Spectral Analysis for Matrix Hamiltonian Operators
In this work, we study the spectral properties of matrix Hamiltonians
generated by linearizing the nonlinear Schr\"odinger equation about soliton
solutions. By a numerically assisted proof, we show that there are no embedded
eigenvalues for the three dimensional cubic equation. Though we focus on a
proof of the 3d cubic problem, this work presents a new algorithm for verifying
certain spectral properties needed to study soliton stability. Source code for

Stimulant treatment history predicts frontal-striatal structural connectivity in adolescents with attention-deficit/hyperactivity disorder

168155.pdf (publisher's version ) (Closed access)Diffusion tensor imaging (DTI) has revealed white matter abnormalities in individuals with attention-deficit/hyperactivity disorder (ADHD). Stimulant treatment may affect such abnormalities. The current study investigated associations between long-term stimulant treatment and white matter integrity within the frontal-striatal and mesolimbic pathways, in a large sample of children, adolescents and young adults with ADHD. Participants with ADHD (N=172; mean age 17, range 9-26) underwent diffusion-weighted MRI scanning, along with an age- and gendermatched group of 96 control participants. Five study-specific white matter tract masks (orbitofrontal-striatal, orbitofrontal-amygdalar, amygdalar-striatal, dorsolateral-prefrontal-striatal and medialprefrontal- striatal) were created. First we analyzed case-control differences in fractional anisotropy (FA) and mean diffusivity (MD) within each tract. Second, FA and MD in each tract was predicted from cumulative stimulant intake within the ADHD group. After correction for multiple testing, participants with ADHD showed reduced FA in the orbitofrontal-striatal pathway (p=0.010, effect size=0.269). Within the ADHD group, higher cumulative stimulant intake was associated with lower MD in the same pathway (p=0.011, effect size=-0.164), but not with FA. The association between stimulant treatment and orbitofrontal-striatal MD was of modest effect size. It fell short of significance after adding ADHD severity or ADHD type to the model (p=0.036 and p=0.094, respectively), while the effect size changed little. Our findings are compatible with stimulant treatment enhancing orbitofrontal-striatal white matter connectivity, and emphasize the importance of the orbitofrontal cortex and its connections in ADHD. Longitudinal studies including a drug-naive baseline assessment are needed to distinguish between-subject variability in ADHD severity from treatment effects
Cloning, in silico structural characterization and expression analysis of MfAtr4, an ABC transporter from the banana pathogen Mycosphaerella fijiensis
ABC transporters are membrane proteins that use the energy released from the hydrolysis of ATP to drive the transport of compounds across biological membranes. In some plants, pathogenic fungi ABC transporters play a role as virulence factors by mediating the export of plant defense compounds or fungal virulence factors. Mycosphaerella fijiensis, the causal agent of black Sigatoka disease in banana, is the main constraint for the banana industry worldwide. So far, little is known about molecular mechanism that it uses to infect the host. In this study, degenerated primers designed from fungal ABC transporters known to be involved in virulence were used to isolate homologs from M. fijiensis. Here, we reported the full cloning of MfAtr4 a putative ortholog of MgAtr4, an ABC transporter of the related Mycosphaerella graminicola with a function in virulence. Similarities and differences with its presumed ortholog MgAtr4 are described, and the putative function of MfAtr4 are discussed. Analysis of MfAtr4 gene expression in field banana samples exhibiting visible symptoms of black Sigatoka disease indicated a higher expression of MfAtr4 during the first symptomatic stages in comparison to the late necrotrophic phases, suggesting a role for MfAtr4 in the early stages of pathogenic development of M. fijiensis.Key words: ABC transporters, virulence factors, MgAtr4 ortholog, Mycosphaerella fijiensis, black Sigatoka, Musa sp
Depressive Symptoms and Amygdala Volume in Elderly with Cerebral Small Vessel Disease: The RUN DMC Study
Introduction. Late onset depressive symptoms (LODSs) frequently occur in elderly with cerebral small vessel disease (SVD). SVD cannot fully explain LODS; a contributing factor could be amygdala volume. We investigated the relation between amygdala volume and LODS, independent of SVD in 503 participants with symptomatic cerebral SVD. Methods. Patients underwent FLAIR and T1 scanning. Depressive symptoms were assessed with structured questionnaires; amygdala and WML were manually segmented. The relation between amygdala volume and LODS/EODS was investigated and adjusted for age, sex, intracranial volume, and SVD. Results. Patients with LODS had a significantly lower left amygdala volume than those without (P = 0.02), independent of SVD. Each decrease of total amygdala volume (by mL) was related to an increased risk of LODS (OR = 1.77; 95% CI 1.02–3.08; P = 0.04).
Conclusion. Lower left amygdala volume is associated with LODS, independent of SVD. This may suggest differential mechanisms, in which individuals with a small amygdala might be vulnerable to develop LODS
101 Dothideomycetes genomes: A test case for predicting lifestyles and emergence of pathogens.
Dothideomycetes is the largest class of kingdom Fungi and comprises an incredible diversity of lifestyles, many of which have evolved multiple times. Plant pathogens represent a major ecological niche of the class Dothideomycetes and they are known to infect most major food crops and feedstocks for biomass and biofuel production. Studying the ecology and evolution of Dothideomycetes has significant implications for our fundamental understanding of fungal evolution, their adaptation to stress and host specificity, and practical implications with regard to the effects of climate change and on the food, feed, and livestock elements of the agro-economy. In this study, we present the first large-scale, whole-genome comparison of 101 Dothideomycetes introducing 55 newly sequenced species. The availability of whole-genome data produced a high-confidence phylogeny leading to reclassification of 25 organisms, provided a clearer picture of the relationships among the various families, and indicated that pathogenicity evolved multiple times within this class. We also identified gene family expansions and contractions across the Dothideomycetes phylogeny linked to ecological niches providing insights into genome evolution and adaptation across this group. Using machine-learning methods we classified fungi into lifestyle classes with >95 % accuracy and identified a small number of gene families that positively correlated with these distinctions. This can become a valuable tool for genome-based prediction of species lifestyle, especially for rarely seen and poorly studied species

Generation of transgenic Mycosphaerella fijiensis expressing reporter genes: tools for pathogenicity and mating studies.
Mycosphaerella fijiensis, the causal agent of black leaf streak disease is the most devastating pathogen of banana and plantains. Detailed studies on the plant pathogen interaction as well as mating behavior could be improved by the use of non-destructive reporter gene assays. M. fijiensis isolates from different global populations and opposite mating types, CIRAD86 (Mat1-1) from Cameroon and CIRAD139A (Mat1-2) from Colombia, were selected for our studies. In order to generate transgenic M. fijiensis strains, two gene constructs carrying either reporter genes gfp (green fluorescent protein) or dsred (Discosoma sp. red fluorescent protein) driven by the constitutive promoter PtoxA were used. Both constructs also carried the hph gene for resistance to the antibiotic hygromycin as selective marker. Transformation procedures mediated by Agrobacterium tumefaciens strain LBA1100 were carried out using macerated mycelium from 3-week-old cultures of CIRAD86 and CIRAD139A. After the cocultivation period, the cultures were transferred to selective medium containing 30 mg/l hygromycin. Transgenic events were identified by fluorescent microscopy observations after 2-3 weeks. In this part of the work, we were able to generate transgenic strains from both isolates expressing either reporter genes, namely CIRAD86::GFP, CIRAD86::DsRed, CIRAD139A::GFP and CIRAD139A::DsRed. Simultaneously, pathogenicity validation screens and mating assays using in vitro leaf fragments are also being conducted in order to characterize the different stages in the pathogenesis on banana
Weak Sequential Composition in Process Algebras
n this paper we study a special operator for sequential composition, which is defined relative to a dependency relation over the actions of a given system. The idea is that actions which are not dependent (intuitively because they share no common resources) do not have to wait for one another to proceed, even if they are composed sequentially. Such a notion has been studied before in a linear-time setting, but until recently there has been no systematic investigation in the context of process algebras.
We give a structural operational semantics for a process algebraic language containing such a sequential composition operator, which shows some interesting interplay with choice. We give a complete axiomatisation of strong bisimilarity and we show consistency of the operational semantics with an event-based denotational semantics developed recently by the second author. The axiom system allows to derive the communication closed layers law, which in the linear time setting has been shown to be a very useful instrument in correctness preserving transformations. We conclude with a couple of examples
Saccade dysmetria indicates attenuated visual exploration in autism spectrum disorder
Background: Visual exploration in autism spectrum disorder (ASD) is characterized by attenuated social attention. The underlying oculomotor function during visual exploration is understudied, whereas oculomotor function during restricted viewing suggested saccade dysmetria in ASD by altered pontocerebellar motor modulation. Methods: Oculomotor function was recorded using remote eye tracking in 142 ASD participants and 142 matched neurotypical controls during free viewing of naturalistic videos with and without human content. The sample was heterogenous concerning age (6–30 years), cognitive ability (60–140 IQ), and male/female ratio (3:1). Oculomotor function was defined as saccade, fixation, and pupil-dilation features that were compared between groups in linear mixed models. Oculomotor function was investigated as ASD classifier and features were correlated with clinical measures. Results: We observed decreased saccade duration (∆M = −0.50, CI [−0.21, −0.78]) and amplitude (∆M = −0.42, CI [−0.12, −0.72]), which was independent of human video content. We observed null findings concerning fixation and pupil-dilation features (POWER =.81). Oculomotor function is a valid ASD classifier comparable to social attention concerning discriminative power. Within ASD, saccade features correlated with measures of restricted and repetitive behavior. Conclusions: We conclude saccade dysmetria as ASD oculomotor phenotype relevant to visual exploration. Decreased saccade amplitude and duration indicate spatially clustered fixations that attenuate visual exploration and emphasize endogenous over exogenous attention. We propose altered pontocerebellar motor modulation as underlying mechanism that contributes to atypical (oculo-)motor coordination and attention function in ASD
