    Does a 7-day restriction on the use of social media improve cognitive functioning and emotional well-being?:Results from a randomized controlled trial

    Introduction Screen time apps that allow smartphone users to manage their screen time are assumed to combat negative effects of smartphone use. This study explores whether a social media restriction, implemented via screen time apps, has a positive effect on emotional well-being and sustained attention performance. Methods A randomized controlled trial (N = 76) was performed, exploring whether a week-long 50% reduction in time spent on mobile Facebook, Instagram, Snapchat and YouTube is beneficial to attentional performance and well-being as compared to a 10% reduction. Results Unexpectedly, several participants in the control group pro-actively reduced their screen time significantly beyond the intended 10%, dismantling our intended screen time manipulation. Hence, we analyzed both the effect of the original manipulation (i.e. treatment-as-intended), and the effect of participants’ relative reduction in screen time irrespective of their condition (i.e. treatment-as-is). Neither analyses revealed an effect on the outcome measures. We also found no support for a moderating role of self-control, impulsivity or Fear of Missing Out. Interestingly, across all participants behavioral performance on sustained attention tasks remained stable over time, while perceived attentional performance improved. Participants also self-reported a decrease in negative emotions, but no increase in positive emotions. Conclusion We discuss the implications of our findings in light of recent debates about the impact of screen time and formulate suggestions for future research based on important limitations of the current study, revolving among others around appropriate control groups as well as the combined use of both subjective and objective (i.e., behavioral) measures

    Beliefs in times of corona: Investigating the relationship between media use and COVID-19 conspiracy beliefs over time in a representative Dutch sample

    We investigated the relationship between different media sources (traditional media, online news media, online health sources, social media) and COVID-19 related conspiracy beliefs, and how these change over time, using four-wave panel data from a representative sample of the Dutch population (N = 1,166). Across waves, 0.1%–3.4% of our sample were certain the selected conspiracy theories were true, though this belief was unstable over time. Random intercept cross-lagged panel models revealed that individuals’ temporary level of conspiracy beliefs did not significantly depend on their temporary level of media use at a previous occasion, or vice versa. However, significant correlations at the group level indicated that more frequent use of health-related and social media sources were associated with higher levels of conspiracy beliefs. These results suggest that relationships between media use and conspiracy beliefs are nuanced. Underlying processes should be investigated to develop tailored communication strategies to combat the ongoing infodemic

    The influence of BRAF and KRAS mutation status on the association between aspirin use and survival after colon cancer diagnosis

    Background: Use of aspirin after diagnosis of colon cancer has been associated with improved survival. Identification of cancer subtypes that respond to aspirin treatment may help develop personalized treatment regimens. The aim of this study was to investigate the influence of BRAF and KRAS mutation status on the association between aspirin use and overall survival after colon cancer diagnosis. Methods: A random selection of 599 patients with colon cancer were analyzed, selected from the Eindhoven Cancer Registry, and BRAF and KRAS mutation status was determined. Data on aspirin use (80 mg) were obtained from the PHARMO Database Network. Parametric survival models with exponential (Poisson) distribution were used. Results: Aspirin use after colon cancer diagnosis was associated with improved overall survival in wild-type BRAF tumors, adjusted rate ratio (RR) of 0.60 (95% CI 0.44-0.83). In contrast, aspirin use in BRAF mutated tumors was not associated with an improved survival (RR 1.11, 95% CI 0.57-2.16). P-value for interaction was non-significant. KRAS mutational status did not differentiate in the association between aspirin use and survival. Conclusion: Low-dose aspirin use after colon cancer diagnosis was associated with improved survival in BRAF wild-type tumors only. However, the large confidence interval of the rate ratio for the use of aspirin in patients with BRAF mutation does not rule out a possible benefit. These results preclude BRAF and KRAS mutation status to be used as a marker for individualized treatment with aspirin, if aspirin becomes regular adjuvant treatment for colon cancer patients in the future

    GlxA is a new structural member of the radical copper oxidase family and is required for glycan deposition at hyphal tips and morphogenesis of <i>Streptomyces lividans</i>

    Streptomyces lividans displays a distinct dependence on copper to fully initiate morphological development. Evidence has accumulated to implicate the participation of an extracytoplasmic cuproenzyme in morphogenesis. In the present study, we show that GlxA fulfils all criteria to be that cuproenzyme. GlxA is membrane associated and has an active site consisting of a mononuclear copper and a cross-linked Y-C cofactor. The domain organization of the tertiary structure defines GlxA as a new structural member of the mono-copper oxidase family, with copper co-ordination geometry similar to, but spectroscopically distinct from fungal galactose oxidase (Gox). EPR spectroscopy reveals that the oxidation of cupric GlxA generates a protein radical residing on the Y-C cross-link. A variety of canonical Gox substrates (including D-galactose) were tested but none were readily turned over by GlxA. A glxA null-mutant leads to loss of glycan accumulation at hyphal tips and consequently a drastically changed morphology both on solid substrates and in liquid-grown environments, a scenario similarly observed in the absence of the neighbouring glycan synthase CslA (cellulase synthase-like protein). In addition the glxA mutant has lost the stimulation of development by copper, supporting a model whereby the enzymatic action of GlxA on the glycan is required for development and morphology. From a biotechnology perspective, the open mycelium morphology observed with the glxA mutant in submerged culture has implications for use as an enzyme production host.</jats:p
