WhpR, an orphan transcriptional regulator of virulence in the pathogen of woody hosts Pseudomonas savastanoi pv. savastanoi
The genome of the olive tree pathogen Pseudomonas savastanoi pv. savastanoi (Psv) NCPPB 3335 encodes a region of about 15 kb named WHOP (from Woody Host and Pseudomonas) which is involved in the catabolism of aromatic compounds and is essential for the virulence of Psv in woody olive plants. This region is shared with other strains of Pseudomonas syringae pathovars infecting woody hosts, but it is absent in strains infecting herbaceous plants. The WHOP region is organized into four operons, antABC (metabolism of anthranilate), catBCA (catabolism of catechol) ipoBCA (oxigenase activity) and dhoAB (degradation of fenolic compounds) and three independently transcribed genes, antR (positive regulator of the antABC operon), PSA3335_3206 (aerotaxis receptor) and whpR (putative AraC family regulator). In this study we identified two domains in WhpR, a DBD (DNA bingding domain), characterised by a classical HTH (Helix-Turn-Helix) motif and an AraC-like bingding domain. BlastP searches showed that no homologs (≥ 60%) of this protein are found outside the P. syringae complex. We also addressed the role of WhpR in virulence by the construction of ΔwhpR mutants in several P. savastanoi strains isolated from olive and oleander (P. savastanoi pv. nerii). Moreover, quantitative real-time PCR (RT-qPCR) analysis of Psv NCPPB 3335 and its ΔwhpR mutant revealed that WhpR is a negative regulator of most of the operons encoded in the WHOP region. Our future aims are to elucidate the mechanism of WhpR-dependent regulation and to determine whether other genes codified outside the WHOP region are also regulated by WhpR.Universidad de Málaga. Inverse Modeling of Human Knee Joint Based on Geometry and Vision Systems for Exoskeleton Applications
Current trends in Robotics aim to close the gap that separates technology and humans, bringing novel robotic devices in order to improve human performance. Although robotic exoskeletons represent a breakthrough in mobility enhancement, there are design challenges related to the forces exerted to the users’ joints that result in severe injuries. This occurs due to the fact that most of the current developments consider the joints as noninvariant rotational axes. This paper proposes the use of commercial vision systems in order to perform biomimetic joint design for robotic exoskeletons. This work proposes a kinematic model based on irregular shaped cams as the joint mechanism that emulates the bone-to-bone joints in the human body. The paper follows a geometric approach for determining the location of the instantaneous center of rotation in order to design the cam contours. Furthermore, the use of a commercial vision system is proposed as the main measurement tool due to its noninvasive feature and for allowing subjects under measurement to move freely. The application of this method resulted in relevant information about the displacements of the instantaneous center of rotation at the human knee joint
Symbiosis of mycorrhizal fungi with Rubus spp., in four farms of the municipality of Pamplona, Norte de Santander
Introduction− The blackberry crop is part of the prioritized productive chains in Norte of Santander, however, there are no research results on the benefits of Arbuscular Mycorrhizal Fungi (AMF) and the level of natural colonization of these with Rubus sp.
Objective− To determine the levels of colonization and mycorrhization of AMF in commercial varieties of blackberry and wild species of Rubus sp. in areas of the municipality of Pamplona Norte de Santander
Methodology− This work was carried out in the Sabaneta Baja, Sabaneta Alta and San Francisco veredas of the municipality of Pamplona, sampling four farms. Root samples were collected from commercial blackberry crops and neighboring wild species to evaluate the percentage of colonization and mycorrhization by AMF. ANOVAS were carried out using the SPSS statistical package.
Results− High levels of AMF symbiosis were found in the three commercial varieties of blackberry (colonization between 92 to 98%) and in the three species of Rubus sp. (colonization between 71 to 84%), as well as the presence of mycorrhizal structures of AMF in blackberry varieties and native species.
Conclusions− The biological variables of the AMF did not vary statistically between the commercial varieties, or wild species, but did between the different farms and in the variety x farm interaction for the commercial varieties. The biological variables of the AMF in the commercial varieties differed from the wild species located on the same farm.
An analysis of the evolving comoving number density of galaxies in hydrodynamical simulations
The cumulative comoving number-density of galaxies as a function of stellar
mass or central velocity dispersion is commonly used to link galaxy populations
across different epochs. By assuming that galaxies preserve their
number-density in time, one can infer the evolution of their properties, such
as masses, sizes, and morphologies. However, this assumption does not hold in
the presence of galaxy mergers or when rank ordering is broken owing to
variable stellar growth rates. We present an analysis of the evolving comoving
number density of galaxy populations found in the Illustris cosmological
hydrodynamical simulation focused on the redshift range . Our
primary results are as follows: 1) The inferred average stellar mass evolution
obtained via a constant comoving number density assumption is systematically
biased compared to the merger tree results at the factor of 2(4) level
when tracking galaxies from redshift out to redshift ; 2) The
median number density evolution for galaxy populations tracked forward in time
is shallower than for galaxy populations tracked backward in time; 3) A similar
evolution in the median number density of tracked galaxy populations is found
regardless of whether number density is assigned via stellar mass, stellar
velocity dispersion, or dark matter halo mass; 4) Explicit tracking reveals a
large diversity in galaxies' assembly histories that cannot be captured by
constant number-density analyses; 5) The significant scatter in galaxy linking
methods is only marginally reduced by considering a number of additional
physical and observable galaxy properties as realized in our simulation. We
provide fits for the forward and backward median evolution in stellar mass and
number density and discuss implications of our analysis for interpreting
multi-epoch galaxy property observations.Comment: 18 pages, 11 figures, submitted to MNRAS, comments welcom
