    Dynamic proteomic changes in soft wheat seeds during accelerated ageing

    Previous research demonstrated that soft wheat cultivars have better post-harvest storage tolerance than harder cultivars during accelerated ageing. To better understand this phenomenon, a tandem mass tag-based quantitative proteomic analysis of soft wheat seeds was performed at different storage times during accelerated ageing (germination ratios of 97%, 45%, 28%, and 6%). A total of 1,010 proteins were differentially regulated, of which 519 and 491 were up- and downregulated, respectively. Most of the differentially expressed proteins were predicted to be involved in nutrient reservoir, enzyme activity and regulation, energy and metabolism, and response to stimulus functions, consistent with processes occurring in hard wheat during artificial ageing. Notably, defense-associated proteins including wheatwin-2, pathogenesis-related proteins protecting against fungal invasion, and glutathione S-transferase and glutathione synthetase participating in reactive oxygen species (ROS) detoxification, were upregulated compared to levels in hard wheat during accelerated ageing. These upregulated proteins might be responsible for the superior post-harvest storage-tolerance of soft wheat cultivars during accelerated ageing compared with hard wheat.     A Single Dose of Baicalin Has No Clinically Significant Effect on the Pharmacokinetics of Cyclosporine A in Healthy Chinese Volunteers

    Despite its narrow therapeutic window and large interindividual variability, cyclosporine A (CsA) is the first-line therapy following organ transplantation. Metabolized mainly by CYP3A and being a substrate of P-glycoprotein (P-gp), CsA is susceptible to drug–drug interactions. Baicalin (BG) is a drug used for adjuvant therapy of hepatitis in traditional Chinese medicine. Since its aglycone baicalein (B) inhibits CYP3A and P-gP, co-administration might affect CsA pharmacokinetics. This study investigated the effect of BG on CsA pharmacokinetics. In a two-period study, 16 healthy volunteers received a single 200 mg oral CsA dose alone (reference period) or in combination with 500 mg BG (test period). Pharmacokinetic evaluation of CsA was carried out using non-compartmental analysis (NCA) and population pharmacokinetics (popPK). Treatments were compared using the standard bioequivalence method. Based on NCA, 90% CIs of AUC and Cmax test-to-reference ratios were within bioequivalence boundaries. In the popPK analysis, a two-compartment model (clearance/F 62.8 L/h, central and peripheral volume of distribution/F 254 L and 388 L) with transit compartments for absorption appropriately described CsA concentrations. No clinically relevant effect of 500 mg BG co-administration on CsA pharmacokinetics was identified and both treatments were well tolerated

    Raw rehmannia radix polysaccharide can effectively release peroxidative injury induced by duck hepatitis A virus

    Background: Duck viral hepatitis (DVH), caused by duck hepatitis A virus (DHAV), is a fatal contagious infectious disease which spreads rapidly with high morbidity and high mortality, and there is no effective clinical drug against DVH.Materials and Methods: Raw Rehmannia Radix Polysaccharide (RRRP), Lycii Fructus polysaccharides and Astragalus Radix polysaccharides were experimented in vitro and in vivo. Mortality rate, livers change, liver lesion scoring, peroxidative injury evaluation indexes in vitro and in vivo, and hepatic injury evaluation indexes of optimal one were detected and observed in this experiment.Results: RRRP could reduce mortality with the protection rate about 20.0% compared with that of the viral control (VC) group, finding that RRRP was the most effective against DHAV. The average liver scoring of the VC, blank control (BC), RRRP groups were 3.5, 0, 2.1. Significant difference (P<0.05) appeared between any two groups, demonstrating that it can alleviate liver pathological change. RRRP could make the hepatic injury evaluation indexes similar to BC group while the levels of the VC group were higher than other two groups in general. The levels of SOD, GSH-Px, CAT of RRRP group showed significant higher than that of VC group while the levels of NOS and MDA showed the opposite tendency, thus, RRRP could release peroxidative injury.Conclusion: RRRP was the most effective against duck hepatitis A virus (DHAV). RRRP could reduce mortality, alleviate liver pathological change, down-regulate liver lesion score, release peroxidative injury and hepatic injury. The antiviral and peroxidative injury releasing activity of RRRP for DHAV provided a platform to test novel drug strategies for hepatitis A virus in human beings.Keywords: Raw Rehmannia Radix Polysaccharide; duck hepatitis A virus; peroxidative injury; hepatic injur

    Gazelle: A Low Latency Framework for Secure Neural Network Inference

    The growing popularity of cloud-based machine learning raises a natural question about the privacy guarantees that can be provided in such a setting. Our work tackles this problem in the context where a client wishes to classify private images using a convolutional neural network (CNN) trained by a server. Our goal is to build efficient protocols whereby the client can acquire the classification result without revealing their input to the server, while guaranteeing the privacy of the server's neural network. To this end, we design Gazelle, a scalable and low-latency system for secure neural network inference, using an intricate combination of homomorphic encryption and traditional two-party computation techniques (such as garbled circuits). Gazelle makes three contributions. First, we design the Gazelle homomorphic encryption library which provides fast algorithms for basic homomorphic operations such as SIMD (single instruction multiple data) addition, SIMD multiplication and ciphertext permutation. Second, we implement the Gazelle homomorphic linear algebra kernels which map neural network layers to optimized homomorphic matrix-vector multiplication and convolution routines. Third, we design optimized encryption switching protocols which seamlessly convert between homomorphic and garbled circuit encodings to enable implementation of complete neural network inference. We evaluate our protocols on benchmark neural networks trained on the MNIST and CIFAR-10 datasets and show that Gazelle outperforms the best existing systems such as MiniONN (ACM CCS 2017) by 20 times and Chameleon (Crypto Eprint 2017/1164) by 30 times in online runtime. Similarly when compared with fully homomorphic approaches like CryptoNets (ICML 2016) we demonstrate three orders of magnitude faster online run-time

    ObjectiveIn the context of “internet + medical health” and emphasis on evaluation mechanism for medical and health talents in China, we design an evaluation index system for doctors on online medical platforms by synthesizing two patterns of existing online medical platforms, which is the first step to enhance the capabilities of doctors on online medical platforms.MethodsBased on the doctor evaluation model integrating information systems success model (ISS-DE model) and grounded theory, the evaluation indicators were obtained through expert interviews, offline medical institutions investigation, online platforms investigation, and literature research, and were assigned weights using the analytic hierarchy process (AHP) method. A working group composed of 23 experts was set up to review and determine the competency standards of doctors on the online medical platforms.ResultsA new indicator framework covering 3 dimensions of system quality, service quality and information quality was constructed in this study. The index system included 3 first-level indicators, 8 s-level indicators and 60 third-level indicators, and each indicator was given different weightage.ConclusionThe complete index system constructed by the Delphi method in this study is suitable for China’s online medical platforms, which will help to improve the quality of platforms and the ability of doctors, thus promoting the process of internet medical integration.</p

    Evolution Mode, Influencing Factors, and Socioeconomic Value of Urban Industrial Land Management in China

    (1) Background: Accurate measurement of the matching relationship between urban industrial land change and economic growth is of great value for industrialized and re-industrialized countries to perform land resource management in territorial spatial planning. (2) Methods: Based on the combination of the Boston Consulting Group matrix, Geodetector, and decoupling model, we constructed a new method integrating “model evolution + driving mechanism + performance evaluation + policy design” in this paper, and conducted an empirical study on the economic value of urban industrial land management in the Yangtze River Delta. (3) Results: The evolution modes of urban industrial land in the Yangtze River Delta are divided into four types: stars, cows, dogs, and question, distributed in structures ranging from an “olive” shape to a “pyramid” shape, with high spatial heterogeneity and agglomeration and low autocorrelation. The government demand led by driving economic growth and making large cities bigger is the key factor driving the change in urban industrial land and the influence of transportation infrastructure and the business environment has remained stable for a long time. The mechanisms of industrialization, globalization, and innovation are becoming increasingly complicated. Industrial land change and value-added growth in most cities have long been in a state of strong and weak decoupling, with progressive decoupling occurring alongside the unchanged stage and regressive decoupling. The government outperforms the market in terms of urban industrial land management, and the degradation of the synergy between urban industrial land and corporate assets emerges as a new threat to sustainable and high-quality development of the region. (4) Conclusions: This paper establishes a technical framework for zoning management and classification governance of urban industrial land to divide the Yangtze River Delta into reduction-oriented transformation policy zoning, incremental high-quality development zoning, incremental synchronous growth zoning, and reduction and upgrading development zoning. It also proposes an adaptive land supply governance strategy for quantitative and qualitative control, providing a basis for territorial spatial planning and land resource management
