    Characterization of Platelet Function and Hemostasis in the Cib1-/- Mouse

    Platelet activation initiates platelet-platelet aggregation, platelet-extracellular matrix interactions and thrombus formation. Platelet aggregation requires activation of the αIIbβ3 integrin, an event regulated by the integrin cytoplasmic tails. CIB1 binds to the cytoplasmic tail of the integrin αIIb subunit. CIB1 has been demonstrated to regulate αIIbβ3 activation in an acute setting. We studied the Cib1- /- mouse to determine if the chronic loss of CIB1 causes a defect in hemostasis. Plasma analysis exposed several significant differences in protein levels between the Cib1+/+ and Cib1-/- plasma. Most notable were the increases in von Willebrand factor and soluble P-selectin. To understand the effect of two pro-thrombotic proteins circulating at higher levels in the plasma, we examined bleeding time and arterial thrombus formation discovered no significant difference in thrombus formation or stability between the Cib1+/+ and the Cib1-/- mice. Previous overexpression and knockdown studies in murine megakaryocytes from our lab demonstrated that CIB1 inhibits platelet integrin αIIbβ3 activation. We analyzed Cib1-/- mice to determine the function of CIB1 in platelets in vivo. We found that although these mice had no overt platelet phenotype, mRNA level of CIB1 homolog CIB3 was increased in Cib1-/- megakaryocytes. In vitro binding experiments demonstrate that recombinant CIB1, -2 and -3 bound specifically to an αIIb cytoplasmic tail peptide. Subsequent protein modeling experiments indicated that CIBs 1-3 each have a highly conserved hydrophobic binding pocket. Therefore, the potential exists for compensation by these CIB family members for loss of CIB1, thereby preventing pathologic thrombus formation in Cib1-/- mice. Interestingly, the Cib1-/- mouse does have a phenotype that may provide a model for studying critical levels of plasma proteins required for normal hemostasis.Doctor of Philosoph

    El objetivo principal del presente trabajo de tesis es estudiar los procesos fotosensibilizados por Lumazina sobre biomoléculas y células eucariotas. Para ello, en primer lugar, se realizó la caracterización de algunas propiedades fotofísicas y fotoquímicas de Lumazina. A continuación se mencionan los objetivos particulares que trazaron el rumbo de las tareas y experiencias descriptas en esta tesis: Estudiar y caracterizar las propiedades fotoquímicas y fotofísicas de Lumazina. - Caracterización de los estados electrónicos excitados singletes y tripletes. - Evaluación del comportamiento fotoquímico de Lumazina bajo condiciones de irradiación continua con radiación UV. - Evaluar la capacidad que presenta Lumazina de generar especies reactivas de oxígeno, en particular, oxígeno singlete, anión superóxido y peróxido de hidrógeno. Analizar la participación de Lumazina en diferentes procesos u oxidaciones fotosensibilizadas de implicancia biológica. - Estudio de procesos de degradación de nucleótidos purínicos fotoinducida por Lumazina, y dilucidación de los mecanismos implicados. Análisis de la influencia del pH en los mecanismos de reacción. Caracterización de los fotoproductos generados en la oxidación fotosensibilizada.     Chemical and photochemical properties of chloroharmine derivatives in aqueous solutions

    Thermal and photochemical stability (ΦR), room temperature UV-vis absorption and fluorescence spectra, fluorescence quantum yields (ΦF) and lifetimes (τF), quantum yields of hydrogen peroxide (ΦH2O2) and singlet oxygen (ΦΔ) production, and triplet lifetimes (τT) have been obtained for the neutral and protonated forms of 6-chloroharmine, 8-chloroharmine and 6,8-dichloroharmine, in aqueous media. When it was possible, the effect of pH and oxygen concentration was evaluated. The nature of electronic transitions of protonated and neutral species of the three investigated chloroharmines was established using Time-Dependent Density Functional Theory (TD-DFT) calculations.     The Effect of Ventricular Assist Devices on Post-Transplant Mortality An Analysis of the United Network for Organ Sharing Thoracic Registry

    ObjectivesThis study sought to determine the relationship between pre-transplant ventricular assist device (VAD) support and mortality after heart transplantation.BackgroundIncreasingly, VADs are being used to bridge patients to heart transplantation. The effect of these devices on post-transplant mortality is unclear.MethodsPatients 18 years or older who underwent first-time, single-organ heart transplantation in the U.S. between 1995 and 2004 were included in the analyses. This study compared 1,433 patients bridged with intracorporeal and 448 patients bridged with extracorporeal VADs with 9,455 United Network for Organ Sharing status 1 patients not bridged with a VAD with respect to post-transplant mortality. Because the proportional hazards assumption was not met, hazard ratios (HRs) for different time periods were estimated.ResultsIntracorporeal VADs were associated with an HR of 1.20 (95% confidence interval [CI]: 1.02 to 1.43; p = 0.03) for mortality in the first 6 months after transplant and an HR of 1.99 (95% CI: 1.44 to 2.75; p < 0.0001) beyond 5 years. Between 6 months and 5 years, the HRs were not significantly different from 1. Extracorporeal VADs were associated with an HR of 1.91 (95% CI: 1.53 to 2.37; p < 0.0001) for mortality in the first 6 months and an HR of 2.93 (95% CI: 1.19 to 7.25; p = 0.02) beyond 5 years. The HRs were not significantly different from 1 between 6 months and 5 years, except for an HR of 0.23 (95% CI: 0.06 to 0.91; p = 0.04) between 24 and 36 months.ConclusionsExtracorporeal VADs are associated with higher mortality within 6 months and again beyond 5 years after transplantation. Intracorporeal VADs are associated with a small increase in mortality in the first 6 months and a clinically significant increase in mortality beyond 5 years. These data do not provide evidence supporting VAD implantation in stable United Network for Organ Sharing status I patients awaiting heart transplantation

    Drosophila APC2 and APC1 Play Overlapping Roles in Wingless Signaling in the Embryo and Imaginal Discs

    The regulation of signal transduction plays a key role in cell fate choices, and its disregulation contributes to oncogenesis. This duality is exemplified by the tumor suppressor APC. Originally identified for its role in colon tumors, APC family members were subsequently shown to negatively regulate Wnt signaling in both development and disease. The analysis of the normal roles of APC proteins is complicated by the presence of two APC family members in flies and mice. Previous work demonstrated that, in some tissues, single mutations in each gene have no effect, raising the question of whether there is functional overlap between the two APCs or whether APC-independent mechanisms of Wnt regulation exist. We addressed this by eliminating the function of both APC genes simultaneously. We find that APC1 and APC2 play overlapping roles in regulating Wingless signaling in the embryonic epidermis and the imaginal discs. Surprisingly, APC1 function in embryos occurs at levels of expression nearly too low to detect. Further, the overlapping functions exist despite striking differences in the intracellular localization of the two APC family members

    Coral Bleaching Independent of Photosynthetic Activity

    SummaryThe global decline of reef-building corals is due in part to the loss of algal symbionts, or “bleaching,” during the increasingly frequent periods of high seawater temperatures [1, 2]. During bleaching, endosymbiotic dinoflagellate algae (Symbiodinium spp.) either are lost from the animal tissue or lose their photosynthetic pigments, resulting in host mortality if the Symbiodinium populations fail to recover [3]. The >1,000 studies of the causes of heat-induced bleaching have focused overwhelmingly on the consequences of damage to algal photosynthetic processes [4–6], and the prevailing model for bleaching invokes a light-dependent generation of toxic reactive oxygen species (ROS) by heat-damaged chloroplasts as the primary trigger [6–8]. However, the precise mechanisms of bleaching remain unknown, and there is evidence for involvement of multiple cellular processes [9, 10]. In this study, we asked the simple question of whether bleaching can be triggered by heat in the dark, in the absence of photosynthetically derived ROS. We used both the sea anemone model system Aiptasia [11, 12] and several species of reef-building corals to demonstrate that symbiont loss can occur rapidly during heat stress in complete darkness. Furthermore, we observed damage to the photosynthetic apparatus under these conditions in both Aiptasia endosymbionts and cultured Symbiodinium. These results do not directly contradict the view that light-stimulated ROS production is important in bleaching, but they do show that there must be another pathway leading to bleaching. Elucidation of this pathway should help to clarify bleaching mechanisms under the more usual conditions of heat stress in the light

    Light-induced full aromatization and hydroxylation of 7-methoxy-1-methyl-3,4-dihydro-2H-pyrido[3,4-b]indole alkaloid: Oxygen partial pressure as a key modulator of the photoproducts distribution

    Full-aromatic and partially hydrogenated β-carboline (βC) derivatives constitute a group of alkaloids widely distributed in a great variety of living systems. In plants and bacteria, tetrahydro-βCs are the primary product of the Pictet-Spengler enzymatically catalyzed condensation. Tetrahydro-βC skeleton is further modified giving rise to the formation of a vast set of derivatives including dihydro- and full-aromatic βCs. However, in most of the cases, the later processes still remain unclear and other sources, such as photo-triggered reactions, deserve to be explored. In this context, the photophysic and photochemistry of 7-methoxy-1-methyl-3,4-dihydro-2H-pyrido[3,4-b]indole or harmaline (Hlina) in aqueous solution is reported herein. UV–visible absorption and fluorescence emission spectroscopy coupled with multivariate data analysis (PARAFAC), HPLC and HRESI-MS techniques were used for both quantitative and qualitative analysis. The formation singlet oxygen and hydrogen peroxide reactive oxygen species (ROS) was quantified and their role together with the influence of pH and oxygen partial pressure on the photochemical degradation of HlinaH+ was assessed. We report herein the first study on photochemical full-aromatization of a dihydro-βC derivative.
