Since my works are about my experience and reactions toward my life, cultural differences between east and west culture have a tremendous impact on my works. Moving to America was a turning point to me these years. The surrounding is totally different and everything is new to me. I found that I’m not only learning art but also learning how to live in a foreign place. Once my life was occupied by these unknown things, my works manifest every emotions I have in every moment. This is not me trying to unify art and life, It is them integrating with each other spontaneously. Somehow I think that art is life, and life is art

    Loss of vesicular dopamine release precedes tauopathy in degenerative dopaminergic neurons in a Drosophila model expressing human tau.

    While a number of genome-wide association studies have identified microtubule-associated protein tau as a strong risk factor for Parkinson's disease (PD), little is known about the mechanism through which human tau can predispose an individual to this disease. Here, we demonstrate that expression of human wild-type tau is sufficient to disrupt the survival of dopaminergic neurons in a Drosophila model. Tau triggers a synaptic pathology visualized by vesicular monoamine transporter-pHGFP that precedes both the age-dependent formation of tau-containing neurofibrillary tangle-like pathology and the progressive loss of DA neurons, thereby recapitulating the pathological hallmarks of PD. Flies overexpressing tau also exhibit progressive impairments of both motor and learning behaviors. Surprisingly, contrary to common belief that hyperphosphorylated tau could aggravate toxicity, DA neuron degeneration is alleviated by expressing the modified, hyperphosphorylated tau(E14). Together, these results show that impairment of VMAT-containing synaptic vesicle, released to synapses before overt tauopathy may be the underlying mechanism of tau-associated PD and suggest that correction or prevention of this deficit may be appropriate targets for early therapeutic intervention

    HIV-1 encoded candidate micro-RNAs and their cellular targets

    MicroRNAs (miRNAs) are small RNAs of 21–25 nucleotides that specifically regulate cellular gene expression at the post-transcriptional level. miRNAs are derived from the maturation by cellular RNases III of imperfect stem loop structures of ~ 70 nucleotides. Evidence for hundreds of miRNAs and their corresponding targets has been reported in the literature for plants, insects, invertebrate animals, and mammals. While not all of these miRNA/target pairs have been functionally verified, some clearly serve roles in regulating normal development and physiology. Recently, it has been queried whether the genome of human viruses like their cellular counterpart also encode miRNA. To date, there has been only one report pertaining to this question. The Epstein-Barr virus (EBV) has been shown to encode five miRNAs. Here, we extend the analysis of miRNA-encoding potential to the human immunodeficiency virus (HIV). Using computer-directed analyses, we found that HIV putatively encodes five candidate pre-miRNAs. We then matched deduced mature miRNA sequences from these 5 pre-miRNAs against a database of 3' untranslated sequences (UTR) from the human genome. These searches revealed a large number of cellular transcripts that could potentially be targeted by these viral miRNA (vmiRNA) sequences. We propose that HIV has evolved to use vmiRNAs as a means to regulate cellular milieu for its benefit

    Effect and Evaluation of an Ultrasonic Atomizer With Large Vibration Amplitude

    An ultrasonic atomizer can produce large vibration amplitude is designed. Different from the structure of the usually seen ultrasonic spray nozzle, the atomizer is fundamentally constructed with a hollow tube encircled with several pieces of sectional type piezoelectric actuators, which can radially oscillate the tube to generate desired vibration profile. Atomization is formed on the surface around the liquid outlet of the tube where maximum vibration amplitude occurs. In search of resonance frequency and vibration amplitude, modal and harmonic analyses of the ultrasonic atomizer are carried out by ANSYS. In comparison the simulated results with the experimental results, both are in good agreement. A measurement system is set up for detecting the atomization droplets and calculating the droplet size and distribution. An attempt is to design an ultrasonic atomizer can produce high distribution and small diameter droplets for some application-level requirements, droplet diameter around 20�60 lm is assumed to be the specification for performance verification of the proposed atomizer. In experiment, it is found nearly 90% of atomized droplets fit for the requirement. Besides the most important factor of operating frequency, a relation of amplitude is found to include in the well-known Lang and Rayleigh�s equation

    A feeder-free culture using autogeneic conditioned medium for undifferentiated growth of human embryonic stem cells: Comparative expression profiles of mRNAs, microRNAs and proteins among different feeders and conditioned media

    <p>Abstract</p> <p>Background</p> <p>Human embryonic stem (hES) cell lines were derived from the inner cell mass of human blastocysts, and were cultured on mouse embryonic fibroblast (MEF) feeder to maintain undifferentiated growth, extensive renewal capacity, and pluripotency. The hES-T3 cell line with normal female karyotype was previously used to differentiate into autogeneic fibroblast-like cells (T3HDF) as feeder to support the undifferentiated growth of hES-T3 cells (T3/HDF) for 14 passages.</p> <p>Results</p> <p>A feeder-free culture on Matrigel in hES medium conditioned by the autogeneic feeder cells (T3HDF) was established to maintain the undifferentiated growth of hES-T3 cells (T3/CMHDF) for 8 passages in this investigation. The gene expression profiles of mRNAs, microRNAs and proteins between the undifferentiated T3/HDF and T3/CMHDF cells were shown to be very similar, and their expression profiles were also found to be similar to those of T3/MEF and T3/CMMEF cells grown on MEF feeder and feeder-free Matrigel in MEF-conditioned medium, respectively. The undifferentiated state of T3/HDF and T3/CMHDF as well as T3/MEF andT3/CMMEF cells was evidenced by the very high expression levels of "stemness" genes and low expression levels of differentiation markers of ectoderm, mesoderm and endoderm in addition to the strong staining of OCT4 and NANOG.</p> <p>Conclusion</p> <p>The T3HDF feeder and T3HDF-conditioned medium were able to support the undifferentiated growth of hES cells, and they would be useful for drug development and toxicity testing in addition to the reduced risks of xenogeneic pathogens when used for medical applications such as cell therapies.</p

    Impact of Enhancer of Zeste Homolog 2 on T Helper Cell-Mediated Allergic Rhinitis

    Enhancer of zeste homolog 2 (Ezh2) has been shown to play a role in the differentiation of T helper (Th) 1 and 2 cells in mice studies using Ezh2-deficient T cells. However, the results have been inconsistent, and the function of Ezh2 in human Th1 and Th2 cell differentiation and its association with disease remains controversial. We measured the expression of Ezh2 in Th1 and Th2 cells in peripheral blood mononuclear cells after acute challenge with house dust mite using flow cytometry in patients with allergic rhinitis (AR) and controls. The role of Ezh2 was further explored by adding the p38 inhibitor to see if this affected allergen-induced Th1 and Th2 differentiation. The expression of Ezh2 in the Th1 and Th2 cells was significantly lower in the patients than in the controls and was negatively correlated with serum IL-17A levels in the patients. Ex vivo allergen challenge resulted in rapid Th2 cell differentiation, which was negatively associated with the Ezh2 expression in Th2 cells. Inhibiting p38 activity increased the expression of Ezh2 in Th2 cells and reduced the number of differentiated Th2 cells. Our findings suggest that Ezh2 expression is potentially associated with AR development

    Bicycling has become a mainstream activity among the environmental aware generation. Bicycling communities have gradually shown interests in quantitative data of the bicycling experiences such as road roughness, inclination, pollution, etc. Bikers utilize these data to infer the possible stamina cost and quality of surroundings. This supports them to make a better decision. This study assumes that fitness level indexed by stamina cost could enhance a biker's sense of control. The prototype in this paper was developed to provide stamina cost information, which is inferred from the terrain patterns of a biking route. In the system evaluation, participants took a positive attitude toward this prototype and approved the importance of stamina cost feedback. This paper also concluded several key issues about designing the stamina cost feedback system for bikers

    Gene therapy for follistatin mitigates systemic metabolic inflammation and post-traumatic arthritis in high-fat diet-induced obesity

    Obesity-associated inflammation and loss of muscle function play critical roles in the development of osteoarthritis (OA); thus, therapies that target muscle tissue may provide novel approaches to restoring metabolic and biomechanical dysfunction associated with obesity. Follistatin (FST), a protein that binds myostatin and activin, may have the potential to enhance muscle formation while inhibiting inflammation. Here, we hypothesized that adeno-associated virus 9 (AAV9) delivery of FST enhances muscle formation and mitigates metabolic inflammation and knee OA caused by a high-fat diet in mice. AAV-mediated FST delivery exhibited decreased obesity-induced inflammatory adipokines and cytokines systemically and in the joint synovial fluid. Regardless of diet, mice receiving FST gene therapy were protected from post-traumatic OA and bone remodeling induced by joint injury. Together, these findings suggest that FST gene therapy may provide a multifactorial therapeutic approach for injury-induced OA and metabolic inflammation in obesity

    A Moral Education Lesson to Enhance Self-Affirmative Consciousness in Japan and Taiwan

    This paper addresses clinical research on moral education lessons to enhance the self-affirmative consciousness of students in Japan and Taiwan. We conducted four lessons for fifth and sixth graders under the same conditions. The lesson was about the natural environment and was organized as a cooperative learning experience. The Association Method was employed as an assessment.  Prompted by the image of , response words such as “water,” “air,” and “environment” appeared in four classes after the lesson. Through the 60-minute lesson, more than half of the response words changed. This indicates that the children concentrated deeply on the lesson, and that it made an impression on them.  The results from the Japanese A and B and Taiwanese C and D schools were diverse. We compared the response words before andafter the lesson, using the Association Method by cue word , which shows self-affirmative consciousness. In class A, words that recognize the importance of nature appeared even in self-consciousness, and in class B, positive words increased and negative words decreased significantly (p < .05). In class C, consciousness of “oneself” and “family” increased, and in class D, “life” and “friend” increased. We interpreted these differences as a result of the children’s reflections on their own inner character and the historical background they have learned and not learned
