    A combined approach involving ampelographic description, berry oenological traits and molecular analysis to study native grapevine varieties of Greece

    A combined approach involving phenotypical characterization (ampelographic description and oenological traits) and molecular analysis was applied on 91 accessions of native Greek grape varieties plus 3 references, all conserved in the Ampelographic Collection of the Aristotle University of Thessaloniki. The accessions were described in accordance to 48 OIV descriptors. Their berry oenological traits were determined at maturity to detect a high juice sugar concentration in most of the assessed varieties, whereas the titratable acidity was found to be extremely low, particularly in the white accessions. Moreover, skin anthocyanin and phenolic content fluctuated from 0.09 to 39.4 mg∙g-1 f.w. and from 2.05 to 30.65 mg∙g-1 f.w. respectively, whereas seed phenolic content was in the range of 2.83 and 32.72 mg∙g-1 f.w. Finally, the discriminative SSR analysis confirmed the differences and similarities among the analyzed varieties as can be evinced from the phylogenetic analysis where close genetic relationship has been detected between 'Fokiano' and 'Armeletousa', 'Moschato Spinas' and 'Moschato Samou', 'Vilana' and 'Asprouda Patron', and 'Mouchtouri' and 'Mavro Spetson'. In all these occasions, the parts of each pair possess similar morphological characteristics

    The grapevine intra-specific variability captured an increasing interest during the last decades, as demonstrated by the number of recently funded European projects focused on the grapevine biodiversity preservation. However, nowadays, crop plants are mainly characterized by genotyping methods. The present work summarizes the phenotype data collected among 20 ampelographic collections spread over 15 countries, covering most of the viticultural areas in the Euro-Asiatic region: from Portugal to Armenia and from Cyprus to Luxembourg. Together with agro-climatic characterization of the experimental site, over two years about 2,400 accessions were described. A common experimental protocol mainly focused on the carpological and oe-nological traits was followed, obtaining a general overview of the distribution of the considered phenotypic traits in the cultivated Vitis vinifera species. The most replicated cultivars were selected and, for the subset of these reference cultivars, their behavior in the different environmental conditions over sites and years was described by ANOVA methods

    O objetivo do presente trabalho foi avaliar os efeitos do ácido giberélico, do bioestimulante Crop Set e do anelamento sobre o aumento do tamanho de bagas e a produtividade da uva 'Thompson Seedless' no Vale do São Francisco. O experimento foi conduzido durante dois ciclos de produção (2001 e 2002), no Campo Experimental de Bebedouro, pertencente à Embrapa Semi-Árido, em Petrolina-PE. Utilizou-se do delineamento experimental de blocos ao acaso, com 12 tratamentos e 3 repetições. Os tratamentos corresponderam à aplicação de ácido giberélico em cinco fases de desenvolvimento da videira, nas doses de 10 + 15 + 15 +50 + 50 mg.L-1, do bioestimulante Crop Set em duas doses de 0,1 e 0,2% e do anelamento no caule, isolados e combinados entre si. Os tratamentos combinados de anelamento + ácido giberélico e anelamento + ácido giberélico + Crop Set destacaram-se como aqueles que promoveram os maiores peso e tamanho de cachos e de bagas, com diferenças significativas em relação à testemunha. Entretanto, o anelamento não cicatrizou completamente, causando a morte de plantas, recomendando-se cautela na sua realização. Apesar de não se observar efeito significativo dos tratamentos sobre a produtividade, pode-se notar um aumento de 63% para o tratamento anelamento + ácido giberélico em relação ao ciclo de produção de 2001.<br>The present research was aimed to evaluate the effects of the gibberellic acid, the biostimulant Crop-Set and girdling applied during bloom and post-bloom stage to improve yield and berry size of the grape cv. Thompson Seedless in the São Francisco River Valley, Northeast of Brazil. The trial was carried out throughout two growing seasons (2001 and 2002) in the Bebedouro Experimental Station, Embrapa Semi-Árido, Petrolina, PE. The trial was laid out in a randomized complete block design with three replicates, each replicate consisting of a four-tree plot. The treatments were: gibberellic acid aplied during five phases of development (10 + 15 + 15 + 50 + 50 mg.L-1 doses), Crop-Set in two doses 0,1 and 0,2% and trunk girdling, isolated or combined to each other. The combination treatments of girdling + gibberellic acid and girdling + gibberellic acid + Crop Set showed the best results, promoting the largest weight of bunches and size of berries with significative diferences compared to control. On the other hand, girdling caused damage in the tissue cicatrization and death of plants, so it is recomended caution to practice it. The yield was increased 63% in the combination girdling + gibberellic acid in the growing season of 2001, although it had not been observed significative diferences among treatments

    First results of the European grapevine collections' collaborative network: Validation of a standard eno-carpological phenotyping method

    A standard protocol for phenotyping structural and compositional aspects of the grape berry has been adopted by 18 East and West European germplasm collections during one season by testing a total of 469 accessions, including reference cultivars as well as local and minor germplasm accessions of specific interest. The protocol consists in the collection of triplicates for 26 phenotypic traits, from biological samples, each formed by 10 berries collected from 9 representative bunches from every analyzed accessions. The protocol concatenates the data from measurements and acquisitions, with the objective to generate new derived variables, which are expressed with different units (%; content per kg of grapes, per berry, per g of tissue). For each variable, the Least Significant Differences (LSD), to contrast a pair of single accession mean values, and the Confidence Intervals (CI), to estimate each single accession mean value, were computed. The application of the protocol revealed satisfactory results with high accuracy and efficiency in estimation of phenotypic traits of each accession. The whole data set will be useful for researchers, breeders and viticulturists in yield evaluation of grapevine cultivars, as well as in comparative analyses of environment-variety interaction

    Grapevine N fertilization may affect and be affected by arbuscular mycorrhizal (AM) fungal colonization and change berry composition. We studied the effects of different N fertilizers on AM fungal grapevine root colonization and sporulation, and on grapevine growth, nutrition, and berry composition, by conducting a 3.5-year pot study supplying grapevine plants with either urea, calcium nitrate, ammonium sulfate, or ammonium nitrate. We measured the percentage of AM fungal root colonization, AM fungal sporulation, grapevine shoot dry weight and number of leaves, nutrient composition (macro- and micronutrients), and grapevine berry soluble solids (total sugars or degrees Brix) and total acidity. Urea suppressed AM fungal root colonization and sporulation. Mycorrhizal grapevine plants had higher shoot dry weight and number of leaves than non-mycorrhizal and with a higher growth response with calcium nitrate as the N source. For the macronutrients P and K, and for the micronutrient B, leaf concentration was higher in mycorrhizal plants. Non-mycorrhizal plants had higher concentration of microelements Zn, Mn, Fe, and Cu than mycorrhizal. There were no differences in soluble solids (degrees Brix) in grapevine berries among mycorrhizal and non-mycorrhizal plants. However, non-mycorrhizal grapevine berries had higher acid content with ammonium nitrate, although they did not have better N nutrition and vegetative growth
