    Post-Disaster Reconstruction Training Effectiveness

    Training in a post-disaster environment offers an opportunity to build resilience within high-risk communities. Education research amasses a field of study that is large in both depth and breath, but there is a considerable lack of focus in post-disaster contexts, specifically the effectiveness of post-disaster training programs. Addressing this gap meant exploring recovery efforts in the Philippine region of Eastern Samar following Super Typhoon Haiyan, regarded as the strongest tropical cyclone ever recorded at landfall. The purpose of this research first explores expanding education theories into the post-disaster context and second, examines the practical implementation of training programs in the wake of the 2013 typhoon. A mixed methods approach combined qualitative data derived from accounts of community members and aid organizations with quantitative data that delineated community members learning style preferences in respect to experiential learning theory (ELT). Findings show that aid organizations administered training largely in lecture format, aligning with the reflective observation mode of ELT, but lacked diversity in formats represented in other poles of ELT. Moreover, analysis revealed that community members showed a preference toward divergent learning styles. Since aid organizations provided predominantly lecture based training, this partially aligned with community learning preferences, but fell short in cultivating other forms of knowledge acquisition. Based on this research, the application of existing learning theories will improve construction training as it applies to a post-disaster environment.National Science Foundatio

    Characterizing Post-Disaster Reconstruction Training Methods and Learning Styles

    Large disasters damage or destroy infrastructure that is then reconstructed through programmes that train community members in construction techniques that reduce future risks. Despite the number of post-disaster reconstruction programmes implemented, there is a dearth of research on education and training in post-disaster contexts. To address this gap, we applied a mixed methods approach based upon experiential learning theory (ELT) to three shelter programmes administered in Eastern Samar, Philippines following Typhoon Haiyan. First, we characterize post-disaster training programmes based on learning modes and then, compared this to the learning styles of community members. To assess learning modes of training programmes, we analysed qualitative data from interview accounts of community members and aid organizations; and, to delineate community member’s learning style preferences, we analysed quantitative data from survey questionnaires. Findings show that aid organizations administered training largely in lecture format, aligning with the reflective observation mode of ELT, but lacked diversity in formats represented in other poles of ELT. Moreover, analysis revealed that community members tended to grasp new information in accordance with the concrete experimentation mode, then preferred transforming newly acquired knowledge via the reflective observation mode. The lecture-based training predominately administered by aid organizations partially aligned with community learning preferences, but fell short in cultivating other forms of knowledge acquisition known to enhance long-term learning.National Science Foundatio

    High-entropy mixtures of pristine fullerenes for solution-processed transistors and solar cells

    The solubility of pristine fullerenes can be enhanced by mixing C-60 and C-70 due to the associated increase in configurational entropy.     A High Throughput Substrate Binding Assay Reveals Hexachlorophene as an Inhibitor of the ER-resident HSP70 Chaperone GRP78

    Glucose-regulated protein 78 (GRP78) is the ER resident 70 kDa heat shock protein 70 (HSP70) and has been hypothesized to be a therapeutic target for various forms of cancer due to its role in mitigating proteotoxic stress in the ER, its elevated expression in some cancers, and the correlation between high levels for GRP78 and a poor prognosis. Herein we report the development and use of a high throughput fluorescence polarization-based peptide binding assay as an initial step toward the discovery and development of GRP78 inhibitors. This assay was used in a pilot screen to discover the anti-infective agent, hexachlorophene, as an inhibitor of GRP78. Through biochemical characterization we show that hexachlorophene is a competitive inhibitor of the GRP78-peptide interaction. Biological investigations showed that this molecule induces the unfolded protein response, induces autophagy, and leads to apoptosis in a colon carcinoma cell model, which is known to be sensitive to GRP78 inhibition

    Factors associated with shunt dynamic in patients with cryptogenic stroke and patent foramen ovale: an observational cohort study

    <p>Abstract</p> <p>Background</p> <p>As previously reported there is evidence for a reduction in right to left shunt (RLS) in stroke patients with patent foramen ovale (PFO). This occurs predominantly in patients with cryptogenic stroke (CS). We therefore analysed factors associated with a shunt reduction on follow-up in stroke patients suffering of CS.</p> <p>Methods</p> <p>On index event PFO and RLS were proven by transesophageal echocardiography and contrast-enhanced transcranial Doppler-sonography (ce-TCD). Silent PE was proved by ventilation perfusion scintigraphy (V/Q) within the stroke work-up on index event; all scans were re-evaluated in a blinded manner by two experts. The RLS was re-assessed on follow-up by ce-TCD. A reduction in shunt volume was defined as a difference of ≥20 microembolic signals (MES) or the lack of evidence of RLS on follow-up. For subsequent analyses patients with CS were considered; parameters such as deep vein thrombosis (DVT) and silent pulmonary embolism (PE) were analysed.</p> <p>Results</p> <p>In 39 PFO patients suffering of a CS the RLS was re-assessed on follow-up. In all patients (n = 39) with CS a V/Q was performed; the median age was 40 years, 24 (61.5%) patients were female. In 27 patients a reduction in RLS was evident. Silent PE was evident in 18/39 patients (46.2%). Factors such as atrial septum aneurysm, DVT or even silent PE were not associated with RLS dynamics. A greater time delay from index event to follow-up assessment was associated with a decrease in shunt volume (median 12 vs. 6 months, <it>p </it>= 0.013).</p> <p>Conclusions</p> <p>In patients with CS a reduction in RLS is not associated with the presence of a venous embolic event such as DVT or silent PE. A greater time delay between the initial and the follow-up investigation increases the likelihood for the detection of a reduction in RLS.</p

    Heat shock protein Grp78/BiP/HspA5 binds directly to TDP-43 and mitigates toxicity associated with disease pathology

    Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease with no cure or effective treatment in which TAR DNA Binding Protein of 43 kDa (TDP-43) abnormally accumulates into misfolded protein aggregates in affected neurons. It is widely accepted that protein misfolding and aggregation promotes proteotoxic stress. The molecular chaperones are a primary line of defense against proteotoxic stress, and there has been long-standing interest in understanding the relationship between chaperones and aggregated protein in ALS. Of particular interest are the heat shock protein of 70 kDa (Hsp70) family of chaperones. However, defining which of the 13 human Hsp70 isoforms is critical for ALS has presented many challenges. To gain insight into the specific Hsp70 that modulates TDP-43, we investigated the relationship between TDP-43 and the Hsp70s using proximity-dependent biotin identification (BioID) and discovered several Hsp70 isoforms associated with TDP-43 in the nucleus, raising the possibility of an interaction with native TDP-43. We further found that HspA5 bound specifically to the RNA-binding domain of TDP-43 using recombinantly expressed proteins. Moreover, in a Drosophila strain that mimics ALS upon TDP-43 expression, the mRNA levels of the HspA5 homologue (Hsc70.3) were significantly increased. Similarly we observed upregulation of HspA5 in prefrontal cortex neurons from human ALS patients. Finally, overexpression of HspA5 in Drosophila rescued TDP-43-induced toxicity, suggesting that upregulation of HspA5 may have a compensatory role in ALS pathobiology

    Decrease in shunt volume in patients with cryptogenic stroke and patent foramen ovale

    <p>Abstract</p> <p>Background</p> <p>In patients with patent foramen ovale (PFO) there is evidence supporting the hypothesis of a change in right-to-left shunt (RLS) over time. Proven, this could have implications for the care of patients with PFO and a history of stroke. The following study addressed this hypothesis in a cohort of patients with stroke and PFO.</p> <p>Methods</p> <p>The RLS volume assessed during hospitalisation for stroke (index event/T0) was compared with the RLS volume on follow-up (T1) (median time between T0 and T1 was 10 months). In 102 patients with a history of stroke and PFO the RLS volume was re-assessed on follow-up using contrast-enhanced transcranial Doppler/duplex (ce-TCD) ultrasound. A change in RLS volume was defined as a difference of ≥20 microembolic signals (MES) or no evidence of RLS during ce-TCD ultrasound on follow-up.</p> <p>Results</p> <p>There was evidence of a marked reduction in RLS volume in 31/102 patients; in 14/31 patients a PFO was no longer detectable. An index event classified as cryptogenic stroke (P < 0.001; OD = 39.2, 95% confidence interval 6.0 to 258.2) and the time interval to the follow-up visit (P = 0.03) were independently associated with a change in RLS volume over time.</p> <p>Conclusions</p> <p>RLS volume across a PFO decreases over time, especially in patients with cryptogenic stroke. These may determine the development of new strategies for the management in the secondary stroke prevention.</p
