    Elucidating the effect of tomato leaf surface microstructure on Botrytis cinerea using synthetic systems

    For some pathogenic fungi, sensing surface topography is part of their infection strategy. Their directional growth and transformation to a new developmental stage is influenced by contact with topographic features, which is referred to as thigmo-response, the exact functionality of which is not fully understood. Research on thigmo-responses is often performed on biomimetically patterned surfaces (BPS). Polydimethylsiloxane (PDMS) is especially suitable for fabrication of BPS. Here, we used synthetic BPS surfaces, mimicking tomato leaf surface, made from PDMS with the pathogenic fungus Botrytis cinerea to study the influence of structural features of the leaf surface on the fungus behavior. As a control, a PDMS surface without microstructure was fabricated to maintain the same chemical properties. Pre-penetration processes of B. cinerea, including the distribution of conidia on the surface, germination, and germ tube growth were observed on both leaf-patterned and flat PDMS. Microstructure affected the location of immediate attachment of conidia. Additionally, the microstructure of the plant host stimulated the development of germ tube in B. cinerea, at a higher rate than that observed on flat surface, suggesting that microstructure plays a role in fungus attachment and development.Peer Reviewe

    The effect of biochar on plant diseases: what should we learn while designing biochar substrates?

    The increasing demand for soilless substrates and rising environmental concerns about the use of non-renewable resources such as peat has led to the search for alternative constituents of growing mixtures for containerized plants. In this report we reviewed the works concerning biochar as constituent of growing media, targeting its influence on plant growth and plant disease. Biochar mostly has positive or neutral influences on plant growth compared with peat media when present in concentrations higher than 25% (v:v). However, studies of biochar influence on plant disease reveals that while lower concentrations (≤1%) of biochar often suppressed several diseases, higher concentrations (≥3%) were mostly ineffective or induced plant disease. For use as horticultural peat replacement, it is recommended that biochar feedstocks and concentrations be standardized and the potential effect of biochar on plant disease be considered, so that growers can rely on consistent and reproducible biochars for desired effects

    The biochar effect: plant resistance to biotic stresses

    Biochar (charcoal) is the solid co-product of pyrolysis, the thermal degradation of biomass in the absence of oxygen. Pyrolysis also yields gaseous and liquid biofuel products. There is a growing interest worldwide in the pyrolysis platform, for at least four reasons: (i) pyrolysis can be a source of renewable biofuels; (ii) many biomass waste materials can be treated by pyrolysis and thus converted into a fuel resource; (iii) long-term sequestration of carbon dioxide which originated in the atmosphere may result from adding biochar to soil; and (iv) biochar soil amendment contributes to improved soil fertility and crop productivity. Currently, however, very little biochar is utilized in agriculture, in part because its agronomic value in terms of crop response and soil health benefits have yet to be quantified, and because the mechanisms by which it improves soil fertility are poorly understood. The positive effects of biochar on crop productivity under conditions of extensive agriculture are frequently attributed to direct effects of biochar-supplied nutrients and to several other indirect effects, including increased water and nutrient retention, improvements in soil pH, increased soil cation exchange capacity, effects on P and S transformations and turnover, neutralization of phytotoxic compounds in the soil, improved soil physical properties, promotion of mycorrhizal fungi, and alteration of soil microbial populations and functions. Yet, the biochar effect is also evident under conditions of intensive production where many of these parameters are not limited. Biochar addition to soil alters microbial populations in the rhizosphere, albeit via mechanisms not yet understood, and may cause a shift towards beneficial microorganism populations that promote plant growth and resistance to biotic stresses. In addition to some scant evidence for biochar-induced plant protection against soilborne diseases, the induction of systemic resistance towards several foliar pathogens in three crop systems has been demonstrated. There are indications that biochar induces responses along both systemic acquired resistance (SAR) and induced systemic resistance (ISR) pathways, resulting in a broad spectrum controlling capacity in the canopy. This review examines the effects of biochar soil amendment on the different soil-plant-microbe interactions that may have a role in plant health.     Gene Editing of the Decoy Receptor LeEIX1 Increases Host Receptivity to Trichoderma Bio-Control

    Fungal and bacterial pathogens generate devastating diseases and cause significant tomato crop losses worldwide. Due to chemical pesticides harming the environment and human health, alternative disease control strategies, including microorganismal bio-control agents (BCAs), are increasingly sought-after in agriculture. Bio-control microorganisms such as Trichoderma spp. have been shown to activate induced systemic resistance (ISR) in the host. However, examples of highly active bio-control microorganisms in agricultural settings are still lacking, due primarily to inconsistency in bio-control efficacy, often leading to widespread disease prior to the required ISR induction in the host. As part of its plant colonization strategy, Trichoderma spp. can secrete various compounds and molecules, which can effect host priming/ISR. One of these molecules synthesized and secreted from several species of Trichoderma is the family 11 xylanase enzyme known as ethylene inducing xylanase, EIX. EIX acts as an ISR elicitor in specific plant species and varieties. The response to EIX in tobacco and tomato cultivars is controlled by a single dominant locus, termed LeEIX, which contains two receptors, LeEIX1 and LeEIX2, both belonging to a class of leucine-rich repeat cell-surface glycoproteins. Both receptors are able to bind EIX, however, while LeEIX2 mediates plant defense responses, LeEIX1 acts as a decoy receptor and attenuates EIX induced immune signaling of the LeEIX2 receptor.     Botrytis spp. are efficient pathogens, causing devastating diseases and significant crop losses in a wide variety of plant species. Here we outline our review of these pathogens, as well as highlight the major advances of the past 10 years in studying Botrytis in interaction with its hosts. Progress in molecular genetics and the development of relevant phylogenetic markers in particular, has resulted in the characterisation of approximately 30 species. The host range of Botrytis spp. includes plant species that are members of 170 families of cultivated plants

    Flavobacteria are important members of aquatic and terrestrial bacterial communities, displaying extreme variations in lifestyle, geographical distribution and genome size. They are ubiquitous in soil, but are often strongly enriched in the rhizosphere and phyllosphere of plants. In this study, we compared the genome of a root-associated Flavobacterium that we recently isolated, physiologically characterized and sequenced, to 14 additional Flavobacterium genomes, in order to pinpoint characteristics associated with its high abundance in the rhizosphere. Interestingly, flavobacterial genomes vary in size by approximately two-fold, with terrestrial isolates having predominantly larger genomes than those from aquatic environments. Comparative functional gene analysis revealed that terrestrial and aquatic Flavobacteria generally segregated into two distinct clades. Members of the aquatic clade had a higher ratio of peptide and protein utilization genes, whereas members of the terrestrial clade were characterized by a significantly higher abundance and diversity of genes involved in metabolism of carbohydrates such as xylose, arabinose and pectin. Interestingly, genes encoding glycoside hydrolase (GH) families GH78 and GH106, responsible for rhamnogalacturonan utilization (exclusively associated with terrestrial plant hemicelluloses), were only present in terrestrial clade genomes, suggesting adaptation of the terrestrial strains to plant-related carbohydrate metabolism. The Peptidase/GH ratio of aquatic clade Flavobacteria was significantly higher than that of terrestrial strains (1.7±0.7 and 9.7±4.7, respectively), supporting the concept that this relation can be used to infer Flavobacterium lifestyles. Collectively, our research suggests that terrestrial Flavobacteria are highly adapted to plant carbohydrate metabolism, which appears to be a key to their profusion in plant environments
