Rapid, simple, and sensitive detection of the ompB gene of spotted fever group rickettsiae by loop-mediated isothermal amplification
BACKGROUND: Spotted fever caused spotted fever group rickettsiae (SFGR) is prevalent throughout China. In this study, we describe a rapid, simple, and sensitive loop-mediated isothermal amplification (LAMP) assay targeting the ompB gene of spotted fever group rickettsiae ideal for application in China. The LAMP assay has the potential to detect spotted fever group rickettsiae early in infection and could therefore serve as an alternative to existing methods. METHODS: A set of universal primers which are specific 7 common species of spotted fever group rickettsiae in China were designed using PrimerExplorer V4 software based on conserved sequences of ompB gene. The sensitivity, specificity and reproducibility of the LAMP were evaluated. The LAMP assay for detecting SFGR was compared with conventional PCR assays for sensitivity and specificity in early phase blood samples obtained from 11 infected human subjects. RESULTS: The sensitivity of the LAMP assay was five copies per reaction (25 ÎźL total volume), and the assay did not detect false-positive amplification across 42 strains of 27 members of the order Rickettsiales and 17 common clinical pathogens. The LAMP assay was negative to typhus group rickettsiae including R. prowazekii and R. typhi for no available conserved sequences of ompB was obtained for designing primers. To evaluate the clinical applicability of the LAMP assay, a total of 11 clinical samples, 10 samples confirmed serologically (3 cases), ecologically (1 case), by real-time polymerase chain reaction (PCR; 2 cases), ecologically and by real-time PCR (1 case), and serologically and by real-time PCR (3 cases) were analyzed by the ompB LAMP assay. Data were validated using a previously established nested PCR protocol and real-time PCR. A positive LAMP result was obtained for 8 of the 10 confirmed cases (sensitivity, 73%; specificity, 100%), while none of these samples were positive by nested PCR (sensitivity, 0%; specificity, 100%). CONCLUSIONS: The LAMP assay described here is the most reliable among the three methods tested and would be an ideal choice for development as a rapid and cost-effective means of detecting SFGR in China
An evaluation study of miniature dielectric crossed compound parabolic concentrator (dCCPC) panel as skylights in building energy simulation
The potential of miniature dielectric crossed compound parabolic concentrator (dCCPC) panel as skylights for daylighting control has drawn a considerable research attention in the recent years, owing to its feature of variable transmittance according to the sun position, but the viability of using it as skylights in buildings has not been explored yet comprehensively. This paper aims to study the feasibility of utilizing miniature dCCPC panel as skylight in different locations under various climates in terms of energy saving potential besides its daylighting control function. The transmittance of dCCPC panel varies at every moment according to the sky condition and sun position. Due to this specific property, this study novelly implemented a polynomial formula of the dCCPC transmittance in the Grasshopper platform, from which EnergyPlus weather data can be called to calculate the hourly transmittance data of dCCPC skylight panel throughout the whole year. An hourly schedule of transmittance is generated according to the hourly sky condition determined by the daylight simulation through Radiance and Daysim, and is then input to EnergyPlus simulation to predict the energy consumption of a building with dCCPC skylight. Fourteen locations around the world are therefore compared to find the most appropriate place for using miniature dCCPC panel as skylights. The energy saving in cooling, heating and lighting with use of dCCPC skylight panel are investigated and compared with low-E and normal double glazing. The results show that the dCCPC skylight panel can reduce cooling load by mitigating solar heat gain effectively although its performance is affected by several criteria such as sky conditions and local climates. It is generally more suitable for the locations with longer hot seasons, e.g., Log Angeles, Miami, Bangkok and Manila, in which dCCPC could provide up to 13% reduction in annual energy consumption of building. For the locations having temperate and continental climates like Beijing, Rome, Istanbul and Hong Kong, a small annual energy saving from 1% to 5% could be obtained by using dCCPC skylight panel
New Broad-Spectrum Viral Fusion Inhibitors Act by Deleterious Effect on the Viral Membrane through the Production Singlet Oxygen Molecules
Chitosan oligosaccharides (COS), the degraded products of chitosan, have been demonstrated to have versatile biological functions. In primary studies, it has displayed significant adjuvant effects when mixed with other vaccines. In this study, chitosan oligosaccharides with different deacetylation degrees were prepared and conjugated to porcine circovirus type 2 (PCV2) subunit vaccine to enhance its immunogenicity. The vaccine conjugates were designed by the covalent linkage of COSs to PCV2 molecules and administered to BALB/c mice three times at two-week intervals. The results indicate that, as compared to the PCV2 group, COS-PCV2 conjugates remarkably enhanced both humoral and cellular immunity against PCV2 by promoting lymphocyte proliferation and initiating a mixed T-helper 1 (Th1)/T-helper 2 (Th2) response, including raised levels of PCV2-specific antibodies and an increased production of inflammatory cytokines. Noticeably, with the increasing deacetylation degree, the stronger immune responses to PCV2 were observed in the groups with COS-PCV2 vaccination. In comparison with NACOS (chitin oligosaccharides)-PCV2 and LCOS (chitosan oligosaccharides with low deacetylation degree)-PCV2, HCOS (chitosan oligosaccharides with high deacetylation degree)-PCV2 showed the highest adjuvant effect, even comparable to that of PCV2/ISA206 (a commercialized adjuvant) group. In summary, COS conjugation might be a viable strategy to enhance the immune response to PCV2 subunit vaccine, and the adjuvant effect was positively correlated with the deacetylation degree of COS.</p
Experimental analysis of defrosting and heating performance of a solar-assisted heat pump integrated phase change energy storage
This thesis investigates a novel solar-assisted heat pump integrated phase change energy storage system. The defrosting performance of this system was studied experimentally and the results were compared with two traditionally used methods: reverse cycle defrosting (RCD) method and hot gas bypass defrosting (HGBD) method. The results show that the phase change energy storage system has superior performance compared with traditional defrosting methods. The indoor temperature drop recorded was relatively small and the defrosting time was 75% of the RCD system and 53% of HGBD system. The phase change energy storage system increased the condensation temperature which consequently increased the temperature difference of heat transfer resulting in higher conductivity in the defrosting progress. Compared with the method of RCD and the method of HGBD, the recovery time of the system was shortened by 90 and 160âseconds, respectively. The system works with low-temperature heat source and circulating water, which considerably reduces energy consumption, thereby improving the performance of the defrosting system. A further experimental study was also conducted on the heating performance and the results also indicated that the value of COP can reach up to 3.6 in daytime, and the indoor temperature can be stably maintained above 18°C throughout the day
Genome-wide association for milk production and lactation curve parameters in Holstein dairy cows
The aim of this study was to identify genomic regions associated with 305-day milk yield and lactation curve parameters on primiparous (n = 9,910) and multiparous (n = 11,158) Holstein cows. The SNP solutions were estimated using a weighted single-step genomic BLUP approach and imputed high-density panel (777k) genotypes. The proportion of genetic variance explained by windows of 50 consecutive SNP (with an average of 165 Kb) was calculated, and regions that accounted for more than 0.50% of the variance were used to search for candidate genes. Estimated heritabilities were 0.37, 0.34, 0.17, 0.12, 0.30 and 0.19, respectively, for 305-day milk yield, peak yield, peak time, ramp, scale and decay for primiparous cows. Genetic correlations of 305-day milk yield with peak yield, peak time, ramp, scale and decay in primiparous cows were 0.99, 0.63, 0.20, 0.97 and -0.52, respectively. The results identified three windows on BTA14 associated with 305-day milk yield and the parameters of lactation curve in primi- and multiparous cows. Previously proposed candidate genes for milk yield supported by this work include GRINA, CYHR1, FOXH1, TONSL, PPP1R16A, ARHGAP39, MAF1, OPLAH and MROH1, whereas newly identified candidate genes are MIR2308, ZNF7, ZNF34, SLURP1, MAFA and KIFC2 (BTA14). The protein lipidation biological process term, which plays a key role in controlling protein localization and function, was identified as the most important term enriched by the identified genes
Expert-based development of a generic HACCP-based risk management system to prevent critical negative energy balance in dairy herds
The objective of this study was to develop a generic risk management system based on the Hazard Analysis and Critical Control Point (HACCP) principles for the prevention of critical negative energy balance (NEB) in dairy herds using an expert panel approach. In addition, we discuss the advantages and limitations of the system in terms of implementation in the individual dairy herd. For the expert panel, we invited 30 researchers and advisors with expertise in the field of dairy cow feeding and/or health management from eight European regions. They were invited to a Delphi-based set-up that included three inter-correlated questionnaires in which they were asked to suggest risk factors for critical NEB and to score these based on 'effect' and 'probability'. Finally, the experts were asked to suggest critical control points (CCPs) specified by alarm values, monitoring frequency and corrective actions related to the most relevant risk factors in an operational farm setting. A total of 12 experts (40 %) completed all three questionnaires. Of these 12 experts, seven were researchers and five were advisors and in total they represented seven out of the eight European regions addressed in the questionnaire study. When asking for suggestions on risk factors and CCPs, these were formulated as 'open questions', and the experts' suggestions were numerous and overlapping. The suggestions were merged via a process of linguistic editing in order to eliminate doublets. The editing process revealed that the experts provided a total of 34 CCPs for the 11 risk factors they scored as most important. The consensus among experts was relatively high when scoring the most important risk factors, while there were more diverse suggestions of CCPs with specification of alarm values and corrective actions. We therefore concluded that the expert panel approach only partly succeeded in developing a generic HACCP for critical NEB in dairy cows. We recommend that the output of this paper is used to inform key areas for implementation on the individual dairy farm by local farm teams including farmers and their advisors, who together can conduct herd-specific risk factor profiling, organise the ongoing monitoring of herd-specific CCPs, as well as implement corrective actions when CCP alarm values are exceeded
