    Effet de la pression interstitielle sur la réponse sismique des sols : modélisation numérique 1D-3 Composantes

    International audienceDuring strong quakes, the propagation of seismic waves in soil layers involves nonlinearities changing with the excitation level. A nonlinear hysteretic law is necessary to describe the variations of the stiffness and the energy dissipation during the seismic shaking. Furthermore, the influence of the pore pressure (cyclic mobility and liquefaction) cannot be neglected for saturated soils under strong quakes. Starting from a FEM formulation describing 1D propagation and three-dimensional loading ("1D-3 components approach"), the influence of the water is accounted for through a relation between the pore pressure and the work of the shear stress initially proposed by Iai. This model describes the variations of the pore pressure from the three-dimensional stress state of the soil. It has been validated through comparisons to laboratory tests (cyclic triaxial tests on saturated sands) and an analysis under three-dimensional excitations (seismic loading polarized along the 3 directions of space). The results involving 3 simultaneous excitation components and a single component in 3 separated analyses show the influence of the loading path on the seismic response and the pore pressure build-up

    A new approach for the heliometric optics

    The heliometer of Fraunhofer in Koenigsberg (1824) is a refractor in which the lens is split into two halves to which is applied a linear displacement along the cut. Later in 1890s a variation of the heliometer has been realized in Goettingen using a beam splitting wedge: these methods were both subjected to chromatic and refractive aberrations; the second configuration being much less affected by thermal fluctuations. The reflector version of the heliometer conceived at the Observatorio Nacional of Rio de Janeiro overcome these problems: the two halves of the vitrified ceramic mirror split at a fixed heliometric angle produce the two images of the Sun exempt of chromatisms and distortions. The heliometer of Rio is a telescope which can rotate around its axis, to measure the solar diameter at all heliolatitudes. A further development of that heliometer, now under construction, is the annular heliometer, in which the mirrors are concentric, with symmetrical Point Spread Functions. Moreover the location of the Observatory of Rio de Janeiro allows zenithal observations, with no atmospheric refraction at all heliolatitudes, in December and January

    Parallaxes of southern extremely cool objects III : 118 L and T dwarfs

    We present new results from the Parallaxes of Southern Extremely Cool dwarfs program to measure parallaxes, proper motions and multiepoch photometry of L and early T dwarfs. The observations were made on 108 nights over the course of 8 yr using the Wide Field Imager on the ESO 2.2m telescope. We present 118 new parallaxes of L and T dwarfs of which 52 have no published values and 24 of the 66 published values are preliminary estimates from this program. The parallax precision varies from 1.0 to 15.5mas with a median of 3.8mas. We find evidence for two objects with long term photometric variation and 24 new moving group candidates. We cross-match our sample to published photometric catalogues and find standard magnitudes in up to 16 pass-bands from which we build spectral energy distributions and H-R diagrams. This allows us to confirm the theoretically anticipated minimum in radius between stars and brown dwarfs across the hydrogen burning minimum mass. We find the minimum occurs between L2 and L6 and verify the predicted steep dependence of radius in the hydrogen burning regime and the gentle rise into the degenerate brown dwarf regime.     Viral discovery and diversity in trypanosomatid protozoa with a focus on relatives of the human parasite <i>Leishmania</i>.

    Knowledge of viral diversity is expanding greatly, but many lineages remain underexplored. We surveyed RNA viruses in 52 cultured monoxenous relatives of the human parasite &lt;i&gt;Leishmania&lt;/i&gt; ( &lt;i&gt;Crithidia&lt;/i&gt; and &lt;i&gt;Leptomonas&lt;/i&gt; ), as well as plant-infecting &lt;i&gt;Phytomonas&lt;/i&gt; &lt;i&gt;Leptomonas pyrrhocoris&lt;/i&gt; was a hotbed for viral discovery, carrying a virus (Leptomonas pyrrhocoris ostravirus 1) with a highly divergent RNA-dependent RNA polymerase missed by conventional BLAST searches, an emergent clade of tombus-like viruses, and an example of viral endogenization. A deep-branching clade of trypanosomatid narnaviruses was found, notable as &lt;i&gt;Leptomonas seymouri&lt;/i&gt; bearing Narna-like virus 1 (LepseyNLV1) have been reported in cultures recovered from patients with visceral leishmaniasis. A deep-branching trypanosomatid viral lineage showing strong affinities to bunyaviruses was termed " &lt;i&gt;Leishbunyavirus&lt;/i&gt; " (LBV) and judged sufficiently distinct to warrant assignment within a proposed family termed " &lt;i&gt;Leishbunyaviridae&lt;/i&gt; " Numerous relatives of trypanosomatid viruses were found in insect metatranscriptomic surveys, which likely arise from trypanosomatid microbiota. Despite extensive sampling we found no relatives of the totivirus &lt;i&gt;Leishmaniavirus&lt;/i&gt; (LRV1/2), implying that it was acquired at about the same time the &lt;i&gt;Leishmania&lt;/i&gt; became able to parasitize vertebrates. As viruses were found in over a quarter of isolates tested, many more are likely to be found in the &gt;600 unsurveyed trypanosomatid species. Viral loss was occasionally observed in culture, providing potentially isogenic virus-free lines enabling studies probing the biological role of trypanosomatid viruses. These data shed important insights on the emergence of viruses within an important trypanosomatid clade relevant to human disease

    MDL28170, a Calpain Inhibitor, Affects Trypanosoma cruzi Metacyclogenesis, Ultrastructure and Attachment to Rhodnius prolixus Midgut

    BACKGROUND: Trypanosoma cruzi is the etiological agent of Chagas' disease. During the parasite life cycle, many molecules are involved in the differentiation process and infectivity. Peptidases are relevant for crucial steps of T. cruzi life cycle; as such, it is conceivable that they may participate in the metacyclogenesis and interaction with the invertebrate host. METHODOLOGY/PRINCIPAL FINDINGS: In this paper, we have investigated the effect of the calpain inhibitor MDL28170 on the attachment of T. cruzi epimastigotes to the luminal midgut surface of Rhodnius prolixus, as well as on the metacyclogenesis process and ultrastructure. MDL28170 treatment was capable of significantly reducing the number of bound epimastigotes to the luminal surface midgut of the insect. Once the cross-reactivity of the anti-Dm-calpain was assessed, it was possible to block calpain molecules by the antibody, leading to a significant reduction in the capacity of adhesion to the insect guts by T. cruzi. However, the antibodies were unable to interfere in metacyclogenesis, which was impaired by the calpain inhibitor presenting a significant reduction in the number of metacyclic trypomastigotes. The calpain inhibitor also promoted a direct effect against bloodstream trypomastigotes. Ultrastructural analysis of epimastigotes treated with the calpain inhibitor revealed disorganization in the reservosomes, Golgi and plasma membrane disruption. CONCLUSIONS/SIGNIFICANCE: The presence of calpain and calpain-like molecules in a wide range of organisms suggests that these proteins could be necessary for basic cellular functions. Herein, we demonstrated the effects of MDL28170 in crucial steps of the T. cruzi life cycle, such as attachment to the insect midgut and metacyclogenesis, as well as in parasite viability and morphology. Together with our previous findings, these results help to shed some light on the functions of T. cruzi calpains. Considering the potential roles of these molecules on the interaction with both invertebrate and vertebrate hosts, it is interesting to improve knowledge on these molecules in T. cruzi

O uso do silício e de microrganismos benéficos (bioagentes) mostram-se como alternativas de controle sustentåvel das doenças do arroz. Tendo em vista a implantação de um manejo integrado para o controle da brusone, este trabalho teve como objetivo comparar o efeito da adubação silicatada em interação com bioagentes na supressão da brusone foliar, no cultivo de arroz de terras altas em sistema agroecológico
