    Suggestive evidence of a multi-cytokine resistin pathway in humans and its role on cardiovascular events in high-risk individuals

    In cells and tissues resistin affects IL-1β, IL-6, IL-8, IL-12 and TNF-α expression, thus suggesting the existence of a multi-cytokine "resistin pathway". We investigated whether such pathway does exist in humans and, if so, if it is associated with cardiovascular risk factors and with major adverse cardiovascular events (MACE). Serum cytokines were measured in 280 healthy subjects from the Gargano Study 2 (GS2) whose BMI, waist circumference, HOMA IR, triglycerides, HDL-cholesterol, systolic and diastolic blood pressure data were available and in 353 patients with type 2 diabetes and coronary artery disease from the Gargano Heart Study (GHS)-prospective design (follow-up 5.4 ± 2.5 years; 71 MACE). In GS2, cytokines mRNA levels in white blood cells were also measured. In GS2, resistin mRNA was correlated with all cytokines expression (all p < 0.001), but IL-12B. Consistently, serum resistin was correlated with all serum cytokines (all p < 0.001), but IL-12. Expression (eRPS) and serum (sRPS) resistin pathway scores (excluding IL-12) were each other correlated (p < 0.001) and both associated with cardiovascular risk factors (all p < 0.01). In GHS, sRPS was independently associated with MACE (HR = 1.44, 95% CI = 1.10-1.90). Our data indicate the existence of a resistin pathway, which is associated with cardiovascular risk factors and which strongly and independently predicts MACE

    Neutron Star Radius-to-mass Ratio from Partial Accretion Disc Occultation as Measured through Fe Kα\alpha Line Profiles

    We present a new method to measure the radius-to-mass ratio (R/M) of weakly magnetic, disc-accreting neutron stars by exploiting the occultation of parts of the inner disc by the star itself. This occultation imprints characteristic features on the X-ray line profile that are unique and are expected to be present in low mass X-ray binary systems seen under inclinations higher than ~65 degrees. We analyse a NuSTAR observation of a good candidate system, 4U 1636-53, and find that X-ray spectra from current instrumentation are unlikely to single out the occultation features owing to insufficient signal-to-noise. Based on an extensive set of simulations we show that large-area X-ray detectors of the future generation could measure R/M to ~2{\div}3% precision over a range of inclinations. Such is the precision in radius determination required to derive tight constraints on the equation of state of ultradense matter and it represents the goal that other methods too aim to achieve in the future.Comment: 17 pages, 8 figures; this is a pre-print edition of an article that has been accepted for publication in the Astrophysical Journa

    Higher levels of osteoprotegerin and immune activation/immunosenescence markers are correlated with concomitant bone and endovascular damage in HIV-suppressed patients

    HIV-infected patients appear to have a significantly greater risk of non-AIDS comorbidities such as osteoporosis and atherosclerosis. Subjects with osteoporosis are at a higher risk of developing cardiovascular disease than those with normal bone mass, therefore a possible relation between these two conditions can be hypothesized. In the setting of HIV infection, several factors might contribute to bone disease and endothelial dysfunction. The aim of our study was to evaluate the relationship between bone and cardiovascular disease and to investigate the role of traditional factors, T-cell phenotype and osteoprotegerin in HIV positive subjects on effective antiretroviral therapy. We included 94 HIV positive subjects on antiretroviral therapy with virological suppression and 41 healthy subjects matched for age and gender as a control group. Carotid-Intima Media Thickness (c-IMT) and bone mineral density (BMD) were performed by ultrasound and DEXA, respectively. CD4+/CD8+ T-cell activation, senescence and osteoprotegerin plasma levels were measured by flow-cytometry and ELISA, respectively. Among HIV positive patients, 56.4% had osteopenia/osteoporosis and 45.7% had pathological c-IMT (>0.9mm). Subjects with pathological c-IMT and BMD exhibited higher CD4+ and CD8+ activated, CD8+ senescent and osteoprotegerin than subjects with normal c-IMT and BMD. HIV positive subjects with osteopenia/osteoporosis had higher c-IMT than subjects with normal BMD, and linear regression analysis showed a negative correlation between BMD and c-IMT. Several factors are implicated in the pathogenesis of non-AIDS comorbidities in HIV positive patients. Osteoprotegerin together with inflammation and immunosenescence in HIV positive patients could affect bone and vascular system and could be considered as a possible common link between these two diseases

    Meat quality in three pork commercial lines of different slaughter weights

    O objetivo deste trabalho foi determinar as características de qualidade da carne de suínos de diferentes linhagens genéticas, em diferentes pesos de abate. Neste estudo foram utilizados 88 suínos por linhagem, fêmeas e machos castrados, com idade e peso médio iniciais de 74 dias e 30kg, respectivamente, pertencentes a três linhagens genéticas distintas, designadas de AgroceresPic, Dalland e Seghers. A etapa experimental foi dividida em quatro fases (Crescimento I, Crescimento II, Terminação I e Terminação II). Ao final de cada etapa, foram abatidos 60 animais (10 por linhagem/sexo), para as análises de qualidade da carne. O pH e a temperatura foram determinados a 1 e 24 horas post mortem. Foram retiradas amostras para as determinações de cor, perda de água por exsudação (PAE), perda de água por cocção (PAC) e força de Cisalhamento. As amostras provenientes das carcaças dos animais da linhagem AgroceresPic e Dalland apresentaram valores médios de PAE superiores aos da linhagem Seghers. Os valores médios de PAC foram diferentes entre as diferentes fases. Na fase Crescimento II, as carnes provenientes das carcaças das fêmeas Dalland foram mais duras, ou seja, com maiores valores de força de cisalhamento, que as carnes dos machos. Entretanto, na linhagem Seghers, as carnes dos machos foram mais duras. Observou-se diferença de L* entre os sexos da linhagem AgroceresPic nas fases Terminação I e II e, na linhagem Dalland, na Terminação II. As linhagens Dalland e Seghers apresentaram carnes com resultados superiores de a*, parâmetro característico da cor vermelha (a*>0). Pode-se concluir, portanto, que as características de qualidade da carne de suínos (pH, perda de água por exsudação, cor e maciez) podem variar entre grupos genéticos, entre sexos e entre diferentes pesos ao abate.The aim of this study was to determine the meat quality characteristics of swine sire lines from different slaughter weights. In this study were used 88 castrated piglets (castrated males and females) per sire line (AgroceresPic, Dalland and Seghers), with initial age of 74 days and live weight of 30kg. The experiment was divided in four phases (Growing I, Growing II, Finishing I and Finishing II). At the end of each experimental phase, 60 animals were slaughtered (10 by lineage/sex) for meat quality determinations. The pH and the temperature were determined 1 and 24 hours post mortem. Meat samples were collected for color, water loss by exudation (WLE), water loss by cooking (WLC) and shear force determinations. Samples of AgroceresPic and Dalland carcasses presented average WLE values superior than those of Seghers. The average values of WLC were different between the phases. In Growth II phase, the meat from Dalland female carcasses were harder, this is, with higher shear force values than the meats from the males. However, for Seghers, the meat from males was harder. Difference in the parameter L* was observed between sex for AgroceresPic animals in Termination I and II phases and, in Dalland animals in Termination II. The Dalland and Seghers animals presented meats with higher a* values, parameter which is characteristic of redness (a*>0). Therefore, it can be concluded that pig meat quality characteristics (pH, loss of water, color and texture) can vary between genetic groups, between sex and between different slaughter weights

    O objetivo do presente estudo é verificar a metodologia aplicada em pesquisas realizadas no âmbito das instituições de ensino superior brasileiras e publicadas pela Revista de Ciências da Administração (RCA) da Universidade Federal de Santa Catarina (UFSC), no período de 2011 a 2015. A citada Revista tem como objeto o desenvolvimento de temas na área de administração e áreas correlatadas. Neste contexto, este estudo analisa as metodologias empregadas nas pesquisas relatadas por tal periódico, utilizando, para tanto, a pesquisa bibliográfica, documental e descritiva, com abordagem qualitativa, além de elementos de estudo de caso. Os resultados obtidos indicam que, apesar do esforço dos autores na abordagem dos mais variados temas, a metodologia empregada por eles não apresenta diversidade significativa, havendo prevalência dos estudos com abordagem qualitativa e com objetivos exploratórios ou descritivos, o que se constitui em características típicas de pesquisas na área das ciências sociais aplicadas, em que se enquadra a Administração. Por fim, este artigo expõe a amplitude da metodologia científica, apesar de revelar a predileção para a adesão a métodos e técnicas típicas da abordagem qualitativa, confirmando a tendência apontada pela teoria para à área de estudo a que se referem os artigos analisados – a Social aplicada

    3D Microfluidic model for evaluating immunotherapy efficacy by tracking dendritic cell behaviour toward tumor cells

    Immunotherapy efficacy relies on the crosstalk within the tumor microenvironment between cancer and dendritic cells (DCs) resulting in the induction of a potent and effective antitumor response. DCs have the specific role of recognizing cancer cells, taking up tumor antigens (Ags) and then migrating to lymph nodes for Ag (cross)-presentation to naïve T cells. Interferon-α-conditioned DCs (IFN-DCs) exhibit marked phagocytic activity and the special ability of inducing Ag-specific T-cell response. Here, we have developed a novel microfluidic platform recreating tightly interconnected cancer and immune systems with specific 3D environmental properties, for tracking human DC behaviour toward tumor cells. By combining our microfluidic platform with advanced microscopy and a revised cell tracking analysis algorithm, it was possible to evaluate the guided efficient motion of IFN-DCs toward drug-treated cancer cells and the succeeding phagocytosis events. Overall, this platform allowed the dissection of IFN-DC-cancer cell interactions within 3D tumor spaces, with the discovery of major underlying factors such as CXCR4 involvement and underscored its potential as an innovative tool to assess the efficacy of immunotherapeutic approaches

    TECNOLOGIA DA INFORMAÇÃO NO AMBIENTE UNIVERSITÁRIO: UMA CONTRIBUIÇÃO PARA A GESTÃO DO CONHECIMENTO

    Um dos papeis relevantes que a tecnologia da informação (TI) exerce na gestão do conhecimento é ampliar o alcance e intensificar a velocidade de transferência da informação e do conhecimento. Porém, vale mencionar que a TI fornece apenas a infra-estrutura necessária à gestão do conhecimento. Em outras palavras, visa oferecer suporte as atividades de geração, codificação ou transferência do conhecimento. O intuito é disponibilizar o conhecimento para todas as pessoas na organização. Nesse sentido, o presente artigo visa discutir como a tecnologia da informação no ambiente universitário pode contribuir para a gestão do conhecimento. Diversas tecnologias têm sido utilizadas para auxiliar a gestão do conhecimento no contexto organizacional, entre elas destacam-se: internet, intranet, extranet, workflow, groupware, gerenciamento eletrônico de documentos, mapas do conhecimento, data mining, data warehousing, entre outras. A esse respeito, sabe-se que esse novo contexto está exigindo das Instituições de Ensino Superior (IES) uma maior responsabilidade no gerenciamento da informação e do conhecimento. Nesse sentido, entende-se que os dirigentes universitários precisam dispor de uma gestão do conhecimento a fim de detectar oportunidades de interação com a sociedade, bem como obter o aumento da vantagem competitiva. Para dar subsídios a essa discussão, optou-se por um artigo de revisão, isto é, será realizada uma pesquisa bibliográfica envolvendo diversos autores

    Multiple AGN in the crowded field of the compact group SDSSJ0959+1259

    We present a multi-wavelength study of a newly discovered compact group (CG), SDSS J0959+1259, based data from XMM-Newton, SDSS and the Calar Alto optical imager BUSCA. With a maximum velocity offset of 500 km s1^{-1}, a mean redshift of 0.035, and a mean spatial extension of 480 kpc, this CG is exceptional in having the highest concentration of nuclear activity in the local Universe, established with a sensitivity limit LX>4×_{X}>4\times 1040^{40} erg s1^{-1} in 2--10 keV band and R-band magnitude MR<19M_R < -19. The group is composed of two type-2 Seyferts, one type-1 Seyfert, two LINERs and three star forming galaxies. Given the high X-ray luminosity of LINERs which reaches 1041\sim 10^{41} erg s1^{-1}, it is likely that they are also accretion driven, bringing the number of active nuclei in this group to to 5 out of 8 (AGN fraction of 60\%). The distorted shape of one member of the CG suggests that strong interactions are taking place among its galaxies through tidal forces. Therefore, this system represents a case study for physical mechanisms that trigger nuclear activity and star formation in CGs.Comment: 8 pages, 4 figures and 4 tables.     Concentration of steroid hormones in the follicular fluid of mature and immature ovarian follicles of patients with polycystic ovary syndrome submitted to in vitro fertilization

    OBJETIVO: avaliar a concentração dos hormônios esteroides no fluido folicular (FF) de folículos pequenos (10-14 mm) e grandes (&gt; 18 mm) de mulheres com síndrome dos ovários policísticos (SOP) submetidas à hiperestimulação ovariana controlada (HOC) e ciclos de fertilização in vitro (FIV). MÉTODOS: estudo caso-controle foi conduzido em 13 mulheres inférteis com SOP (17 ciclos) e 31 mulheres inférteis por fator masculino - Grupo Controle (31 ciclos). Os FF foram aspirados individualmente e dividos em 4 grupos: G1 (FF pequeno do Grupo Controle), G2 (FF pequeno do grupo SOP), G3 (FF grande do Grupo Controle) e G4 (FF grande do grupo SOP). A metodologia utilizada para as dosagens de estradiol, progesterona e &#946;-hCG foi a quimioluminescência, e de testosterona e androstenediona o radioimunoensaio. Para a análise das dosagens hormonais no FF entre os grupos SOP e Controle utilizou-se o teste t não-pareado, e para a comparação entre os quatro grupos, o ANOVA. Para a taxa de gravidez, foi utilizado o teste exato de Fisher. RESULTADOS: os folículos pequenos dos dois grupos tiveram valores menores de progesterona (8.435±3.305 ng/mL) comparados aos grandes (10.280±3.475 ng/mL), com valor de p<0,01. Os níveis de progesterona de todos os folículos do grupo SOP (8.095±4.151 ng/mL) foram inferiores ao Controle (9.824±3.128 ng/mL), com valor de p=0,03. Os níveis de testosterona diferiram entre G1 (326,6±124,4 ng/dL) e G3 (205,8±98,91 ng/dL), com valor de p<0,001, e entre G3 (205,8±98,91 ng/dL) e G4 (351,10±122,1 ng/dL), com valor de p<0,001. Os folículos pequenos (508,9±266 ng/dL) apresentaram valores superiores de testosterona comparados aos grandes (245,10±123 ng/dL), com valor de p<0,0001. As taxas de gravidez não diferiram entre os grupos SOP (5/13, 38,5%) e Controle (9/31, 40,9%), com valor de p=072. CONCLUSÕES: mulheres com SOP apresentam altas concentrações de testosterona no FF, independentemente do estágio de desenvolvimento folicular, e níveis de progesterona diminuídos, sugerindo que fatores parácrinos podem inibir sua secreção pelas células foliculares. As taxas de gravidez mostraram que o tratamento de HOC e FIV é uma boa opção para mulheres com infertilidade secundária à SOP.PURPOSE: to evaluate the concentration of steroid hormones in follicular fluid (FF) of small (10-14 mm) and large (&gt; 18 mm) follicles of women with polycystic ovary syndrome (PCOS) submitted to controlled ovarian hyperstimulation (COH) and in vitro fertilization (IVF) cycles. METHODS: a case-control study was conducted on 13 infertile women with PCOS (17 cycles) and 31 infertile women due to male factor - Control Group (31 cycles). FF was aspirated individually and divided into four groups: G1 (FF of small follicles of the Control Group), G2 (FF of small follicles of the PCOS group), G3 (FF of large follicles of the Control Group) and G4 (FF of large follicles of the PCOS group). Estrogen, progesterone and &#946;-hCG were determined by chemiluminescence, and testosterone and androstenedione by radioimmunoassay. The unpaired t-test was used to compare the hormone determinations in the FF of the PCOS and Control Groups, and the four groups were compared by ANOVA. Fisher's exact test was used to compare the pregnancy rates. RESULTS: the small follicles of the two groups had lower progesterone levels (8,435±3,305 ng/mL) than large follicles (10,280±3,475 ng/mL), p-value <0.01. The progesterone levels of all follicles of group PCOS (8,095±4,151 ng/mL) were lower than Control (9,824±3,128 ng/mL), p-value =0.03. Testosterone differed between G1 (326.6±124.4 ng/dL) and G3 (205.8±98.91 ng/dL), p-value <0.001, and between G3 (205.8±98.91 ng/dL) and G4 (351.10±122.1ng/dL), p-value <0.001. Small follicles had higher testosterone levels (508.9±266 ng/dL) than large follicles (245.10±123 ng/dL), p-value <0.0001. The pregnancy rates did not differ between the PCOS (5/13, 38.5%) and the Control groups (9/31, 40.9%), p-value =072. CONCLUSIONS: women with PCOS had high testosterone concentrations in the FF, regardless of the stage of follicle development, and reduced progesterone levels, suggesting that paracrine factors may inhibit the secretion of the latter by follicular cells.
