Myeloid-Epithelial-Reproductive Receptor Tyrosine Kinase and Milk Fat Globule Epidermal Growth Factor 8 Coordinately Improve Remodeling After Myocardial Infarction via Local Delivery of Vascular Endothelial Growth Factor.
BACKGROUND: In infarcted heart, improper clearance of dying cells by activated neighboring phagocytes may precipitate the transition to heart failure. We analyzed the coordinated role of 2 major mediators of efferocytosis, the myeloid-epithelial-reproductive protein tyrosine kinase (Mertk) and the milk fat globule epidermal growth factor (Mfge8), in directing cardiac remodeling by skewing the inflammatory response after myocardial infarction. METHODS AND RESULTS: We generated double-deficient mice for Mertk and Mfge8 (Mertk(-/-)/Mfge8(-/-)) and challenged them with acute coronary ligature. Compared with wild-type, Mertk-deficient (Mertk(-/-)), or Mfge8-deficient (Mfge8(-/-)) animals, Mertk(-/-)/Mfge8(-/-) mice displayed greater alteration in cardiac function and remodeling. Mertk and Mfge8 were expressed mainly by cardiac Ly6C(High and Low) monocytes and macrophages. In parallel, Mertk(-/-)/Mfge8(-/-) bone marrow chimeras manifested increased accumulation of apoptotic cells, enhanced fibrotic area, and larger infarct size, as well as reduced angiogenesis. We found that the abrogation of efferocytosis affected neither the ability of circulating monocytes to infiltrate cardiac tissue nor the number of resident Ly6C(High) and Ly6C(How) monocytes/macrophages populating the infarcted milieu. In contrast, combined Mertk and Mfge8 deficiency in Ly6C(High)/Ly6C(Low) monocytes/macrophages either obtained from in vitro differentiation of bone marrow cells or isolated from infarcted hearts altered their capacity of efferocytosis and subsequently blunted vascular endothelial growth factor A (VEGFA) release. Using LysMCre(+)/VEGFA(fl/fl) mice, we further identified an important role for myeloid-derived VEGFA in improving cardiac function and angiogenesis. CONCLUSIONS: After myocardial infarction, Mertk- and Mfge8-expressing monocyte/macrophages synergistically engage the clearance of injured cardiomyocytes, favoring the secretion of VEGFA to locally repair the dysfunctional heart
Genetic and Pharmacological Inhibition of TREM-1 Limits the Development of Experimental Atherosclerosis.
BACKGROUND: Innate immune responses activated through myeloid cells contribute to the initiation, progression, and complications of atherosclerosis in experimental models. However, the critical upstream pathways that link innate immune activation to foam cell formation are still poorly identified. OBJECTIVES: This study sought to investigate the hypothesis that activation of the triggering receptor expressed on myeloid cells (TREM-1) plays a determinant role in macrophage atherogenic responses. METHODS: After genetically invalidating Trem-1 in chimeric Ldlr-/-Trem-1-/- mice and double knockout ApoE-/-Trem-1-/- mice, we pharmacologically inhibited Trem-1 using LR12 peptide. RESULTS: Ldlr-/- mice reconstituted with bone marrow deficient for Trem-1 (Trem-1-/-) showed a strong reduction of atherosclerotic plaque size in both the aortic sinus and the thoracoabdominal aorta, and were less inflammatory compared to plaques of Trem-1+/+ chimeric mice. Genetic invalidation of Trem-1 led to alteration of monocyte recruitment into atherosclerotic lesions and inhibited toll-like receptor 4 (TLR 4)-initiated proinflammatory macrophage responses. We identified a critical role for Trem-1 in the upregulation of cluster of differentiation 36 (CD36), thereby promoting the formation of inflammatory foam cells. Genetic invalidation of Trem-1 in ApoE-/-/Trem-1-/- mice or pharmacological blockade of Trem-1 in ApoE-/- mice using LR-12 peptide also significantly reduced the development of atherosclerosis throughout the vascular tree, and lessened plaque inflammation. TREM-1 was expressed in human atherosclerotic lesions, mainly in lipid-rich areas with significantly higher levels of expression in atheromatous than in fibrous plaques. CONCLUSIONS: We identified TREM-1 as a major upstream proatherogenic receptor. We propose that TREM-1 activation orchestrates monocyte/macrophage proinflammatory responses and foam cell formation through coordinated and combined activation of CD36 and TLR4. Blockade of TREM-1 signaling may constitute an attractive novel and double-hit approach for the treatment of atherosclerosis

L’inflammation joue un rôle crucial dans les processus de réparation du muscle cardiaque consécutifs à un infarctus du myocarde (IM). En particulier, les monocytes (MO) et les macrophages (Mf) assument une fonction majeure dans la régulation de la taille d’infarctus et de la fibrose du coeur infarci. Dans un premier travail, nous avons analysé le rôle coordonné de la myeloidepithelial- reproductive protein tyrosine kinase (Mertk) et du milk fat globule epidermal growth factor (Mfge8) dans la propension des MO et des Mf à éliminer les débris cellulaires. Nous avons notamment émis l'hypothèse que ces médiateurs pourraient jouer un rôle clé dans la synchronisation de l’efférocytose dans le tissu cardiaque infarci. Les souris dépourvues de Mertk et Mfge8 présentaient un phénotype cardiaque défectueux par rapport aux souris contrôles. Fait intéressant, au 3ème jour après l’IM, le niveau de protéine cardiaque d’un facteur pro-angiogénique majeur, le VEGF-A, était plus faible chez les souris chimères Mertk-/ -Mfge8-/-par rapport aux contrôles. À partir de ces données, nous avons postulé qu'une altération du taux de la protéine VEGF-A pourrait être liée à une réduction de la capacité des MO/Mf cardiaques à libérer ces facteurs et donc à participer au remodelage bénéfique du coeur infarci. De fait, après un IM, la suppression du VEGF-A dans les cellules myéloïdes réduit la fonction cardiaque. Ces effets étaient associés à une taille d'infarctus supérieure et à un nombre limité de capillaires dans les coeurs infarcis des souris présentant une invalidation conditionnelle du VEGF-A dans les cellules myéloïdes. Ainsi, le VEGF-A dérivé des cellules myéloïdes joue un rôle essentiel dans la régulation de la fonction cardiaque, probablement grâce à l'activation du processus angiogénique dans la zone ischémique. Dans un deuxième travail, nous nous sommes intéressés au rôle de l’hepcidine dans les fonctions réparatrices des MO et des Mf. L'hepcidine est une hormone qui régule activement le trafic du fer. De façon inattendue, la déficience complète en hepcidine restaure considérablement la fonction cardiaque après un IM. L'hepcidine est exprimée par les Mf cardiaques. En outre, les souris ayant une moelle déficiente en hepcidine (Hamp -/-) ou présentant une invalidation conditionnelle dans les cellules de la lignée myéloide (LysMCre +/Hampf/f) ont une fonction cardiaque fortement améliorée. Cet effet a été associé à une réduction de la taille de l'infarctus, de la fibrose et à une stimulation imprévue du renouvellement des cardiomyocytes. De même, dans un modèle de régénération cardiaque induite par résection apicale chez des souris nouveaux nés, nous avons révélé que les macrophages déficients en hepcidine favorisent la prolifération des cardiomyocytes. L'impact des Mf déficients en hepcidine repose sur l'altération de leur métabolisme du fer et sur la libération d'IL-4 et d'IL-13. D’ailleurs, la suppression génétique de l'IL-4 et de l'IL-13 dans les Mf dépourvus d'hepcidine annihile leur effet bénéfique sur la fonction et la réparation cardiaque. Ainsi, l'hepcidine commande la réparation et la régénération cardiaque induite par les Mf par une voie liée à l'IL-4 / IL-13. Ces travaux ont permis de démontrer l’importance des Mf dans la revascularisation et la régénération post-ischémique et de souligner l’efficacité potentielle d’un traitement basé sur la modulation de l’activité des cellules immunitaires dans les pathologies ischémiques cardiaques.No abstrac

Activation of CB1R Promotes Lipopolysaccharide-Induced IL-10 Secretion by Monocytic Myeloid-Derived Suppressive Cells and Reduces Acute Inflammation and Organ Injury.
Cannabis sativa and its principal components, Δ9-tetrahydrocannabinol (Δ9-THC) and cannabidiol, are increasingly being used to treat a variety of medical problems, including inflammatory conditions. Although studies suggest that the endocannabinoid system has immunomodulatory properties, there remains a paucity of information on the effects of cannabinoids on immunity and on outcomes of infection and injury. We investigated the effects and mechanism(s) of action of cannabinoid receptor agonists, including Δ9-THC, on inflammation and organ injury in endotoxemic mice. Administration of Δ9-THC caused a dramatic early upregulation of plasma IL-10 levels, reduced plasma IL-6 and CCL-2 levels, led to better clinical status, and attenuated organ injury in endotoxemic mice. The anti-inflammatory effects of Δ9-THC in endotoxemic mice were reversed by a cannabinoid receptor type 1 (CB1R) inverse agonist (SR141716), and by clodronate-induced myeloid-cell depletion, but not by genetic invalidation or blockade of other putative Δ9-THC receptors, including cannabinoid receptor type 2, TRPV1, GPR18, GPR55, and GPR119. Although Δ9-THC administration reduced the activation of several spleen immune cell subsets, the anti-inflammatory effects of Δ9-THC were preserved in splenectomized endotoxemic mice. Finally, using IL-10-GFP reporter mice, we showed that blood monocytic myeloid-derived suppressive cells mediate the Δ9-THC-induced early rise in circulating IL-10. These results indicate that Δ9-THC potently induces IL-10, while reducing proinflammatory cytokines, chemokines, and related organ injury in endotoxemic mice via the activation of CB1R. These data have implications for acute and chronic conditions that are driven by dysregulated inflammation, such as sepsis, and raise the possibility that CB1R-signaling may constitute a novel target for inflammatory disorders

An injury-responsive mmp14b enhancer is required for heart regeneration.
Mammals have limited capacity for heart regeneration, whereas zebrafish have extraordinary regeneration abilities. During zebrafish heart regeneration, endothelial cells promote cardiomyocyte cell cycle reentry and myocardial repair, but the mechanisms responsible for promoting an injury microenvironment conducive to regeneration remain incompletely defined. Here, we identify the matrix metalloproteinase Mmp14b as an essential regulator of heart regeneration. We identify a TEAD-dependent mmp14b endothelial enhancer induced by heart injury in zebrafish and mice, and we show that the enhancer is required for regeneration, supporting a role for Hippo signaling upstream of mmp14b. Last, we show that MMP-14 function in mice is important for the accumulation of Agrin, an essential regulator of neonatal mouse heart regeneration. These findings reveal mechanisms for extracellular matrix remodeling that promote heart regeneration

Splenic Marginal Zone B Lymphocytes Regulate Cardiac Remodeling After Acute Myocardial Infarction in Mice
International audienceBackground. Mature B lymphocytes alter the recovery of cardiac function after acute myocardial infarction (MI) in mice. Follicular B cells and marginal zone B (MZB) cells are spatially distinct mature B cell populations in the spleen and exert specific functional properties. miR21/Hypoxia-inducible factor (HIF)α-related pathways have been shown to govern B cell functions. Objectives. We aimed to unravel the distinct role of MZB cells and that of endogenous activation of miR21/ HIFα signalling in MZB cells during post-ischemic injury. Methods. Acute MI was induced by permanent ligation of the left anterior descending coronary artery in mice. Cardiac function and remodeling were assessed using echocardiography and immunohistochemistry. To determine the specific role of MZB cells, we used mice with B cell lineage-specific conditional deletion of Notch signaling, which leads to selection deficiency of MZB cells. To evaluate the role of HIF-1α isoform, we generated mice with MZB cell lineage specific conditional deletion of Hif1a. Results. Acute MI prompted miR21-dependent increase of HIF-1α, particularly in splenic MZB cells. MZB cell deficiency and MZB cell-specific deletion of miR21 or Hif1a improved cardiac function after acute MI. miR21/HIF-1α signaling in MZB cells was required for Tolllike receptor dependent expression of the monocyte chemo-attractant protein CCL7, leading to increased mobilization of inflammatory monocytes to the ischemic myocardium and to adverse post-ischemic cardiac remodeling. Conclusions. This work reveals a novel function for miR21/HIF-1α pathway in splenic MZB cells with potential major implications for the modulation of cardiac function after acute MI
