    A Genomic-Based Approach Combining In Vivo Selection in Mice to Identify a Novel Virulence Gene in Leishmania

    Parasites of the genus Leishmania cause a variety of human diseases that range from destructive skin lesions caused by L. major to visceral infections of the liver and spleen caused by L. donovani that result in death. The Leishmania genes responsible for these different pathologies are not known. In the present study, we used a comparative genome-based approach to introduce and over-express L. donovani genes in L. major to determine whether this results in increased virulence of L. major in visceral organs of infected mice. Through this approach, a novel gene termed Li1040 was identified that is potentially involved in protein transport and was shown to increase pathogenesis in the visceral organs in mice. The Li1040 gene may therefore represent a Leishmania virulence gene that has the potential to regulate the pathology of infection in the mammalian host. These observations help to define how Leishmania causes fatal infections in humans and therefore provide a parasite-specific target for therapy

    Anion Distribution, Structural Distortion, and Symmetry-Driven Optical Band Gap Bowing in Mixed Halide Cs2SnX6 Vacancy Ordered Double Perovskites.

    Mixed anion compounds in the Fm3Ě…m vacancy ordered perovskite structure were synthesized and characterized experimentally and computationally with a focus on compounds where A = Cs+. Pure anion Cs2SnX6 compounds were formed with X = Cl, Br, and I using a room temperature solution phase method. Mixed anion compounds were formed as solid solutions of Cs2SnCl6 and Cs2SnBr6 and a second series from Cs2SnBr6 and Cs2SnI6. Single phase structures formed across the entirety of both composition series with no evidence of long-range anion ordering observed by diffraction. A distortion of the cubic A2BX6 structure was identified in which the spacing of the BX6 octahedra changes to accommodate the A site cation without reduction of overall symmetry. Optical band gap values varied with anion composition between 4.89 eV in Cs2SnCl6 to 1.35 eV in Cs2SnI6 but proved highly nonlinear with changes in composition. In mixed halide compounds, it was found that lower energy optical transitions appeared that were not present in the pure halide compounds, and this was attributed to lowering of the local symmetry within the tin halide octahedra. The electronic structure was characterized by photoemission spectroscopy, and Raman spectroscopy revealed vibrational modes in the mixed halide compounds that could be assigned to particular mixed halide octahedra. This analysis was used to determine the distribution of octahedra types in mixed anion compounds, which was found to be consistent with a near-random distribution of halide anions throughout the structure, although some deviations from random halide distribution were noted in mixed iodide-bromide compounds, where the larger iodide anions preferentially adopted trans configurations

    Differential contribution of psycholinguistic and cognitive skills to written composition in Chinese as a second language

The final version is available from Springer via the DOI in this record.This study examined the contribution of the constructs of orthographic processing (orthographic choice and orthographic choice in context), syntactic processing (grammaticality and sentence integrity), and verbal working memory (two reading span indicators) to written Chinese composition (narration, explanation, and argumentation) in 129 fifteen-year-old L2 learners. A matrix task was also administered as a control task to tap cognitive flexibility. Structural Equation Modeling (SEM) analysis with written composition as a latent variable revealed orthographic processing and working memory as two significant, independent contributors, whereas the unique contribution of syntactic processing was not significant. Subsequent SEM analysis with narration, explanation, and argumentation as separate endogenous variables found varied patterns of the contribution of each latent predictor to written composition in different genres. These patterns are discussed in light of the importance of attention to learners’ developmental stage and genre-sensitive measures to capture the psycholinguistic and cognitive underpinnings of written composition in L2 Chinese

    A2 gene of Old World cutaneous Leishmania is a single highly conserved functional gene

    BACKGROUND: Leishmaniases are among the most proteiform parasitic infections in humans ranging from unapparent to cutaneous, mucocutaneous or visceral diseases. The various clinical issues depend on complex and still poorly understood mechanisms where both host and parasite factors are interacting. Among the candidate factors of parasite virulence are the A2 genes, a family of multiple genes that are developmentally expressed in species of the Leishmania donovani group responsible for visceral diseases (VL). By contrast, in L. major determining cutaneous infections (CL) we showed that A2 genes are present in a truncated form only. Furthermore, the A2 genomic sequences of L. major were considered subsequently to represent non-expressed pseudogenes [1]. Consequently, it was suggested that the structural and functional properties of A2 genes could play a role in the differential tropism of CL and VL leishmanias. On this basis, it was of importance to determine whether the observed structural/functional particularities of the L. major A2 genes were shared by other CL Leishmania, therefore representing a proper characteristic of CL A2 genes as opposed to those of VL isolates. METHODS: In the present study we amplified by PCR and sequenced the A2 genes from genomic DNA and from clonal libraries of the four Old World CL species comparatively to a clonal population of L. infantum VL parasites. Using RT-PCR we also amplified and sequenced A2 mRNA transcripts from L. major. RESULTS: A unique A2 sequence was identified in Old World cutaneous Leishmania by sequencing. The shared sequence was highly conserved among the various CL strains and species analysed, showing a single polymorphism C/G at position 58. The CL A2 gene was found to be functionally transcribed at both parasite stages. CONCLUSION: The present study shows that cutaneous strains of leishmania share a conserved functional A2 gene. As opposed to the multiple A2 genes described in VL isolates, the CL A2 gene is unique, lacking most of the nucleotide repeats that constitute the variable region at the 5'end of the VL A2 sequences. As the variable region of the VL A2 gene has been shown to correspond to a portion of the protein which is highly immunogenic, the present results support the hypothesis of a possible role of the A2 gene in the differential tropism of CL and VL leishmania parasites

    A bibliometric analysis of research related to ocean circulation

    [[abstract]]This study is a bibliometric analysis on ocean circulation-related research for the period 1991-2005. Selected documents included "ocean circulation, sea circulation, seas circulation, marine circulation, and circulation ocean" as a part of the title, abstract or keywords. Analyzed parameters included the document type, the article output, the article distribution in journals, the publication activity of countries, and institutes and the authorship. An indicator, citation per publication (CPP) was applied to evaluate the scientific impact of a publication. The relationship between cumulative articles and the year was modeled. Three dominant categories were picked out, and their output increase was modeled. The USA was found to be leading the research with 47% share of total articles, with a CPP up to 5.9. Woods Hole Oceanography Institute in the USA was the most productive institute with a CPP of 6.8. In the citation analysis, a 5th year citation mode was found. A paper life model was applied to compare the cumulative citations increasing rates of different years

    Fast and Accurate Camera Covariance Computation for Large 3D Reconstruction

    Estimating uncertainty of camera parameters computed in Structure from Motion (SfM) is an important tool for evaluating the quality of the reconstruction and guiding the reconstruction process. Yet, the quality of the estimated parameters of large reconstructions has been rarely evaluated due to the computational challenges. We present a new algorithm which employs the sparsity of the uncertainty propagation and speeds the computation up about ten times \wrt previous approaches. Our computation is accurate and does not use any approximations. We can compute uncertainties of thousands of cameras in tens of seconds on a standard PC. We also demonstrate that our approach can be effectively used for reconstructions of any size by applying it to smaller sub-reconstructions.Comment: ECCV 201

    Channel discord and distortion

    Discord, originally notable as a signature of bipartite quantum correlation, in fact can be nonzero classically, i.e. arising from noisy measurements by one of the two parties. Here we redefine classical discord to quantify channel distortion, in contrast to the previous restriction of classical discord to a state, and we then show a monotonic relationship between classical (channel) discord and channel distortion. We show that classical discord is equivalent to (doubly stochastic) channel distortion by numerically discovering a monotonic relation between discord and total-variation distance for a bipartite protocol with one party having a noiseless channel and the other party having a noisy channel. Our numerical method includes randomly generating doubly stochastic matrices for noisy channels and averaging over a uniform measure of input messages. Connecting discord with distortion establishes discord as a signature of classical, not quantum, channel distortion
