of what it is to be present among students while honouring the topic’s own being and becoming--staying open to its future possibilities not yet known. The pedagogical turn away from predetermined outcomes to reciprocity, interconnectedness, and relationships is examined in how they help us understand teaching and learning.  Keywords: Three Sisters; Kimmerer; hermeneutics; curriculum, erfahrung; reciprocity, education; pre-service teacher

    Analysis of putative resistance gene loci in UK field populations of Haemonchus contortus after six years of macrocyclic lactone use

    Sheep farmers in the UK rely on strategic anthelmintic use to treat and control gastrointestinal roundworms in their flocks. However, resistance to these drugs is now widespread and threatens the sustainability of sheep production. The mechanisms underlying resistance to the most commonly used class, the macrocyclic lactones, are not known and sensitive diagnostic tools based on molecular markers are not currently available. This prohibits accurate surveillance of resistance or assessment of strategies aimed at controlling its spread. In this study, we examined four UK field populations of Haemonchus contortus, differing in macrocyclic lactone treatment history, for evidence of selection at ‘candidate gene’ loci identified as determining macrocyclic lactone resistance in previously published research. Individual worms were genotyped at Hc-lgc-37, Hc-glc-5, Hc-avr-14 and Hc-dyf-7, and four microsatellite loci. High levels of polymorphism were identified at the first three candidate gene loci with remarkably little polymorphism at Hc-dyf-7. While some between-population comparisons of individual farms with and without long-term macrocyclic lactone use identified statistically significant differences in allele frequency and/or fixation index at the Hc-lgc-37, Hc-glc-5 or Hc-avr-14 loci, we found no consistent evidence of selection in other equivalent comparisons. While it is possible that different mechanisms are important in different populations or that resistance may be conferred by small changes at multiple loci, our findings suggest that these are unlikely to be major loci conferring macrocyclic lactone resistance on UK farms or suitable for diagnostic marker development. More powerful approaches, using genome-wide or whole genome sequencing, may be required to define macrocyclic lactone resistance loci in such genetically variable populations

    Presence and species identity of rumen flukes in cattle and sheep in the Netherlands

    The purpose of the study was to gain knowledge about the prevalence and identity of rumen flukes (RF) in cattle and sheep in the Netherlands. Routine faecal examinations of diagnostic submissions between May 2009 and September 2014 showed a mean annual herd or flock RF prevalence of 15.8% for cattle and 8.0% for sheep. Prevalence in cattle was higher after 2012 than before, which may reflect a change in detection method as well as an increase in true prevalence. During November and December 2014, an abattoir survey was conducted to allow for scoring of rumen fluke burden and to obtain specimens for molecular species characterization. Over 8 visits to 5 abattoirs in areas deemed to pose a high risk for trematode infection, 116 cows and 41 sheep from 27 herds and 10 flocks were examined. Prevalence of RF was higher in beef cattle than in dairy cattle and higher in cattle than in sheep. Median fluke burden was >100 specimens per animal for most positive animals. Using a semi-quantitative RF density score as a gold standard, sensitivity and specificity of a modified quantitative Dorsman egg counting method were estimated at 82.6% and 83.3%, respectively. Of 14 collected adult rumen flukes, twelve (8 bovine and 4 ovine specimens) were identified as Calicophoron daubneyi. The other two, of bovine origin, were identified as Paramphistomum leydeni, which was unexpected as in other European countries all recently collected rumen flukes in both cattle and sheep were identified as C. daubneyi. The findings implicate that multiple rumen fluke species, intermediate host species and transmission cycles may play a role in rumen fluke infections in the Netherlands

    The emergence of resistance to the benzimidazole anthlemintics in parasitic nematodes of livestock is characterised by multiple independent hard and soft selective sweeps

    Anthelmintic resistance is a major problem for the control of parasitic nematodes of livestock and of growing concern for human parasite control. However, there is little understanding of how resistance arises and spreads or of the “genetic signature” of selection for this group of important pathogens. We have investigated these questions in the system for which anthelmintic resistance is most advanced; benzimidazole resistance in the sheep parasites Haemonchus contortus and Teladorsagia circumcincta. Population genetic analysis with neutral microsatellite markers reveals that T. circumcincta has higher genetic diversity but lower genetic differentiation between farms than H. contortus in the UK. We propose that this is due to epidemiological differences between the two parasites resulting in greater seasonal bottlenecking of H. contortus. There is a remarkably high level of resistance haplotype diversity in both parasites compared with drug resistance studies in other eukaryotic systems. Our analysis suggests a minimum of four independent origins of resistance mutations on just seven farms for H. contortus, and even more for T. circumincta. Both hard and soft selective sweeps have occurred with striking differences between individual farms. The sweeps are generally softer for T. circumcincta than H. contortus, consistent with its higher level of genetic diversity and consequent greater availability of new mutations. We propose a model in which multiple independent resistance mutations recurrently arise and spread by migration to explain the widespread occurrence of resistance in these parasites. Finally, in spite of the complex haplotypic diversity, we show that selection can be detected at the target locus using simple measures of genetic diversity and departures from neutrality. This work has important implications for the application of genome-wide approaches to identify new anthelmintic resistance loci and the likelihood of anthelmintic resistance emerging as selection pressure is increased in human soil-transmitted nematodes by community wide treatment programs

    Bacteria - mineral - nanoparticle interactions in water and wastewater

    With a rising demand for nanomaterials and their continual increase in production, the release of nanoparticles (NPs) into the environment is inevitable (Petosa et al., 2010). Problematically, NPs can have a wide range of toxic effects, which are exacerbated by their size (at least one dimension smaller than 100 nm)(Engineering., 2004). Detrimental effects include brain, intestinal and respiratory injury, delayed embryonic development, DNA damage which ultimately lead to increased mortality (Trouiller et al., 2009), (Handy et al., 2008). Natural and manufactured NPs also have the ability to bind and transport chemical pollutants, thus enhancing their toxicity (Moore, 2006). While an array of techniques are available for in situ remediation of numerous groundwater contaminants, there are currently none for in situ remediation of nanoparticles. This fundamental technology gap means we are poorly prepared to deal with nanoparticle pollution events. The aim of this PhD was to develop mechanisms to immobilise and remove nanoparticles from water and waste water in order to prevent the transport of nanoparticles to sites where they have the potential to cause harm. Experiments conducted demonstrate the potential of microbially mediated mineral formation to immobilise nanoparticles from water. The ureolytic bacteria Sporosarcina pasteurii was used to induce calcium carbonate precipitation in batch and column experiments. Nanoparticle immobilisation was tested as a function of nanoparticle size and surface charge. The results demonstrate the successful immobilisation of negatively charged nanoparticles (both large and small, 150 and 35 nm respectively), while failing to remove positively charged nanoparticles from solution. In order to capture positively charged nanoparticles a second mineral, struvite, was tested. The precipitation of struvite successfully immobilised positively charged nanoparticles. However, in comparison to the calcite precipitation experiments the removal of positively charged nanoparticles was found to be pH and ionic strength dependant. Finally, the ability of Bacillus subtilis, a common groundwater bacterium and wastewater treatment biofilm to adsorb and remove nanoparticles from solution was examined. Here both biosorbent materials were highly efficient at removing positively charged nanoparticles from solution whilst negatively charged nanoparticles remained in suspension. The research presented here demonstrates that microbially induced mineral precipitation may be used as a tool to immobilise nanoparticles from contaminated groundwater. In addition, bacteria and wastewater treatment biofilm were found to be highly efficient biosorbents of positively charged nanoparticles. These findings hold implications for the fate and transport of nanoparticles through environmental systems and wastewater treatment plants

    Multiplexed-tandem PCR for the specific diagnosis of gastrointestinal nematode infections in sheep : an European validation study

    Background: Traditional methods of detecting and identifying gastrointestinal nematode infections in small ruminants, including sheep and goats, are time-consuming and lack in sensitivity and specificity. Recently, we developed an automated multiplexed-tandem (MT)-PCR platform for the diagnosis and identification patent infections with key genera/species of gastrointestinal nematodes of sheep and validated this approach in detailed experiments carried out in Australia. In the present study, we deployed this diagnostic platform in Scotland and Belgium to test samples from naturally infected sheep in these environments and to validate the MT-PCR platform relative to traditional diagnostic methods routinely used by local laboratories. Results: MT-PCR detected all microscopy positive samples and there was a significant agreement between the results of the different test methods in terms of the species detected and their relative proportion in a test sample, however, for some samples, there were discrepancies between the results of the different test methods. Selective sequencing of purified MT-PCR products demonstrated the results to be 100% specific. Conclusions: The MT-PCR platform is an advanced method for the species/genus-specific diagnosis of gastrointestinal nematode infections in small ruminants and has demonstrated utility when deployed in different countries and climatic zones. The platform is user-friendly due to the largely automated procedure and has high versatility in that it can achieve a specific diagnosis from different types of sample templates, including larval culture and faecal samples. With appropriate modifications of the primers used, the MT-PCR platform also provides potential for the diagnosis of a variety of other pathogens of veterinary or medical importance

    Prevalence of liver fluke (Fasciola hepatica) in wild Red Deer (Cervus elaphus): coproantigen ELISA is a practicable alternative to faecal egg counting for surveillance in remote populations

    Red deer (Cervus elaphus) are hosts of liver fluke (Fasciola hepatica); yet, prevalence is rarely quantified in wild populations. Testing fresh samples from remote regions by faecal examination (FE) can be logistically challenging; hence, we appraise frozen storage and the use of a coproantigen ELISA (cELISA) for F. hepatica surveillance. We also present cELISA surveillance data for red deer from the Highlands of Scotland. Diagnoses in faecal samples (207 frozen, 146 fresh) were compared using a cELISA and by FE. For each storage method (frozen or fresh), agreement between the two diagnostics was estimated at individual and population levels, where population prevalence was stratified into cohorts (e.g., by sampling location). To approximate sensitivity and specificity, 65 post-slaughter whole liver examinations were used as a reference. At the individual level, FE and cELISA diagnoses agreed moderately (κfrozen = 0.46; κfresh = 0.51), a likely reflection of their underlying principles. At the population level, FE and cELISA cohort prevalence correlated strongly (Pearson’s R = 0.89, p < 0.0001), reflecting good agreement on relative differences between cohort prevalence. In frozen samples, prevalence by cELISA exceeded FE overall (42.8% vs. 25.8%) and in 9/12 cohorts, alluding to differences in sensitivity; though, in fresh samples, no significant difference was found. In 959 deer tested by cELISA across the Scottish Highlands, infection prevalence ranged from 9.6% to 53% by sampling location. We highlight two key advantages of cELISA over FE: i) the ability to store samples long term (frozen) without apparent loss in diagnostic power; and ii) reduced labour and the ability to process large batches. Further evaluation of cELISA sensitivity in red deer, where a range of fluke burdens can be obtained, is desirable. In the interim, the cELISA is a practicable diagnostic for F. hepatica surveillance in red deer, and its application here has revealed considerable geographic, temporal, sex and age related differences in F. hepatica prevalence in wild Scottish Highland red deer

    Exploring the fate of cattle herds with inconclusive reactors to the tuberculin skin test

    Bovine tuberculosis (TB) is an important animal health issue in many parts of the world. In England and Wales, the primary test to detect infected animals is the single intradermal comparative cervical tuberculin test, which compares immunological responses to bovine and avian tuberculins. Inconclusive test reactors (IRs) are animals that demonstrate a positive reaction to the bovine tuberculin only marginally greater than the avian reaction, so are not classified as reactors and immediately removed. In the absence of reactors in the herd, IRs are isolated, placed under movement restrictions and re-tested after 60 days. Other animals in these herds at the time of the IR result are not usually subject to movement restrictions. This could affect efforts to control TB if undetected infected cattle move out of those herds before the next TB test. To improve our understanding of the importance of IRs, this study aimed to assess whether median survival time and the hazard of a subsequent TB incident differs in herds with only IRs detected compared with negative-testing herds. Survival analysis and extended Cox regression were used, with herds entering the study on the date of the first whole herd test in 2012. An additional analysis was performed using an alternative entry date to try to remove the impact of IR retesting and is presented in the Supplementary Material. Survival analysis showed that the median survival time among IR only herds was half that observed for clear herds (2.1 years and 4.2 years respectively; p < 0.001). Extended Cox regression analysis showed that IR-only herds had 2.7 times the hazard of a subsequent incident compared with negative-testing herds in year one (hazard ratio: 2.69; 95% CI: 2.54, 2.84; p < 0.001), and that this difference in the hazard reduced by 63% per year. After 2.7 years the difference had disappeared. The supplementary analysis supported these findings showing that IR only herds still had a greater hazard of a subsequent incident after the IR re-test, but that the effect was reduced. This emphasizes the importance of careful decision making around the management of IR animals and indicates that re-testing alone may not be sufficient to reduce the risk posed by IR only herds in England and Wales

    An inquiry into the pedagogical practices of high school social studies teachers: Imagining and re-imagining becoming a teacher

    On three days in 2017-18 two university professor-researchers gathered with three high school teachers to talk about the experience of teaching social studies and about the insights, questions, and wonderments therefrom derived. The intent was to research the life and work of social studies teachers. The gatherings were funded by the Brandon school division (Brandon is a small city in the southwestern Manitoba (Canada). The research was conducted under the auspices of the Manitoba Education Research Network (MERN) with support from Manitoba Education and Training (MET). The underlying premise was that teachers, through their lived experience in the classroom and beyond, were able to access understandings unique to teaching and learning. This article chronicles, in part, what one teacher revealed about his conception of being a social studies teacher and his teaching experience and how, thereby, the understanding of social studies teaching is increased and provides a signal as regards what it might be to be a social studies teacher
