    Investigating pathways in tumour cell migration and invasion in response to novel isoquinolinone compounds

    Abstract The ability of cancer cells to disseminate from a primary tumour and invade via the breaching of surrounding tissue vasculature is a hallmark of the metastatic phenotype. Cancer cells are highly dynamic adopting a wide range of mechanisms in order to accomplish this enhanced migrational activity. This study looks at the mechanisms behind enhanced migration and invasion in vitro in two highly aggressive cell lines in response to the heterocyclic aromatic organic Isoquinolinone compounds. Here we show that the structurally similar A5 and A3 compounds reduce adhesion of MDA-MB-231 cells to Fibronectin, Matrigel, and Type I collagen which was paralleled in A5 treated HT1080 cells on these matrix components. Compound A5 also induced 2D migration of MDA-MB-231 cells on all matrix substrates. Cell movement of MDA-MB-231 cells through 3D matrices was modestly enhanced by A5 and significantly enhanced by A3. Using gene expression arrays we were able to identify fifteen genes whose expression was regulated by the isoquinolinone compounds and heavily implicated in the process of cell motility. Moreover, we established that these genes are differentially regulated by the compounds over a 24 hour period with expression typically being more enhanced at the earlier time points of 4- and 8- hours and suppressed at 24 hours. The main findings presented in this thesis reveal many proteins involved in cell motility that have not been studied in MDA-MB-231 or HT1080 cells. These results lend support that compound treated cell lines activate several pathways in cell migration and invasion that warrant further investigation to elucidate the precise mechanism of these proteins involved and to develop novel inhibitors for some of the more obscurely studied, but heavily implicated proteins such as RhoQ

    Point Contact Spectra on YBa2_2Cu3_3O7x_{7-x}/La0.7_{0.7}Ca0.3_{0.3}MnO3_3 bilayers

    We present conductance characteristics of point contact junctions realized between a normal Pt-Ir tip and YBa2_2Cu3_3O7x_{7-x}/La0.7_{0.7}Ca0.3_{0.3}MnO3_3 (YBCO/LCMO) bilayers. The point contact characteristics show a zero bias conductance peak, as a consequence of the formation of Andreev bound states at the YBCO Fermi level. The temperature evolution of the spectra reveals a depressed zero bias peak and a reduced superconducting energy gap, both explainable in terms of spin polarization effects due to the LCMO layer.Comment: 4 pages, 4 EPS figures.     O presente Caso Pedagógico foi desenvolvido no âmbito de Marketing Internacional com enfoque no caso de uma conceituada marca de gelados gourmet – Santini. Com mais de 60 anos de história, a marca foi recentemente alvo de reestruturação e reposicionamento, como parte da sua estratégia de expansão. A abertura de uma nova loja na Baixa de Lisboa, em 2008, foi o primeiro de vários passos neste sentido. Sempre priorizando o consumidor final, a marca levou a cabo um conjunto de medidas inovadoras, de forma a ganhar visibilidade entre os consumidores e quota de mercado junto da concorrência. Um exemplo de excelência em todo o processo de expansão, que pode e deve ser analisado como exemplo a seguir por empresas na mesma situação. Sem medo de assumir riscos, apesar de consciente da situação económica e financeira vivida em Portugal, o Santini consegue alargar os seus limites físicos e barreiras emotivas para chegar mais perto dos consumidores e para satisfazer as suas necessidades. O presente Caso Pedagógico faz a análise da marca juntamente com uma análise descritiva e quantitativa do mercado dos gelados e do seu consumo em Portugal. Focado nos principais valores do Santini e na sua brand idea, este Caso será uma referência no estudo do Marketing Estratégico e Branding, oferecendo um contributo bastante relevante dada a componente teórico-prática do tema em análise, como uma situação aplicável a tantas outras empresas e marcas.The present Case Study was developed as a final thesis of International Marketing scope; it is focus on a regional and traditional Portuguese ice cream brand – Santini. The well established brand in gourmet ice cream, has expanded its business, opened a new store, and proceeded with several other strategic changes. Brand managers are extremely focused on maintaining the good reputation of the brand assuming the responsibility of quality assurance, homemade production and excellence of the service provided with familiarity and simplicity, to gain recognition. The Case Study comes up with an analysis of the actual brand, focus on its main values and core idea; above all the main end is to understand how it leads to new opportunities of growth, in times of financial crises and savings, as the current situation that the Portuguese society is going through. Using the information kindle provided by the Company, the Case offers challenges, difficulties and keys to the success, which the reader should understand, analyze and solve, with innovation and creativity. Also, a quality and quantity study of Portuguese industry and consumer behavior regarding ice creams is going to be developed.     The present report is part of the curricular internship of the Master’s degree in Intercultural Studies for Business, from the Porto Accounting and Business School (Instituto Superior de Contabilidade e Administração do Porto). This internship took place at the tourism field related company, Dixo’s Oporto Apartments. Globalization has been increasing at unprecedented rates and with that, the limits and the borders between countries and nations have been reducing. As a result, there has been a marked increase in the need for intercultural communication as it becomes reality and people depend more on this type of communication. The tourism sector is no exception from the inevitable changes imposed by globalization, and it too depends on intercultural communication. Cities such as Porto with a great flux of tourism rely on the finest quality services delivered by leading professionals. High standards can only be achieved by those who are used to intercultural communication and are familiarized with concepts such as globalization and interculturality. The purpose of this report is to describe and understand, at the most precise level the activities carried out during the internship based on the concepts and knowledge learnt during the master’s degree.O presente relatório insere-se no âmbito do estágio curricular do Mestrado em Estudos Interculturais para Negócio, no Instituto Superior de Contabilidade e Administração do Porto (ISCAP).     on the statics of curved masonry structures via numerical models

    Purpose This paper aims to review recent literature results on the equilibrium problem and the strengthening design of masonry vaults. Design/methodology/approach A Lumped Stress Method (LSM) is considered within the Heyman's safe theorem, based on the definition of thrust surface of a masonry curved structure. In particular, the static problem of the vault is formulated by introducing a membrane continuous of the studied masonry structure to associate with a spatial truss through a nonconforming variational approximation of the thrust surface and membrane stress potential. A tensegrity approach based on a minimal mass design strategy, different strengths in tension and compression of the material is discussed within the strengthening strategy of masonry vaults. Findings The numerical results have highlighted the efficacy of the two numerical approaches to assess the vulnerability of existing structures and design optimal strengthening interventions of these structures. Originality/value The presented models can represent fast and useful tools to assess the vulnerability of existing structures and design optimal strengthening interventions with composite materials of these structures

    Manejo Integrado de Plagas para el control de Varroa destructor y sus implicaciones para las colonias de Apis mellifera

    Integrated Pest Management (IPM) is a pest management system that, in the socioeconomic context of farming systems, the associated environment and the population dynamics of the pest species, utilizes all suitable techniques in a compatible manner as possible to maintain the pest population levels below those causing economic injury. This article covers the principal aspects of the interaction between Apis mellifera and Varroa destructor and it describes the classical control forms applied to reduce the mite negative impact on colonies. Some examples of IPM activities that have been done to control this parasite in the southeast of Buenos Aires province, Argentina have shown good results. Several products worldwide have shown good effectiveness as well. Nevertheless, there are certain risks and hazards inherent to their use, such as: their negative impact on human health, resistance phenomena, loss of beneficial insects and native fauna, environmental pollution and drug residues in the hive products harmful for human consumption. The development of acaricide resistance in V. destructor populations and the possibility of incorporating contaminants in colonies by means of this type of treatment have promoted the addition of new molecules to minimize these disadvantages. The application of organic acids, essential oils and their components have become a worthwhile alternative. It can be concluded that to achieve an integrated management of V. destructor entails a change of mind for beekeepers and the active participation of all actors involved in the beekeeping sector to promote scientific activities aimed to discovering and developing new tools to be incorporated in an IPM Program against V. destructor.El Manejo Integrado de Plagas (MIP) es un sistema de manejo que en el contexto socioeconómico de los sistemas agrícolas, asociado al ambiente y a la dinámica poblacional de las especies plaga, utiliza las técnicas apropiadas de una manera compatible para mantener, las poblaciones de la plaga por debajo de los niveles que causan daño económico. Este artículo cubre los principales aspectos de la interacción entre Apis mellifera y Varroa destructor y describe las formas clásicas de control aplicadas para reducir el impacto del ácaro en las colonias. Algunos ejemplos de las actividades de manejo integrado de plagas realizadas para controlar este parásito en el Sureste de la provincia de Buenos Aires, Argentina, mostraron buenos resultados. Existen algunos productos acaricidas de síntesis en el mundo, que presentaron buena efectividad. Sin embargo, el desarrollo de resistencia a los acaricidas en poblaciones de V. destructor y la posibilidad de incorporación de contaminantes en las colonias por este tipo de tratamientos, se han transformado en cuestiones de gravedad. Esto ha promovido la búsqueda de nuevas moléculas para minimizar estas desventajas. Ácidos orgánicos, aceites esenciales y sus componentes se han convertido en una valiosa alternativa. El éxito de la implementación de herramientas para el Manejo Integrado de Varroa involucra un cambio de mentalidad en los apicultores y la participación activa de todos los actores del sector apícola, para promover actividades científicas que ayuden a desarrollar nuevas alternativas para ser incorporadas en Programas Regionales de Manejo Integrado de esta parasitosis.Fil: Ruffinengo, Sergio Roberto.
